1.Application of diffusion-weighted MR imaging in the diagnosis of thyroid disease

Objective To analyze and summarize the characteristics of thyroid diseases on the MR diffusion-weighted imaging.Methods Forty-two patients with thyroid lesions,including 10 males and 32 females [age range 20-72 years,mean age (42 + 13) years] underwent MR DWI before surgery and biopsy.The diagnoses of thyroid lesions were confirmed by pathological results.ADC values of benign and malignant nodules,with different b values (b values was 300,500 and 800 s/mm2),were generated by using post-processing software Functool of GE company(USA).The independent-samples t test was used and ROC curve was made to evaluate the diagnostic efficiency of the ADC values by using statistical software SPSS 12.0.Results Histologically,there were 28 benign lesions and 14 malignant lesions.Benign lesions included 20 cases of thyroid adenoma,6 cases of nodular goiter and 2 cases of Hashimoto's thyroiditis respectively.Malignant lesions consisted of 11cases of thyroid papillary carcinoma,2 cases of follicular thyroid cancer,and 1case of dysplasia.mean ADC value of benign thyroid lesions was (2.39 ±0.38) ×10 -3 mm2/s and mean ADC value of malignant thyroid lesions was ( 1.60 ± 0.56) × 10 -3 mm2/s with b value of 300 s/mm2.The statistical difference was significant between them( t =5.35,P < 0.05 ).The statistical difference of mean A DC values,between benign and malignant nodules with b value of 500 s/mm2 [ (1.85 +0.33 ) × 10 -3 mm2/s and ( 1.65 ± 0.42 ) × 10 -3 mm2/s ],was insignificant ( t =1.70,P > 0.05 ).The statistical difference of mean ADC values,between benign and malignant nodules with b value of 800 s/mm2 [ ( 1.61± 0.30) × 10 -3mm2/s and( 1.44 +0.29) × 10 -3mm2/s],was insignificant (t =1.76,P >0.05 ).ROC curve indicated that the ADC value of 1.98 × 10-3 mm2/s or higher was the cut-off value for differentiating benign from malignant cold thyroid nodules,with a sensitivity of 85.7%,and a specificity of 78.6%.Conclusions The image quality is best with b value of 300 s/mm2.Mean ADC value of benign lesions was significantly higher than mean ADC value of malignant lesions with b value of 300 s/mm2.
2.Application of Action Learning in Ethics Teaching For Operating Room Nursing Students

Objective:To explore the application effect of action learning for nursing ethic teaching in operating room.Methods:Total 94 nursing students were divided into two groups to compare the testing scores and the inci-dence of hidden security events .The control group underwent the traditional teaching method , and the experimental group taken Action learning .Results:The testing scores and the accurate rates of case analysis problem of the ex-perimental group were higher than those in the control group (P<0.05).While the rate of hidden danger events of the experimental group was significantly lower than that in the control group ( P <0 .05 ) .Conclusion: Action learning can stimulate students′learning potential , improve the effect of learning , and reduce the incidence of secu-rity events.
3.Research on the Payment Pricing Mechanism of Medicare Drugs

OBJECTIVE:To provide suggestions for the government decision of payment price of medicare drugs. METHODS:Investigation was conducted for the beginning of payment price policy of medicare drugs,effect of the implementation of policy in other countries and the encountered problems. The payment pricing mechanism was explored based on the combination of new medi-cal reform with theoretical analysis and empirical research. RESULTS:The reform of payment pricing mechanism of medicare drugs will have effect on current drug price,especially for R&D oriented pharmaceutical industries. The policy can decrease drug expendi-ture in medicare expenditure,and it is not sure whether it can decrease total medicare expenditure. CONCLUSIONS:It is suggest-ed to notice the related measures and government should keep balance between drug availability and encouraging R&D innovation.
4.The effect of N-cad-Fc chimera protein on neuronal differentiation of P19 cells

