Aleutian mink disease parvovirus (AMDV) causes a persistent infection associated with immune complex disease, hypergammaglobulinemia, and high levels of antiviral antibodies. Despite the presence of an antibody, the virus is not cleared in vivo. Pre-existing antibodies may enhance viral infections, by Fc-receptor-mediated antibody-dependent enhancement (ADE), but the mechanism that underlies ADE has not been fully defined. Three models have been proposed, including: (1) interactions between antibody and FcR, complement C3 fragment and CR, or between C1q and C1qR, which promotes viral attachment to cells; (2) suppression of IFN-gamma-mediated host-cell antiviral gene expression by the upregulation of negative regulators of pathogen pattern recognition; and (3) the promotion of early IL-10 secretion. In addition, the role of cytokine IL-6 in ADE mediated disease development is discussed, to facilitate a better understanding of the pathogenesis of AMDV infection, as well as give insights into rational vaccine design approaches.

Chinese Journal of Tissue Engineering Research 2005;9(3):232-233
BACKGROUND:It has been known that there exists a close relationship between leptin and obesity. Then, how are serum leptin and insulin related to obesity-related hypertension(ORH)? OBJECTIVE:To study the changes of serum leptin and insulin in elderly men with ORH,and investigate the relationship between leptin resistance and ORH in elderly men. DESIGN: Retrospective controlled study based on diagnosis. SETTINGS:The Department of Neurology and the Department of Nuclear Medicine,Affiliated Union Hospital of Tongji Medical College, Huazhong University of Science and Technology. PARTICIPANTS: Altogether 62 elderly male inpatients or outpatients aged from 60 to 82 years in the Affiliated Union Hospital of Tongji Medical College from October 2001 to June 2002 were selected. The diagnosis of hypertension was based on the diagnostic criteria established by the WHO in 1999:systolic blood pressure(SBP) ≥ 140 mm Hg(1 mm Hg=0.133 kPa) and(or) diastolic blood pressure(DBP) ≥ 90 mm Hg. INTERVENTIONS:The content of serum leptin and insulin in 41 hypertensive and 21 normotensive elderly men was determined by the method of radioimmunoassay. MAIN OUTCOME MEASURES:Blood pressure, body mass index (BMI),serum leptin and insulin level. RESULTS:The content of serum leptin and insulin in the hypertensive obese men were increased significantly by 1.8 μ g/L and 2.7 mIU/L,respectively,as compared with that in the normotensives (t=2.212,2.395,P< 0.01). The level of serum leptin and insulin in the ORH patients was increased by 2.7 μ g/L and 4.7 mIU/L,respectively,as compared with that of the normotensives with significant differences (t=3.348,5.113,all P< 0.001). The level of serum leptin in the hypertensive patients older than 70 years was significantly increased by 1.7 μ g/L compared with that of those aged 60 to 70 years(t=2.767,P< 0.05). CONCLUSION:Resistance to leptin and insulin exists in obesity-related hypertension of elderly men and leptin is closely associated with obesity-related hypertension.

Chlorogenic acid displays several important roles in the therapeutic properties of many herbs, such as antioxidant activity, antibacterial, antiviral, scavenging free radicals and exciting central nervous system. Only about one-third of chlorogenic acid was absorbed in its prototype, therefore, its gut metabolites play a more important role in the therapeutic properties of chlorogenic acid. It is necessary to consider not only the bioactivities of chlorogenic acid but also its gut metabolites. This review focuses on the potential activities and mechanisms of chlorogenic acid and its gut metabolites on central nervous system diseases.

Objective To clarify the protective effect of nrsodeoxycholic acid ( UDCA ) on endoplasmic reticulum stress-mediated apoptosis in pancreatic β-cell of streptozotocin ( STZ )-induced diabetic rats. Methods Rats( n =40) received a single injection STZ( 50 mg/kg) intra-peritoneally and formed a β-cell injury model.Weight-matched normal rats( the control group,n =10 ) were injected with the buffer alone.STZ-treated rats with persistent random blood glucose higher than 16.7 mmol/L for 1 week were considered as diabetic status( n=14 ),then divided randomly into STZ-induced diabetes mellitus ( DM ) group ( n =7 ) and UDCA-treated DM group ( n =7 ).UDCA (40 mg· kg- 1,d-1 ) was administered daily by intragastric intubations throughout the experimental period (30 d).During the entire experiment, blood glucose in all rats was assessed.By the end of the experiment, all rats were sacrificed with the pancreas removed and the blood sample collected immediately.Fasting insulin levels were assayed by radioimmunoassay. The morphological changes of pancreatic β-cells apoptosis were determined by TUNEL assay.RNA in pancreas was abstracted and microarray containing 89 pieces of apoptosis related genes was applied. The related gene expressions were detected by RT-PCR and Western blot. Results The concentration of blood glucose in diabetic rats was gradually decreased after UDCA treatment, but at the end of the experiment it was still higher than that in the normal control group. The treatment with UDCA raised the fasting insulin level in diabetic rats, but this concentration was significantly lower as compared to the control group.Based on TUNEL stained tissue sections, the percentage of β-cell apoptosis of UDCA-treated DM group was significantly lower than that of STZ-induced diabetic group(P<0.05 ).Among 89 genes,42 genes up-regulated and 46 genes down-regulated in diabetic rats, some of which were ameliorated by UDCA treatment. The expressions of Caspase-3,Bax,Bip,and CHOP mRNA in pancreas of DM group were significantly up-regulated as compared with those in the control group ( P < 0.05 ) ; while the expression of Bcl-2 mRNA was markedly down-regulated (P<0.05 ).However, these parameters in the U DCA-treated animals showed a marked improvement. Conclusion Ursodeoxycholic acid seems to protect pancreatic β-cell from apoptosis in STZ-induced diabetes by attenuating the severity of endoplasmic reticulum stress.

AIM: To investigate the time - effect relationship between the expression of rhodopsin and recoverin and photoreceptor damage induced by N - nethl - N -nitrosourea ( MNU) .
METHODS: Thirty-six 7-week old Sprague-Dawley ( SD ) rats were intraperitoneally injected with MNU ( 60mg/kg ) and were put to death by dislocation of cervical vertebra 6, 12, 24h; 3, 7d after injection ( 6 per group) , respectively. As a control, six rats were injected with phosphate buffer saline (PBS) 5mL/kg and sacrificed on d3 after injection. The degree of photoreceptor apoptosis was detected by HE staining, terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling ( TUNEL ) and transmission electron microscope ( TEM ) in the right eyes. The mRNA expressions of rhodopsin and recoverin were detected different time after injection by Western blot and immunohistochemical method in the left eyes.
RESULTS:The dissolution of photoreceptor nucleus and apoptosis body were first perceived at 12h by TEM; most of cells at outer nuclear layer were presented positive reaction. The apoptotic index reached peak ( 29. 7% ±2.3%) at 24h which was coincided with the observation of TEM. The results of immunohistochemistry displayed that rhodopsin and recoverin were on a declining curve with time extension. Furthermore, the results of Western blot indicated that rhodopsin had dramatic decline at 6h after injection (P<0. 05), and extremely significant difference comparing to control group after 12h ( P<0. 01 ); while recoverin dramatic declined at 12h, and extremely significant difference after 24h (P<0. 01).
CONCLUSION:60mg/kg MNU intraperitoneally injection one - time may specifically induce photoreceptor apoptosis, The mechanism of down - regulation of rhodopsin and recoverin may be related to the selected apoptosis of photoreceptors.
