3.The effect of PKCζinhibitor on the proliferation and invasion of skin squamous carcinoma cell line A-431
Tianjin Medical Journal 2017;45(6):566-570
Objective To investigate the effect of protein kinase C (PKC)ζinhibitor T5450996 on the proliferation and invasion of skin squamous carcinoma cell line A-431. Methods PKCζinhibitor T5450996 was screened through Z′-LYTE?kit. Cell proliferation assay and cell cycle analysis were used to observe the effects of T5450996 on the proliferation of skin squamous carcinoma A-431 cells. Scratch assay and invasion assay were used to explore the effects of T5450996 on the migration and invasion of skin squamous carcinoma A-431 cells. Results The PKCζinhibitor T5450996 can inhibit the activity of PKCζkinase, and the IC50 value of T5450996 was about 35μmol/L. Compared to the control group, 35μmol/L and 70 μmol/L concentrations of T5450996 significantly suppressed the proliferation of A-431 cells and blocked the cell cycle of A-431 cells. The results of scratch assay and invasion assay indicated that the migration and invasion capacities of A-431 cells were markedly impaired after the treatments with 35μmol/L and 70μmol/L concentrations of T5450996 (P<0.05). However, 20μmol/L concentration of T5450996 showed no significant effect (P>0.05). Conclusion PKCζinhibitor T5450996 significantly inhibits the proliferation and invasion capacities of skin squamous carcinoma cell line A-431, and which may be a small molecular inhibitor with potential applications in the future.

Objective T_O analyze the electrophysiological features of shoulder-hand syndrome(SHS)fol- lowing stroke and investigate the relationships between peripheral nerve damage and the factors causing SHS.Meth- ods Fifty-eight stroke patients were divided into an SHS group(39 patients with shoulder-hand syndrome)and a model group(19 patients without shoulder-hand syndrome).Standard sensory and motor nerve conduction studies were performed with 58 of the patients, including sensory nerve conduction velocity(SCV),amplitude of the sensory nerve action potential(SNAP),distal motor latency(DMI)and the amplitude of the compound muscle action poten- tial(CMAP)of the median nerve.A needle electromyogram(EMG)test was performed on the abductor pollicis of the affected side with both groups. Results The needle EMG showed abnormal insertion potential, fibrillation po- tential and positive sharp waves in all 39 cases of the SHS group, which was significantly higher than in the model group. The amplitude of the sensory nerve action potential(7.77?4.34 mV)and the amplitude of the compound muscle action potential(10.13?3.15 mV)were significantly lower than those of the model group.Abnormal ampli- tude was more severe in sensory nerves than in motor nerves. Conclusion Peripheral nerve damage was found in the shoulder-hand syndrome patients, and this damage was mainly the dystrophy of axonal neuropathy. The damage severity was more in sensory nerves than in motor nerves. The study could offer an useful clue of prevention and treat- ment of shoulder-hand syndrome.

The metabolism study of radiolabeled drugs plays an important role in the development of new drugs. It provides information on drug absorption, metabolism, tissue distribution and excretion, and plays an irreplaceable role in the metabolite safety evaluation and mass balance of new drugs. The new guidance draft on clinical trials of radiolabeled drugs recently released by the US FDA puts forward higher standards and has been widely concerned by the industry. In recent years, in the research and development of new drugs in China, 14C labeled drugs have been used to carry out clinical metabolism studies, which has overcome key technical bottlenecks and accumulated experience. This paper summarizes the above research progress, analyzes the existing problems, and preliminarily looks forward to the future technological development and application.
