1.Development of bone tissue engineering scaffold materials

BACKGROUND:Bone tissue engineering is the most promising way to treat bone defects at present. The key problem is to construct vascular networks which can provide oxygen and nutrients for new bone, and thereby provide a way for the body metabolism. OBJECTIVE:To review the characteristics of bone tissue engineering scaffold materials and to introduce the development of composite scaffold materials. METHODS:With the key words of“bone tissue engineering, scaffold, vascularization, composite scaffold”in Chinese and in English, respectively, a computer-based search was performed for articles published in CNKI and PubMed databases from January 2001 to January 2014. After the initial screening, the reserved articles were further detailed, summarized and concluded. RESULTS AND CONCLUSION:According to the different sources, the bone tissue engineering scaffold materials can be divided into artificial materials, natural derivatives and composite scaffold materials. Single scaffold is difficult to be the most ideal material for repair of bone defects, while composite scaffold can make up for the defects of the single scaffold to different degrees. Therefore, in recent years, the bone tissue engineering scaffolds have developed from single to composite scaffolds and there is the trend of organic combination of artificial materials and natural derivatives. However, composite scaffolds have many problems to be solved in the clinical application. The main aspect is to control the proportion of the composite scaffold so that the degradation of materials can be matched with growth of tissues and cells. The other one is to keep the porous and high mechanical strength of the composite scaffold.
2.Control Study on Short-range Application of Perioperative Antibacterials for Gynecological TypeⅠ&Ⅱ Incision

OBJECTIVE: To discuss about the meaning of short-range application of gynecological perioperative antibacterials and its efficacy of infection prevention. METHODS: 100 cases of type Ⅰ & Ⅱ incision selective operations during Jun.~Nov., 2006 (control group) and 105 cases during Jun.~Nov., 2007 (short-range group) were collected and analyzed in respect of hospitalization time, hospitalization time after operation, applying time of antibacterials, cost of antibacterials, variety of antibacterials and post-operation infection. RESULTS: In short-rang group costs and applying time of antibacterials reduced significantly, which were significantly different from that in control group. No significant difference was noted in cases of post-operation infection. CONCLUSION: There is no significant difference between short-range medication and long-range medication for preventing infection of gynecological type Ⅰ & Ⅱ incision selective operation.
3.Evaluation of early change of left ventricular systolic function in type 2 diabetes mellitus by ultrasound speckle tracking imaging

Objective To evaluate the early change of left ventricular (LV) systolic function in patients with type 2 diabetes mellitus(T2DM) by ultrasound speckle tracking imaging. Methods Ninety-five subjects were included:40 T2DM with normal LV ejection fraction (LVEF≥50% ,group A),20 T2DM with abnormal LVEF (LVEF<550 %) (group B), and 35 normal controls. Two-dimensional strain images were acquired from LV short-axis view. LV peak systolic global and segmental radial strain and circumferential strain at the levels of mitral annulus, papillary muscle and apex, and LV systolic global and segmental rotation at the levels of mitral annulus and apex and LV peak systolic global twist were assessed. Results In group A, compared with control group, LV peak systolic radial strain parameters were not significant difference (P >0.05), peak systolic circumferential strain of part segments, average peak strain of each plane and global strain were reduced (P <50.05~0.001) ,and LV peak systolic rotation of all apical segments, peak systolic global rotation at the levels of mitral annulus and apex,and peak systolic global twist were significantly increased (P <0.05~0.001). Compared with control group and group A,all LV systolic radial strain and circumferential strain parameters, LV systolic global and segmental rotation and peak systolic global twist in group B were significantly reduced (P <0.05 ~ 0.001). Conclusions Speckle tracking imaging could be used to evaluate the early change of LV torsional deformation and LV systolic function in T2DM.
4.Evaluation of left ventricular rotation and twist in uremia patients by two-dimensional ultrasound speckle tracking imaging

