1.Ursolic acid inhibits migration and invasion of human lung cancer A549 cells by targeting miRNA-133a

AIM:To investigate the effects of ursolic acid ( UA) on the migration and invasion of human lung cancer cell line A549, and to explore its mechanism .METHODS:The cell viability was detected by MTT assay .The ex-pression of miRNA-133a was detected in the A549 cells treated with UA by real-time PCR.The miRNA-133a mimics and inhibitor were transfected into the A 549 cells, and the transfection efficiency was analyzed by real-time PCR.The cell mi-gratory and invasive abilities were determined by wound healing and Transwell methods , respectively .RESULTS:The via-bility of the human lung cancer A549 cells was significantly inhibited by UA in a dose-dependent manner (P<0.05).IC50 of UA (24 h) for lung cancer A549 cells was 31.04 μmol/L.UA treatment significantly inhibited the migratory and inva-sive abilities of A549 cells in a concentration-dependent manner , accompanied by significantly elevation of miRNA-133a expression.The mimics and inhibitor of miRNA-133a significantly upregulated and downregulated the expression of miRNA -133a in the transfected A549 cells, respectively.In addition, the viability of the A549 cells was decreased extremely after tansfected with the miRNA-133a mimics (P<0.01), so did the results of the cell migration and invasion test .The A549 cells tansfected with the miRNA-133a inhibitor showed an opposite changes of the cell viability , migration and invasion . CONCLUSION:UA inhibited the viability , migration and invasion of lung cancer A 549 cells by elevating the expression of miRNA-133a.
2.The effects of the Seven Acupoints of the Cranial Base on health related quality of life for patients with Parkinson’s disease:a randomized controlled trial

Objectives To evaluate the effects of“the Seven Acupoints of the Cranial Base”(SACB) in improving quality of life(QOL)of patients with PD. Methods The study Subjects were seventy(70)patients with a diagnosis of Parkinson's disease. The Subjects were divided into two groups, i.e. acupuncture group and drug group.The former was treated by SACB every two days,while the latter took Levodopa and Benserazide Tablet250mg tid. The subjects all received treatment for 6 months. PDQ-39,UPDRSⅡand PDSS were adopted to measure quality of life at 4 time points. Results The total effective rate of the treatment group was 85.71%, the control group was 71.43%. And there was a statistical difference of level distribution between the two groups(P<0.01). The total score of PDQ-39, UPDRSⅡ and PDSS of patients in the treatment group [(20.41± 11.64), (9.09±5.02), (126.31±8.37)]were significantly improved compared with the control group[(27.48± 8.69), (10.29±4.93), (109.94±8.11), P<0.01]. The 5 dimension score of PDQ-39, and the 4 single item score of UPDRSⅡ in the treatment group[(8.60±5.07), (3.20±2.08), (2.34±1.73), (1.20±1.68), (1.63±1.96),(1.31±0.68), (0.77±0.94), (0.91±0.74), (0.77±0.65)]were significantly improved compared with the control group[(11.69±4.11), (5.09±3.72), (5.37±1.99), (2.86±2.33), (2.74±2.08), (1.74±0.98), (1.31±0.93), (1.43±0.78), (1.20±0.68), P<0.01 or 0.05]. Conclusion SACB can markedly and multi-dimensionally improve the QOL of patients with PD and has a higher efficacy than Levodopa and Benserazide Tablet.
3.Effectiveness of integrative medicine for preventing postoperative recurrence of primary hepatic carcinomas: a systematic review.

OBJECTIVETo systematically evaluate the effectiveness of integrative medicine (IM) for preventing postoperative recurrence of primary hepatic carcinomas (PHC).
METHODSSuch databases as The Cochrane Library, PubMed, EMbase, China National Knowledge Infrastructure (CNKI), VIP Database, Wan-fang Database were retrieved to collect randomized controlled trials (RCTs) on preventing postoperative recurrence of PHC by IM. The retrieval period started from database establishment to June 2013. Literature screening, data extraction, and quality evaluation were performed by two reviewers strictly according to inclusion criteria. Meta-analysis was conducted using RevMan 5.2 Software.
RESULTSA total of 5 RCTs involving 367 patient cases were included. Results of Meta-analysis showed that the IM group was superior to the Western medicine (WM) group in the 1-, 2-, and 3-year recurrence rates (P < 0.05).
CONCLUSIONIn the prevention of postoperative recurrence of PHC, the effectiveness of IM was more significant, when compared with treatment of Western medicine.

