1.The preliminary study of the effect of ritodrine on the change of heart rate in threatened premature labor.

Objective To determine the change of maternal heart rate during the treatment with ritodrine for threatened premature labor. Methods110 cases of threatened premature labor were chosen. The ritodrine 100mg in 5% G. S 500mL was intravenously administered. The initial dosage was 6 drops/min, and the dose was increased until uterine contractions were inhibited. The treatment lasted for 40 to 48 hours after the uterine contractions were inhibited, then the oral tablets was recommended. If necessary, repeated treatment was given to some patients. The maternal heart rate was recorded in the whole process of treatment and was compared with the dosage administered. The labor time, fetal weight and Apgar′s score were also analyzed. Results The maternal basic heart rate was increased at first and then decreased. The increasing significantly increased( P < 0.01 ) when the uterine contraction was inhibited. , but no difference showed among the groups of different gestational age( P>0.05 ). The maternal heart rate was increased with the increasing of ritodrine( max. 140/min). labor time was significantly prolonged with the treatment of ritodrine( P<0.01 ) ,while that of the idiopathic premature labor was prolonged most obviously ( P < 0.05 ). Average fetal weight was (2.42 ± 0.49) kg, and the average Apgar′s score was≥8. Conclusion In the treatment of inhibiting uterine contractions,the increasing of maternal heart rate caused by ritodrine can be used as an indicator of therapeutic effect.

3.Analysis of Plasma Levels of Omentum-1 and Vaspin in Patients With Acute Coronary Syndrome

Objective: To observe the changes of plasma levels of omentin-1 and vaspin in patients with acute coronary syndrome (ACS). Methods: Our research included in 2 groups: ACS group, the patients with conifrmed diagnosis in our hospital from 2013-05 to 2013-09,n=100 including 52 male, 48 female and Control group, the patients with simultaneous coronary angiography excluded CAD,n=88. Plasma levels of omentin-1 and vaspin were examined by ELISA and the differences were compared between 2 groups. Results: Plasma levels of omentin-1 and vaspin in ACS group were lower than Control group (t=2.718,P<0.05) and (t=2.416,P<0.05). Correlation analysis presented that in ACS group, omentin-1 level was negatively related to total cholesterol (TC) (r=-0.702,P<0.05) and C-reactive protein (CRP) (r=-0.714,P<0.05); vaspin was negatively related to TC (r=-0.655, P<0.05) and CRP (r=-0.587,P<0.05); Omentin-1 was positively related to vaspin (r=0.643,P<0.05). Multiple linear stepwise regression analysis showed that plasma levels of omentin-1 and vaspin might be affected by CRP, TC and LDL-C. Conclusion: ACS patients had decreased plasma levels of omentin-1 and vaspin; in addition, omentin-1 and vaspin might be involved in lipid metabolism.
4.Recombinant adenovirus-mediated hypoxia inducible factor 1alpha mutant promotes proliferation of adipose-derived mesenchymal stem cells

BACKGROUND:To improve the survival rate of transplanted tissue, most scholars focus on cel therapy, particularly cel-assisted fat grafting.
OBJECTIVE:To analyze the effect of recombinant adenovirus-mediated hypoxia inducible factor 1alpha (HIF1α) mutant on survival rate of transplanted fat particles through transfection of adipose-derived mesenchymal stem cels.
METHODS:Recombinant adenovirus-mediatedtriple-mutantHIF1αwas inserted into an adenovirus pAdEasy-1 system, folowed by viral packaging and titer determination. Human adipose-derived mesenchymal stem cels were cultured, passaged and identified, and subsequently transfected with three kinds of viruses and blank vector (experimental group with transfection of the triple mutantof HIF1α;positive control group; negative control group; blank control group). Transfection efficiency was determined using enhanced green fluorescent protein labeling. Additionaly, MTT assay was used to detect cel proliferation.
RESULTS AND CONCLUSION:The recombinant adenovirus was successfuly constructed and packaged in line with transfection requirements. Moreover, adipose-derived mesenchymal stem cels were successfuly identified by adipogenic, osteogenic and chondrogenic induction and could be used asseed cels for subsequent experiments. RT-PCR results showed that HIF1α mRNA expression in the experimental group and positive control group was significantly higher than that in the other two groups (P< 0.05). Western blot analysis showed that the relative absorbance value in the experimental group was significantly higher than that in the other three groups (P< 0.05). A significant increase in the cel proliferation was found in the experimental group, significantly different from the other three groups (P<0.05). Therefore, our findings indicate that transfection of adenovirus-mediated triple-mutant HIF1α not only can sustain the expression of target protein in transfected adipose-derived mesenchymal stem cels under normoxic conditions, but also can promote the proliferation of transfected adipose-derived mesenchymal stem cels.
5.Multislice spiral CT of left pulmonary artery sling with tracheobronchial anomaly

Objective To discuss the value of MSCT in diagnosis of left pulmonary artery sling (LPAS) with tracheobronchial anomaly.Methods MSCT was performed in 9 children with LPAS and tracheobronchial anomalies,CT findings were analyzed retrospectively. Results According to the Wells classification of LPAS,which includes two main types and two subtypes,there were 2 patients of type 1A,3 patients of type 1 B,2 patients of type 2A and 2 patients of type 2B.All of children had tracheobronchial stenosis.Local tracheobronchial stenosis was found in 4 patients ( all were type 1 ) and long tracheobronchial stenosis in 5 patients ( 1 patient with type 1 and 4 patients with type 2).Conclusions MSCT is an ideal imaging modality for simultaneously identifying LPAS and associated tracheobronchial anomalies.The Wells classification is very useful for operation planning.
6.Study on polymorphism of human eighth complement component(C8A)by using allele specific amplification method

