1.Warm Needling Moxibustion at Zhongji (CV 3) and Zusanli (ST 36) for Urinary Retention after Gynecological Surgery

Objective:To observe the clinical effect of warm needling moxibustion at Zhongji (CV 3) and Zusanli (ST 36) in treatment of urinary retention after gynecological surgery. <br> Methods: Forty patients with urinary retention after gynecological surgery were randomly divided into two groups by the randomized method, 20 cases in each group. Patients in the treatment group were treated by warm needling moxibustion at Zhongji (CV 3) and Zusanli (ST 36). Patients in the control group were treated by inductive urination. The therapeutic effects of the two groups were observed after one course of treatment. <br> Results:The total effective rate was 95.0%in the treatment group and 70.0% in the control group. The difference in the total effective rate between the two groups was statistically significant (P<0.05). <br> Conclusion: Warm needling moxibustion at Zhongji (CV 3) and Zusanli (ST 36) is more effective than inductive urination in treating urinary retention after gynecological surgery.
3.Qualification Monitoring Techniques and Methods of Steam Sterilizer

OBJECTIVE Employing biological and chemical monitoring methods in steam sterilization qualification tests,to assure the qualification of the steam sterilizers. METHODS BI PCD and B-D test pack for three consecutive testings in empty chamber were used to qualify steam sterilizers. RESULTS Biological and chemical monitoring methods were the most effective monitoring techniques for steam sterility assurance. CONCLUSIONS The qualification tests should be executed exactly with the introduction of steam sterilizer in OR sterile supply division to assure medical safety.
4.Repeatability of Ophtha TOP in measuring corneal parameters and its agreement with Pentacam

Background Accurate measurement of corneal refractive parameters plays an important role in the diagnosis of eye diseases,design of refractive surgery and calculation of intraocular lens (IOL) power.Ophtha TOP is a new IOL calculator,so it is necessary to evaluate the repeatability and accuracy of Ophtha TOP in measuring corneal refractive parameters.Objective This study was to evaluate the repeatability of Ophtha TOP in measuring corneal refractive status in normal eyes and determine the agreement of the measuring outcomes between Ophtha TOP and Pentacam in pre-surgery cataract eyes.Methods A reliability evaluation method of diagnosis test was performed from September 2013 to October 2014.Sixty eyes of healthy volunteers aged (30.83 ±8.66) years old were examined in Tianjin Eye Hospital with Ophtha TOP for more than 5 times to evaluate the stability in measuring corneal refractive parameters.Corneal refractive parameters were measured by both Ophtha TOP and Pentacam for more than twice in 30 pre-surgery cataract eyes to assess the agreement with Pentacam.Keratometry and astigmatism parameters were recorded,including the flattest meridian (Kf),steepest meridian (Ks),mean keratometry (Km) and the astigmatism at J0 and J45.Intra-class correlation coefficients (ICC),repeatability (2.77Sw) and coefficient of variation (CoVs) were used to assess the stability of Ophtha TOP,and Bland-Altman graphs were adopted to value the consistency of measuring outcomes between Ophtha TOP and Pentacam.This study complied with Helsinki declaration,and written informed consent was obtained from each patient prior to any medical examination.Results The ICC of Ks,Kf,Km,J0 and J45 by Ophtha TOP were all ≥0.90,and 2.77Sw were all ≤0.36,The CoVs of Kf,Ks,Km by Ophtha TOP were all ≤0.30.The measuring values of Kf,Ks and Km were significantly higher in the Ophtha TOP than those in the Pentacam,with the mean differences (0.18±0.28) D,(0.24±0.29) D and (0.21± 0.26) D,respectively(t =3.48,4.50,4.49,all at P=0.00).The mean difference of the J0 was (0.07±0.21) D and that of the J45 was (-0.02±0.18)D between Ophtha TOP and Pentacam,showing insignificant differences between them (both at P>0.05).Bland-Altman graphs revealed that the 95% limit of agreement (LoA) of Kf,Ks and Km between the two methods was-0.37 D to 0.73 D,-0.33 D to 0.81 D and-0.30 D to 0.72 D,respectively,and that of J0 and J45 was-0.34 D to 0.48 D and-0.37 D to 0.33 D,respectively.Conclusions Ophtha TOP shows an excellent repeatability.Compared with Pentacam,Ophtha TOP presents a good consistence for measurement of the corneal astigmatism and moderate consistence for corneal curvature.
5.The Process Safety Management of EO Disinfection

OBJECTIVE To explore the process safety management of the details of its application,storage and disinfection.METHODS Poutine managements of every link of the EO disinfection Process such as the responsibility,operation procedure,regularly inspection,etc were carried out.RESULTS There were no accident occured since April,2004,when the EO disinfection machine began to work.Every monitoring sign was within the permitted scope.CONCLUSIONS Strictly management of every key procedure is the safety guarantee for the whole process of EO disinfection.
6.Enhancing Management of Ethylene Oxide Disinfection Using in Operating Theater

