Identification and validation of a new target is one of the most important steps for new antituberculosis (TB) drug discovery. Researches have shown that Mycobacterium tuberculosis (Mtb) encodes 20 CYP450 enzymes which play important roles in the synthesis and metabolism of lipid, cholesterol utilization, and the electron transport of respiratory chain in Mtb. With the critical roles within the organism as well as the protein structures of six Mtb CYP450 enzymes being clarified, some of them have been highlighted as potential anti-tuberculosis targets. In this paper, the phylogenetic analysis, the structural features, and the enzymatic functions of Mtb CYPs, as well as the mechanism of interactions with selective inhibitors such as azole antifungal agents for the CYPs have been reviewed and summarized. The druggability of the CYPs has also been analyzed for their further utility as targets in high throughput screening and rational design of more selective inhibitors.

Objective To explore the effect of ankle strategy stability limit training on balance and gait in recovering stroke patients with hemiplegia. Methods Forty recovering stroke patients were randomized into an intervention group and a control group. The patients in the intervention group were given ankle strategy stability limit training using visual feedback on the static long sets of a Smart Equitest Balance Master (SEBM) machine. Those in the control group practiced routine postural balance training using mirror visual feedback in parallel bars. Both groups of patients practiced balance and posture control for 30 minutes, once daily,6 days a week for two weeks. Both groups were also given routine therapy and other rehabilitation. The patients' balance function was evaluated using the Berg Balance Scale (BBS),and their gait was assessed using the walk across technique (WA). Results There was no significant difference between the two groups with regard to general information,BBS scores or WA results before treatment. After 2 weeks of treatment,BBS scores as well as the step length and pace in the WA improved significantly in both groups, but all improved significantly more in the intervention group. There was no significant difference in width of gait. Conclusion Ankle strategy stability limit training can enhance weight-bearing on stroke patients' affected foot as well as their balance and the symmetry of their steps.

Objective To explore the feasibility of mechanical chest compression to establish a rat model of car?diopulmonary resuscitation ( CPR) . Methods 4?month old healthy male Sprague Dawley rats were randomly divided into control group ( n=6 ) and model group ( n=10 ) . After induction of anaesthesia with 10% chloraldurate ( 3 ml/kg, i. p. ) , tracheal intubation and left femoral artery cannulation were performed. Under electrocardiographic and artery blood pressure monitoring, tracheal obstruction ( TO) was performed to rats in model group. At 2 min after the cardiac arrest ( CA) occurred, CPRs were administered to the rats using a self?made animal chest compressor, which provided chest?com?pression at a rate of 200 bpm. Results Shortly after TO, rats in the model group had respiratory arrest, cyanosis and ar?rhythmia. Electrocardiography indicated that CA occurred within 4-5 min, with a decreased artery systolic blood pressure ( <40 mmHg) and a zero pulse pressure. Return of spontaneous circulation ( ROSC) after the CPR was successfully a?chieved in 8 rats (80%), with a transient reperfusion arrhythmia. Finally, 60% of the rats (n=6) recovered to con?sciousness and survived for 24 hrs. The serum biochemical analysis indicated that there were electrolyte disturbances, aci?dosis, impaired renal functions and increased myocardial enzyme spectrum. Pathological examination revealed cardiac rhab? domyolysis, no?reflow phenomenon in renal glomeruli, decrease of neurons and pulmonary congestion in the model group rats. Conclusions Mechanical chest compression can provide minimal cardiac output for the requirement of CPR incardiac arrestin rats. It is feasible to establish rat CPR model with the mechanical chest compression.

Objective To investigate the effects of aging on the brain stem speech encoding in spontaneous post-menopause women. Methods There were twenty post-menopause women with normal hearing for test and twenty ovariectomized women with normal hearing for contrast.Speech-evoked auditory brainstem response (speech-ABR) was used. The speech syllable /da/ as stimulus sound was transmitted to right ear by the insert earphones in speech-ABR test. Results Response waves of speech-ABR in ovariectomized women were similar to those in post-menopause women, which contained the onset response (peak V and A), the transition (peak C), the frequency following responses (peak D, E and F) and the offset response (peak O).The characteristics of speech-ABR's peak latency and magnitude were similar between the two groups.Except that the bilateral ovariectomized women had a shorter latency of waves O for the transient response (P<0.01), the larger amplitude of peak F for the periodic portion than post-menopause women (P<0.05).No significant differences were found in the amplitude the latency of the other waves, and V/A slope between bilateral ovariectomized women (0.21±0.10 μV /ms)and post-menopause women (-0.20±0.09 μV/ms) was similar and had no statistical significance (P>0.05).After the combination of ovariectomized women and post-menopause women, the age of these subjects was positively correlated with the latency of O wave (P<0.05).Others had no correlation with age in the amplitude and latency of the waves of speech-ABR.Conclusion Aging does not affect on brain stem speech encoding in spontaneous post-menopause women.
