2.Gene detection and clinical study of rhinovirus isolated from children with acute low respiratory tract infection in Shanghai

Objective To understand human rhinovirus (HRV) etiology of acute lower respiratory tract infection (ALRTI) in children in Shanghai area and establish a nested reverse transcription- polymerase chain reaction (nested RT-PCR) assay.Methods Three hundred and forty-two naso- pharyngeal secretion (NPS) samples from ALRTI cases who were hospitalized were collected during January 2005—December 2005.Nested RT-PCR techniques were used to detect HRV-specific RNA.The PCR products were sequenced and data of nucleotides were analyzed.The proportion of HRV infection in children with ALRTI,the distribution of gender,age and season,and clinical char- acteristics were also investigated.Results Forty-six (13.5%) of 342 samples were HRV positive detected by nested RT-PCR.The sequences of 15 positive samples shared high homology of 83%- 97% with HRV sequence in GenBank.Within the 15 positive samples,nucleotide homology varied from 64.4% to 98.4%,and the ratio of genetic variation was from 1.6% to 48.3%./00.These 15 ampli- cons attribute to the two branches of HRV cladogram.The sequences of 15 amplieons were highly varied,in which single nucleotide mutation and several nearby nueleotides mutations were found. Ribonucleotide deletion and insertion in the nucleotide sequence was also found.HRV positive sam- ples were detected in 33 boys and 13 girls,respectively.The ratio of infection cases between boys and girls was 2.5:1.Of 46 HRV infected cases,27 (58.7%) were less than 12 months of age and 38 (82.6%) were less than 3 years old.HRV infected ALRTI occured all the year round and peaked from March to May.Of the patients whose NPS samples were HRV positive detected by nested RT-PCR,45 patients were diagnosed with bronchopneumonia and 1 was diagnosed with asthmatic bronchitis.Fever of most patients was moderate.The peripheral blood leukocyte counts in thirty-nine (84.8%) patients were less than 10?10~9/L.Neutrophil percentages in thirty-seven (80.4%) patients were less than 0.50.C-reactive protein of thirty-six (78.3%) patients were less than 8 mg/L. All of these features were the characteristics of viral pneumonia.The complications were not common and conditions of most patients were not severe.All the children were cured.Conclusions This nes- ted RT-PCR technique is highly specific,rapid and convenient for the detection of HRV RNA in NPS of patients with ALRTI and the genome of HRV viruses is highly variable.The incidence of HRV infection predominates in children in Shanghai area.ALRTI of HRV is short of specificity and condi- tions of most patients are not severe and their prognoses are fine.
3.Clinical analysis of non-Hodgkin lymphoma of palatine tonsil-Report of 70 cases

Objective To investigate the treatment method and prognostic factors of patients with non-Hodgkin lymphoma(NHL)of palatine tonsil.Methods 70 patients with NHL of palatine tonsil were re- viewed.According to Ann Arbor staging classification,12 patients had stageⅠ,39 stageⅡ,15 stageⅢand 4 stageⅣ.Working formulation was used in pathologic classification which was low grade 8 cases,intermedi- ate grade 28 cases and high grade 34 cases.30 cases were treated by chemotherapy alone,3 cases by radio- therapy alone,37 cases by chemotherapy and radiotherapy.Results The overall survival rate was 56.3%. They were 82.5% in 1 year,67.3% in 2 years and 58.5% in 5 years,respectively.The overall survival rates in high grade cases of pathologic classification were 20.0% in chemotherapy alone,0 in radiotherapy alone, 52.6% in chemotherapy and radiotherapy(P
4.Effect of ACE inhibitor on calpain system in infarcted myocardium in rats

AIM: To investigate the contribution of angiotensin-converting enzyme inhibitor (ACEI) to the regulation of calpain system in infarcted myocardium. METHODS: Rat myocardial infarction (MI) model was established by permanent ligation of the left coronary artery. The treatment with the ACEI inhibitor rampril (1 mg?kg~-1 ?d~-1 ) was started 7 days prior to surgery. On day 1, 3, 7 and 14 after MI, protein levels of calpainⅠ, Ⅱ and calpastatin were determined in left ventricular free wall (LVFW), interventricular septum (IS) and right ventricule. RESULTS: CalpainⅠprotein level was increased in IS 14 d post MI, whereas the protein level of calpainⅡ was maximally increased in LVFW 3 d post MI. Rampril decreased protein up-regulation of calpainⅠ and Ⅱ, and reduced infarct size and interstitial fibrosis. Calpastatin protein expression was not affected by ACEI. CONCLUSIONS: CalpainⅠ is involved in cardiac remodelling in the late and calpainⅡ contributes to cardiac tissue damage in the early phase of MI. The heart protective effect of ACEI may be related to the inhibition of calpain system in the pathogenesis of myocardial infarction.
5.Protection of calcium antagonists against cardiomyocyte injury caused by anoxia and reoxygenation

