The immune nutrients play an important role in nutritional support,immune regulation and maintenance of intestinal barrier function.In recent years,with the deepening study,the effect of immune nutrients in the comprehensive cancer treatment has constantly being recognized.Many studies have shown that glutamine (Gln),arginine (Arg) and polyunsaturated fatty acids (PUFA) have effect on the tumor itself and the combination cancer chemotherapy.
2.Sequence analysis of the coat protein gene of Chinese soybean mosaic virus strain SC7 and comparison with those of SMV strains from the USA.

To unveil genetic variations between the predominant soybean mosaic virus (SMV) strains in China and in the USA, as well as to reveal the potential relevance between the similarity of gene sequences and the virulence of the viruses, we isolated and sequenced the coat protein (CP) gene of Chinese SMV strain SC7 by RT-PCR and compared the SC7 sequence with those of SMV strains from the USA. Analysis is showed that the CP gene of SC7 was 795 nucleotides in length and encoded 265 in amino acids'. The CP gene of SC7 and those of the strains from the USA exhibited 4%-5% nucleotide diversity and 1%-2% diversity amino acids. The conserved amino-acid sequence associated with aphid spread in the USA strains was DAG, and corresponded to DAD in SC7. The virulence of SC7 was greater than that of the SMV strains from the USA. Nevertheless, no clear relationships between sequence similarity of the CP genes from different strains and their virulence on differential hosts were found.

3.Expression of loricrin and cytochrome P450 3A5 in oral submucous fibrosis and their significance.

OBJECTIVETo investigate the expression of loricrin (LOR) and cytochrome P450 3A5 (CYP 3A5) in oral submucous fibrosis (OSF) and to evaluate their roles in the defending ability of epithelium mucosae.
METHODSThe expression of LOR and CYP 3A5 was examined in the specimens of 66 OSF and 14 normal buccal mucosa samples by immunohistochemistry, and the protein and mRNA expression of them was detected by Western blot and reverse transcriptase-PCR (RT-PCR).
RESULTSLOR was overexpressed in 42 (63.6%) cases of OSF, and showed a significant difference only between the early and moderately stages of OSF (P < 0.05), but no clear difference between moderately and advanced stages (P > 0.05). All normal buccal mucosa tissues showed positive immunoreactivity for CYP 3A5 protein in the membrane and cytoplasm of spinous epithelial cells and cytoplasm of endothelial cells, 5 (7.6%) cases of OSF showed weak staining of CYP 3A5 in spinous epithelial cells and 33 (50%) showed faint in cytoplasm of endothelial cells. A negative relationship between its expression and pathological stages was found in OSF (P < 0.05). RT-PCR results were fully consistent with the immunohistochemical data. But the results of Western blot only showed the expression of CYP 3A5 was significantly higher in normal buccal mucosa samples than OSF.
CONCLUSIONThe results suggest that the LOR and CYP 3A5 might play a vital role in the change of defending ability of epithelium mucosae as well as the pathopoiesis and carcinogenesis of OSF.

5.Immunigical effect of CpG oligodeoxynucleotide as immune adjuvant of hepatitis B vaccine on pregnant mice and neonatal mice

6.Relationship between expression of triggering receptor-1 on myeloid cells in intestinal tissue and intestinal barrier dysfunction in severe acute pancreatitis

BACKGROUND: Triggering receptor expressed on myeloid cells-1 (TREM-1) in the intestine was upregulated and correlated with disease activity in inflammatory bowel diseases. Membrane-bound TREM-1 protein is increased in the pancreas, liver and kidneys of patients with severe acute pancreatitis (SAP), suggesting that TREM-1 may act as an important mediator of inflammation and subsequent extra-pancreatic organ injury. This study aimed to investigate the relationship between the expression of TREM-1 in intestinal tissue and intestinal barrier dysfunction in SAP. METHODS: Sixty-four male Wistar rats were randomly divided into a sham operation group (SO group, n=32) and a SAP group (n=32). A SAP model was established by retrograde injection of 5%sodium deoxycholate into the bile-pancreatic duct. Specimens were taken from blood and intestinal tissue 2, 6, 12, and 48 hours after operation respectively. The levels of D-lactate, diamine oxidase (DAO) and endotoxin in serum were measured using an improved spectro-photometric method. The expression levels of TREM-1, interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) mRNA in terminal ileum were detected by real-time reverse transcription-polymerase chain reaction (RT-PCR). Specimens of the distal ileum were taken to determine pathological changes by a validated histology score. RESULTS: The serum levels of D-lactate, DAO and endotoxin were significantly increased in each subgroup of SAP compared with the SO group (P<0.01, P<0.05). The expression levels of TREM-1, IL-1β and TNF-α mRNA in the terminal ileum in each subgroup of SAP were significantly higher than those in the SO group (P<0.01, P<0.05). The expression level of TREM-1mRNA was positively correlated with IL-1β and TNF-α mRNA (r=0.956, P=0.044; r=0.986, P=0.015), but the correlation was not found between IL-1β mRNA and TNF-α mRNA (P=0.133). Compared to the SO group, the pathological changes were aggravated significantly in the SAP group. CONCLUSIONS: The expression level of TREM-1 in intestinal tissue of rats with SAP was elevated, leading to the release of inflammatory mediators and intestinal mucosal injury. This finding indicates that TREM-l might play an important role in the development of intestinal barrier dysfunction in rats with SAP.
7.Effect of andrographolide on the mRNA expression of MexAB-OprM efflux pump of Pseudomonas aeruginosa PAOI strain

