1.Optimized formulation of alkanin liposome by a combination of orthogonal design with central composite design/response surface method

AIM:To select an optiomal formulation of liposome entrapped alkanin prepared with ethanol injection in the combination of orthogonal design and central composite design/Response surface method.METHODS:Five influencing factors,including ratio of alkanin to liposome and soyabean lecithin to cholesterol,volume ratio of ethanol phase to water phase,soybean concentration and mannitol concentration were chosen as indices by orthogonal design.Then the method utilized the central composite design,which fitted multivariate linear and second/third order nonlinear regression equation,to find out the optimal formulation.RESULTS:Third order regression equation gained made the good prediction about entrapment(85.4%)and alkanin load(5.16%),and exhibited no significant difference between theoretic value and measured value.CONCLUSION:The combination of orthogonal design with central composite design is a feasible and convenient method to optimize the prescription of alkanin-liposome.

3.Quantitative Determination of Emod in and Chrysophanic Acid in Compound Rhubarb Spray by RP -HPLC

Objective To establish a method for the determi nation of emodin and chrysophanic acid in Compound Rhubarb Spray by HPLC.Methods The ODS column was applied.methanol∶water(87∶13)served as mobile phase.The detectio n wavelength was at 254nm and the flow r ate was 0.65ml /min.Results The average recovery of emodin was 99.48%with relative standard deviation being 1.75%(n=9)and the average recovery of chrysoph anic acid was 101.46%with rela-tive standard deviation being 2.85%(n=9).Conclusion The method was simple,rapid and reli able.It is suitable for the quality control of Compound Rhubarb Spray.
4.Study on micro-emulsions of volatile oil in rhubarb of compound liquid spray by triangular phase diagram

AIM: To optimize the best prescription of micro-emulsions of volatile oil in rhubarb of compound liquid spray by triangular phase diagram METHODS : Poloxamer-108 was used as emulsifiers,absolute alcohol as assistant emulsifiers,volatile oil from Lonicera dasystyla and Curcuma longa as the oil phase,the proportion between the emulsifier and append-emulsifier was established RESULTS : Optimized ratio of Poloxamer-108 and alcohol was 1∶10,the volatile oil in prescription formed micro-emulsions,distributing stably in the spray CONCLUSION : The study of micro-emulsions of volatile oil in compound liquid preparation of traditional Chinese medicine by triangular phase diagram offered a short-cut for resolving its stability. The experiment also suggested that the micro-emulsions could increase solubility of insoluble volatile oil from TCM.
5.Application value of real-time three-dimensional echocardiography in diagnosis of structural heart disease

Objective To explore the application value of real-time three-dimensional echocardiography (RT-3DE) in diagnosis of structural heart disease through a comparison with two dimensional echocardiography (2DE).Methods Ninety-two patients with heart disease were enrolled from October 2009 to February 2014 , all of whom were examined by RT-3DE.By cutting and spiraling structural 3D images of lesions in regions of interest , we made a diagnosis of lesions .In addition, the size of atrial(ASD) and ventricular septal defects (VSD),based on surgical results,was measured and analyzed.By comparing the coherence and difference of diagnosis on structural heart disease examined by 2DE and RT-3DE, the accuracy and superiority of RT-3DE were evaluated.Results (1)The 3D images of 92 patients with heart disease were successfully obtained by RT-3DE.The structure of lesions was displayed directly from various angles .In addition, the position, scope and degree of lesions and the spatial relationship of adjacent structures were accurately confirmed.RT-3DE provided more direct, accurate and abundant information than 2DE.(2)In the diagnosis of congenital heart disease, the diagnostic accordance rate of RT-3DE was the same as that of 2DE, but was slightly higher in the diagnosis of valvular disease and other lesions .( 3 ) Compared with 2DE, the ASD maximum diameters obtained from RT-3DE were more correlaed with surgical findings (r=0.94,0.86),so were the VSD maximum diameters (r=0.93, 0.84).Conclusion RT-3DE is easy to operate and quick to produce image ,which can provide more spatial information than 2DE in structural heart disease diagnosis , increase the accuracy rate of diagnosis , and serve as a supplement to 2DE.
6.The in-hospital mortality and its determinants for very elderly patients with acute myocardial infarction

