1. Silence of Wnt5a gene inhibits migration and invasion of lung squamous carcinoma cells

Objective: To study the influence of Wnt5a gene silence on the proliferation, migration and invasion of lung squamous carcinoma cells. Methods: A recombinant plasmid pH1-siRNAWnt5a- was constructed and used to deliver small interference RNA (siRNA) targeting Wnt5a in SK-MES-1 cells; the transfected cells were screened to establish a stable transgenic cell line. MTT, cell cycle and Transwell assays were employed to evaluate the effect of Wnt5a gene silence on the proliferation, migration and invasion of lung squamous carcinoma cells. Results: Western blotting assay revealed that Wnt5a was lowly expressed in SK-MES-1Wnt5a- (13.6%). The proliferation index (PI) of transgenic cell line was slightly lower than that of the control cell line ([28.3±3.8] % vs [30.5±5.2]%). The migration and invasion capabilities of SK MES-1Wnt5a- cells were (47.3±9.2)% and (39.7±11.7)% of the control cells, respectively. Conclusion: Low Wnt5a expression can significantly inhibit the migration and invasion capabilities of SK-MES-1 cells, indicating that WntSa might be a potential target for gene therapy of lung squamous carcinoma.
2. Adenovirus vector-mediated RNA interference inhibits expression of vascular endothelial growth factor receptor in colon carcinoma cells and tumor growth

Objective: To study the inhibitory effects of an adenovirus (Ad)-based short hairpin RNA (shRNA) expression system on expression of vascular endothelial growth factor receptor (VEGFR) and on growth of colon carcinoma cells in vitro and in vivo. Methods: RNA interference pAd-Easy/VEGFR vector was used to transfect 293 cells via Lipofectamine 2000. The adenoviral vector carrying VEGFR was used to transfect CW2 cells and the transfection efficiency was determined by fluorescent microscope and flow cytometry. The expression of VEGFR was examined by RT-PCR and Western blotting. The cell growth was observed with MTT method and the growth curve was plotted. Nude mouse was transplanted with CW2 cells to establish colon cancer tumor model and the growth of tumor was observed daily. Micro-vessel density (MVD) was detected by immunohistochemistry. Results: The recombinant pAd-Easy carrying shRNA against VEGFR was verified by sequencing. Flow cytometry showed that the transfection efficiency of CW2 cells was 99.7%. RT-PCR and Western blotting showed that the expression of VEGFR in pAd-Easy/VEGFR group was obviously decreased. The growth curve showed that the cell growth in the pAd-Easy/ VEGFR group was obviously slowed down. We also found that the tumor growth in the nude mouse model was obviously inhibited and the MVD was also decreased. Conclusion: pAd-Easy/VEGFR-mediated VEGFR shRNA can effectively inhibit the expression of VEGFR in CW2 cells and suppress tumor growth in vivo.
3.Progress of diagnosis and treatment of hypertensive renal damage by Chinese medicine.

Hypertensive renal damage is based on the extent and duration of hypertension, renal damage caused by varying severity. Hypertensive renal damage due to various causes imbalance of vascular active substances, renal arteriosclerosis, so that the abnormal renal hemodynamic, renal ischemia, low specific gravity of urine, low osmotic pressure and urine. The rapidly increasing incidence of hypertensive renal damage has become one of the most important reasons of end stage renal disease (ESRD). Effective treatment of hypertension is limited by poor compliance and significant adverse reaction of antihypertensive drugs. Therefore, some patients have turned to Chinese medicine (CM), hoping that such treatments might improve the efficiency. The author reviews relevant theory and the latest researches, on the basis of combining diseases and syndrome, discusses state and achievement of hypertensive renal damage with Chinese herbal medicines from fundamental and clinical research and action mechanism from standpoints of Chinese herbal compound and herbal effective chemical composition to take future research for important reference.

6.Surgery of gastrointestinal stromal tumor of the duodenum

Objective To evaluate the results of surgical treatment for duodenal gastrointestinal stromal tumor (GIST).Methods Between January 2000 and January 2013,33 patients underwent surgical resection of duodenal GIST in Peking University Cancer Hospital,including local resection and pancreatoduodenectomy.Clinical data,surgical outcome and prognosis were reviewed.Results The median survival time is 94 months (range:55-132 months).The 1,3,5-year survival rates are 97%,89% and 80%,respectively.The 5 year survival rate of patients after pancreatoduodenectomy and local resection is 78% and 83%,respectively (P =0.091).It shows that tumor size and mitotic count and Fletcher classification influence overall survival.Conclusions When fulfiling proper safe margins,local resection is a reliable and curative option for most duodenal GISTs.
7.Clinical Observation on 100 Cases of E -Type Viral Hepatitis Treated by Differentiation of Symptoms and Signs ( Bian Zheng) : with Comparative Observation on 100 Cases of A-Type Viral Hepatitis as Control