Objective To develop a real-time polymerase chain reaction(PCR)system to determine transcriptional level of MexAB-OprM multidrug efflux pump gene and to investigate the impact of androgra- pholide on MexAB-OprM gene transcription in Pseudomonas aeruginosa.Methods The fragments of mexB gene of mexAB-oprM operon and 30S rRNA gene rpsL were amplified and cloned into two plas- mids respectively.These plasmids were used as external standards for real-time PCR.Real-time PCR was applied to measure the mRNA transcripition of mexB and rpsL gene in Pseudomonas aeruginosa growing in medium with different concentrations of andrographolide.Results The plasmids for standard curve were constructed successfully.The relative mexB mRNA expressions in 50,100,150 and 200?g/mL andrographolide were 0.04?0.03,0.06?0.07,0.09?0.03 and 0.04?0.03 respectively, which were significantly lower than that in the control(0.24?0.04,P0.05).Conclusion Andrographolide can reduce the transcriptional level of MexAB-OprM,which may he one mechanism for its anti-infection effect.
8.Effect of surgically induced bilateral cleft alveolar on maxillary growth in dogs.

OBJECTIVE:
To establish an animal model of bilateral alveolar cleft and to determine the effect of cleft on maxillary growth.
METHODS:
Eight dogs (12 weeks old) were divided into unoperated control groups (n = 4) and the model group (n = 4). The model dogs were operated to establish a bilateral alveolar cleft. All the dogs were killed and the craniofacial morphology on clean skull was analyzed by the direct detection and CT.
RESULTS:
The length, the foreside width, and the foreside height of the maxillary in the model group were shrunken compared with those of the control group (P < 0.05).
CONCLUSION
The alveolar cleft plays an important role in the maxillary aberration.

9.Effect of Neoadjuvant Chemotherapy Treatment on Prognosis of Patients with Advanced Gastric Cancer: a Retrospective Study.

OBJECTIVETo evaluate the prognostic effects of neoadjuvant chemotherapy (NAC) in patients with local advanced gastric cancer.
METHODSWe retrospectively analyzed prognosis in 191 patients with advanced gastric cancer, of whom 71 were treated with NAC and 120 received surgery only between February 2007 and July 2013. Postoperative complication rate was recorded. Survival by clinicopathological features, pathological T and N stages, and histopathological tumor regression was retrospectively compared between the two groups.
RESULTSAccording to Response Evaluation Criteria in Solid Tumors, none of the 71 patients in the NAC followed by surgery group showed complete response, 36 showed partial response, 25 had stable disease, and 10 had progressive disease. The chemotherapy response rate was 50.7%; the disease control rate was 85.9%. Grade 3/4 adverse events were seen in less than 20% patients, with acceptable toxicities. No difference was found in the overall postoperative complication rates between the two groups (7 versus 22 cases, P=0.18). Median survival time was significantly different, at 54 months in the NAC combined with surgery group and 25 months in the surgery-only group (P=0.025).
CONCLUSIONIn patients with operable gastric adenocarcinomas, NAC can significantly improve overall survival without increasing surgical complications.

10.Expression and significance of cartilage oligomeric matrix protein, C-X-C motif 9 and keratin 19 in oral submucosal fibrosis.

OBJECTIVETo investigate the expression and localization of cartilage oligomeric matrix protein (COMP), C-X-C motif 9 (CXCL9) and keratin 19 (KRT19) in oral submucous fibrosis (OSF) and to evaluate their roles in the pathopoiesis of OSF.
METHODSThe expression and localization of COMP, CXCL9 and KRT19 were investigated in the specimens of 66 patients with oral submucous fibrosis and 14 normal controls by immunohistochemistry, and their protein and mRNA expressions were detected by Western blotting and RT-PCR.
RESULTSCOMP was overexpressed in 36 (55%) cases of OSF, and 43 (65%) cases showed positive immunoreactivity for CXCL9 protein in the cytoplasm of inflammatory cells and endothelial cells in OSF. All normal buccal mucosa tissues were stained continuously and strongly for KRT19 in the cytoplasm of basal cells, only 7 (11%) of 66 OSF samples showed faint and fragmented KRT19 staining in the cytoplasm of basal cells. RT-PCR and Western blotting results were fully consistent with the immunohistochemical data.
CONCLUSIONSCOMP, CXCL9 and KRT19 play an important role in the pathopoiesis of OSF.
