1.Effect of Balance Training on Cerebral Cortex and Balance Function of Mouse

Objective To observe the effect of balance training on cerebral cortex and balance function of mouse.Methods Forty-five C57BL/6J female mice were randomly divided into the control group, two-week training group and four-week training group with 15 animals in each group. An new model of balance function training was copied. The balance function, brain index, and the content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in the mouse cerebral cortex were tested after training.Results The total time passing balance beam of the four-week training group significantly shorten compared with the control group and two-week training group ( P<0.01). Compared with the control group, the brain index of two-week training group increased ( P<0.05), and four-week training group also increased significantly ( P<0.01). Compared with the control group, the SOD activity of cerebral cortex significantly increased (P<0.01), and MDA decreased ( P<0.05) in the four-week training group.Conclusion This new balance training can improve balance function, increase the brain index and decrease lipid peroxidation level in the cerebral cortex of the mice.
3.Investigation of clinical stage and surgical method of lung adenocarcinoma of trans-lobe type

Objective To explore the most accurate T staging and optimal surgical method of lung adenocarcinoma of trans-lobe type, and to provide supportive diagnosis as well as therapeutic evidences for this disease. Methods A total of 192 postoperative patients, hospitalized in Tianjin Medical University General Hospital from January 2008 to June 2013, who were diagnosed with lung adenocarcinoma were recruited. Patients were divided into three groups according to the 7th edition of TNM staging criteria issued by the IASLC in 2009. A total of 163 patients with T2 stage were selected as Group T2, and 12 patients with T3 stage were selected as Group T3, both of which were considered as control groups. Other 17 pa?tients who were diagnosed as trans-lobe type of lung adenocarcinoma, were Group trans-lobe. The clinical data and progno?sis were compared between three groups. The trans-lobe type of lung adenocarcinoma was diagnosed based on imaging and pathological examination. Subtypes of trans-lobe lung adenocarcinoma were identified by referring to 2011 international mul?tidisciplinary classification standard of lung adenocarcinoma. Kaplan-Meier method was used to analyze the prognosis of dif?ferent subtypes and surgical modus in patients with lung adenocarcinoma of trans-lobe type. Results By comparison, the postoperative survival rate was significantly lower in patients diagnosed with trans-lobe type of lung adenocarcinoma than that of Group T2 (P<0.05), and no significant difference in survival rate compared with Group T3 (P>0.05). There were no significant differences in survival rates between different surgical modus (P<0.05). Seventeen patients with trans-lobe type of lung adenocarcinoma consisted of four subtypes, including 8 solid predominant, 5 acinar predominant, 3 papillary predomi?nant and 1 invasive mucinous adenocarcinoma. There were no statistical significances in postoperative survival time and sur?vival rates between four subtypes. Conclusion The clinical stage of trans-lobe type of lung adenocarcinoma should be clas?sified as stage T3. Both pulmonary bilobectomy and lobectomy combined with resection of proximal invaded lobe can be used as effective surgical therapies for trans-lobe type of lung adenocarcinoma.
4.Effect of Gegen Qinlian decoction on hepatic cytochrome CYP450 isozymes in rats by HPLC-MS/MS.

To study the effect of Gegen Qinlian decoction and its major effective components on five hepatic microsomal CYP450 isozymes in rats. The in vitro hepatic microsomal incubation technique was used to co-culture Gegen Qinlian decoction and its major effective components together with each probe substrate. HPLC-MS/MS was used to establish the analytical method for metabolites of the five isoform probe substrates of CYP450 isozymes, detect the linearity among micoromal protein concentration, incubation time and metabolite formation amount. And HPLC-MS/MS was applied to determine the formation rate (V) of corresponding metabolites (acetaminophen, 4-OH-chlorzoxazone, dextrophan, 6-OH-chlorzoxazone and 6β-hydroxytestosterone) specific probe substrates of the five isoform probe substrates of CYP450 isozymes (phenacetin, polbutamide, dextromethorphan, chlorzoxazone, testosterone), in order to determine the activity of each isozyme. The result showed good linearity among acetaminophen, 4-OH-tolbutamide, dextrophan, 6-OH-chlorzoxazone and 6β-hydroxytestosterone, satisfactory precision, stability and average recovery, suggesting the method was feasible. The optimized in vitro microsomal incubation conditions conformed to the requirements in the guideline of drug-drug interaction. Gegen Qinlian decoction showed different degrees of inhibitor effect on 5 CYP450 isoforms (CYP1A2, CYP2C11, CYP2D2, CYP2E1, CYP3A1/2). Its major effective component berberine could inhibit each CYP450 isoform at high concentrations (except for CYP1A2, CYP3A1/2).