4.Baculoviruses as Vectors in Mammalian Cells

The Baculoviridae are a large family of enveloped DNA viruses exclusively pathogenic to arthropods. Baculoviruses have been extensively used in insect cell-based recombinant protein expression system and as biological pesticides. They have been deomostrated to be safe to mammals, birds and fish. Recently, baculoviruses has been shown to transduce different mammalian cells in spite of the fact that they cannot replicate in mammalian cells (11, 73, 76). This has resulted in the development of baculoviruses as mammalian expression systems and even as vestors for gene therapy.
5.A clinical study on the correlation between the unstability of carotid plaques by ultrasonic examination and ischemic cerebrovascular diseases

Objective To explore the correlation between the unstability of carotid plaques and ischemic cerebrovascular diseases. Methods 112 cases were divided into 6 types according to their carotid ultrasonic examination, counted the focus of ischemic stroke by CT of each patient, then compared the difference of the quantities a-mong the 6 types. Results There were significant differences between the stable and unstable types in the quantities of focus. Conclusion There is a closed relationship between the unstability of carotid plaques by ultrasonic and ischemic cerebrovascular diseases.
6.Effect of Two Different 1.5T MRI Scanners on the Apparent Diffusion Coefifcient Measurement and its Reproducibility in Diffusion Weighted Imaging of the Liver

Purpose To investigate the effect of two different 1.5T MRI scanners on the apparent diffusion coefficient (ADC) measurement and its reproducibility in diffusion weighted imaging (DWI) of the liver. Materials and Methods Two groups of volunteers (33 people in each group) underwent liver DWI examination twice by using the same DWI sequence with b value of 0 and 800 s/mm2 on different MRI scanners (1.5T Siemens Espree and 1.5T Philips Achieva). On ADC maps, two region of interests were placed on three central slices of the left and right liver lobes, respectively. The ADC measurements of six region of interests on left and right liver lobes were averaged to represent the mean ADC values of the left and right liver lobe, respectively. The reproducibility of the ADC measurements was assessed by Bland-Altman method. Results The ADC values of the left and right liver lobes measured on Siemens MRI scanner were significantly higher than those on Philips MRI scanner (t=2.045-2.713, P<0.05); on both scanners, the ADC values of the left liver lobe were significantly higher than those of the right liver lobe (t=-10.561--7.263, P<0.001). The Bland-Altman results showed that the reproducibility of ADC measurement was similar on both MRI scanners, and both displayed higher reproducibility of right liver lobe than that of left liver lobe (on Siemens MRI scanner: the agreement were 16.65% and 7.55% for the left and right liver lobe, respectively;and on Philips MRI scanner:16.30%and 7.85%, respectively. Conclusion Different MRI scanners may have influence on ADC values, but does not affect its reproducibility. Therefore, when ADC values obtained on different MR scanners are compared in clinical analysis, this influence should be considered.
7.Feasibility of magnetic resonance diffusion-weighted intravoxel incoherent motion imaging and its reproducibility in normal liver

Objective To investigate the feasibility of magnetic resonance diffusion-weighted intravoxel incoherent motion (DW-IVIM)imaging and its reproducibility in normal liver.Methods Thirty-five young healthy volunteers underwent liver DW-IVIM ex-amination twice on a 1 .5 Tesla Philips MR scanner.Parameter maps of D,D* and f were generated by using a manufacturer-sup-plied software.Six Region-of-interests (ROIs)(3 ROIs in left and right liver lobe,respectively)were drawn on three consecutive middle slices,respectively.The average values of D,D* and f were measured and compared between the left and right liver lobe. The reproducibility of D,D* and f measurement were assessed with Bland-Altman method.Results The values of D,D* and f of left liver lobe [D(×10-3 mm2/s):1.250±0.210,D*(×10-3 mm2/s):101.906±15.063 ,f(%):22.954±5.268]were statistically higher than those of right liver lobe[D(×10-3 mm2/s):1.039±0.124,D*(×10-3 mm2/s):100.127±20.757,f(%),18.317± 3.671](D,f,P<0.001,D*,P<0.232-0.605),however ,their reproducibility of left liver lobe [limit of agreement(LOA):22.55%,51.05%,39.3%,respectively]were lower than those of right liver lobe (LOA:20.3%,39.25%,33.9%).The D values had the best reproducibility in both the left and right liver lobe.Conclusion The liver DW-IVIM is feasible.The values of D,D*and f in left liver lobe were higher that those in right liver lobe but with worse reproducibility of measurement.The D value was most reproducible in both liver lobes.
8.Effects of acupuncture and moxibustion on the levels of matrix metalloproteinase-1 and interleukin-1β in articular cartilage during the development of knee osteoarthritis caused by immobilization in rabbits