AIM: To investigate the protective effect of calcium antagonists on anoxia/reoxygenation (A/R) injury of cardiomyocytes. METHODS: Primary-cultured cardiomyocytes were divided into four groups, namely A/R, A/R+nifedipine(Nif), A/R+ruthenium red(Ru)+heparin (Hep) and control groups. The following parameters were measured in all groups: intracellular calcium concentration ([Ca 2+ ]i), cardiac cell viability, ATP content, lactate dehydrogenase (LDH) activity in the medium, PKC and MAPK activity and -Leucine(-Leu) incorporation. 6.Effects of three kinds of calcium activator on the proliferation of cultured vascular smooth muscle cells in rats

AIM: To investigate the effects of intracellular free calcium ([Ca 2+ ]i) from different resources on the proliferation mediated by mitogen activated protein kinase (MAPK) in vascular smooth muscle cells (VSMCs). METHODS: Cultured VSMCs were used in all experiments. Calcium influx was stimulated by angiotension Ⅱ(AngⅡ). The release of intracellular calcium stores was induced by inositol trisphosphate(IP 3) and ryanodine(RY). MAPK activity was measured by [?- 32 P]-ATP incorporation , MAPK protein expression by western blot, VSMCs proliferation by -Leucine(-Leu) and -Thymidine(-TdR) incorporation. RESULTS: Compared to the control VSMCs, AngⅡ, IP 3 and RY significantly increased [Ca 2+ ]i concentration , activity of MAPK and its protein content in VSMCs. 7.Analysis of Mental Health and Correlative Factors of Elderly Patients with Reflux Esophagitis

ObjectiveTo analyze the mental health level and the correlative factors of elderly reflux esophaditis patients.Methods86 elderly reflux esophagitis patients were tested with Symptom Check List-90 (SCL-90) and the Correlative Factor Inquiry List.ResultsThe total SCL-90 scores of the patients was 137.5±27, the number of symptomatic items was 29.9±17, which was significantly higher than that of domestic norm ( P<0.01). Besides of psychopathic and phobic, other factor scores of SCL-90 were also significantly higher than that of domestic norm ( P<0.05~0.01). The somatization, depression and anxiety were first three factors according to significant level. The constitution of patients showed the normal distribution at the level of total SCL-90 score.ConclusionMental health level of elderly reflux esophaditis patients is lower than that of normal people. The two primary reasons are long-term trend and stress of disease.
8.Linkage disequilibrium and mutation rate analysis of sixteen X-STR loci.

OBJECTIVE:
To assess the patterns of linkage disequilibrium (LD) of 16 STR loci on X chromo- some and investigate the genetic stability.
METHODS:
Genomic DNA samples extracted from blood stains from 500 unrelated individuals and 885 lineage members from Eastern Chinese Han population were genotyped through multiplex amplification using IDtyperX-16 kit by our independent research followed by capillary electrophoresis. LD was assessed by PowerMarker v3.25 software and mutation rate of every locus was analyzed.
RESULTS:
LD were not found at the 16 X-STR loci. Allele mutations were observed at 10 loci. Among them, mutation rates of DXS6809 and DXS7132 were both up to 0.0048.
CONCLUSION
When the 16 X-STR loci included in IDtyperX-16 kit were used for parentage testing, product princi- ples can be applied to calculate the likelihood, but mutation should be taken into consideration in the case that the genotypes do not meet the genetic law (especially at DXS6809 and DXS7132).

In this paper, biotin photoaffinity labeled molecule with propargyl group 1 has been designed and synthesized, followed it's labeling of N2-acetyl-2'-O-propargyl guanosine 9 by "click chemistry". This technology presents delight development potential in labeling of second messenger cyclic nucleotide, antisense oligonucleotide or siRNA.

10.Relation between cryptogenic ischemic stroke and patent foramen ovale in young and middle-aged adults

Objective To compare the prevalence of patent foramen ovale (PFO) in young and middle-aged patients with cryptogenic ischemic stroke (CS) and in normal population.Methods The casecontrol study included consecutive 318 young and middle-aged patients with CS and 336 normal control subjects with matched age and sex for group comparisons.Stroke risk factors including hypertension,diabetes mellitus,hyperlipidemia,ischemic heart disease,atrial fibrillation,carotid atherosclerosis plagues and smoking,etc.were studied.Transesophageal echocardiography (TEE) were performed to detect the presence of PFO.The prevalence of PFO and difference of risk factor levels between the groups was compared.Then the odds ratios (OR) of a PFO was estimated in CS patients versus control subjects.Results The prevalence of PFO was significant higher in patients with CS than in control subjects (145/318,45.6 % versus 46/336,13.7%,P <0.001).The odds ratio(OR) for PFO in CS for patients versus control subjects was 5.3 (confidence interval,3.6 to 7.8).The mean size of PFO was larger in stroke group than that in control group (P < 0.001).There were no significant differences in levels of other stroke risk factors between two groups.Conclusions PFO may play an important role in etiology of CS in young and middleaged adults.Larger and longer PFOs may be more concomitant with ischemic attacks.More efforts should be employed in patients with CS to detect PFO for further treatment.