5.Study on the correlation between human papillomavirus infection and cervical cytokines

Objective To study the correlation between human papillomavirus(HPV) infection and cervical immune function.Methods A total of 209 women with HPV positive results in HPV-infected group,40 women with HPV negative results in control group,cervical samples were detected and genotyped with HPV GenoArray Diagnostic Kit.Concentrations of cytokines including interleukin(IL)-2,IL-4,interferon(IFN)-γ,and transforming growth factor(TGF)-β in cervical specimens were measured by ELISA.Results IL-2,IL-4 and IFN-γ levels in the HPV-infected group were different with those of the control group(P=0.000 1,0.001 0,0.000 1),TGF-β in HPV-infected group and control group showed no significant difference(P=0.680 0).IFN-γ in high-risk HPV group was significant lower than that in the low-risk HPV group(P=0.007 0).Conclusion HPV infection might be one of the main reasons of cervical local immune dysfunction.
6.Effects of light and plant growth regulators on growth of normal and hairy root of Lycium barbarum in vitro.

OBJECTIVETo study effects of the cycle of light and exogenous plant growth regulators on growth of normal root and hairy root of Lycium barbarum and establish a long-term system of in vitro culture of L. barbarum roots.
METHODUsing normal roots from aseptic seedlings and hairy roots originated from Agrobacterium rhizogenes A4-mediated genetic transformation of L. barbarum, the growth of root segments was examined after 20 days of culture under different culture conditions.
RESULT AND CONCLUSIONWhen cultured on the MS medium with different plant growth regulators (IAA, IBA, NAA), normal root segments of L. barbarum could elongate and branch, and the addition of 1 mg x L(-1) IBA into the medium was the most suitable for long-term in vitro culture of L. barbarum root. When cultured on MS medium without hormone, light promoted the elongation and lateral root formation of type II hairy root, but not of type I hairy root. Among three tested hormones, IAA at low concentration was the best for growth of type I hairy root, while 1 mg x L(-1) IBA was the most suitable for growth of type II hairy root. With the increase of hormone level added into MS medium, callusing frequencies of type I, II hairy roots were increased, at the same time, the formation of lateral root from hairy root or callus was inhibited.

Objective The purpose of this study was to investigate the value of serum galactomannan (GM) in the diagnosis of invasive pulmonary aspergillosis (IPA) in patients with chronic obstructive pulmonary disease (COPD).Methods We enrolled 60 COPD patients in the study, including 19 IPA and 41 non-IPA cases.We examined serum GM of the patients by ELISA, evaluate the value of serum GM test for the diagnosis of IPA in patients with COPD, and compared the GM values before and after treatment.Results With 0.5 as the positive cutoff value, the sensitivity, specificity, positive predictive value, and negative predictive value of serum GM were 57.9%, 95.3%, 84.6%, and 83.0%, respectively, with a high specificity and a low sensitivity.The 7 IPA cases showed a significantly decreased GM value after treatment as compared with the baseline (0.30±0.21 vs 1.48±1.37, P=0.004).Conclusion The serum GM test has a limited value in the diagnosis of IPA in patients with COPD, but dynamic monitoring of the changes of the serum GM value may help evaluate the patient's condition.
9.Clinical value of the serum long noncoding RNA BC200 in the diagnosis and treatment of gastric cancer

Objective To detect serum level of long noncoding RNA ( lncRNA) BC200 in gastric cancer(GC) patients, and investigate its relationship with clinical features , and evaluate its diagnostic value for GC.Methods A case-control study was performed.From November 2014 to July 2015, serum levels of lncRNA BC200 were detected by real-time quantitative polymerase chain reaction in 124 patients with GC , 41 patients with atrophic gastritis and 59 normal controls who were hospitalized in Qilu Hospital of Shandong University.Meanwhile , serum carcinoembryonic antigen ( CEA ) and carbohydrate antigen 72-4 ( CA72-4 ) were detected by electrochemical luminescence immunoassay .Serum levels of lncRNA BC200, before and 3, 7, 10, 30, 100 days after radical operation in another 31 patients with GC were determined.The sensitivity and specificity of serum lncRNA BC200, CEA and CA72-4 were analyzed by using of the receiver operating characteristic ( ROC) curve.The comparison between two groups was performed with Mann-Whitney U test and the comparison among many groups was conducted with Kruskal-Wallis H test.Results Serum levels of lncRNA BC200 in GC patients with stage Ⅰ and Ⅱ[1.041(0.794,1.462)] and stage Ⅲ and Ⅳ[1.290 (0.978,1.794)]were significantly higher than those in patients with precancerous lesion [0.969(0.699, 1.219)]and normal controls[0.801(0.556,1.599)](H =54.68,P<0.000 1).Compared with pre-operation[1.120 (0.859,1.663)], the serum BC200 levels decreased significantly in 10 days [0.903 (0.724,1.182)](U=55.0,P<0.000 1), 30 days[0.759(0.671,1.037)](U=299.0,P=0.026 1), and 100 days[0.478(0.378,0.635)](U=41.0,P<0.000 1) after surgery.The area under the receiver operating characteristics curve ( AUC) of serum lncRNA BC200 was 0.865 for GC diagnosis, which was significantly higher than that of serum CA 72-4 ( AUC =0.699 ) or CEA ( AUC =0.807 ) .The AUC of combined detection of three tests was 0.934.Conclusion Serum lncRNA BC200 levels are significantly increased in GC patients , which may be used as a potential biomarker in GC diagnosis and monitoring .
