3.Research progresses of Mycobacterium tuberculosis cytochrome P450s as a potential drug target.

Identification and validation of a new target is one of the most important steps for new antituberculosis (TB) drug discovery. Researches have shown that Mycobacterium tuberculosis (Mtb) encodes 20 CYP450 enzymes which play important roles in the synthesis and metabolism of lipid, cholesterol utilization, and the electron transport of respiratory chain in Mtb. With the critical roles within the organism as well as the protein structures of six Mtb CYP450 enzymes being clarified, some of them have been highlighted as potential anti-tuberculosis targets. In this paper, the phylogenetic analysis, the structural features, and the enzymatic functions of Mtb CYPs, as well as the mechanism of interactions with selective inhibitors such as azole antifungal agents for the CYPs have been reviewed and summarized. The druggability of the CYPs has also been analyzed for their further utility as targets in high throughput screening and rational design of more selective inhibitors.

4.The co-vaccination effects of BCG and Poly I:C on the development of T cell subsets in neonatal BALB/c mice

Objective To explore the effects of BCG and Poly I:C co-vaccination on the development of spleen T cell subsets of neonatal BALB/c mice. Methods Neonatal BALB/c mice were inoculated with BCG and/or Poly I:C intraperitoneally within 2-3 d after birth. Four weeks later, spleen cells of mice were isolated and the percentage of CD3+ CD8+ IFN-γ+,CD3+ CD8-IFN-γ+,CD3+ CD8+ IL-4+,CD3+ CD8- IL-4+,CD4+ Foxp3+ T cells,which represent Tc1,TH1,Tc2,TH2,Treg cells,respectively,were tested by flow cytometry at single cell level,and the ratios of TH 1/TH 2 and Tc1/Tc2 were calculated. Results The percentages of TH1 and Tc1 cells of BCG-vaccinated mice,Poly I:C-vaccinated mice and BCG plus Poly I:C-vaccinated mice were significantly higher than that of control mice(P<0.05 or P<0.01),and there was no difference among the three vaccinated group. The ratios of TH1/TH2 and total IFN-γ/IL-4 of the three vaccinated groups were higher than that of control group,but not the ratio of Tc1/Tc2. The TH1/TH2 ratio of BCG plus Poly I:C-vaccinated group was higher than that of BCG-vaccinated group(P<0.05).The percentages of Trge cells showed no difference among the four groups(P>0.05). Conclusion BCG and Poly I:C co-vaccination can significantly increase the number of Tc1 and TH 1 cells and TH 1/TH2 ratio in spleen cells. BCG and Poly I:C vaccination may have a synergistic effect on TH 1/TH2 ratio of spleen cells in neonataI mice. The percentage of CD4+ Foxp3+ T cells among four groups showed no significant difference.
5.Thinking on the application and students' feedback of classic PBL in a medical microbiology course

Classic problem-based learning ( PBL ) was applied in a medical microbiology course.The results of questionnaire survey showed most of the students were satisfied with the whole learning program.They thought the case was appropriate,and the discussing could increase their understanding of the knowledge.Based on the problems faced during the application and the students' feedback,the duty of the tutors,summary of the learning topic,arrangement of the course,application of different teaching methods,and selection of the tutors were discussed in detail.
6.Analysis on Medication Rules of Using Chinese Medicine in Reducing Recurrence and Metastasis of Stage II and III Colorectal Cancer among Postoperative Patients

This study was aimed to analyze the living status and medication rules of traditional Chinese medicine (TCM) treatment of postoperative patients with stage II and III colorectal cancer by Prof. Y ang Y ufei. A total of 70 postoperative patients with stage II and III colorectal cancer were enrolled in the cohort study at the Xiyuan Hospital from September 2007 to August 2009. The out-patient records were reviewed and follow-ups were given for the analysis. The results showed that until July 2013, the loss to follow-up rate was 8.6%, which was less than 15%. The four-year postoperative recurrence and metastasis rates of the high and low exposure group were 7.1% and 25.0%, respectively. The Fisher exact probability test showed that the high exposure and long period therapy of TCM treatment can significantly reduce the recurrence and metastasis rate of patients (P < 0.05). The analysis of Prof. Y ang Y ufei's medication rules in TCM treatment of postoperative colorectal cancer showed that the syndrome differ-entiation pattern with the highest frequency was spleen-stomach deficiency pattern; and the liver-kidney yin defi-ciency pattern was in the second place. The prescriptions used with the highest frequency were Si-Jun-Zi Decoction and Liu-Wei-Di-Huang Decoction. It was concluded that the comprehensive TCM treatment can improve the prog-nosis of stage II and III colorectal cancer. The treatment also has certain meaning in the reducing of the recurrence and metastasis rate of patients, and prolonging the time of the recurrence and metastasis.
7.Study on determining method of dl-tetrahydropalmatine by TLCS

