2.Comparison of effect of lanceolata and astragalus on electrocardiogram of rats with experimental myocardial ischemia

BACKGROUND: Lanceolata and astragalus can improve myocardial ischemia, which are widely used in clinics.OBJECTIVE: To compare the effects of extract of lanceolata and astragalus on electrocardiogram (ECG) of rats with experimental myocardial ischernia.DESIGN: Completely randomized and controlled study.SETTING: Pharmacological College of Harbin Commerce University.MATERIALS: The experiment was carried out at the Laboratory of Pharmacological College of Harbin Commerce University from September to December 2002. Totally 30 healthy adult Wistar rats were divided randomly into 3 groups: control group, lanceolata group and astragalus group with 10 in each group.METHODS: ① After anesthesia, lead ECG Ⅱ of rats was recorded.30 g/kg saline, 30 g/kg lanceolata extract and 30 g/kg astragalus extract [extract of lanceolata and astragalus (raw drug: 1 g/mL)] were injected into rats in the control group, lanceolata group and astragalus group respectively. After 1-3 minutes, 0.5 U/kg pituitrin was injected intravenously (for model of myocardial ischemia), and then saline was used to dilute from 0.1to 0.2 mL for 10-second injection. Changes of ECG after injection of pituitrin were recorded with LMA-2B Ⅱ physiological recorder (migration of T wave and ST section and change of heart rate). ② Differences of data were compared with t test.MAIN OUTCOME MEASURES: Height of T wave and Migrations of ST section and changes of heart rate of ECG of rats before and after medication.RESULTS: Data of totally 30 rats was entered the final analysis. ①Height of T wave: The height was higher after medication than that before medication in three groups (P < 0.01), but that in astragalus group was obviously lower than that in lanceolata group [(1.63±3.81), (2.18±2.93) mm,P < 0.01]. ② Heart rate: The rate was lower after medication than that of before medication in three groups (P < 0.01), and that in astragalus group was obviously lower than that in lanceolata group [(212.21 ±5.01),(221.18±5.66) time/minute, P < 0.01]. ③ Height of ST section: There were not significantly different in three groups (P > 0.05), and the difference was not obvious before and after medication (P > 0.05).CONCLUSION: ① Both astragalus and lanceolata can protect experimental myocardial ischemia induced by pituitrin. ② Effect of lanceolata on reduction of T-wave height is stronger than that of astragalus, but the effect of lanceolata on reduction of heart rate is weaker than that of astragalus.
3.Comparison of Radix Codonopsis and Radix Astragali on central nervous system

Object To study the difference between Radi x Codonopsis (RC) and Radix Astragali (RA) on the central nerv ous system. Methods Sleeping was induced by pentobarbital sodium and ether. The water maze was used. Results Both RC and RA could prolong the sleeping time induced by pentobarbital sodium and ether. RC and RA had effect on the scopolamine-indu ced study and memory disturbance of mice. Conclusion The effect of RC on prolonging the sleeping time induced by pentobarbital sodium and ether is better than that of RA. On the study and memory disturbance induced by sc opolamine, RC is also better tha n RA.
4.Role of ERKⅠ/Ⅱ in the induction and maintenance of the spinal LTP

AIM: The role of external-signal regulated kinaseⅠ/Ⅱ(ERKⅠ/Ⅱ) in the induction and maintenance of spinal long-term potentiation(LTP) is evaluated.METHODS: The C-fiber evoked field potentials were recorded at the superficial layers of spinal dorsal horn at the lumbar enlargement.RESULTS:(1) PD98059(100 ?mol/L) or SL327(200(?mol/L)),a selective MEK inhibitor,completely blocked LTP induction when applied at 30 min prior to tetanic stimulation.(2) PD98059 or SL327 reversed spinal LTP in a time-dependent manner.At 15 min after LTP induction,PD98059(100 ?mol/L) or SL327(200 ?mol/L) reversed LTP completely,and at 30 min after LTP induction,the inhibitory rate of spinal LTP inhibited by PD98059 or SL327 was 62.5% and 75.0%, respectively.At 1 h after LTP induction,however,the same concentration of PD98059 or SL327 did not affect the spinal LTP.CONCLUSION: Activation of ERKⅠ/Ⅱ in spinal dorsal horn may be crucial for the induction and the early-phase maintenance of LTP of C-fiber evoked field potentials.
6. Mechanisms of Ertong Huichun Granules in treatment of respiratory virus infection diseases by network pharmacology and molecular docking

