1.Development of Parental Punishment Style Questionnaire

Objective: To develop parental punishment style the questionnaire. Methods: Based upon previous literature about parental punishment style and expert interview, data were collected for the parental punishment style questionnaire. A sample of 1028 junior high school students were asked to complete the questionnaire and 158 of them was given retest after a month. Results: Taken from the questionnaire by exploratory factor analysis four factors were compatible. The results of confirmatory factor analysis showed a good fit to the data. The internal consistency reliability of father and mother questionnaire was satisfactory, with Cronbach ? coefficient 0.91; and the test-retest reliability was also satisfactory, with test-retest correlation coefficient 0.88 and 0.89, which showed good content validity and high construct validity. Conclusion: The questionnaire has good reliability and validity, and meets the need of psychometrics.
2.Analysis of effect of topographical conditions on content of total alkaloid in Coptidis Rhizoma in Chongqin, China.

To study ecology suitability rank dividing of the total alkaloid content of Coptis Rhizoma for selecting artificial planting base and high-quality industrial raw material in Chongqing province. Based on the investigation of PCB and DEM data of Chongqing province, the relationship between the total alkaloid content in Coptis Rhizoma and topographical conditions was analyzed by statistical analysis. The geographic information systems (GIS)-based assessment and landscape ecological principles were applied to assess eco logy suitability areas of Coptis Rhizoma in Chongqing. slope, aspect and altitude are main topographical factors that affect the content of the total alkaloid content in Coptis Rhizoma The total alkaloid content in Coptis Rhizoma is higher in the lower altitude, shady slope and bigger slope areas. The total alkaloid content is higher in the south areas of Chongqing province and lower in the northeast. Terrain conditions of the southern region of Chongqing are most suitable for The accumulated of total alkaloid Coptis Rhizoma content.

4.The change and relationship of thrombopoietin and platelet parameter, megakaryocyte in immune vasculitis of weanling rabbits

Objective To study the change and relationship among thrombopoietin (TPO), platelet parameter and megakaryocyte in immune vasculitis of weanling rabbits. The role of platelet parameter, megakaryocyte, thrombopoietin (TPO) in kawasaki disease (KD) was accessed. Methods An experimental model of weanling rabbits for KD was reproduced by bovine serum. The blood platelet counts (PLT), mean platelet volume (MPV), platelet distribution width (PDW), platelet hematocrit (PCT), megakaryocyte and TPO were determined ev-ery four days. Pathological analysis of coronary artery, liver, spleen, kidney, brain was also done. Results PLT, PCT, PDW and the per-centage of total number of megakaryocyte and middle board megakaryocyte in the experimental group had obviously changes at the 12th, 16th, 20th, 24th, 28th day (P <0.05). MPV in the experimental group had obviously changes in the 12th, 24th, 28th day(P <0.05). TPO in the experimental group was higher than that in control group (P < 0.05). TPO in both groups was positively correlated with PLT, PCT, the percentage of total number of megakaryocyte and middle board megakaryocyte (P < 0.05). In experimental group, small arteries showed obvious inflammatory injury at the 17th day, medium-sized arteries showed obvious inflammatory injury at the 28th day, but the aortas showed mild changes. Conclusion Platelet, megakaryocyte and TPO took part in the pathogenesis of KD, and they had interaction and co-ordination in the course of regulation, which suggested that they could be used to monitor the changes in the KD and guide the treatment.

6.Recent Advance in Studies on Angelica sinensis

Angelicae Sinensis Radix (ASR) is the root of Angelica sinensis which is a fragrant and perennial herb native to China,Japan,and Korea.In traditional Chinese medicine (TCM),the plant is useful for replenishing and invigorating blood,relieving pain,and moistening the intestines,resulting in its application for the treatment of menstrual disorders,and as an emollient and laxative for chronic constipation of the aged and debilitated.An in-depth review of the literature brings to light a great number of chemical constituents that have been isolated from ASR as well as both preclinical (in vivo and in vitro) and clinical studies,which over the years,have sought to investigate the medicinal relevance of some of these phytoconstituents and/or extract(s) prepared from ASR.The purpose of this review is therefore to present some major pharmacological and pharmacokinetic research findings on some selected phytoconstituents of ASR with emphasis on the current trends in terms of research techniques or design.This review would also provide a wealth of information for users/practitioners of TCM regarding the use of ASR or its products for maximum efficiency and minimal toxicity or side effects.
7.Inhibitory effect of siRNA-mediated MSK1-silencing on the proliferation of CNE2 cells and its mechanisms

