1.Comparison of two extraction techniques for larger chemically aceilular nerve allografts

[Objective]To compare two preparing procedures for larger chemically acellular nerve allografts (CANA). [Methods]The sciatic nerves of pigs were exposed by a muscle-splitting incision and were isolated free of the underlying fascia. The 60-mm-long segments of the nerve were obtained. They were treated according to the following decellularization processes.In group I,the nerve segments were treated with 7% Triton-100 solution and 7% sodium deoxycholate for two times.In group II,another protocol was created with the detergents Triton X-200,sulfobetaine-16,and sulfobetaine-10 for two times. The degrees of decellularization,activity of laminin,degrees of demyelination,and integrity of the nerve fiber tube were observed under microscope and were assessed by a scoring system.[Results]In both experimental groups the activity of laminin was present and the degrees of decellularization were complete. As for the demyelization of the nerve segments,the myelin sheath in Group II was partially preserved,but it completely disappeared in Group Ⅰ. The structure of the nerve fiber tube in Groups Ⅰ and Ⅱ were not as integral as that in the normal group.[Conclusion]It may be a better method for the larger CANA,to be treated with TritonX-100 and sodium deoxycholate during the decellularization procedure.
3.Clinical effect of autohemotherapy combined loratadine on 120 cases of ASST positive chronic idiopathic urticaria

Objective To investigate the efficacy and safety of autohemotherapy therapy in ASST positive chronic idiopathic ur‐ticaria .Methods One hundred and twenty cases of chronic idiopathic urticaria treated in our department from April ,2012 to Janu‐ary ,2013 were divided into treatment group and control group ,60 cases in each group .Patients in treatment group were given oral ioratadine (10 mg/d) ,supplemented by autohemotherapy ;patients in control group were only given oral ioratadine (10 mg/d) .Both of the two groups were treated with twelve weeks ,then observed the effectiveness and safety of two kinds of treatment .Results The effective rate of treatment group and control group were 68 .33% and 48 .33% respectively ,and there was significant difference between the two groups (P<0 .01) .Conclusion Autohemotherapy combined antihistamine was of high efficiency and safety in the treatment of ASST positive chronic idiopathic urticaria ,while the long‐term curative effect remains to be observed .
4.Effects of estrogen, clomiphene on gamma-aminobutyric acid immunoreactive cell and ?1 subunit of GABA_A receptor in hippocampus of epileptic rats

Objectives To find out the effects of estrogen and clomiphene on behavior of epileptic rats induced by kainic acid (KA) and probe into some mechanisms. Methods Ovariectomized Sprague-Dawley female rats were treated with estrogen (E) or estrogen and clomiphene (C). Their behaviors when they were induced seizures were observed and compared. Indirect immunofluorescence method was used to measure the alterations of gamma-aminobutyric acid (GABA) immunoreactive cells and ?1 subunits of GABA_A receptors in the hippocampus of all groups. Results The latency and time at reaching 4/5 degrees in KA+E group ((24.63?11.44) minutes and (41.50?16.22) minutes, respectively) were reduced greatly than KA group ((46.75?14.61) minutes and (65.13?12.99) minutes), while the latency of (KA+)E+C group (adding estrogen and clomiphene, (43.50?5.75) minutes) became prolonged significantly than in KA+E group. Conclusion High-level estrogen should be proconvulsant and the clomiphene might have some antiepileptic effects, which may be related with some alterations of GABA energic function in the brain.
5.Effect of triethanolamine cream in treatment of wound healing and its effect on level of IL-2 , IL-8 and TNF-α

Objective To study the clinical effect of triethanolamine cream in treatment of traumatic wounds and its effect on level of interleukin-2 (IL-2), interleukin-8 (IL-8) and tumor necrosis factor-α(TNF-α).Methods 88 patients with traumatic wounds were selected in our hospital from February 2015 to January 2016, those patients were divided into observation group and control group according to coin method.The control group was treated with alginate dressing, observation group were given triethanolamine cream for treatment.The levels of IL-2, IL-8, TNF-αand visual analogue scale(VAS) between the two groups were compared.The clinical curative effect of the two groups was evaluated.Results After treatment, the total effective rate of the observation group(90.91%) was significantly higher than the control group (68.18%).There were no significant differences in VAS scores between the two groups before treatment, 5, 10,20 days after treatment, the VAS scores of the two groups were significantly lower than before treatment (P<0.05), at 5,10,20 days after treatment, the VAS scores in the observation group were significantly lower than the control group (P<0.05).The level of IL-2 in the observation group were significantly higher than the control group, the levels of IL-8,TNF-αwere significantly lower than the control group (P<0.05).Conclusion Triethanolamine cream can increase the level of IL-2 and decrease the levels of IL-8 and TNF-αin patients with traumatic wounds, it can relieve the pain of patients and has good clinical curative effect.
6.Effect of tetramethylpyrazine injection on serum ion and β2-GPI/ox-LDL in patients with IgA nephropathy

