1.Inverted Papilloma of the Nose and Paranasal Sinuses:Analysis of CT Misdiagnosis

Objective To analyses the misdiagnostic reasons of inverted papilloma of the nose and paranasal sinuses by CT.Methods 10 patients with inverted papilloma of the nose and paranasal sinuse,to be misdiagnosed by CT before surgery were analysed.Results The CT findings of inverted papilloma of the nose and paranasal sinuses were the crawling growths of soft-tissue mass in the one side of the nose and paranasal,there were slight to moderate enhancement and probably with destruction of bone and osteosclerosis.Conclusion The rate of CT misdiagnosis of inverted papilloma in nose and paranasal sinuse can be reduced if we are strictly followed the operational rules and to do CT contrast enhance scan.
2.Therapeutic effect of Xuebijing injection on acute lung injury in a rat model of cecal ligation puncture

Objective To investigate the therapeutic role of Xuebijing injection on acute lung injury (ALI) in a rat model of cecal ligation puncture (CLP) and the potential mechanism.Methods A total of 110 SD rats were divided into sham group (n =30),CLP group (n =40) and Xuebijing treatment group (n =40),according to the random number table.Survival was observed up to 7 days.Rats in each group were sacrificed 12 h after the CLP surgery.Blood samples were obtained by abdominal aorta puncture and inflammatory cytokines and blood gas were detected.Samples of the lung tissue were excised to measure mRNA expression of cytokines,activation of signaling molecules,wet to dry ratio and histopathological changes.Peripheral neutrophils were isolated in vitro and divided into negative control group,high-dose Xuebijing (1 ∶ 30 dilution) group,lipopolysaccharide (LPS,100 ng/ml) group,LPS plus low-,middle-and high-dose Xuebijing (1∶ 300,1∶ 100,1∶30 dilution respectively) groups.Supernatant levels of tumor necrosis factor (TNF-α),interleukin (IL)-6 and neutrophil elastase (NE) in each group were detected.Activation of intracellular signaling molecules was measured as well.Results In animal survival observation,the mortality was 65 % in CLP model group and reduced to 30% in Xuebijing treatment group (P < 0.01).Compared to sham group,serum levels of TNF-α,IL-6,PaCO2 and lactate in CLP group were increased markedly to (32.213 ± 9.802) pg/ml,(21.592 ± 5.427) ng/ml,(39.575 ± 5.295) mmHg and (3.438 ± 0.192) mmol/L respectively,mRNA expressions of TNF-α and IL-6 in CLP group were increased to 5.223 ±0.525 and 12.097 ±2.535 respectively,activation of IκB-α,p38,extracellular regulated protein kinases (ERK) and c-Jun N-terminal kinase (JNK)in CLP group were significantly up-regulated,and water content of rat lung tissue in CLP group were significantly increased to (78.226 ±0.894) % (all P <0.01).Moreover,histopathological appearance of bleeding,exudation and increased infiltration of neutrophils to the alveolar septum were detected in the lung tissue of CLP rats.Compared to CLP group,Xuebijing treatment reduced the serum levels of TNF-α [(21.592 ± 5.427)pg/ml],IL-6 [(2.252 ± 0.905) ng/m],PaCO2 [(34.088 ± 2.260) mmHg] and lactate [(2.563 ± 0.250)mmol/L],suppressed the up-regulation of TNF-α and IL-6 mRNA expressions [6.866 ± 1.604,3.398 ± 0.334],and reduced the water content of rat lung tissue [(77.159 ± 0.626)%] (P <0.05 or 0.01).Compared to CLP group,Xuebijing treatment ameliorated the histological appearance of ALI.In vitro study,negative control group and high-dose Xuebijing group showed basal levels of TNF-α,IL-6 and neutrophil elastase (NE).Levels of TNF-α,IL-6 and NE in cell supernatant were elevated in LPS group [(7 102.520 ± 528.026) pg/ml,(3 109.598 ± 191.291) pg/ml and (63.985 ± 2.823) ng/ml respectively],with significant differences from these detected in negative control group (P <0.01).While the release of TNF-α,IL-6 and NE was significantly inhibited in LPS plus middle-and high-dose Xuebijing groups,compared to LPS group (P < 0.05 or P <0.01).LPS also increased the degradation of IκB-α and phosphorylation of p38 and ERK in neutrophils,and Xuebijing in higher concentration markedly suppressed the degradation of IκB-α and phosphorylation of p38 and ERK.Conclusions CLP surgery results in the appearance of typical ALI in rats.Administration of Xuebijing can effectively inhibit the proinflammatory response in CLP rats and attenuate ALI.And such effects may be attributed to its inhibition on the proinflanunatory activation of neutrophils in lung tissue.
3.CT Study of Type-Ⅲb Aortic Dissection

Objective To explore the CT manifestations and study the value in diagnosing type Ⅲb aortic dissection.Methods CT scanning was performed in 16 cases with type-Ⅲb aortic dissection.The imaging findings were analyzed retrospectively.Results All the cases survived beyond 5 years,12 cases were showed crescent-shaped valve,the false lumen in left was obtained in 15 cases,thrombosis in 9 cases were demonstrated in false lumen and 6 cases showed leakage complications.Conclusion The rate of five years survival is high in cases with type-Ⅲb aortic dissection.CT feature of type-Ⅲb aortic dissection is the crescent-shaped valve with the false lumen in the left.
4.Design and preliminary experiment of an intelligentized physiologic pulsatile flow cardiac support system.