Objective Abnormal activation of mitogen-and stress-activated kinase (MSK1) plays an important role in the development of various cancers.This study was to explore the effect of small interfering RNA (siRNA)-mediated MSK1-silencing on the proliferation of human nasopharyngeal carcinoma (NPC) cells and its underlying mechanism.Methods The siRNA vector targeting MSK1 was constructed and transfected into CNE2 cells, and the NPC cell line stably expressing MSK1 was established.Then the cells were divided into a blank control (without transfection of the plasmid), a negative control (with stable transfection of the negative control plasmid), and an experimental group (with stable transfection of the positive recombinant plasmid).The expressions of MSK1 mRNA and protein were detected by real-time quantitative PCR and Western blot, respectively, the proliferation of the cells determined by CCK-8 and colony formation assays, the cell cycles analyzed by flow cytometry, the level of histone H3 phosphorylation at Ser10 examined by Western blot, and The transcriptional activity and expression of the c-jun protein measured by dual-luciferase reporter gene assay and Western blot.Results Compared with the blank control, the inhibition rates of cell proliferation at 48, 72 and 96 hours were significantly reduced in the experimental group (P<0.05), and so were the colony formation ability of the cells (P<0.01) and the expression and transcriptional activity of the c-jun protein (P<0.05).In comparison with the negative control, the experimental group showed significant decreases in the rate of cell growth after 24 hours, the inhibition rates of cell proliferation at 48, 72 and 96 hours (P<0.05), the number of formed colonies ([221.00±20.08] vs [99.67±15.57] / 300 cells, P<0.01), the proportion of S-phase cells (P<0.01), and the expression of the c-jun protein in the CNE2 cells ([100.00±0.00] vs [48.77±10.71] %, P<0.05), but a remarkable increase in the percentage of G0/G1-phase cells (P<0.01).Furthermore, histone H3 phosphorylation at Ser10 was markedly reduced (P<0.01) but no significant change was observed in the expression of the total c-jun protein in the experimental group.Conclusion Knockdown of MSK1 using siRNA can significantly inhibit the growth and proliferation of CNE2 cells, which may be closely related to the decreased phosphorylation of histone H3 and subsequently down-regulated transcriptional activity of c-jun.

9.Level investigation on coptidis rhizoma processing methods and product specifications.

The existing processing methods, commercial specification and grades, and the marketing of Coptidis Rhizoma were systematical researched, referring to ancient, modern and contemporary medical literatures with the combination of our fieldwork on main origins of Coptidis Rhizoma and some herbs markets. Results showed that Coptidis Rhizoma processing methods mainly included sun-dried method and baked method anciently. For now, the processing methods become various, including thin paper-covering under the sun drying, direct drying, oven drying and microflame-fry drying. In the literatures, the main chemical constituent berberine was determined of its content to analyze the processing methods, finding that the sun drying and baked drying affect the berberine content, so the temperature must be controlled when drying. The thin paper-covering drying method is so cumbersome for large quantities of medicinal herbs and in wind conditions that it has been eliminated. Eventually, direct drying, oven drying and microflame-fry drying are chosen to the large-scale socialized production for their convenient and simple operation, making up the current main processing methods. Coptidis Rhizoma products have 3 specifications of Weilian, Yalian, Yunlian, of each specification there are 2 levels, but in fact the market of Weilian commodities overtakes the Yunlian, which only sold in parts of Yunnan, and the no-sale Yalian. The mainstream commercial Weilian are often sold in general way, gradeless and uniformly-priced. There are regional differences of the processing methods on Coptidis Rhizoma, which needs unified research and development of relevant standard operating procedures to ensure the quality of medicine, urgently. Coptidis Rhizoma product specifications and the intrinsic quality-grade correlation need to be further improved, which could provide a more reliable standard for quality at competitive prices, and it is recommended that "medicinal commercial specification and grade standard" content be increased into the Herbs part of Chinese Pharmacopoeia.

10.The effect of post-pyloric feeding on the prognosis of critically ill patients with acute gastrointestinal injury grade II

Objective:To explore the effect of post-pyloric feeding by spiral nasoenteric tubes on the prognosis of critically ill patients with acute gastrointestinal injury (AGI) grade Ⅱ.Methods:A retrospective study was performed to analyze the clinical data of critically ill adult patients with AGI grade Ⅱ, who were enrolled in three randomized controlled trials conducted by Guangdong Provincial People's Hospital for post-pyloric tube placement between April 2012 and May 2019. Data including demographic characteristics, serological indicators of nutrition, the tube tip position confirmed by abdominal X-ray 24 h after tube insertion, and intensive care unit (ICU), 28-day and hospital mortality were collected. Patients were divided into the post-pyloric feeding group and gastric feeding group according to the tube tip position. Propensity score matching method was used to perform 1:1 matching, and the differences of each index between the two groups were compared after matching. Then the influencing factors of P<0.1 were included in multivariate logistic regression analysis to investigate the potential ICU mortality risk factors of critically ill patients with AGI gradeⅡ. Factors with 0.1 level of significance from the univariate analysis were considered in the multivariate analysis. Results:There were 90 patients in post-pyloric feeding group and 90 patients in the gastric feeding group. Demographics and clinical characteristics of study population were well balanced between the two groups after matching. ICU, 28-day and hospital mortality in the post-pyloric feeding group were significantly lower than those in the gastric feeding group (4.4% vs 15.6%, 14.4% vs 27.8%, 6.7% vs 17.8%, all P < 0.05). Multivariate logistic regression analysis indicated that post-pyloric feeding was an independent protective factor [odds ratio ( OR)=0.295, 95% confidence internal (95% CI): 0.091-0.959, P=0.042] and APACHEⅡ score was an independent risk factor ( OR=1.111, 95% CI: 1.025-1.203, P=0.010) for ICU mortality of critically ill patients with AGI gradeⅡ. Conclusions:Post-pyloric feeding for critically ill patients with AGI grade Ⅱ could decrease ICU mortality and is an independent protective factor against mortality.