For the purpose of detecting the polymorphism of the human eighth complement component(C8A),an allele specific amplification method were established for detecting two alleles of C8A based on a nucleotide exchange of the C8? cDNA sequence.100 DNA blood samples from unrelated individuals of Han population in Chengdu were typed by this method.The result of comparison with the data of protein typing of the same samples using SDS PAGE method proved that the described method is efficient,correct and reliable for C8A genotypes.This methtod is valuable for further application in population genetic study and forensic science practice.
7.Study on the construction of standard D12S375 allelic ladder and its genetic polymorphism in Chinese Han, Hui and Wei populations.

s:To resolve the problem of the accuracy and standardization of STR PCR typing in forensic science practice,we have designed a new method to produce standard D12S375 allelic ladder.Seven different PCR amplified D12S375 allelic fragments were isolated from the gel,eluted into the distilled water and reamplified by PCR.The purified allelic fragments were then blunt end subcloned individually into the pUC plasmid vectors and transfected into competent E.coli DH5? TM cells.The sequencing results confirmed that the size and the constructure of the inserts were correct.The recombinant plasmids DNA with 7 inserts were then used as templates for reamplification to generate D12S375 standard ladder, with which the genetic polymorphisms of D12S375 locus in Chinese Han population in Chengdu,Hui population in Gansu and Wei population in Xinjiang were studied.
8.Comparison of Radix Codonopsis and Radix Astragali on central nervous system

Object To study the difference between Radi x Codonopsis (RC) and Radix Astragali (RA) on the central nerv ous system. Methods Sleeping was induced by pentobarbital sodium and ether. The water maze was used. Results Both RC and RA could prolong the sleeping time induced by pentobarbital sodium and ether. RC and RA had effect on the scopolamine-indu ced study and memory disturbance of mice. Conclusion The effect of RC on prolonging the sleeping time induced by pentobarbital sodium and ether is better than that of RA. On the study and memory disturbance induced by sc opolamine, RC is also better tha n RA.
9.The application of the multi-slice CT based OMENS classification in diagnosing pediatric hemifacial microsomia

Objective To assess the value of the multi-slice CT based OMENS classification in diagnosing pediatric hemifacial microsomia. Methods Eleven patients including 9 boys and 2 girls with suspicion of hemifacial microsomia underwent MSCT. Multi-planar reformation (MPR), maximum intensity projection (MIP) and volume-rendered image (VR) reconstruction were performed at the workstation. The malformations of the facial structures were analyzed respectively with the OMENS classification system.Results On CT reconstruction images, hemifacial microsomia in all 11 patients was demonstrated clearly.Nine patients were found to have unilateral lesion, including 6 on the left side and 3 on the right side. Two patients presented bilateral involvement with various severity degree. The OMENS classification system can be perfectly used to analyze the deformities. Respectively, the orbit and zygomatic arch deformity involved 8 sides in 8 cases, the mandible deformity involved 10 sides in 9 cases, the auricular deformity involved 13 sides in 11 cases, and the soft tissue deformity involved 12 sides in 11 cases. Conclusion MSCT based OMENS classification system can make accurate and comprehensive evaluation of the morphological changes in hemifacial microsomia, which provide strong imaging evidence to clinical evaluation.
10.Comparison of effect of lanceolata and astragalus on electrocardiogram of rats with experimental myocardial ischemia

BACKGROUND: Lanceolata and astragalus can improve myocardial ischemia, which are widely used in clinics.OBJECTIVE: To compare the effects of extract of lanceolata and astragalus on electrocardiogram (ECG) of rats with experimental myocardial ischernia.DESIGN: Completely randomized and controlled study.SETTING: Pharmacological College of Harbin Commerce University.MATERIALS: The experiment was carried out at the Laboratory of Pharmacological College of Harbin Commerce University from September to December 2002. Totally 30 healthy adult Wistar rats were divided randomly into 3 groups: control group, lanceolata group and astragalus group with 10 in each group.METHODS: ① After anesthesia, lead ECG Ⅱ of rats was recorded.30 g/kg saline, 30 g/kg lanceolata extract and 30 g/kg astragalus extract [extract of lanceolata and astragalus (raw drug: 1 g/mL)] were injected into rats in the control group, lanceolata group and astragalus group respectively. After 1-3 minutes, 0.5 U/kg pituitrin was injected intravenously (for model of myocardial ischemia), and then saline was used to dilute from 0.1to 0.2 mL for 10-second injection. Changes of ECG after injection of pituitrin were recorded with LMA-2B Ⅱ physiological recorder (migration of T wave and ST section and change of heart rate). ② Differences of data were compared with t test.MAIN OUTCOME MEASURES: Height of T wave and Migrations of ST section and changes of heart rate of ECG of rats before and after medication.RESULTS: Data of totally 30 rats was entered the final analysis. ①Height of T wave: The height was higher after medication than that before medication in three groups (P < 0.01), but that in astragalus group was obviously lower than that in lanceolata group [(1.63±3.81), (2.18±2.93) mm,P < 0.01]. ② Heart rate: The rate was lower after medication than that of before medication in three groups (P < 0.01), and that in astragalus group was obviously lower than that in lanceolata group [(212.21 ±5.01),(221.18±5.66) time/minute, P < 0.01]. ③ Height of ST section: There were not significantly different in three groups (P > 0.05), and the difference was not obvious before and after medication (P > 0.05).CONCLUSION: ① Both astragalus and lanceolata can protect experimental myocardial ischemia induced by pituitrin. ② Effect of lanceolata on reduction of T-wave height is stronger than that of astragalus, but the effect of lanceolata on reduction of heart rate is weaker than that of astragalus.
