1.Efficacy and prognosis of tetramethylpyrazine injection as an adjuvant of 1,6-diphosphate on neonatal hypoxic-ischemic encephalopathy

Objective To probe into clinical application value of tetramethylpyrazine injection as an adjuvant of 1,6-diphosphate in the treatment of neonatal hypoxic-ischemic encephalopathy.Methods From January 2012 to December 2012,98 neonatal hypoxic-ischemic encephalopathy children patients were selected in the Second Affiliated Hospital of Shenyang Medical College.The children patients were divided into observation group and control group randomly.There were 49 cases in each group.Both two groups adopted 1 ,6 diphosphate - fructose,and the observation group adopted ligustrazine injection on the basis of control group.The clinical treatment and prognosis of the children patients in two groups were compared and analyzed.Results The rates of significant efficiency and total efficiency were 67.35%and 93.88%in observation group,which were higher than those in control group (51.02%,71.43%),and there were statistically significant (P <0.05 );compared with control group,the NBNA scores of observation group were improved significantly after 3rd,7th and 14th days treatment,and there were statistically significant (P<0.05);compared with control group,the ratio of cerebral palsy,epilepsy,mental retardation and death in observation group showed different degree of reduction,but there were no statistically significant.Conclusion The effect of tetramethylpyrazine injection adjuvant 1 ,6-diphosphate in treatment of neonatal hypoxic-ischemic encephalopathy was effective and significant.It has positive role in promoting the improvement and upgrading of the clinical efficacy and prognosis.
2.Isolation of CD44~+/CD24~(-/low) and side population cells from MDA-MB-453 cells and the analysis of their activation of Wnt and Notch pathway

Objective:To explore stem cells phenotype of human epidermal growth factor receptor 2(HER2)-overexpressing breast cancer cells,MDA-MB-453 by isolation of CD44+CD24-/low and side population(SP).Analyze the relationship between SP cells and the gene amplification levels of the key proteins,?-catenin and Notch-1 in Wnt and Notch signal pathways.And study the correlation of SP cells and the gene amplification of HER2 and human epidermal growth factor receptor 3(HER3).Methods:CD44+CD24-/low and SP phenotype were studied in MDA-MB-453(HER2-overexpressing cell line)and MCF7(HER2 negative cell line)respectively by flow cytometry.The gene amplification of ?-catenin and Notch-1 in Wnt and Notch pathways were also studied by reverse transcriptase polymerase chain reaction(RT-PCR).The gene amplification levels of the proteins mentioned,HER2 and HER3 were roughly compared in MDA-MB-453 cells and their SP by RT-PCR.Results:There was no CD44+CD24-/low subpopulation in MDA-MB-453 cells,However,SP could be observed in MDA-MB-453.The gene amplification levels of ?-catenin and Notch-1 in Wnt and Notch pathways were both observed in MDA-MB-453 and MCF7.Their gene amplification levels were more evident in SP of MDA-MB-453 than in MDA-MB-453 cells.However,the similar gene amplification levels of HER2 and HER3 were observed.Conclusion:SP can be enriched in breast cancer stem cells of MDA-MB-453.Wnt and Notch pathways are activated in MDA-MB-453,which may be related to maintenance and activity of breast cancer stem cells of MDA-MB-453.

The development of EIT is summarized based on the concept and advantages of the Electrical impedance tomography (EIT) from the following three aspects: multi-frequency electrical impedance tomography (MEEIT), induced current electrical impedance tomography (ICEIT)and magnetic induction tomography (MIT). The advantages of the developing methods are expatiated and compared with the traditional EIT . The future application trend of them is pointed out. EIT is one of the key projects of biomedical engineering today. It is an ideal medical imaging technology with enticing prospect.
4.Enhanced recovery after laparoscopic radical gastrectomy for gastric cancer

Objective To explore the safety and effectiveness of enhanced recovery after surgery (ERAS) after laparoscopic radical gastrectomy for gastric cancer.Methods From January 2014 to December 2015,167 gastric cancer patients undergoing laparoscopic radical gastrectomy with D2 lymphadenectomy were divided into the enhanced recovery after surgery group (ERASG,67 cases) and 102 cases the conventional surgery group (CSG,102 cases).The general status,tumor stage,postoperative recovery,complications,hospitalization time and 30 day readmission were compared between ERASG and CSG.Results The first exhaust time in ERASG and CSG were (2.6 ± 1.1) days and (3.8 ± 1.3) days (t =-6.494,P < 0.01).Statistical analysis showed that there was no significant difference in the postoperative hospitalization time,30 day readmission rate,complications,anastomotic leakage,wound infection,pneumonia,gastric retention and intestinal obstruction between of ERASG and CSG (all P > 0.05).Conclusions Patients in ERASG have quicker postoperative recovery than those in CSG,with similar postoperative complications.
5.Application of Automatic Unit-dose Tablet Sorting and Counting Machine in Our Hospital

OBJECTIVE:To provide reference for the generalization of automatic unit-dose tablet sorting and counting machine (ATM) in hospital pharmacy. METHODS: The preparative work and the working procedure of the ATM were introduced, and the advantages, the causes of medication errors and the problems related to the application of the ATM were analyzed. RESULTS: ATM helped to enhance the dispensing efficiency, improve the sanitation of drugs, ensure drug safety to great extent and improve the drug control. However, sometimes it might bring about medication errors. 236 (0.48%) of the total 49 059 medical orders analyzed in our study were found to be of medication errors. CONCLUSIONS: ATM has great application value. The wide use of ATM is consistent with the tendency of pharmacy automation, yet its use remains to be further improved.

