Objective To investigate effects of cerebral isehemic preconditioning on neurological function,in- farct volume, and the expression of caspase-3 in brain issue after brain ischemia again, and to investigate the brain protection mechanism produced by cerebral ischemia preconditioning. Methods 48 healthy male Sprague-Dawley (SD)rats(weighted 200~250g)were randomly divided into 2 groups:ischemia group(n=24),ischemia precondi- tioning group(namely preconditioning group,n=24).Each group was divided into 4 subgroups according to 6h,1d, 2d,4d after ischemia again. Results At the same point of time after ischemia, neurological deficit in preconditioning group was much less than that in ischemia group, the difference was significant(P
4.Intervention of Tert-butyl Hydroperoxide-Induced Injury in Human Umbilical Cord Derived Mesenchymal Stem Cells with Ginkgo Biloba Extract
Journal of Guangzhou University of Traditional Chinese Medicine 2015;(3):458-463
Objective To observe the intervention effect of Ginkgo biloba extract EGb761 on tert-butyl hydroperoxide ( t-BHP)-induced injury in human umbilical cord-derived mesenchymal stem cells ( hUC-MSCs) . Methods Proliferation of hUC-MSCs after primary culture was detected by methyl thiazolyl tetrazolium (MTT) assay, and then the optimal concentrations of t-BHP and EGb761 for oxidative stress injured MSC model were screened. Cell apoptosis was assessed by flow cytometry after Annexin V-fluorescein isothiocyanatel propidium iodide ( Annexin V-FITC/PI) staining. Content of malondialdehyde ( MDA) and superoxide dismutase (SOD) activity in hUC-MSCs were evaluated, and the expression levels of p53 and p21Cip1/Waf1 were analyzed by real-time fluorescence PCR. Results Pretreatment with 10~200 mg/L of EGb761 for 3 hours reduced the sensitivity of hUC-MSCs to t-BHP ( 100 μmol/L) induced proliferation inhibition, while EGb761 over 100 mg/L had no significant effect on enhancing the protection of hUC-MSCs . EGb761 at 100 mg/L prohibited hUC-MSCs apoptosis and MDA accumulation in hUC-MSCs induced by 100μmol/L of t-BHP acting for 6 hours, maintained the enzymatic activity of SOD, and decreased the expression of p53 and p21Cip1/Waf1 in hUC-MSCs with t-BHP-induced injury. Conclusion EGb761 is capable of protecting hUC-MSCs against oxidative stress injury, and its mechanism is probably related with the modulation of p53/p21 signal pathway.

Journal of Leukemia & Lymphoma 2015;24(3):169-172
Objectives To analysis the expression difference of eIF4E in bone marrow between the acute myeloid leukemia (AML) and the non-tumor patient, before and after induction therapy, and the different subtypes of AML,and to explore the relation between eIF4E and other molecular biology abnormalities in AML.Methods The bone marrow specimens of newly diagnosed AML and non-tumor control patients were collected. The expression level of eIF4E was detected by RT-PCR.Results The positive rate of eIF4E was 65.2 % (101/155) in AML.eIF4E turned to negative in 12 patients (17.6 %) who got complete remission after induction therapy.eIF4E was negatively expressed in bone marrow of the nontumor control patients. The positive rates of eIF4E were 75.0 % (15/20) and 80.8 % (21/26) in M4 and M5 type AML,respectively, and was 59.6 % (65/109) in other subtype AML (P > 0.05).FLT3/ITD gene mutation was found in 26 cases of newly diagnosed AML.The eIF4E expression and FLT3-ITD gene mutation were independent each other (P > 0.05).Conclusions eIF4E is positive in most of the newly diagnosed AML and turns to negative in part of AML achieving complete remission. The expression of eIF4E is no difference among different subtype of AMLs.eIF4E and FLT3-ITD gene mutation are independent each other.

Objective To study the mechanism of calcium oxalate monohydrate and calcium oxalate dihydrate stone formation. Methods 258 urinary stones were examined by infrared spectrophotometry, from which, 20 patients with calcium oxalate monohydrate stones (COM) and 10 patients with calcium oxlate dihydrate stones (COD) were selected for the study of some urinary parameters. The statistical study was carried out with SPSS t test . Results The urinary excretions of both calcium and phosphate varied obviously between the two groups of patients. In the COM group, the urinary calcium was (4.83?1.98)mmol/24h whereas in COD group it was (9.88?4.28)mmol/24h,( P

Objective To evaluate the modified POSSUM surgical scoring system in urologic surgery patients over 70. Methods 180 patients over 70 were retrospectively analyzed with the mo dified POSSUM surgical scoring system.A comparison between the observed and the predicted morbidity was made. Results In the complication group and noncomplication group, the mean age of the patients were 74.9?4.1 and 74.7?3.6,the mean length of stay were (42.1?4.2)d and (28.7? 14.2 )d,respectively. In the complication group, both the physiological scores and the operative severity scores of POSSUM scoring system were significantly higher than the no complication group ( 22.0 ? 4.1 vs 20.1?3.9 in pysiological score and 11.7?4.3 vs 10.3?3.5 in operative severity score, P

Objective To explore the clinical characteristics of coronary heart disease (CHD) companied by type-2 diabetes mellitus (DM) and features of their coronary angiographies. Methods The data of clinical characteristics and coronary angiography results of 108 CHD cases with T2DM and 103 CHD cases without T2DM were compared and analyzed. Results Compared with the cases of CHD without diabetes mellitus, the cases of CHD with T2DM exhibited significantly increased incidence rates of hypertension and dyslipidemia. The body mass index and serum uric acid level also increased obviously. The main manifestation of dyslipidemia of CHD patients with T2DM was the higher levels of triglycerides and lower level of serum high-density lipoprotein cholesterol. And the severe coronary artery disease and lower left ventricular ejection fraction were often found in CHD patients with T2DM Conclusion CHD patients with type-2 DM tend to have multiple cardiovascular-risk factors, and often have multi-branches lesion and more severe lesions. To treat cardiovascular risk factors actively is very important to prevent the CHD and improve its prognosis.

Journal of Integrative Medicine 2005;3(3):203-6
OBJECTIVE: To explore the pathological changes of lower extremity arteries of patients with metabolic syndrome. METHODS: Fifty patients with metabolic syndrome (MS) and 30 normal volunteers were included in this study. The inner diameters, peak systolic velocities and intima-media thicknesses of the lower extremity arteries, and the sizes and numbers of the plaques in the lower extremity arteries were obtained with color Doppler ultrasound (CDU). RESULTS: In the fifty MS patients, the intima-media thicknesses of forty-one were thicker than 1.1 mm and thirty-three had plaques which had brought stenosis in different degrees. The intima-media thicknesses of the common femoral, popliteal and posterior tibial arteries in the MS patients were significantly thicker than those in the normal volunteers (P<0.01 or P<0.05), and a greater number of plaques in the lower extremity arteries were observed in the patients, as compared with the normal volunteers (P<0.01). CONCLUSION: Various pathological changes had taken place in the lower extremity arteries of the MS patients. The sites and degrees of the lower extremity arterial diseases in MS patients can be definitely detected with CDU.
