1.Quality assessment of iodized salt and a survey of urinary iodine level in Shijiazhuang city from 2004 to 2008

Objective To assess the quality of iodized salt and investigate the urinary iodine level of pupils and women after implementation of universal salt iodization in Shijiazhuang city. Methods Between 2004 and 2008, nine salt samples were randomly collected from wholesale enterprises of 5 different directions of east,south, west, north and center in Shijiazhuang city every month. Meanwhile, eight salt samples, four urine samples of women, were randomly collected in each of the 8 households of 210 villages selected from 23 counties. Between 2004 and 2008, twenty urine samples of 8 - 10 year old pupils were collected in each of 160 schools from the 23 counties. Direct titration method was used for salt iodine determination, iodine concentration in urine was detected by the method of ammonium persulfate digestion-As3+-Ce4 + catalytic spectrophotometry. Results At wholesale level, the qualified rate of iodized salt was above 99%. At household level, the consuming rate of iodized salt was above 95%, and the rate of consuming non-iodized salt was less than 5%. The rate of qualified iodized salt was above 90% in every year except 2004(87.10%), and the rate of consuming qualified iodized salt was above 90% in those years except 2004(83.08%). Urinary median iodine level of both the pupils and the women was higher than 100 μg/L, and the rate of urinary iodine level that less than 50 μg/L was below 10%. Conclusions The quality assessment of iodized salt from wholesale companies and related indices of urinary iodine level of pupils, women and household iodized salt have already reached the national standard for eliminating iodine deficiency disorders.
2.Application of transanus ilues tube in the case of left-semicolon cancer with intestinal obstruction

Objective To investigate the case of left-semicolon cancer with intestinal obstruction for the methods of one-stage resection and anastomosis.Methods The clinical data of ten patients with left-semicolon can- cer with intestinal obstruction treated by transanus ilues tube,were restrospectively analyzed from October 2004 to December 2006.Results No postoperative anastormotic leakage was found and the patients were clinically cured. Conclusion The technical problem that left-semicolon cancer with obstruction for one-stage resection and anastomo- sis was resolved by the application of transanus ilues tube.

6.Inhibitory effect of resveratrol on the growth of human colon cancer ls174t cells and its subcutaneously transplanted tumor in nude mice and the mechanism of action.

OBJECTIVETo explore the effect and mechanism of resveratrol against human colon cancer ls174t cells in vitro and the growth of colon cancer in tumor-bearing nude mice.
METHODSMTT method was used to test the inhibiting effect of resveratrol on the growth and proliferation of ls174t cells. Transmission electron microscopy was used to observe the morphological changes of cell apoptosis, and FCM assay was performed to measure the changes of cell apoptosis rate and cell cycle. RT-PCR method was used to detect the expression of bcl-2 and bax mRNA, and Western blot was used to detect the expression of bcl-2 and bax protein.
RESULTSMTT test revealed that resveratrol showed significant inhibiting effect on ls174t cells in a concentration- and time-dependent manner. In the concentration range of 25, 50, 100, 200 and 400 micromol/L, the inhibition rate after resveratrol treatment for 24 hours was respectively 1.0%, 9.1%, 17.4%, 27.8% and 66.5%, while the inhibition rate after treatment for 48 hours was respectively 3.6%, 13.7%, 30.2%, 58.4% and 86.1%, and the inhibition rate after treatment for 72 hours was 18.1%, 33.0%, 48.6%, 61.2% and 89.4%, respectively, showing a very significant difference (P < 0.01). Typical ultrastructural apoptotic changes were observed in resveratrol-treated ls174t cells. It was found through FCM assay that resveratrol caused apoptosis in ls174 cells and blocked the cell cycle at S phase. RT-PCR and Western blot test showed that after the treatment of colon cancer cells with resveratrol at different concentrations (25, 50, 100 and 200 micromol/L), the expression level of bcl-2 was decreased, while expression level of bax was increased. The highest inhibition rate was 47.9%. In 200 mg/kg and 800 mg/kg resveratrol treatment groups, the weight of subcutaneously transplanted tumors in nude mice was 4.10 +/- 0.18 g and 3.05 +/- 0.35 g, respectively, the difference was significant compared with that of the control group (P < 0.01).
CONCLUSIONResveratrol can inhibit the growth of ls174t cells through apoptosis induction. The mechanism is probably related to inhibition of anti-apoptotic factor bcl-2 and enhancement of expression of apoptotic factor bax.