Objective To investigate the effect of tetramethylpyrazine injection on serum ion and β2-GPI/ox-LDL in patients with IgA nephropathy.Methods 58 IgA nephropathy patients were randomly divided into two gorups with 29 cases in each group.The control group were treated by benazepril and conventional treatment of dipyridamole, the experiment group were treated on the basis of the control group with tetramethylpyrazine injection.10 days of 1 courses, 2 courses of treatment.The serum ion,β2-GPI/ox-LDL and renal function related indexes were tested.Results Compared with before treatment, the serum inorganic phosphorus,β2-GPI/ox-LDL, Scr, BUN, CysC levels and 24 h urinary protein quantitation were lower( P<0.05), the blood calcium level were higher(P<0.05).Compared with the control group, the serum inorganic phosphorus,β2-GPI/ox-LDL, Scr, BUN, CysC levels and 24 h urinary protein quantitation were lower(P<0.05), the blood calcium level were higher(P<0.05), the clinical effect were higher (P<0.05).Conclusion Tetramethylpyrazine injection can effectively decrease the serum phosphorus, β2-GPI/ox-LDL, SCR, BUN, CysC levels and urinary protein amount of patients with IgA nephropathy, elevate blood calcium levels, significantly improve renal function and with less adverse reactions.
8.The influence of bFGF gene transfected BMSCs on inflammatory cytokines expression of COPD rat

Objective To study the influence of bFGF gene transfected bone marrow-derived mesenchymal stem cells (BMSCs) on the inflammatory cytokines of COPD rat. Methods The BMSCs were separated from SD rat and cultured and then bFGF gene was imported to BMSCs by liposome transfection method. The samples were prepared into six groups: normal control group, COPD group (A), BMSCs group (B), pcDNA3.1-BMSCs group (C), bFGF-pcDNA3.1-BMSCs group (D), and bFGF group (E). The expressions of TNF-α and IL-1β by QRT-PCR were detected. Results Compared with COPD group, TNF-α and IL-1β genes from groups B to D dropped significantly (P < 0.05). The changes of TNF-α and IL-1β genes among groups B to D showed no significant difference (P > 0.05). Conclusion BFGF transfected BMSCs, sample BMSCs and pcDNA3.1 transfected BMSCs can inhibit the expression of inflammatory cytokines of TNF-α and IL-1β, but there is no obvious advantage in comparison to bFGF transfected BMSCs and sample BMSCs in respect of inhibiting the expression of inflammatory cytokines of TNF-α and IL-1β.
9.Surface roughness and hardness of macromolecule ocular prosthesis materials

BACKGROUND:The physical properties of macromolecule ocular prosthesis materials, such as density, hardness, mechanical strength, can affect the working accuracy of ocular prosthesis and can also affect the surface roughness of ocular prosthesis, thus affecting the comfort when the patients wear.
OBJECTIVE:To study the differences of different macromolecule ocular prosthesis materials in bubble generation rate, hardness and surface roughness.
METHODS:Thirty-seven test pieces, 30 mm×30 mm×2 mm, were made and then divided to seven groups according to their brands, caled groups A, B, C, D, E, F, G, each group of five pieces. One surface of each test piece was polished in accordance with the principle of coarse to fine, the surface roughness of the two surfaces and hardness on the polished surface were measured, and then the bubble formation rate of the material was calculated.
RESULTS AND CONCLUSION:The surface roughness values of groups A-G were (0.078±0.016), (0.074±0.019), (0.075±0.022), (0.066±0.020), (0.075±0.017), (0.068±0.015), and (0.067±0.017) μm, respectively. There was no significant difference in the surface roughness between the groups (P > 0.05). The hardness values of groups A-G were 766.92±3.71, 771.84±14.51, 791.20±9.64, 804.50±4.49, 779.00±17.92, 772.20±19.18, 704.00±7.23, respectively. There was a significant difference in the hardness between groups D, G and the other groups (P < 0.05) as wel as between group D and group G (P < 0.05), but there was no significant difference between the other groups (P > 0.05). The bubble generation rates of groups A-G were (8.87±0.29)%, (8.29±1.02)%, (6.94±0.43)%, (4.83±0.20)%, (7.59±0.19)%, (8.61±0.25)%, (4.89±0.17)%, respectively. There was a significant difference in the bubble generation rate between groups D, G and the other groups (P < 0.05), but there was no significant difference between group D and group G (P > 0.05), as wel as between the other groups (P > 0.05). To a certain extent, the smaler the bubble formation rate is, the greater the hardness and the smaler the surface roughness are.
10.Serum level of B cell activating factor correlates with disease activity in dermatomyositis

Objective To investigate the correlation between serum levels of B cell activating factor (BAFF) and disease activity in the patients with dermatomyositis (DM).Methods Serum BAFF levels of 61 patients with DM and 25 matched healthy controls were measured by ELISA.The results of the two groups were compared using unpaired Mann-Whitney U test and the relevance was analyzed using Spearman correlation analysis.Results Serum levels of BAFF in DM patients were significantly higher compared to healthy controls [(274 7±264 6) pg/ml 与 (832±170) pg/ml,Z=-5.492,P<0.01].A cross-sectional assessment revealed that serum BAFF levels were positively correlated with global disease activity (r=0.501,P<0.001),muscle disease activity (r=0.303,P<0.05),and cutaneous disease activity (r=0.467,P<0.01).High serum BAFF levels were associated with increased incidence of interstitial lung disease (x2=17.238,P<0.01).The longitudinal study showed modest correlations between serum BAFF levels and global disease activity (r=0.658,P<0.01),muscle disease activity (r=0.307,P<0.05),as well as cutaneous disease activity (r=0.565,P<0.01).Conclusion Serum levels of BAFF correlate with disease activity in DM patients.The results of this study suggest that BAFF is a serological biomarker for DM disease activity.