AIM: To study the determination of dl-tetrahydropalmatine (THP) in Chi nese traditional patent medicines by TLC-Scanning from the viewpoint of the pro cess control of manufacture. METHODS: To select the 5 kinds of representative drugs and its prep arations contained, THP was extracted with solvent method. The mobile phase contains diethylamine. THP w as separated with high-efficiency silica gel plate or handcrafted silica gel p late. Iodine was used for chronogenic agent and fluorescence defectiion for dete rmination. TLC-Scanning results were compared with HPLC. RESULTS: The method is practicable, the result is correlative with the HPLC. CONCLUSION: TLC-Scanning is fit for the quality control of the Chi nese traditional patent medicine in the process of manufacture.

9.Influence of nodule enhanced viewing of dual-source CT on efficacy of detecting pulmonary nodule in low-dose CT

Objective To evaluate efficacy in detecting lung nodules at low-dose CT(LDCT) by nodule enhanced viewing(NEV).Methods One hundred and twenty seven patients who were referred to undergo low-dose CT (LDCT) for the evaluation of pulmonary metastasis or screening lung cancer were selected randomly.Two radiologists with at least 10 years experience read the images with normal clinical reading speed to find actionable nodules ≤ 2.0 cm in maximum diameter,and their consensus result was referred as Standard.NEV was adopted to detect the pulmonary nodules.Two residents with experience of less than three years read first detected suspicious nodules and recorded reading time,first consensus and mean time were recorded.Then,they made second decisions on the images with the help of NEV and the results and the reading time were recorded and analyzed by using wilcoxon test.The sensitivity and accuracy of NEV,residents and residents with NEV were analyzed.Results Standard,resident,NEV and resident with NEV detected 570,404,768 and 593 lung nodules ≤2.0 cm in maximum diameter,respectively.More than 60% nodules were less than 0.5 cm in maximum diameter.The performance of NEV in detecting nodules ≤2.0 cm as well as nodules < 0.5 cm in maximum diameter was significantly higher than that of the resident(Z =-6.887,P <0.01 and Z =-7.235,P <0.01),and the performance of resident with NEV indetecting nodules ≤2.0 cm as well as nodules < 0.5 cm in maximum diameter was significantly higher than that of resident without NEV (Z =-6.606,P < 0.01 and Z =-6.657,P < 0.01).The resident,NEV and the resident with NEV detected nodules < 20 mm in maximum diameter with sensitivities of 61.4%,86.3% and 95.3%,and with accuracy of 56.1%,58.1% and 87.6%,respectively.The resident achieved sensitivities of 51.4%,88.1% and 94.8%,and accuracy of 47.0%,56.9% and 87.5% for nodules <5 mm in maximum diameter,respectively.The resident,NEV and resident with NEV spent 120-444 s,85-262 s and 131-1512 s per case to read the CT scans,respectively.The reading time of resident with NEV in was significantly higher than that of resident without NEV(Z =-9.781,P < 0.01).The resident spent 23 s per NEV mark.Conclusion NEV considerable improves the resident's performance in lung nodule detection,especially in maximum diameter < 0.5 cm nodule detection.
10.Construction of eukaryotic expression vector of iASPP and its identification of biological function

Objective Construct the eukaryotic expression vector of inhibitory member of the ASPP family (iASPP) and trans-fect it into colon carcinoma cell lines SW480 and Lovo by liposome .Then observe the expression of iASPP and detect the cell apop-tosis by flow cytometry .Methods The amplified PCR product was digested and inserted into pMD19-T simple vector and sub-cloned into eukaryotic expression vector pcDNA3 .1(+ ) .The recombinant eukaryotic expression plasmid pcDNA3 .1(+ )-iASPP was transfected into colon carcinoma cell lines SW480 and Lovo by liposome ,the iASPP expression was analyzed by RT-PCR .The cell apoptosis was detected by FCM .Results The eukaryotic expression plasmid pcDNA3 .1(+ )-iASPP was constructed success-fully ,the gene squence of iASPP was consistent with that reported (gi 60457962) in GenBank .The mRNA expression levels of iASPP gene of SW480 and Lovo cell lines which transfect the positive plasmid were increased ,and the cell apoptosis rates were de-creased .Conclusion We successfully constructed the recombinant expression plasmid pcDNA 3 .1(+ )-iASPP ,and the plasmid were successfully expressed in colon carcinoma cell lines SW 480 and Lovo ,the cell apoptosis rates of those cell lines were decreased .These facts indicated that reducing the high expression of iASPP may be a new strategy to renew the abilities of P 53 tumor suppressor .
