Objective To study the effect of varicocele(VC) on the expressions of hypoxia-inducible factor-1?(HIF-1?) and apoptosis-associated gene Bax in germ cells of adolescent rats,and investigate the mechanism of the infertility resulting from varicocele. Methods The varicocele model was established by partial ligation of the left renal vein in adolescent male Wistar rats. A total of 25 rats were randomly divided into 2 groups,namely varicocele group(n=15) and sham operation group (SOG,n=10). All the animals were killed 30 d after operation. The left testis was collected and the expression of HIF-1? and Bax was detected by immunohistochemical method. Objective To explore the proliferative and apoptotic effect of 3 different siRNAs targeting survivin in human bladder cancer cell line EJ28. Methods Three siRNAs targeting survivin and 1 fluorescence-labeled siRNA as a negative control were chemically synthesized. Transfetion efficiency was observed under fluorescence microscope after transfecting the fluorescence-labeled siRNAs. EJ28 cells were divided into 6 groups: siRNA164 group, siRNA167 group, siRNA389 group, and negative control group, lipofectin group and cell control group. The proliferation of EJ28 cell was detected by MTT assay at 24, 48 and 72 h after the transfection. Apoptotic rate was detected by Annexin V-FITC-FCM assay at 48 h.Results All the sequence-specific siRNAs targeting survivin efficiently suppressed the proliferation of EJ28 cells. siRNA 164 had the most powerful effect because of its highest inhibitory rate of (41.32?2.54)% and the highest apoptotic rate of (13.20?0.25)% at 48 h after transfection. Conclusion RNAi targeting survivin has a potential value in the gene therapy of bladder cancer.

Abstract：In recent years，the outbreak and prevalence of respiratory infectious diseases in the world seriously endanger human health，among which the respiratory infectious diseases caused by viral infection account for a large proportion. The use of vaccines and common antiviral drugs is an effective way to fight viral infection，but there are also problems such as lag and drug resistance. Monoclonal antibodies against respiratory viral infections provide a new strategy for clinical treatment. This paper reviews the development of monoclonal antibody against respiratory virus and its application in respiratory viral infectious diseases. A pot experiment was conducted to study effect of drought stress on leaf physiological characteristics and growth of one year old Stellaria dichotoma seedlings. The result showed that plant height and shoot dry weight significantly decreased with decrease in soil water content; however, root length and root dry weight increased at light drought stress and decreased at severe drought stress. The result also showed that with the decrease of soil water content, proline content in S. dichotoma leaves decreased then increase, while solube protein content decreased. Activities of SOD and POD in S. dichotoma leaves significantly decreased as soil water content decreased, while activity of CAT significantly decreased at severe drought stress. Membrane permeability in S. dichotoma leaves increased, while MDA content decreased then increased as soil water decreased. These results suggest that S. dichotoma had osmotic stress resistance ability and reactive oxygen scavenging capacity at light drought stress, which caused S. dichotoma growth was no inhibited at a certain extent drought stress.

OBJECTIVETo explore the clinical application value of Provider-Initiated HIV Testing and Counseling (PITC) by analyzing the positive rate of HIV tests for people in need of PITC and that of routine HIV tests.

METHODSWe retrospectively analyzed the demographic and epidemiologic data about the patients seeking PITC services or undergoing routine HIV tests in Nanjing Drum Tower Hospital between January and December 2013.

RESULTSThe positive rate of initial HIV screening was 1.98% in the PITC group and 0.24% in the routine test group, while that of confirmed HIV was 0. 40% in the former and 0.07% in the latter, both with statistically significant differences between the two groups (P < 0.01). The positive rate of HIV was markedly higher in males than in females, particularly in the PITC group.

CONCLUSIONPITC has a high clinical value in HIV detection for targeted subjects and therefore deserves general application in dermatology.

Objective To obtain differential expression genes from colorectal cancer cells derived from colo205 for further research. Methods RNA from colo205 cells,CD133+cells and CD133-cells were sequenced and analyzed by bioinformatics software. Results One hundred and twenty four differential expression genes were obtained, which involves 32 metabolic pathways. Conclusions Large quantities of differential genes can be found among different groups of cells derived from colo205 cells , which can provide epigenetic evidence for colorectal cancer research.

Objective To investigate SLC25A13gene mutation in neonatal intrahepatic cholestasis caused by citrin defi-ciency (NICCD). Method A total of 17 children with NICCD were collected. PCR-RFLP method was used to analyze the most common eight mutations of SLC25A13 gene in Chinese populations and results were analyzed together with routine laboratory examinations. Results In the 17 NICCD patients, there were six cases of homozygous mutation, three cases of compound heterozy-gous mutation and eight cases of single heterozygous mutation in SLC25A13 gene. Three kinds of mutations detected were 851del4 (73.1%), 1638ins23 (11.5%) and IVS6+5G>A (15.4%). The seventeen cases showed classical NICCD symptoms of low birth weight, pathological jaundice. And laboratory data suggested liver dysfunction, hyperbilirubinemia, hyperbileacidemia, hy-poproteinemia, hypoglycemia, coagulation disorders, hyperlactacidemia and hyperammonemia. Conclusions 851del4, 1638ins23 and IVS6+5G>A are hot spots of SLC25A13 gene mutation in Chinese populations. PCR-RFLP is a rapid, convenient and reliable technology for NICCD molecular diagnosis.

Objective:To evaluate the impression accuracy of different impression methods for flabby ridge.Methods:5 patients with flabby ridge were included.Conventional technique,perforation technique and window opening technique were performed to make im-pressions and then final models were prepared.The optical models of the flabby ridge area were obtained both in patients'mouths and on the master casts by intraoral optical scanner.Data handling and computations were made by using 3D inspection and metrology soft-ware,regarding SD,Mean +and Mean -,with subsequent analysis.Results:The 3D deviation of the models obtained by conventional technique was the biggest,that obtained by window opening technique was the least.Conclusion:Window opening technique can de-crease the deformation of flabby ridge impression.

Objective To explore the aesthetic results of medial upper Iip repair for skin and soft tissue defects using local flap.Methods According to the location and the size of upper lip defect,the modified rhomboid flap of 60 ° above the defect was designed to repair the wound in philtrum;the O-L flap along philtral column to vermilion border was designed to cover the wound close to the peak and lateral to philtral column.Results Twenty-one postoperative patients that underwent pigmented nevus removal (12 female,9 male;ages 16-33 years) had defects in philtrum for 10 cases and close to the peak and philtral column for 11 cases.The diameter of the defect ranged from 0.5 cm to 1.0 cm.All flaps survived primarily without any complications and follow-up was for 1 to 18 months with excellent outcomes in all cases.All patients were satisfied with contour and functions.No pigmented nevus recurrence occurred.Conclusions The flap designed according to the aesthetic principle could be used to repair medial upper lip defect of medium-sized,which not only obtains functional reconstruction,but also satisfies aesthetic results,with aesthetic subunits saved and scar concealed.