Objective To analyze the left ventricular(LV) rotation and twist by two-dimensional ultrasound speckle-tracking imaging(STI),and evaluate the correlation between twist and other components of LV contraction in uremia patients. Methods Twenty-nine uremia patients and twenty-nine healthy volunteers were included. High frame rate two-dimensional images were recorded from the left ventricular short-axis views both of the basal and apical planes. Segmental and global LV rotation were recorded at both The peak systolic LV twist was significantly decreased in uremia patients(P <0. 001). The loss of LV twist occurred mainly by the impairment of maximal global counterclockwise apical rotation(P <0. 001),and was augmented by somewhat less reduction in global clockwise basal rotation although the differences was not significantly reduced(P<0.01), while only the posterior and inferior of the basal plane was significantly fraction(r=0.645,P=0.000), LV end-diastolic volume (r=-0.549,P=0.002), LV end-systolic volume(r=-0.673, P = 0.000) in uremia patients. Conclusions STI enables quantification of segmental and global LV rotation in an objective manner in uremia patients,and provides a new method for evaluation of segmental and global LV systolic function.
5.Two-dimensional strain echocardiography to assess left ventricular systolic function in patients with type 2 diabetes mellitus

Objective To assess the short-axis and long-axis systolic function of the left ventricle (LV) in patients with type 2 diabetes mellitus and without coronary artery disease using two-dimensional strain echocardiography.Methods Eighty-nine subjects with normal ejection fraction were included:37 with diabetes mellitus(DM group),22 with both diabetes and left ventricular hypertrophy(DH group), and 30 normal controls.Two-dimensional strain images were acquired from the LV short-axis view at the levels of mitral annulus, papillary muscle and apex, and the apical four-chamber view, two-chamber view and long-axis views of the LV respectively.The longitudinal strain was measured in the apical views.The radial strain and the rotation were measured in the LV short-axis views.The LV twist was defined as the apical rotation relative to the base.Results The average peak systolic longitudinal strain of different segments and the peak longitudinal global strain in DM group and DH group were significantly reduced when compared with those in control groups ( P<0.001 ),and these parameters in DH group were significantly lower than those in DM group (P<0.001 or P<0.05).There was no significant difference in the average peak systolic radial strain of different segments and peak radial global strain among all groups ( P>0.05).The LV rotations and the LV twist were significantly higher both in DM group and in DH group than in control group ( P<0.001 or P<0.01),and the basal LV rotation and the LV twist were larger in DH group than in DM group ( P<0.01, P<0.05).Conclusions Two-dimensional strain echocardiography may be used to identify the LV systolic function in patients with type 2 diabetes mellitus.
6.Evaluation of right ventricular global longitudinal function in patients with tetralogy of fallot by two-dimensional ultrasound speckle tracking imaging.

Quantification of right ventricular (RV) volume and function remains a challenge because of RV complex geometry by conventional echocardiography. The purpose of this study was to assess RV global longitudinal function in patients with tetralogy of Fallot (TOF) by 2-dimensional ultrasound speckle tracking imaging (STI). Thirty-eight patients with TOF were enrolled in this study and divided into child group (n=25) and adult group (n=13) according to age. Thirty-eight age- and sex-matched normal subjects were selected as control groups including child control group (n=25) and adult control group (n=13). RV global longitudinal peak systolic strain (GLS), strain rate (GLSR(s)), early diastolic strain rate (GLSR(e)) and late diastolic strain rate (GLSR(a)) were measured in all subjects by STI from the apical 4-chamber view and compared between groups. Furthermore, the main factors affecting the RV global longitudinal functional parameters were assessed. Compared with those in controls, RV GLS, GLSR(s) and GLSR(e) were significantly reduced in patients with TOF (P<0.05 for all). RV GLSR(s) was significantly decreased in adult patients with TOF as compared with that in child patients (P<0.05). There was no significant difference in RV GLS, GLSR(e) and GLSR(a) between child and adult TOF groups (P>0.05). The diameter of right ventricle, main pulmonary artery and ventricular septum defect had correlations with RV GLSR(s) (r ( 1 )=-0.490, r ( 2 )=0.580, r ( 3 )=-0.528, respectively, P<0.05 for all). Tricuspid annular plane peak systolic velocity (Sm) was the independent predictor of RV global strain and strain rate (beta(1)=0.355, P (1)=0.031, beta(2)=0.307, P (2)=0.021). RV global longitudinal function is decreased in patients with TOF, especially in adult patients. STI is a sensitive and accurate technique in RV global functional assessment.
7.An in situ gel system for nasal delivery of menthol: preparation and safety evaluation