ObjectiveTo explore the in-hospital mortality and its determinants for very eldly (80 + years of age) patients with acute myocardial infarction (AMI).MethodsA retrospective cohort method was used.The 499 study subjects were very eldly patients with newly diagnosed AMI consecutively admitted into our department between January 1,2002 and February 22,2010.ResultsNinety-seven out of 499 patients died during hospitalization period,with total in-hospital mortality of 19.4%.Multivariable logistic regression analysis showed the independent determinants for mortality of very elderly AMI patients were cardiac Killip grades,complete A-V block,renal dysfunction,stent implant,and the type of AMI.Conclusions The independent determinants for mortality of elderly AMI patients are as following,cardiac Killip grade,complete A-V block,renal dysfunction,stent implant,and the type of MAI.Urgent PCI is safe and effective for some very elderly with AMI,which could improve their survival rate within hospitalization period.
7.Intestional absorption and mechanism of tiliani in Caco-2 cell model.

OBJECTIVETo observe the uptake of tiliani in Caco-2 Cell.
METHODA human intestinal epithelial cell model Caco-2 cell in vitro cultured was applied to study the kinetics of uptake, transport and efflux kinetics of tiliani at small intestine. The effect of time, pH, drug concentration and inhibitors on the uptake of tiliani were investigated. The determination of tiliani was performed by HPLC.
RESULTTiliani in Caco-2 cell uptake was time-dependent. Tiliani in Caco-2 cell uptake was concentration-dependent at 4-16 mg x L(-1) consistent with passive diffusion process. The acid condition was good for the uptake of tiliani at pH 5-8. Compared with the control group, tiliani cell uptake was significantly higher after additional treatmeant with verapamil (1.545 +/- 0.010) mg x g(-1), (P < 0.05), and tiliani cell uptake was significantly lower after additional treatmeanet with sodium azide (0.994 +/- 0.003) mg x g(-1) (P < 0.05), with 2,4-dinitrophenol (1.174 +/- 0.030) mg x g(-1) (P < 0.05), and with phloridzin (1.098 +/- 0.021) mg x g(-1) (P < 0.05). Compared with the control group, tiliani cell uptake was not significantly after additional treatmeant with lactose (1.470 +/- 0.025) mg x g(-1), Papp of Basolateral to Apical was much more than that of Apical to Basolateral (1.10 Fold).
CONCLUSIONP-glycoproteins and SGLT1 participate in the conveying process of tiliani in Caco-2 cells. The uptake of tiliani has no relationship to LPH. passive transport and carrier-mediated transport participate in the uptake process of tiliani in Caco-2 cells.

8.Optimization of extraction process for compund kaliziran gel with orthogonal design.

OBJECTIVETo optimize the extraction process of compund kaliziran gel.
METHODOrthogonal design was used to optimize the extracting process of eight herb medicines with the extract yield, the content of psoralen, isopsoralen and osthole as indexes, and to optimize the extracting process of Polygonum multflorumi and Seman sinapis albae with the extract yield and the content of 2,3,5,4'-tetrehydroxy-stilbene glucoside as indexes.
RESULTThe optimum extraction progress was as follows:eight herb medicines were extract 2 times for 1 hour with 8 times of 60% alcohol; P. multflorumi and S. sinapis albae were extract 2 times for 1 hour with 8 times of 40% alcohol.
CONCLUSIONThe optimum extraction processes are stable and feasible.

OBJECTIVETo investigate the chest X-ray manifestations of SARS cases.
METHODSA retrospective study was conducted among 52 clinically confirmed SARS patients from February 9 to May 10, 2003. Chest X-ray scanning was performed at a interval of 1 - 3 days according to the requirements. The manifestations and special features of SARS in X-ray were analyzed.
RESULTSSmall or large patchy shadows with intensive density in both lungs were observed in 31 cases, ground-glass like opacification in 16, small patchy shadows in one lung lobe or one lung segment in 18, nodular shadows in one lung segment in 1, and increased lung marking in lung interstitial tissues in 2. Rapidly changing consolidations revealed in chest X-ray images were found to be associated with SARS infections, and they were not affected by treatment with antibiotics.
CONCLUSIONChest X-ray provides a sensitive and specific method for the diagnosis and treatment of SARS, and those present with symptoms and signs should undergo chest X-ray scanning every 1 - 3 days.