Objective To investigate the effects of warming-needle acupuncture on the levels of matrix metalloproteinase-1 (MMP-1) and interleukin-1β (IL-1β) in the articular cartilage in knee osteoarthritis induced by immobilization in rabbits.MethodsA total of 30 adult male Japanese white rabbits were randomly assigned into three groups by random number table method: a normal group, a model group and a treatment group, with 10 rabbits in each group. Knee osteoarthritis was induced by 6 weeks of immobilization. The rabbits in the treatment group received warming-needle acupuncture after modeling at points of “Neixiyan” (EX-LE4), “Dubi” (ST35), “Zusanli” (ST36) and “Yanglingquan” (GB34) for 4 weeks. The level of MMP-1 and IL-1β in the articular cartilage of the knee were determined with radioimmunological assay.ResultsThere were significant difference in the levels of MMP-1(0.16 ± 0.02 ng/mg, 0.37 ± 0.02 ng/mg, and 0.28 ± 0.03 ng/mg;F=258.251) and IL-1β (0.21 ± 0.01 pg/mg, 0.34 ± 0.02pg/mg, and 0.31 ± 0.04 pg/mg;F=127.112) among the normal group, model group and treatment group. The levels of MMP-1 and IL-1β both in the model group and treatment group were significant higher than those in the normal group (allP<0.01), while the levels of MMP-1 and IL-1β in the treatment group were significant lower than those in the model group (allP<0.01).ConclusionWarming-needle acupuncture can effectively reduced the levels of MMP-1 and IL-1β in the articular cartilage in knee osteoarthritis induced by immobilization in rabbits.
9.Follow-up study on biochemical and structural response in progressive radioactive io-dine-refractory differentiated thyroid cancer patients treated with apatinib

Objective:To evaluate the biochemical and structural changes of apatinib in patients with progressive radioactive iodine-re-fractory differentiated thyroid cancer (RAIR-DTC). Methods:The participants (n=10) were followed up since March 2016. Treatment ef-fect was evaluated in using both biochemical [thyroglobulin (Tg) and thyroglobulin antibody (Tg-Ab)] and structural responses (target lesions, TL). Adverse events were also recorded over time. Results:The median follow-up was 7.9 months. The Tg level declined rapid-ly within 6 weeks after apatinib treatment, and the average decline ranged from 60%to 90%, indicating the immediate biochemical re-sponse of apatinib in progressive RAIR-DTC. The Tg level tended to stabilize thereafter. However, the Tg level rebounded by 4%–135%when withdrawal was performed for 3–14 days. The number of TLs decreased rapidly within 8 weeks, and the average decreased ranged from 40%to 60%, indicating the presence of rapid structural responses. Thereafter, the number of TLs continued to stabilize. TLs, in contrast to Tg, were not significantly affected by drug withdrawal. The rate of change in Tg (Tgvn) was positively correlated with the rate of change in TL (TLvn) [TLvn=0.17×Tgvn+0.50 (r=0.56, P<0.05)]. The apatinib dose was adjusted due to adverse events, which could be relieved after 3 to 14 days of withdrawal. Apatinib can effectively control the disease even at a reduced dose of 250 mg/d. Conclusion:Apatinib treatment showed a fast and sustainable biochemical and structural responses. Tg could be regarded as an objec-tive indicator. Tgvn is positively correlated with TLvn, and the response of Tg is more sensitive than that of TLs.
10.Posterior cervical open-door expansive laminoplasty with mini-titanium plate:correlation between cervical sagittal alignment and repair effect

BACKGROUND:Posterior cervical open-door expansive laminoplasty is one of the simple and effective methods to treat cervical spinal diseases, has satisfactory results but low complications. However, the imbalance of sagittal plan, loss of lordosis or axial pain is often reported recently. OBJECTVE:To analyze the correlation between cervical sagittal alignment and clinical outcomes after laminoplasty. METHODS:Between January 2011 and June 2015, 122 cases of cervical spinal disease, who were treated with open-door expansive laminoplasty with mini-titanium plate, were retrospectively analyzed. Clinical outcomes were evaluated by Japanese Orthopaedic Association, Neck Disability Index and Visual Analogue Scale. Radiographic results were assessed by C2-7 Cobb angle, C2-7 sagittal vertical axis and C7 slope. The correlation between sagittal alignment and clinical outcomes were analyzed. RESULTS AND CONCLUSION:(1) Al patients were fol owed up for 7-32 months. (2) Japanese Orthopaedic Association, Neck Disability Index and Visual Analogue Scale scores were improved in the final fol ow-up (al P<0.001). No significant difference in upper limb Visual Analogue Scale scores was determined (P=0.142). In the final fol ow-up, C2-7 sagittal vertical axis increased (P=0.036), but neither the C2-7 Cobb angle nor the C7 slope significantly changed (P=0.092, P=0.184). (3) There were no correlations between sagittal alignment parameters (C2-7 Cobb angle, C2-7 sagittal vertical axis and C7 slope) and clinical outcomes (Japanese Orthopaedic Association, Neck Disability Index and Visual Analogue Scale) (al P>0.05). (4) These findings indicated that posterior cervical open-door expansive laminoplasty with mini-titanium plate can significantly improve the neurological function of patients. However, there is no correlation between cervical sagittal alignment and clinical outcomes.