Objective To prepare an in situ gel system for nasal delivery of menthol and to evaluate the safety of this formulation.Methods Menthol in situ gel was prepared with deacetylatedgellan gum.The nasal mucocilia toxicities of this formulation was evaluated using in situ toad palate model.Guinea pig skin sensitization test and the rabbit skin irritation test were conducted.Skin allergy and irritation reaction were monitored and scored.Results No significant effect on nasal mucosa ciliary movement and the morphology of rat nasal mucosa were observed.The formulation did not induce any dermal irritation in rabbits.Skin allergic reaction was not found in guinea pigs.Conclusion The preparation of menthol in situ nasal gel with low ciliary toxicity was easily achieved.This gel has good physiological flexibility.The further investigation was warranted for this formulation as an intranasal drug delivery system.
8.Efficacy analysis of ruptured posterior communicating artery aneurysms with stent-assisted vs.non-stent-assisted coil embolization

Objective To compare the safety and efficacy of stent-assisted coil and non-stent-assisted coil for the treatment of ruptured posterior communicating artery aneurysms.Methods A total of 121 consecutive patients with ruptured posterior communicating artery aneurysm treated at the Department of Neurosurgery,Heze Municipal Hospital between June 2014 and June 2017 were enrolled retrospectively.They divided into either a stent group (n =63) or non-stent group (n =58) according to whether they used stent treatment or not.The two groups were compared,and their clinical data,surgical related complications,the degree of embolism immediately after aneurysm surgery,and the follow-up results of clinical and imaging were analyzed.Results Interventional embolization therapy was successfully performed in all the patients.(1) The proportion of wide-necked aneurysms of the stent group was higher than that of the non-stent group.There was significant difference (92.1% [58/63] vs.8.6% [5/58],x2 =84.249,P < 0.01).There were no significant differences in age,aneurysm size,sex,and HuntHess grade (all P > 0.05).(2) The incidences of intraoperative aneurysm rupture of the stent group and non-stent group were 4.8% (3/63) and 3.4% (2/58) respectively.There was no significant difference (x2 =0.132,P =0.717).The incidence of thromboembolie complication was 12.7% (8/63) and 5.2% (3/58)respectively.There was no significant difference (x2 =2.070,P =0.150).In the stent group,1 patient disabled and 1 died,and in the non-stent group,there were no cases with surgical related disability,and 1 died.(3) The comparison of immediate results after procedure between the stent group and non-stent group:there were no significant differences in the complete embolic rate (39.7% [25/63] vs.37.9%[22/58],x2 =0.039),aneurysm neck residual rate (25.4% [16/63] vs.39.7% [23/58],x2 =2.811],and residual rate of aneurysm (34.9% [22/63] vs.22.4% [13/58],x2 =2.298,all P > 0.05).(4) The recurrence rate of aneurysms (4.3% [2/46]) of the stent group was lower than that of the non-stent group (31.8% [14/44]).There was significant difference between the two groups (x2 =112.610,P =0.01).No rebleeding occurred in all aneurysms after procedure.Conclusion Compared with the non-stent-assisted coil embolization,the stent assisted embolization of ruptured posterior communicating artery aneurysms in acute phase may reduce the recurrence rate of aneurysms and without significantly increasing the incidence of surgery-related complications.
9.Cuprous oxide nanoparticles effect on the epithelial-mesenchymal transition of B16 cells

Objective To research the epithelial-mesenchymal transition (EMT) effects of cuprous oxide nanoparticles (CONPs) on melanoma.Methods Cuprous oxide nanoparticles were prepared hydrothermally.The B16 cells were cultured with cuprous oxide nanoparticlesat different concentrations (5,25,50 μg/ml).The changes of the morphology of the B16 cell were observed under the inverted microscope.The effects of CONPs on B16 cell migration ability were detected through the Wound healing assay and the Transwell assay.Then cell immunofluorescence and western blotting were used to test the EMT related molecular markers, including E-cadherin, N-cadherin, Cytokeratin, and Vimentin.Results The synthesized cuprous oxide nanoparticles distribute uniformly with a diameter of 40 nm.Our study indicated that CONPs inhibited the EMT of B16 cell.A conversion process was discovered in this study.In B16 cells, CONPs inhibited B16 cell migration, promoted the expression of E-cadherin, Cytokeratin and Desmoplakin, while the expression of N-cadherin and Vimentin was repressed in protein level.Conclusion Cuprous oxide nanoparticles can significantly restrain the invasion and metastasis of melanoma cells and inhabit the EMT of B16 cells.