7.Clinical anatomy of iliac bone grafts used for mandibular reconstruction and dental implantation.

OBJECTIVETo investigate the anatomical features of iliac bone grafts used for mandibular reconstruction and dental implantation.
METHODSSixteen cadavers were dissected. The length, width, height and cortical thickness of the iliac bone were measured with respect to points determined by the relative dimensions of the bone.
RESULTSThe length of iliac bone graft was (77.2 ± 6.1) mm. The height was from (38.2 ± 4.2) mm to (41.9 ± 4.7) mm. The width decreased from iliac crest to base line. The least width 10 mm and 15 mm away from iliac crest were (8.4 ± 2.2) mm and (6.5 ± 2.1) mm respectively. The greatest mean cross-sectional cortical thickness at the intermediate line of the iliac crest was (3.4 ± 0.8) mm.
CONCLUSIONSAnatomical features of iliac bone are suitable for designing bone grafts for mandibular reconstruction followed by dental implantation.

8.Determination of serum total glycerol by isotope dilution liquid chromatography tandem mass spectrometry

Objective To develop a method for measuring serum total glycerol by isotope dilution (ID)liquid chromatography tandem mass spectrometry(LC/MS/MS).Methods Serum samples were mixed with [13C3]-glycerol(internal standard)and treated with alcoholic potassium hydroxide to hydrolyze serum glycerides to glycerol.The natural glycerol and the internal standard were benzoylated with the Schotten-Baumann reaction and the benzoates were analyzed by LC/MS/MS.Results The correlation coefficients between the peak area ratios and glycerol concentrations were 0.999 9 and higher.The withinrun coefficients of variation(CV)for serum total glycerol analysis averaged 0.94%(ranged 0.21%~2.62%)and the total CV 1.15%(0.62%~2.00%).Results on certified reference materials(SRM 909b Level Ⅰ and LevelⅡ;GBW 09146 and GBW 09147)showed an averaged bias of 0.20%(-0.20%~1.06%).Conclusions An ID-LC/MS/MS method for serum total glycerol has been developed.The method iS highly specific and precise and may be used as a candidate reference method for serum total glycerol measurements.

Objective To study the animal toxicity test on skin of the Chinese herbal drugs liquid paste (CHDLP).Methods Acute and multiple times skin stimulation test and skin allergy test were performed on healthy guinea pig by using CHDLP.Results The acute skin stimulation score and intensity score were all zero.Multiple skin stimulation score and pathological examination score were all zero.So were the skin allergy score and the sensiti- zation rate.Conclusion CHDLP is a weak sensitizer and arose no skin stimulation on guinea pig.It's safe when ex- ternal use.
10.Toxicokinetics of paraquat in rabbits.

OBJECTIVEto develop a high performance liquid chromatography method (HPLC) for the determination of paraquat in rabbit plasma and study its toxicokinetics in rabbits.
METHODStwelve rabbits were randomly divided into 2 groups with giving oral and intravenous administration of paraquat at a single dose of 60 mg/kg and 6 mg/kg respectively. The plasma paraquat concentrations were determined by HPLC and calculated by DAS pharmacokinetics program.
RESULTSthe linear range of paraquat in plasma was 0.05 ∼ 50.00 mg/L (r = 0.9998). The relative recoveries of the assay were 99.41% ∼ 102.32%. The absolute recoveries of the assay were 83.72% ∼ 90.48%. Both the intra-day and inter-day validations were less than 10%. For oral administration, the toxicokinetics parameters of paraquat were as follows: Cmax (14.46 ± 2.35) mg/L, Tmax (1.63 ± 0.31) h, AUC(0-t) (177.61 ± 14.62) mg × h/L, AUC(0-∞) (182.24 ± 14.54) mg × h/L, While for intravenous administration, the toxicokinetics parameters of paraquat: Cmax (35.13 ± 5.53) mg/L, Tmax 0.05 h, AUC(0-t) (121.74 ± 12.30) mg × h/L, AUC(0-∞) (125.12 ± 12.17) mg × h/L, The difference of these parameters between the two groups had statistical significance (P < 0.05). The oral bioavailability was (14.66 ± 1.55)%.
CONCLUSIONthe oral bioavailability of paraquat is relatively low. The biological half life of paraquat is relatively long and there is no significant difference between oral administration and intravenous on biological half life. This method is simple, sensitive and accurate. It can be used for the investigation of paraquat in rabbits.
