Objective To investigate the relationship between serum surfactant protein-A (SP-A) and -D (SP-D) in patients with systemic lupus erythematosus (SLE) complicated with interstitial lung disease.(SLE-ILD) and its clinical significance.Methods Serum SP-A and SP-D levels of SLE patients and controis were assessed using a sensitive enzyme-linked immunosorbent assay (ELISA).The relationship between SP-A and SP-D and SLE-ILD,the correlation between SP-A and SP-D with age,disease activity index,pulmonary function test and high-resolution computed tomography (HRCT) score were analyzed.Results The level of serum SP-A and SP-D in patients with SLE is higher than that in healthy controls(P＜0.05).The level of serum SP-A and SP-D in patients with SLE complicated with ILD was higher than that in patients without ILD and healthy controls (P＜0.05).The level of serum SP-D in patients with SLE-ILD was correlated positively with HRCT score (r=0.508,P=0.004) and interstitial score (r=0.468,P=0.009),and it correlated inversely with vital capacity (%VC) (r=-0.590,P=0.001 ) and diffusing capacity of carbon monoxide (%DLCO)(r=-0.588,P=0.001 ).There was no correlation between the level of serum SP-A with SLE-ILD and HRCT score,%VC and %DLCO.The level of serum SP-D in patients with SLE-ILD correlated positively with serum IgG (r=0.376,P=0.040),and the level of serum SP-A in patients with SLE-ILD correlated positively with serum C reactive protein (CRP)(r=0.403,P=0.027).There was a positive correlation between the level of serum SP-D in patients with SLE and age (r=0.352,P=O.001 ).Conclusion The levels of serum SP-A and SP-D may be useful markers for ILD in patients with SLE.There is correlation between the level of serum SPD and HRCT score,pulmonary function test,and it is positively correlated with age and disease activity index.

Aberrant microRNA(miRNA) expression has been found in most of human cancers.miRNA-related regulation pathways,including the interaction between miRNA and protein-coding gene and the interaction between miRNA and IncRNA,have been shown correlated with the initiation and progression of human cancers.miRNA might be a therapeutic target in the treatment of malignancies.

Objective To investigate the effect of plant estrogen on serum hormone levels, NOS and oxidative stress products in patients with uterine fibroids with iron deficiency anemia.Methods 80 cases of hysteromyoma patientswith uterine fibroids with iron deficiency anemia were selected, according to the different medication were randomly divided into experiment group and control group,40 cases in each group.Two groups were treated with anemia correction , mifepristone oral ,and the experiment group were treated on the base of the control group with plant estrogen, 12 weeks for a course.serum sex hormone, NOS, SOD and MDA level were detected.ResuIts Compared with the control group, the indexes of patients in the experimental group were lower(P<0.05), level of serum NOS were higher(P<0.05), level of serum SOD were higher(P<0.05), level of serum MDA were lower ( P<0.05 ) .ConcIusion Plant estrogen can improve the serum levels of serum hormone, NOS and oxidative stress levels of the treatment of uterine fibroids, worthy of clinical promotion.

OBJECTIVETo investigate the correlation of the autophagy-associated gene Atg5 with the pathogenesis of prostate cancer.

METHODSUsing real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) and immunohistochemistry, we detected the expression of Atg5 in 50 cases of prostate intraepithelial neoplasm (PIN), 69 cases of prostate cancer (PCa), and 30 cases of benign prostatic hyperplasia (BPH).

RESULTSThe expression level of Atg5 mRNA was significantly higher in PIN (5.270 ± 0.230) and PCa (5.131 ± 0.252) than in the BPH tissue (1.723 ± 0.017) (P <0.01), and so was the positive rate of the Atg5 expression in the patients of the PIN group (94%) and PCa group (88.4%) than in those of the BPH group (6.7%) (P<0.01), but with no statistically significant differences between the PIN and PCa groups (P >0.05). No significant correlation was observed between the expression of Atg5 and the Gleason score of PCa (P >0.05).

CONCLUSIONThe upregulated expression of Atg5 might play a role in the tumorigenesis of prostate cancer.

Objective To screen microRNA (miRNA) that inhibit expression of the metastasisrelated gene ezrin in ovarian cancer cells and explore their correlation to the invasion and metastasis of ovarian cancer. Methods The differential expression of ezrin in two paired high-metastatic and lowmetastatic cell lines were examined by real time reverse transcription (RT)-PCR and western blot. A functional screen with microarray was employed to identify miRNA that were differentially expressed between SKOV3 and SKOV3ip cell lines. Three programs, TARGETSCAN ( http://www. targetscan. org ),MICROCOSM ( http ://www. ebi. ac. uk/enright-srv/microcosm/htdocs/targets/v5/) and PICTAR (http://www. pictar. mdc-berlin. de), were employed to identify all miRNA, which may inhibit the expression of ezrin and were differentially expressed between SKOV3 and SKOV3ip cells. To test the repressive potential of these miRNA, synthetic mimetics were transfected individually into SKOV3ip cells and endogenous ezrin expression levels monitored by western blot and real-time RT-PCR. Results ( 1 ) The mRNA average level of ezrin were (81.74 ± 5.34) -fold higher expression level in SKOV3ip versus SKOV3 cells ( P ＜ 0. 01 ), while (2. 61 ±0. 14)-fold in HO-8910PM versus HO-8910 cells (P ＜0. 01 ). Elevated protein level of ezrin were observed in SKOV3ip cells compared with that in SKOV3 cells, and the same that in HO-8910PM cells compared with HO-8910 cells. Paired SKOV3 and SKOV3ip cells were employed to study the more significant difference in ezrin expression between them. (2) By a functional screen using miRNA microarray combined with bioinformatics analysis,the miR-183 and miR-22 were indentified as two candidate miRNA,which may have the potential regulatory role in ezrin expression. Real time RT-PCR assays revealed that miR-183 and miR-22 were, respectively, an average of (5.84 ± 0.66)-fold and(6.67 ± 0.67)-fold higher expression level in SKOV3ip versus SKOV3 cells (P ＜0. 01 ), which were in agreement with the microarray data. A subsequent validation by western blot and real time RT-PCR revealed that over-expression of miR-183 and miR-22 could both lead to an obvious decrease in ezrin protein level,while there were not signicant difference in the level of ezrin mRNA( P ＞0. 05 ). Conclusion Increased expression of miR-183 and miR-22 may both repress the protein level of ezrin,indicating that miR-183 and miR-22 may bear a potential role in inhibiting ovarian cancer metastasis in a ezrin-mediated way.

Based on the specific disease condition and the patient's tolerance, nutritional supports (with proper nutritional substances and administrative route) should be provided individually.

Objective To evaluate the feasibility of leucovorin(LCV) rescue protocol defined by us,we compared the plasma concentrations,toxicity,LCV doses of different high doses methotrexate(HD-MTX).Methods Seventeen children with acute lymphoblastic leukemia and 1 children with non-Hodgkin′s lymphoma were randomly treated with total 43 courses of HD-MTX.MTX plasma concentrations were measured by fluorescence polarization immuno-assay.Different LCV rescue protocols were prospectively defined for 3 kinds of HD-MTX protocols.Adjusting LCV dose by plasma MTX concentrations.Results No irreversible MTX-related toxicity was observed in all patients.Significant differences of mean steady-state plasma concentrations(Cpss) and total rescue doses were found between 3 groups(P
