1.The effect of combination treatment using iron chelator deferasirox and cisplatin on proliferation and apoptosis in non-small cell lung cancer

Objective To investigate the combination effect using iron chelators deferasirox and cisplatin on A549 cell proliferation and apoptosis and to provide evidences to explore an effective way to treat lung cancer. Methods Lung adeno?carcinoma cells were cultured by conventional way, with administration of different concentrations of deferasirox and cisplat?in. Cell growth inhibition was observed under an inverted microscope. Proliferation inhibition was evaluated by MTT assay. Morphological changes of cell apoptosis was detected using DAPI,AO/EB straning and flow cytometry. Results After a cer?tain time of incubation with different concentrations of the combined drugs,the cell number reduce significantly, which was counted under invert microscope. Cells were dispersed with each other and adherent cells appear shrunken and poor in re?fractivity. MTT assay showed that inhibition of cell proliferation was in a concentration-time-dependent manner. Chromatin condensation, nuclear condensation and other typical apoptotic morphology were detected after DAPI and AO/EB straning. Flow cytometry showed that apoptosis increased with rising drug concentration. So combination therapy was significantly pro-apoptotic. Conclusion Deferasirox has the ability to inhibit proliferation of A549 cells and can promote tumor cell apoptosis and enhances cancer cell tolerance when combined with cisplatin. It can also reduce the amount and toxicity of cis?platin. It provides a basis for finding an effective way to treat lung cancer.
2.Feasibility of low radiation dose and iodine contrast medium in 70 kVp abdominal CTA in low body mass index patients

Objective To explore the feasibility of reducing radiation dose and iodine contrast medium in 70 kVp abdominal CTA imaging in low-body mass index (BMI;≤22 kg/m2) patients.Methods The 48 patients with suspected abdominal vascular diseases and low BMI (≤22 kg/m2) underwent abdominal CTA.All patients were divided into two groups according to tube voltage and contrast intake randomly.The parameters of group A (n=27) were contrast intake 300 mgI/kg,70 kVp,the conditions of group B (n=21) were contrast intake 500 mgI/kg,120 kVp,and the other conditions were the same in both groups.The image quality of right renal arterial images was evaluated by two observers simultaneously (5-scale).The consistency and difference between the two observers were analyzed.The CT values of abdominal aorta,celiac trunk,superior mesenteric artery,common hepatic artery,pancreatic artery and erector spinae,SD values of erector spinae on the level of right renal hilus were measured respectively in both groups.The CT values of abdominal aorta and its main branches,the CNR of abdominal aorta,the volume CT dose index (CTDIvol),dose-length product (DLP) and overall iodine intake in each group were compared.Results The scoring of right renal arterial images in both groups were 5.00 (1.00),the results showed a good consistency (Kappa=0.932,0.911).The CT values of abdominal aorta,superior mesenteric artery,common hepatic artery and pancreatic artery in group A were all more than those of in group B (all P<0.05).The CNR of group A was higher than that of group B (P<0.05).The CTDIvoland DLP in group A decreased by 73.36% and 74.41% compared with group B.The overall iodine intakes in group A and group B were (46.33±6.43)ml and (84.31±10.71)rnl,respectively.The overall iodine intake of group A decreased by 45.05% compared with group B.Conclusion For patients with low BMI (≤22 kg/m2),a 70 kVp tube current abdominal CTA scanning can significantly increase the contrast of images of abdominal artery and its branches.Meanwhile,the radiation dose and overall iodine intake can obviously decrease.
3.Establishment of Method and Verification for Leukocyte Differential Count of Sysmex Hematology Analyzer

Objective To calibrate the leukocyte differential counts and compare between all sysmex hematology analyzers, making the results accurate and reliable.Methods Used the manual visual microscope method as a reference method for leu-kocyte differential count to detect the fresh blood and then did assignment.The target value,calibrating the leukocyte differ-ential counts of Sysmex XE-2100,XS-800i and XN-20 (A1)with the manual value fresh blood,was calibrated and then com-pared hematology analyzers with another fresh blood.Results The leukocyte differential counts of all hematology analyzers were within the 95% confidence interval of the manual visual microscope method.Comparing with the reference system,the correlation coefficient(r)of NEUT%,LYM%,MON%,EOS% and BASO% of XS-800i analyzer were 0.99,0.99,0.98, 0.98 and 0.59,respectively.The correlation coefficient (r)of NEUT%,LYM%,MON%,EOS% and BASO% of XN-20 (A1)analyzer were 0.99,0.99,0.97,0.98 and 0.55,respectively.XS-800i and XN-20 (A1)NEUT%,LYM%,MON% and EOS% were within confidence interval 100%.XS-800i BASO% were within confidence interval 95%.XN-20 (A1)BASO%were within confidence interval 90%.Conclusion This scheme could meet the WS/T 246-2005 demands.In order to ensure the accuracy and the results of mutual recognition of the leukocyte differential counts,the comparison analysis of the results of the manual visual microscope method and hematology analyzer were made.
4.Simultaneous Determination of Five Saponins in Yao-Bi-Tong Capsules by QAMS