OBJECTIVE To explore the methods of management of ethylene oxide disinfection and evaluate its effects.METHODS The biological and chemical indicatons were used to monitor the effects in the disinfection process.The operating procedures were carried out strictly.RESULTS In 150 times disinfection processes 4.6% were not up to the standards in the two kinds of indicators.The main reason was incorrect operations in the whole process.CONCLUSIONS Strictly management and correct monitoring are very important to improve the disinfection quality for safety operation.
7.Effects of recombinant human endostatin on proliferation, cell cycle and expression of KDR/FIK-1 and VEGF in hemangioma endothelial cells in vitro

Objective To observe inhibitory effect of recombinant human endostatin on the proliferation of vascular tumor endothelial cells and to investigate its possible mechanism.Methods Hemangioma endothelial cells were cultured in vitro and different concentrations of endostatin on hemangioma endothelial cell proliferation inhibition were detected by MTT method.Effect of recombinant human endostatin on endothelial cell cycle was detected by flow cytometry.The expression of VEGF, KDR mRNA in hemangioma endothelial cell were detected by Real-time RT PCR. Results Recombinant human endostatin concentrations in 24 h, 48 h and 72 h after three period during which the hemangioma endothelial cells inhibited significantly( P <0.01 ) , and there was a clear dose dependence, IC50 was 355 μg/mL.Recombinant human vascular endostatin ( 250 μg/mL ) intervented for 24 hours, the proportion of cells in G0/G1 phase(94.23 ±1.66)%, compared with control group (90.63 ±1.14)%, had significantly difference (P<0.05).Compared with the control group, the difference of the expression of vascular endothelial growth factor (VEGF) was statistically significant (P<0.05) as well as Flk-1 (P <0.05).Conclusion Recombinant human endostatin does not only have inhibitory effect of hemangioma endothelial cell cycle, but also can inhibit the expression of VEGF and FLK-1.

9.The effects of laminin and fibronectin on human lens epithelial cells growth characters and vimentin expression

AIM: To observe the morphologic characters and the expression of vimentin on human lens epithelial cells (hLECs)cultured on laminin and fibronectin.METHODS: The primarily and secondarily hLECs were cultured on plates treated with laminin or fibronectin. The feature of hLECS was observed and recorded using an inverse microscope with digital camera and statistical analysis was applied. MTT assay was used to show the growth curve of subcultured cells (the 3rd passage). Immunofluorescent staining was used to demonstrate the morphologic change in vimentin expression in the 3rd passaged hLECs.RESULTS: The hLECs in laminin or fibronectin group grew out significant earlier than the untreated group. The cells could be subcultured fluendly in those two treated groups,while only a few survival cells were found in the untreated group. But the hLECs could undergo apoptosis in laminin group at the terminal phase of their life cycle, while lentoid bodies emerged in fibronectin treated group. There were more viable cells in the fibronectin group from the 4th to the 7th day by MTT reaction. Immunofluorescent staining showed a clear morphological difference between the two groups.CONCLUSION: Laminin provide a suitable growth microenvironment for the hLECs while fibronectin promotes the cells proliferation and differentiation.
10.Effect of high-sugar and high-fat diet on the morphology of rabbit knee articular cartilage

BACKGROUND: There is a positive correlation between the serum cholesterol and osteoarthrosis incidence. However, it is still unclear whether the high-sugar and high-fat diet participates in the process of articular cartilage degeneration leading to osteoarthrosis, and the participation mechanism is unknown. OBJECTIVE: To investigate the changes in knee cartilage morphology of New Zealand white rabbits fed with high-sugar and high-fat diet and to explore the effect of high-sugar and high-fat diet on articular cartilage degeneration. DESIGN, TIME AND SETTING: The randomized controlled experiment was performed in Animal Laboratory, Xiangya Medical College from January to September 2008. MATERIALS: Forty healthy adult male New Zealand white rabbits, weighing about 2 kg, were randomly divided into 4 groups with 10 in each group. METHODS: Male New Zealand white rabbits were divided into four groups: ①normal diet group(NG), ②high-sugar diet (mixture of 37% cane sugar and 63% normal fodder) group(SG), ③high-fat diet (mixture of 20% lard and 80% normal fodder) group(FG), ④high-sugar and high-fat diet (mixture of 37% cane sugar, 10% lard and 53% normal fodder) group(SFG), MAIN OUTCOME MEASURES: Fasting blood samples were extracted every 4 weeks. The levels of blood sugar, triglyceride, cholesterol and insulin were detected, and body mass was recorded. The femoral condyle cartilage were dyed with toluidine blue, observed with transmit electron microscope after general observation of rabbit knee joints at the 28th weeks. RESULTS: ①Blood sugar, triglyceride, total cholesterol in FG and SFG were obviously higher than that in NG, the level of blood insulin in FG and SFG increased first, and then decreased significantly (P 0.05). ② By light microscope and transmit electron microscope observation, the FG and SFG demonstrated that tide line disappeared, cartilage cells were ranged in disorder, collagen fibers fractured, cartilage cells and a change in shape, such as shrinkage. These changes of morphology were not observed in SG. CONCLUSION: Long-term high-fat diet or high-sugar and high-fat diet may induce or aggravate the disorder of articular cartilage, suggested that it may take part in the development of osteoarthritis.
