Objective To improve recognition and imaging diagnosis of aneurysmal bone cyst secondary to a giant cell tumor. Methods To collect the dates of 12 patients with aneurysmal bone cyst secondary to a giant cell tumor were proved by operation and pathology from January 2003 to October 2006. Analyzed and summarized their imaging manifestations and correlation with pathohistology. Results Six lesions were located in epiphysis and metaphysic regions of long bone.Six lesions were located in pelvis. All cases showed a cystic lesion with expanded and osteolytic, eccentric 10 cases and centric 2 cases.Four cases display trabeculate, the margin is well define with a rim of bone sclerosis in 2 cases.Magnetic resonance imaging(MRI)scans were available in 10 patients. All case showed cystic, dilated lesions with solid areas. Eight cases manifested single or multitude solid nodules in big cystic wall. Two cases appeared solid masses with multitude cysts. The sign of multitude fluid-fluid level, best seen on T_2-weighted images, was present in all patients.Seven cases emerged soft-tissue masses.MR found indicative of large amounts of hemosiderin in one cases.Eight cases were examined by spiral CT with plain scanning and enhancement scanning. Reconstructed image were CTA and 3D-MPR(three dimensions multiplanar reconstruction)imaging. All cases showed cystic, dilated lesions with solid areas. The sign of multitude fluid-fluid level was present in 6 patients. The solid areas and cystic-wall of lesions showed contrast enhancement in 8 patients.3D-MPR imaging showed supply blood vessel of tumors in 3 cases.Arteriovenous malformation did not found in all patients. The surgeons'operative findings and the gross specimens were studied in all patients. All lesions were composed of solid areas and cystic areas. The diagnosis of pathology were ABC with GCT(grade Ⅱ)in 10 cases and ABC with GCT(grade Ⅲ).Conclusion Aneurysmai bone cyst secondary to a giant cell tumor is not rare.Adequately recognizing the pathologic basis of ABC,and selecting imaging techniques correctly (X-ray and MRI,or X-ray and CT)is especially important to diagnose a giant-cell tumor with secondary aneurysmai bone cyst. When an eccentric, expanded, lytic tumor with a cystic-solid lesion in epiphysis of long bone or pelvis shows multiple fluid levels,a giant-cell tumor with secondary aneurysmai bone cyst components should be sufficiently considered.

The highly-pathogenic EV71 strain is the primary cause of mortality in hand-foot-and-mouth disease (HFMD) associated with neurological symptoms, for which no clinically effective drugs or vaccines exist. This study aimed to construct infectious cDNA clones of the EV71 highly-pathogenic strain and the cell-culture adapted strain using a reverse genetics approach. The genomic RNAs of EV71 parent strains were used as the templates for RT-PCR amplification, and then the PCR products that overlapped the full-length genome were connected into the pBR322 vector to produce infectious clones of pEV71 (HP) and pEV71 (CCA), respectively. The results showed that the HP strain propagated much more quickly than the CCA strain. The rescued viruses derived from the infectious clones not only maintained their consistency with their parent strains in terms of genomic sequences, but also retained their respective biological phenotypes. This research will contribute to our understanding of EV71 pathogenesis and the development of novel vaccines against HFMD.

AIMTo study the pharmacokinetics of spinosin in rat plasma after oral administration of Suanzaoren extract using sulfamethoxazole (SMZ) as internal standard by RP-HPLC method.
METHODSPlasma samples were deproteined with acetonitrile, followed by evaporation of the acetonitrile to dryness. The residual was then resolved in mobile phase and HPLC separation was achieved on a Hypersil C18 (5 microns, 200 mm x 4.6 mm ID) column at 35 degrees C. The mobile phase consisted of acetonitrile-water-acetic acid (15:85:1) at a flow rate of 0.7 mL.min-1. The UV detection wavelength was set at 334 nm.
RESULTSThe calibration curve was shown to be linear over the range from 18.1 to 903.5 micrograms.L-1 (r2 > or = 0.995). Mean recovery was 94.5%. Within-day and between-day precisions RSD were less than 9.0%. The limit of quantitation was 18.1 micrograms.L-1. The plasma spinosin was stable at -20 degrees C.
CONCLUSIONThe simple, sensitive and accurate HPLC method developed has been applied to determine the pharmacokinetics of spinosin in rat plasma after having taken Suanzaoren extract at a single dose.

Objective To investigate the effects of amphiregulin antibody on the axial length, diopter and posterior sclera thickness in eyes of bilateral lens-induced myopic guinea pigs. Methods A total of 60 healthy three-colored short-hair guinea pigs were randomly divided into 5 groups:myopic model group, low dose group, medium dose group, high dose group and normal control group,12 rats in each group, and both eyes of the guinea pigs in the former four groups were induced by-10 D lens, while the normal group did not make any treatment. After wearing of goggles for 2 weeks, the right eyes of guinea pigs were intravitreally administrated with 5 μg,10 μg,20 μg anphiregulin antibody for the low, medium and high dose group, respectively. At the same time, the left eyes of these groups were given the same dose of Ringer' s buffer (buffer group).Continuous wearing of the lenses before and after injection of antibody was allowed.Diopter and axial length of guinea pigs were measured before wearing of the lenses,2 weeks after wearing lenses and 5 weeks after the experiment (after 3-times injections of antibody). Moreover, the thicknesses of retina, choroid and sclera in the posterior pole were detected by PAS staining. Finally, the expression of amphiregulin and EGFR mRNA and protein was detected by real time quantitative PCR and immunofluorescence. Results After 2 weeks of lens induction, the axial length of the myopic model group increased, but the diopter decreased when compared with the normal control group, and the differences were statistically significant (both P < 0.05).Compared with the buffer group, after intravitreal injection of amphiregulin antibody, the axial length in the low, medium, and high dose groups decreased, while the diopter increased, and the scleral thickness at the posterior pole increased significantly in a dose-dependent manner, with statistically significant differences (all P < 0.05).However,there was no significant change in retinal thickness at the posterior pole of all groups.Real-time quantitative PCR and immunofluorescence showed the expression of amphiregulin mRNA and protein in the retina was upregulated the myopic model group and buffer group, but downregulated in the high, medium, and low dose groups.Furthermore, when compared with the buffer group, the expression level of epidermal growth factor receptor in the retina of the low dose group was decreased (t =2.606,P =0.022),but there was no significant difference in the other groups. Conclusion After injection of amphiregulin antibody into the eyes of bilateral lens-induced myopic guinea pigs, the diopter increases, but the axial length is significantly shortened, and the posterior sclera is thickened, which may involve a decrease in the expression of amphiregulin.