Objective To observe the effect of intercellular adhesion molecule on neuronal differentiation of P19 cells in vitro.Methods Monolayer P19 cells were cultured on the plates coated with 0.1% gelatin, 2.5 mg/L and 10 mg/L N-cad-Fc (a fusion protein of the N-cadherin extracellular domain and the IgG Fc domain), and then 5 × 10-7mol/L retinoic acid (RA) was used to induce neuronal differentiation of P19 cells. Neuron-specific class Ⅲβ-tubulin (Tuj-1) antibody was examined by immunofluorescence staining. The protein expressions of oct3/4 in stem cells, nestin in precursor cells, ngnl and map2 in neurons and gfap in glial cells were detected by reverse transcription-polymerase chain reaction (RT-PCR).Results RA at 5 × 10-7mol/L could induce P19 cells to differentiate into neurons. The average differentiation rates on 0. 1% gelatin, 2. 5 mg/L and 10 mg/L N-cad-Fc were 7. 8%, 28. 6% and 26.8%, respectively. Cells cultured on N-cad-Fc coated plates dramatically up-regulated the expressions of neural-specific proteins ngnl and map2. The expression of stem cell specific protein oct3/4 was decreased ahead of schedule and the expression of glial cell specific protein gfap could not be found by RT-PCR. Conclusions RA can induce P19 cells to differentiate into neuronal cells. Intercellular adhesion molecule has promotional effect on cell differentiation.
5.Effects of acupuncture and moxibustion on the levels of matrix metalloproteinase-1 and interleukin-1β in articular cartilage during the development of knee osteoarthritis caused by immobilization in rabbits

Objective To investigate the effects of warming-needle acupuncture on the levels of matrix metalloproteinase-1 (MMP-1) and interleukin-1β (IL-1β) in the articular cartilage in knee osteoarthritis induced by immobilization in rabbits.MethodsA total of 30 adult male Japanese white rabbits were randomly assigned into three groups by random number table method: a normal group, a model group and a treatment group, with 10 rabbits in each group. Knee osteoarthritis was induced by 6 weeks of immobilization. The rabbits in the treatment group received warming-needle acupuncture after modeling at points of “Neixiyan” (EX-LE4), “Dubi” (ST35), “Zusanli” (ST36) and “Yanglingquan” (GB34) for 4 weeks. The level of MMP-1 and IL-1β in the articular cartilage of the knee were determined with radioimmunological assay.ResultsThere were significant difference in the levels of MMP-1(0.16 ± 0.02 ng/mg, 0.37 ± 0.02 ng/mg, and 0.28 ± 0.03 ng/mg;F=258.251) and IL-1β (0.21 ± 0.01 pg/mg, 0.34 ± 0.02pg/mg, and 0.31 ± 0.04 pg/mg;F=127.112) among the normal group, model group and treatment group. The levels of MMP-1 and IL-1β both in the model group and treatment group were significant higher than those in the normal group (allP<0.01), while the levels of MMP-1 and IL-1β in the treatment group were significant lower than those in the model group (allP<0.01).ConclusionWarming-needle acupuncture can effectively reduced the levels of MMP-1 and IL-1β in the articular cartilage in knee osteoarthritis induced by immobilization in rabbits.
6. Effect and mechanism of salvianolic acid B on apoptosis of HK-2 cells induced by iopromide

Objective: To investigate the effect and mechanism of salvianolic acid B on the apoptosis of human renal proximal tubular epithelial cells (HK-2) induced by iopromide. Methods HK-2 cells were divided into eight groups: control group, model group, different concentrations of salvianolic acid B (1, 10, 50, 100, and 200 μmol/L) treatment groups and salvianolic acid B control group (200 μmol/L). The effect of salvianolic acid B on the proliferation of HK-2 cells induced by iopromide was detected by CCK-8 method. The changes of nuclear morphology were observed by DAPI staining. Levels of ROS in different groups were observed by fluorescence microscope and flow cytometry. The expression levels of Bax, Bcl-2, cleaved Caspase-3, p-Akt, p-ERK1/2, and Klotho were detected by Western blotting. Results:Compared with control group, the cell viability of HK-2 cells in model group was decreased significantly (P < 0.01), some nucleus appeared apoptotic characteristics such as chromatin condensation and nuclear division, the level of ROS and the expression of Bax, cleaved Caspase-3, p-ERK1/2 were increased significantly (P < 0.05, 0.01); Meanwhile, the expression of Bcl-2, p-Akt, and Klotho were decreased remarkably(P < 0.05, 0.01). However, the above effects of iopromide can be partially reversed by salvianolic acid B at 50, 100, and 200 μmol/L. But low concentration of salvianolic acid B (1 and 10 μmol/L) showed no obvious protective effect on the injury of HK-2 cells induced by iopromide. Conclusion: Salvianolic acid B can inhibit the apoptosis of HK-2 cells induced by iopromide, the mechanism may be related to anti-oxidative stress, activation of Akt, inhibition of ERK pathway and up-regulation of Klotho expression.
7.ALEX1 expression in cervical cancer tissues and effect of ALEX1 on cervical cancer cell biology behavior

