1.Ongoing improvement of medical services for large-scale activities as guided by patient satisfaction

A satisfaction survey carried out by Beijing Chao-Yang Hospital during the Beijing 2008 Olympic Games, found differences of patient satisfaction between locals and overseas patients in consulting environment, outpatient procedure, quality of service, quality of diagnosis and treatment. The patient satisfaction of overseas patients was lower than locals. Especially noteworthy is the low satisfaction of the former for the hospital coloring, privacy in the sector, registration, vocal communication, patient condition statement, nurse attitude and service quality. Real-time improvement in these aspects of medical services has harvested obvious progress. Results of the satisfaction survey provided valuable reference materials for large-scale activities, and ongoing improvement of medical services in the long run.
2.Investigation of pharmacy postgraduate students' career planning at Peking University

We carried out a questionnaire survey on the career planning situation of pharmacy postgraduate students at Peking university, so as to collect information for career planning education of pharmacy postgraduate students, which is expected to promote their employment on completion of degree. by. The results revealed the status quo of career planning and the demand for guidance in this aspect. Recommendations for career planning education of pharmacy postgraduate students were made thereby.

Objective To explore the possible mechanisms of Arsenic Trioxide in treating rheumatoid arthritis(RA)by observing the changes of HE staining and NF-KB expression as well as the apoptosis of syn- oviocytes in adjuvant-induced arthritis rats.Methods After the animal model was set up on Wistar rats sue- cessfully,they were randomly divided into AA model group and arsenic trioxide treatment group.The treat- ment group were injected with 4 mg'kg~(-1)9?d~(-1)arsenic trioxid fluid for 7 days.All of the rats were killed 3 days after the complete of injections.The joint specimens were exposed,fixed,decalcified,wrapped and cut into slices.All slices were examined by HE stain and immunohistological evaluation.Results HE staining showed that when compared with the normal control group,the layers of synoviocytes of the AA group were increased to 6-8,and the arrangement of synoviocytes was disordered and heavy inflammatory cell infiltration were found in the AA group.In the arsenic trioxide treatment group,the layers of synoviocytes increased to 3~4,and medi- um amount of inflammatory cell infiltration were found.The intensity of synovial NF-kB(p65)positive stain in AA model group was significantly higher than that in the normal control group.The synovial expression and ac- tivation of NF-kB in the treatment group were decreased markedly,and did not return to normal level.The average gray scale calculation showed that there were significant differences between the three groups(P
4.Characteristics of GFP-hMPV infection in BALB/c and SCID mice

Objective To compare the characteristics of hMPV infection in BALB/c and SCID mice.Methods BALB/c and SCID mice were infected intranasally with GFP-rhMPV,and sacrificed on day 3,5,7,9 and 14 post inoculation.Heart,liver,spleen,lungs,kidneys and brain of the animals were used for viral isolation,titration,pulmonary histopathology and detection of GFP-hMPV mRNA expression by RT-PCR and real-time PCR.Results Live viruses were successfully isolated from the lungs of infected mice.Viral titers peaked on the 5th day post inoculation.Viruses remained to be detectable on the 14th day post inoculation in SCID mice,but not in BALB/c mice,whereas genomic RNA of GFP-rhMPV was detectable by PCR targeting F gene in infected BALB/c mice.Live viruses were not able to be isolated from heart,liver,spleen,kidney and brain,neither was genomic RNA of hMPV able to be detected on the 5 th day post inoculation by RT-PCR and real-time PCR.Histopathology of lungs was characterized by interstitial pneumonia on 5 days post inoculation.Lung pathology score of BALB/c mice group was slightly lower than SCID,and the difference was not statistically significant.Conclusion GFP-rhMPV can only replicate in the immunocompetent and immunodeficient mouse lungs,but not in other organs.As compared to that in BALB/c mice,the viral replication appears to be more efficiently and for longer time in SCID mouse lungs probably due to the absence of host cellular and humaral immunity,but this does not necessarily result in more severe pathological lesion.
5.Gene expression profiles in human bladder epithelial cells stimulated with uropathogenic Escherichia coli

Objective To investigate the interaction between uropathogenic Escherichia coli (UPEC) and host uroepithelial cells, define the role uroepithelial cells play in initiating and modulating the host response to infection with UPEC strain. Methods The human bladder transitional epithelial EJ cells were evaluated for their capacities to allow the adherence and invasion by UPEC132, a clinical strain isolated from Tianjin, China, and a cDNA microarray for 22 000 human genes was used to identify the gene expression differences between EJ cells infected with UPEC132 and uninfected EJ cells. Results Microscope observation showed that UPEC132 could adhere to EJ cells with the adherence rate of (73.20 ± 5.26)%. And visualization by confocal microscope revealed that this microorganism could be seen within the cells. EJ cells infected with UPEC132 changed mRNA expression of a total of 29 genes, including 28 genes up-regulated and 1 gene down-regulated. Of these, regulators of growth and proliferation, cytokines, and modulators of apoptotic responses were the most prominent. Conclusion The gene expression profiling of EJ cells is affected by the infection of UPEC strain. The differentially expressed genes may contribute to further investigate the interaction of UPEC and uroepithelial cells.
6.A Clinical Dynamic Observation and Correlative Study of TCD and fVEP in Acute ICH