A patent cardiac support system which is used as a bridge treatment for acute myocardial infarction has been designed and tested in vitro and in two dogs in vivo. This is an easy-to-use intelligentized pulsatile flow cardiopulmonary bypass device to replace the function of heart. The device consists of two identical pumps and perfusion chambers, a sensing and control system, a gas exchanger between the vein and pump, two one way valves between pump and veins or arteries. Arterial pressure and EKG feedback mechanisms are used for maintaining blood pressure and coordinating the pumping activity with heart contraction. A prototype of the device was built to perform hydraulic in vitro tests with aims of verifying the new device's pumping behavior. Functional evaluation of the device was carried out by using it in a model circuit made with standard CPB components plus a mock hydraulic pipeline. This system demonstrated easy manipulation, good controllability, and provided a 65+/-2ml x beat(-1) flow volume. There was a linear correlation between peak pressure value and pulsatile frequency. In the two in vivo experiments, the primary objective was to determine whether the device could work well in dog, whether physiologic pulsatility could be achieved and whether the blood supply to heart should be sufficient during asystole status by drugs. The results suggest that all the goals have been achieved.

5. Study on iTRAQ-based differential expression protein analysis in nuclear cataract lens with different axial length

Objective:
To identify and verify differentially expressed proteins in lens of nuclear cataract eyes with different axial lengths.
Methods:
Forty-eight nuclear cataract patients with the hardness grade Ⅲ to Ⅳ were enrolled from January 2016 to December 2017 in the east area of Xining city.The lens nucleus was obtained during extracapsular cataract extraction (ECCE) surgery.The samples were divided into six groups according to age and axial length, with 8 samples in each group.Seven normal transparent human lens nuclei were obtained from donors during corneal transplantation surgery and served as normal control.Water-soluble, water-insoluble and water-insoluble-urea-soluble protein fractions were extracted separately and merged to analyze by using 8-plex isobaric tags for relative and absolute quantitation (iTRAQ) labeling combined with 2D-LC/MS/MS.The data were processed with ProteinPilot software.Two technical replicates of iTRAQ analyses were carried out.Up-regulated and down-regulated proteins were selected in two replicates as candidate biomarkers.Twenty lens nuclei from 20 nulear cataract patients were divided into long axis young age group, normal axis young age group, long axis old age group and normal axis old age group.Western blot was used to confirm the most different proteins.This study protocol was approved by the Ethics Committee of TangDu Hospital of Air Force Medical University (TDLL2015046). Written consent was obtained from each participant.
Results:
Eighty proteins were identified.Six proteins were selected as differentially-expressed proteins.Sorbitol dehydrogenase (SDH) was elevated whereas beta-crystallin B2 (CRYBB2) was decreased in the long axis groups.The relative expression of SDH protein in the long axis low age group was 1.204±0.142, which was significantly higher than 0.711±0.072 in the normal axis low age group, the relative expression level of SDH in the long axis old age group was 1.092±0.334, which was significantly higher than 0.617±0.089 in the normal axis old age group, the difference was statistically significant (all at

Interferons (IFNs) are cytokines with fundamental roles in resistance to infections, cancer and other diseases. Type-I IFNs, interferon (IFN-) and interferon (IFN-), act through a shared receptor complex (IFNAR) comprised of IFNAR1 and IFNAR2 subunits. Binding of type-I IFN to IFNAR1 will robustly activate Janus activated kinase-signal transducer and activator of transcription (JAK-STAT) signaling pathway. Aberrant activation of the type-I IFN response results in a spectrum of disorders called interferonopathies. The purpose of this research is to develop an assay for high-throughput screening (HTS) of small molecule inhibitors of the type-I IFN signaling pathway. Inhibition of type-I IFN signaling can be beneficial in terms of therapeutic use and understanding the underlying mechanism of action. We report here a HTS campaign with the secreted embryonic alkaline phosphatase (SEAP) reporter gene assay against 32,000 compounds which yielded 25 confirmed hits. These compounds were subsequently characterized for their cytotoxicity, effects on STAT phosphorylation and activities in IFN regulatory factor (IRF) transcription.