7.A Combined Protocol of Chinese Herbal Medicine and Acupuncture for Chronic Gastritis

[Objective] To evaluate the clinical curative effect of a combined protocol using Chinese herbal medicine and acupuncture for chronic gastritis.[Methods] 23 patients used the Chinese medicine only(group 1)and 65 patients received the combined protocol(2).Clinical symptoms and gastroscopic findings before and after treatment were recorded and analyzed.[Results] The clinical symptoms improved greatly in 15 of the group 1,with an overall curative rate 65%,and in 62 of the group 2 with an overall curative rate 95%.The difference between the 2 groups was significant.[Conclusion] The combined use of Chinese medicine and the needle medicine is preferable to single use of Chinese medicine for chronic gastritis.
8.Therapeutic observation on superficial needling with different frequencies for intractable facial paralysis

Objective:To obverse the therapeutic effect of superficial needling with different frequencies for intractable facial paralysis. Methods: A total of 120 patients with intractable peripheral facial paralysis were allocated into a superficial needling with high frequency group (150 times/min), a moderate frequency group (100 times/min) and a low frequency group (50 times/min) according to the random number table method. The Toronto facial grading system (TFGS) was used to evaluate facial nerve functions before treatment and after 2 weeks and 4 weeks of treatment respectively. The electromyography (EMG) test of the mandibular branch of facial nerve was used to compare the motor nerve conduction velocity (MCV), sensory nerve conduction velocity (SCV) and monophasic action potential (MAP) among different groups, and was done before treatment and after 4 weeks of treatment. The clinical efficacy was also compared. Results: After 2 weeks and 4 weeks of treatment, the changes of TFGS scores in the three groups all showed statistical significance (all P<0.05), and the TFGS score in the low frequency group was substantially higher than that in the other two groups. After treatment, the changes of the MCV and SCV in the three groups all showed statistical significance (all P<0.05), and the results in the low frequency group were higher than those in the other two groups; the change of MAP in the three groups showed no statistical significance (P>0.05). The total effective rate was 65.0%, 80.0% and 95.0% in the high frequency group, moderate frequency group and low frequency group respectively, and the between-group differences showed statistical significance (P<0.05). Conclusion: Compared with the superficial needling with high and moderate frequencies, superficial needling with low frequency can produce more significant clinical efficacy for intractable facial paralysis.
9.Comparison of Two Commercial Enzyme-linked Immunosorbent Assays for Detection of IgG Antibody to Diphtheria

Objective To compare the performance of two commercially available ELISA kits for detection of diphtheria IgG antibody to provide data for the proper kit choice. Methods Thirty-three serum samples from staff of the capital airdrome and two standard sera were tested for the evaluation of the kits.Commercial ELISAs were compared with respect to their reproducibility(intra-assay variation and inter-assay variation)and accuracy.Results For the detection of standard serum,intra-assay variation(Coefficient of Variation,CV)was 13.5507% for IBL kit and 5.9541% for Virion kit,the inter-assay variation(CV)was 9.9890% for IBL and 2.4728% for Virion kit.For the serum sample,intra-assay variation(CV)was 14.5270% for IBL and 11.0612% for Virion,the inter-assay variation(CV)11.0039% for IBL and 8.1506% for Virion kit.For the detection of standard serum,inter-assay variation(CV)was 9.9890% for IBL and 2.4728% for Virion.The accuracy was analyzed by variation between the actual concentration and measured concentration of standard serum,the CV was 17.6889% for IBL and 1.3966% for Virion.Conclusion The intra-assay and inter-assay variation of the two kits are generally acceptable(CV

Objective To design siRNA for Caveolin-1(CAV1) and construct eukaryotic expression vector Dsilence4.1-CMV-neo-CAV1, which is transfected into gastric cancer cell line SGC7901 to study the effects of inhibition of CAV1 expression with siRNA on biological behavior of gastric cell line SGC7901.Methods Design and chemosynthesize three pairs of double strands siRNA based on the neucleotide sequence of CAV1 gene provided by GeneBank.The siRNAs were transient transfected into SGC7901.The CAV1 expression of transfected cells were detected respectively by semi-quantity RT-PCR to sieve a valid siRNA sequence.Design and synthesized DNA sequence which can express small hairpin RNA(shRNA),construct RNA interfering eukaryotic expression vector of CAV1 gene.The SGC7901 Was stably transfected and was detected the expression of CAV1 gene by RT-PCR.The influence on the gastric cell growth and pmliferation after CAV1 was inhibited Was detected by cell growth curve, clone formation test and FCM.Results The CAV1 RNA interfering eukaryotic expression vector psilence4.1-CMV-neo-CAV1 was successfully constructed,which can significantly reduce the expression of CAV1 after stably transfected to target cells,and in contrast to Dsilence4.1-CMV-neo transfection group,PI and colony for mution rate were increased(t=7.98,P<0.05;t=13.19,P<0.01),the growth speed Was up.Conclusion The growth and proliferation ability of tumor is remarkablv accelerated by inhibiting the expression of CAV1 of gastric cell by RNAi.