Objective To investigate the cerebral regularity of dynamic charge and the correlation with intracranial pressure(ICP),clinical seriousness and prognosis on acute stage of intracerebral hemorrhage(ICH) patients by transcranial Doppler(TCD).Methods 54 patients with first acute supratentorial ICH 24h after onset were studied.All patients were dynamic evaluated by beside TCD on 1st,7th,21st day.The correlation of TCD with ICP,MESSS were studied.Results On the 1st day the TCD abnormity of the acute phase of the ICH occurred mainly in the anterior circulation of cerebrum,in which the cardinal manifestation was high-resistance,low-flow velocity blood waveforms,especially on the bleeding side.Continuous TCD monitoring revealed the low velocity of the cerebral blood flow increased;meanwhile the high value of PI decreased gradually and the TCD monitoring was within the normal range three weeks after the attack.The ICP had negative correlation with the VmMCA,positive correlation with the PI value.MESSS scale had negative correlation with the VmMCA,positive correlation with the PI value.Conclusions The TCD examination can be used to monitor the cerebral hemodynamics of the patients suffering from intracerebral hemorrhage.The combination of fVEP and TCD facilitate the evaluation of the dynamic changes of ICP so as to instruct lowering the ICP.
7.Contribution of motA gene in pathogenesis-associated chemotaxis and colonization of Campylobacter jejuni

Objective To determine the role of flagellar motor protein MotA in the pathogenesisassociated chemotaxis and colonization of Campylobacter jejuni. Methods The motA gene as well as Kan~r gene and plus-motA gene segments for motA gene knock-out were amplified by PCR and the target amplification fragments were sequenced after cloning. A suicide plasmid(pBlueskrit-Ⅱ-SK~(motA-kan)) and a motA gene knock-out mutant (motA~-) were constructed based on homologious recombination. By using semisolid plate migration test, hard agar plus (HAP)-based chemotactic test towards sodium deoxycholate (SDC) in vitro, and jejunal colonization test in BALB/c-ByJ mice were performed to determine the differences of flagellar motility, chemotaxis towards SDC and colonization in murine jejunum between motA~- mutant and wild-type strain. Results The nucleotide and amino acid sequences of the cloned motA gene were 100% identical to the reported corresponding sequences. The results of PCR, sequencing and continuous passage culture in antibiotics-contained medium demonstrated that both suicide plasmid and motA~- mutant were successfully generated. The diameters of clonies on semisolid plate and 0.2 mol/L SDC-induced chemotactic tings in HAP as well as the bacterial numbers adhering to the surface of murine jejunal mucosa and in jejunal content of motA~- mutant were significantly less than those of wild-type strain(P<0.05). Conclusion A motA gene knock-out mutant of C. jejuni was successfully constructed in this study, motA is an essential gene for flagellax motility, pathogenesis-associated chemotaxis and colonization of C. jejuni.
8.Acetyl-11-keto-beta-boswellic acid and arsenic trioxide regulate the productions and activities of matrix metalloproteinases in human skin fibroblasts and human leukemia cell line THP-1.

In order to reveal the treatment mechanism of Chinese medicine with the effect of activating blood and resolving putridity, we selected acetyl-11-keto-beta-boswellic acid (AKBA) and arsenic trioxide (ATO), the main monomeric components of frankincense and arsenolite which are two most commonly used Chinese medicine with effect of activating blood and resolving putridity. We combined AKBA and ATO as a compound, and explored its regulatory role in productions and activities of matrix metalloproteinase (MMP)-1, MMP-2 and MMP-9 in human skin fibroblasts (HSFbs) and human acute monocytic leukemia cell line THP-1 in inflammatory state.
9.The nursing of the surgery for the dysembryoplastic neuroepithelial tumors

Objective To explore the observation and nursing of the surgery of dysembryoplastic neuroepithelial tumors (DNT) in the function zone of brain.Methods A retrospective analysis of the clinic data of 5 patients receiving the surgery for the dysembryoplastic neuroepithelial tumors (DNT) in the function zone of brain was carried out,the clinical nursing experience was summarized.Results The EEG was re-examined 1 week after surgery.4 cases showed roughly normal EEG,one case with rare slow wave.The re-examined EEG 3 months after surgery suggested 3 cases were normal and 2 cases were almost normal.No epilepsia occurred after 6-month follow-up.Conclusions For DNT patients,microsurgical resection of the tumor is effective and enhancing the patients' perioperative observation and nursing has a significant meaning to increase the success of surgery,reduce complications,promote the rehabilitation of patients and improve the quality of life of patients.
10. Screening and identification of human anti-C3d single chain antibodies

Objective: To screen human single chain antibodies against human C3d from phage-displayed single-chain variable fragment(scFv)library, and analyze their binding activities. Methods: The phage display library was exposed to 3-round selections in immunotubes coated with recombinant C3d at a decreasing concentration range and progressively stringent washing conditions. The positive clones were identified by ELISA, followed with sequencing to determine the specific genes which were then cloned intoex-pression vectors. The single chain antibodies were expressed in the FreeStyleTM 293-F system and harvested by affinity purification. The binding activities with recombinant C3d were determined by using Bio-Layer Interferometry technology. Results: These experiments resulted in three novel single chain antibodies(that is, A1, A3 and B6)against C3d with high affinity ranging from 22.7 to 171 pmol/ L. Conclusion: The high affinity human single chain antibodies against human C3d were obtained.