This study was aimed to establish a quality evaluation method, quantitative analysis of multi-components with a single-marker (QAMS) to determine the contents of five saponins in Yao-Bi-Tong (YBT) capsules. Ginseno-side Rg1 was used as the internal standard; the relative correction factor (RCF) of notoginsenoside R1, ginsenoside Re, ginsenoside Rb1 and ginsenoside Rd were calculated and evaluated. The contents of 5 saponins were determined by the external standard method and QAMS, respectively. Rationality, feasibility and repeatability of the QAMS method were verified by comparing the results obtained from two different methods. The results showed that RCFs of notoginsenoside R1, ginsenoside Re and ginsenoside Rb1 to ginsenoside Rg1 against YBT capsules were 0.999, 1.228, 0.990 and 1.094, respectively, indicating good reproducibility. These results of two methods had no significant dif-ference. It was concluded that the QAMS method can be accurately, rapidly and reasonably used as a new quality assessment model for ginsenosides in YBT capsules.

Conclusions The 3-year survival rate was dramatically increased with combined therapy mainly by cisplatin, the dose of 60~80mg is tolerant for the elderly aged above seventy years, and perioperation complications can be cured.
8.Preliminary study of gastric carcinoma volume measurement using computed tomography for N staging evaluation

Objective To evaluate the value of volume measured by multi-slice spiral CT in preoperative N staging of gastric canc-er.Methods CT data of 1 93 cases of gastric cancer proven pathologically were collected and analyzed.Volume of the tumor was cal-culated in the portal phase,and the correlation between the results and N staging was evaluated.ROC curve was used to get diagnos-tic value to differentiate N stages.Results Intra-observer Kappa value was 0.77 (P < 0.05 ),0.72 (P < 0.05 ),Inter-observer Kappa value was 0.69 (P <0.05).The tumor volume data was positively correlated with different N stages (r=0.568,P <0.05). ROC curve showed that the volume could help differentiate between stage N0 and stage N1 - N3 (cutoff 12.06 cm3 ,sensitivity 55%,specificity 95%),stage N0-N1 and stage N2-N3 (cutoff 22.35 cm3 ,sensitivity 66%,specificity 86%),stage N0-N2 and stage N3 (cutoff 25.95 cm3 ,sensitivity 62%,specificity 89%)respectively.Conclusion The volume of gastric cancer measured by CT plays an important part in predicting lymph node metastasis staging and optimizing individualized clinical strategy for patients.

Objective To explore the mechanism of opioid-psychic dependence involving the aspects of pre-receptor and receptor by observing the changeable expressions of glutamate neurotransmitter and NR2B of N-methyl-D-aspartic acid (NMDA) receptor in the neuroanatomical circuit of ventral temental area, nucleus accumbens and prefrontal cortex (VTA-Nac-PFC) of rats subjected to morphine-induced conditioned place preference (CPP). Methods The models of CPP were validated by escalating doses of morphine in rats (n=16). The colorimetry and immunohistochemistry ways were applied to detect the contents of glutamic acid and the expression level of NR2B in VTA, Nac and PFC. Results As compared with those in the control group physiological saline), the prolonged detention time of white compartment in the model group was notably observed (P<0.05), and increased content of glutamic acid and expression level of NR2B in fields of VTA, Nac and PFC in the model group were significantly detected (P<0.05). Conclusion Increased level of giutamic acid and expression level of NR2B in nuroanatomieal circuit of VTA, Nac and PFC could play key roles in inducing morphine-psychic dependent rats.
10.Effect of curcumin on radiosensitization of CNE-2 cells and its mechanism.

OBJECTIVETo investigate the effect of curcumin (Cur) on radiosensitivity of nasopharyngeal carcinoma cell CNE-2 and its mechanism.
METHODThe effect of curcumin on radiosensitivity was determined by the clone formation assay. The cell survival curve was fitted by Graph prism 6. 0. The changes in cell cycle were analyzed by flow cytometry (FCM). The differential expression of long non-coding RNA was detected by gene chip technology. Part of differentially expressed genes was verified by Real-time PCR.
RESULTAfter 10 micro mol L-1 Cur had worked for 24 h, its sensitization enhancement ratio was 1. 03, indicating that low concentration of curcumin could increase the radiosensitivity of nasopharyngeal carcinoma cells; FCM displayed a significant increase of G2 phase cells and significant decrease of S phase cells in the Cur combined radiation group. In the Cur group, the GUCY2GP, H2BFXP, LINC00623 IncRNA were significantly up-regulated and ZRANB2-AS2 LOC100506835, FLJ36000 IncRNA were significantly down-regulated.
CONCLUSIONCur has radiosensitizing effect on human nasopharyngeal carcinoma CNE-2 cells. Its mechanism may be related to the changes in the cell cycle distribution and the expression of long non-coding IncRNA.