Three hundred cases of E type viral hepatitis were observed with 100 cases of A type as control. We held that the etiology and pathogenesisof E and A types of viral hepatitis were wind, dampness, heat, poisons and stasis, each with its predominance; For A type, its predominant etiology being wind, heat and stasis, while that of E type, dampness, poisons and stasis. For their therapy, the principle of "similar treatment for different diseases " should, adopted. After treatment, ACT, TB, and a markedly decreased, signs and symptoms markedly improved, Epr, IgG also improved to different extents, the cured rate ( rate of negative virus exam) being 80% and 78% for A type and E type respectively. The effect was especially good for simple infection, and rather poor for mixed infection type.
8.Study on correlativity of development of illness state with changes of biochemical indexes in the cases of hepatitis B and E with overlapping infections of the liver and gallbladder dampness-heat type

As the patient of hepatitis B and E overlapping infection of liver and gallbladder damp - heat type compared with the patient of simple hepatitis E of liver and gallbladder damp-heat type, attack was very rapid, jaundice exacerbated progressively and hepatic function damaged severely. Aftertreatment the state of illness restored more slowly and the duration of illness was obviously prolonged. At the same time, it was found that the biochemical indexes (hyaluronic acid, total bile acid) increased significantly at acute stage, and the indexes in the patient of overlapping infection in the time for recovering normality at restoration stage was longer than that in the patient of simple hepatitis E, with a significantly difference between the two groups. It is indicated that lesion of the liver in the patient of overlapping infection of hepatitis B and E was more severe than the simple hepatitis E, with no good prognosis.
9.Clinical Study on Treatment of Psoriasis Based on Differentiation of Symptoms and Signs of the Lung, An Analysis of 486 Cases

Xuan Fei Run Zao Decoction was used for treatment of 486 cases of psoriasis with a total effective rate of 90. 3%, so as to test and verify the TCM theory of the lung being in charge of the hair and the lung being closely related with derma tosis. Clinical study found that in patients of psoriasis there was humoral immunological hyperfunction, manifesting increases of IgG and IgA; trace elements copper and ferrum levels increased and zinc, manganese and calcium decreased In the hair: packed cell volume and viscosity of whole blood increased, and erythrocyte sedimentation rate increased. It is suggested that psoriasis is possibly related to changes of immunity, blood rheology and trace elements in the body.
10.Abnormal patterns of functional connectivity between right temporal parietal junction and whole brain in children with attention deficit hyperactivity disorder

Objective To investigate the pathophysiological distinction of the functional connectivity between right temporal parietal junction(rTPJ) and whole brain in children with attention-deficit/hyperactivity disorder(ADHD) on resting-state functional magnetic resonance imaging(fMRI) and explore the neurological mechanisms of ADHD at the point of functional connectivity.Methods Resting-state functional magnetic resonance imaging scans were obtained in 30 children with ADHD from The Third Affiliated Hospital of Soochow Univcrsity and another 33 with age and gender matched controls from a normal primary school over the same period, and then,blood oxygenation level dependent(BOLD) signal was acquired to calculate the functional connectivity of rTPJ with other brain regions.Significant differences of connectivity intra-class and between groups were analyzed.Also, the correlation between the connectivity strength and the ADHD rating scales were tested.Results The pattern of functional connectivity of the rTPJ for the ADHD group was similar to that of the normal control group.Compared with the normal controls, the left superior temporal gyrus (MNI coordinates-33,6,48) , the right anterior c ingulate coltex(MNI coordinates 6,39,-3) , the right inferior parietal lobule (MNI coordinates 42,-33,30) , the left medial frontal gyrus(MNI coordinates-3,63,-18) in children with ADHD sbowed an increased resting-state functional connectivity with the whole brain ,while the right middle temporal gyrus(MNI coordinates 57,-33,-12) , the right cingulate gyrus(MNI coordinates 18,-21,36) showed a decreased resting-state functional connectivity with the whole brain.The average connection strength between the rTPJ and the whole brain showed positive correlation with the ADHD scores,while in normal controls,it showed negative.The connection strength between the rTPJ and those 6 brain areas with significant statistical difference was relevant to the ADHD scores,but they had no statistical significance.Conclusion The functional connection abnormalities between tbc rTPJ and the whole brain in children with ADHD may be relevant to the clinical symptom of attention-deficit.