Objective: To explore the mechanism of Ertong Huichun Granules in treatment of respiratory virus infection diseases by network pharmacology and molecular docking. Methods: TCMIP, TCMSP and BATMAN-TCM databases were used to search and summarize the meridian and components of medicinal materials from Ertong Huichun Granules. PubChem and Swiss Target Prediction database were used to query the targets corresponding to the active components, and the respiratory virus infection-related targets were inquired in the GeneCards database. The KEGG pathway and GO biological process enrichment were analyzed by DAVID database, and the relevant network was constructed by Cytoscape software. According to the analysis results of the network diagram, components and targets were selected for molecular docking. Results: The herbs in Ertong Huichun Granules were most attributed to lung, stomach, liver meridian; combining database and literature retrieval. A total of 126 Chinese native medicine ingredients and 55 targets were ensured for constructing network. KEGG pathways mainly included cancer, hepatitis related pathways, PI3K-Akt signaling pathway, TNF signaling pathway, and HIF-1 signaling pathway, which were mainly involved in inflammatory response, transmembrane receptor protein tyrosine kinase signaling pathway, hypoxia response, and other biological processes. Molecular docking results showed that 7,8-didehydrocimigenol, cimicifugoside, and cimigenol from Cimicifuga foetida had good binding capacity with PTGS2, MAPK, ACE2, and 3CL hydrolase (3CLpro), and maybe have potential anti-respiratory virus effects. Conclusion: Gallic acid, 7,8-didehydrocimigenoll, ccimicifugoside, baicalin, and other ingredients from Ertong Huichun Granules can be through PTGS2, EGFR, MAPK, IL2, ACE2, and 3CLpro targets for reducing inflammatory reaction, interference virus invasion of the host and inhibiting the virus replication to play the role of treating the respiratory virus infection diseases.
7.Comparison of Radix Codonopsis and Radix Astragali on ECG of experimental myocardial ischemia rats

Object To compare the effects of Radix Codonopsis (RC) and Radix Astragali (RA) on ECG of experimental myocardial ischemia rats. Methods The model of myocardia ischemia was made by injection of pituitrin (Pit) in rats. The changes of ECG were observed with three parameters, viz. the heart rate, the locomotions of T wave and S T section. Results Both RC and RA can decrease the run up of T wave in ECG of rats with experimental myocardial ischemic injury in which RC is superior to RA. While RA has stronger effect on the slowering of the heart rate than RC.Conclusion Both RC and RA have protective effect on experimental myocardial ischemic injury induced by Pit.
8.The effect of TMS on motor function in patients with acute brain infarction

ObjectiveWe discussed the effect of TMS on the motor conduction pathway and studied the function of TMS in the motor function of patients with stroke. Methods65 patients with acute brain infarction were divided into two groups, treatment group given TMS once a day and contral group. Motor nerve center was stimulated twice or three times each day. 14 days were one period of treatment. MEP, CMCT, force and Fugl-Meyer scores were recorded on the first and the last days. Results Before treatment, there was no difference between two groups on CMCT, force and Fugl-Meyer scores. After treatment, the patients in the treatment group got much better improvement in movement ability than the patients in the control group. But there was no difference between two groups on MEP neither before nor after treatment. Conclusions TMS can help patients with stroke to regain movement ability, and improve the recovery of force and fine movement, and also reduce the CMCT distinguishably. TMS is easy to use and no evidently uncomfortable influence on patients happens.
9.Analysis of Clinical Features of Massive Cerebral Infarction with Different Concious States

ObjectiveTo explore the difference on clinical features of massive cerebral infarction with different concious states.MethodsPatients diagnosed as massive cerebral infarction of middle cerebral vascular or internal carotid artery were divided into group without conscious disorder and group with conscious disorder according to concious state. Their gender, age, past disease history, conscious state, location of ischemic area, blood pressure, heart rate, temperature, complications, white blood cell, blood glucose, lipemia, and electrolyte were recorded.ResultsThere are more hyperlipemia and diabetes, more complications and fast heart rate in the group with conscious disorder.ConclusionAbnormity of blood glucose and lipemia for a long time may affect the concious state of massive cerebral infarction patients.
10.Changes of Ambulatory Blood Pressure before and after Stent Treatment

Objective To study the changes of ambulatory blood pressure parameters after endovascular treatment.Methods The ambulatory blood pressure of 7 patients with artery stenosis was tested before and after endovascular treatment,and the coefficient of variation(CV)was calculated.Results All patients' blood pressures decreased after endovascular treatment.The CV of blood pressure of patients with internal carotid stent decreased,but the CV of blood pressure in patients with vertebro-basilar artery stent increased.Conclusion The effects of stent on blood pressure is far from clear.