Aim To investigate ALEX1 gene expres-sion in cervical cancer tissues and adjacent non-can-cerous tissues, and to explore the ALEX1 genetic influ-ence on cell proliferation,cycle and apoptosis of human cervical cancer cell line HeLa. Methods ALEX1 protein expression in cervical cancers and in non-can-cerous cervical tissues was evaluated using immunohis-tochemical method. A small interference RNA targeting ALEX1 gene was transfected into HeLa cells′, and the effect of ALEX1 interference on HeLa cells′ cycle and apoptosis was analysed by flow cytometry. The effect of ALEX1 interference on HeLa cells′ proliferation and sensitivity to resveratrol was analysed by CCK-8 assay. Results ALEX1 protein expression was significantly increased in cervical cancer tissues compared with non-cancerous tissues. HeLa cells′proliferation was inhibi-ted compared with control group and blank group. He-La cells′ sensitivity to resveratrol was enhanced com-pared with control group blank group. Conclution SiRNA silencing of ALEX1 gene could significantly in-hibit HeLa cells′ proliferation and enhance resveratrol ability of inhibiting HeLa cells′proliferation.
8.Assessment of choroid in diabetes mellitus patients by using integrated backscatter

Objective Objective To investigate the relationship between choroidopathy and integrated backscatter(IBS) of choroid in diabetes mellitus(DM) patients.Methods Eighty DM patients of 158 eyes were divided into 3 groups by the course of diabetic retinopathy(DR)-DM patients without DR group,DM patients with the background DR,DM patients with the proliferative DR.80 normal persons of 160 eyes were the control group.Their IBS values were measured on nose side,temple side and middle side of choroid by HP Sonos 5500,and the correction IBS values(IBS%) were calculated.Results With the deterioration of DR,the IBS and IBS% of choroid increased.The statistical significance difference were found in various groups(P < 0.01).Conclusions With the deterioration of diabetic choroidopathy,the IBS and IBS% increased.The IBS technique is useful method to assess the diabetic choroidopathy.
9.Analysis of prostatic fluid cytology in patients with elevated PSA

ObjectiveTo study the value of the exfoliative cytology of prostatic fluid obtained from patients with elevated PSA in prostate cancer diagnosis.MethodsProstatic fluid was obtained from 130 patients with elevated PSA before prostate biopsy and then the Wright's stain and cytological class were done.Each cytological class and patient's age,PSA,total prostate volume,prostatic fluid volume and the number of leukocyte in the prostatic fluid were recorded.The relationship of leukocyte number and patient's age,PSA and prostate volume was analyzed by Spearman correlation test.ResultsProstate biopsy pathology results showed that there were 77 (59.2%) cancer cases and 53 (40.8%) non-cancer cases.Patient numbers in cytological class 1 to 5 were 28 (21.5%),32 (24.6%),22 (16.9%),36 (27.7%),12(9.2%),respectively.The prostate fluid cytology had a specificity of 100% and high sensitivity of 62.5%(10/16) in patients with PSA≥20 μg/L.PSA value had significant difference between class 1,2,3,4and 5.Significant correlation was found among the prostatic fluid volume,total leukocyte number and prostate volume.Prostate volume,leukycyte density and total leukycyte number was significant higher in noncancer patients than in prostate cancer patients.ConclusionsThe exfoliative cytology of prostatic fluid is a valuable method in detecting prostate cancer,particularly in patients with high PSA levels.It has the advantages of non-invasion and less injury than prostate biopsy.There is a relationship between elevated PSA value and high leukocyte numbers.

Ferroptosis is a nonapoptotic form of cell death and differs considerably from the well-known forms of cell death in terms of cell morphology,genetics,and biochemistry.The three primary pathways for cell ferroptosis are system Xc-/glutathione peroxidase 4(GPX4),lipid metabolism,and ferric metabolism.Since the discovery of ferroptosis,mounting evidence has revealed its critical regulatory role in several diseases,especially as a novel potential target for cancer therapy,thereby attracting increasing attention in the fields of tumor biology and anti-tumor therapy.Accordingly,broad prospects exist for identifying ferroptosis as a potential therapeutic target.In this review,we aimed to systematically summarize the activation and defense mechanisms of ferroptosis,highlight the therapeutic targets,and discuss the design of nanomedicines for ferroptosis regulation.In addition,we opted to present the advantages and disadvantages of current ferroptosis research and provide an optimistic vision of future directions in related fields.Overall,we aim to provide new ideas for further ferroptosis research and inspire new strategies for disease diagnosis and treatment.
