1.RNA interference mediated inhibition of epidermal growth factor receptor expression and its significance in different human cancer cell lines

Background and purpose:The epidermal growth factor receptor (EGFR) is commonly overexpressed in a variety of solid tumors, and has important roles in cancer pathogenesis and progression. EGFR thus provides a rational target for cancer therapy. We studied siRNA-mediated inhibition of epidermal growth factor receptor expression and its biologic effects in different human cancer cell lines (A431, HeLa and SPC-A-1). Methods:Cells were transfected with chemically synthesized siRNA-EGFR. EGFR mRNA was quantified by real-time PCR and was detected by immunofluorescence staining and flow cytometry. The biologic effects on cell growth were assessed by colony-formation assay.Results:siRNA-EGFR significantly decreased mRNA level of EGFR by 73.9 %, 44.6 % and 57.7 %, protein expression of EGFR by 77.0 %, 61.3 % and 65.2 %, and reduced colony number by 27.2 %, 53.9 % and 59.1 % in A431, HeLa and SPC-A-1, respectively.Conclusions:Our data suggested that RNA interference could downregulate EGFR and inhibit colony forming ability and EGFR expression at mRNA/protein levels in human cancer cell lines with different pathological types. siRNA could be one of the promising strategies in future targeted cancer therapy.
2.THE CLINICAL EFFICACY OF OMEPRAZOLE IN THE TREATMENT OF PEPTIC ULCER

We have trcated 40 cases of active peptic ulcer with Omeprazolc from Jun. 1990 to Mar. 1991. Compared with it, We have treated 40 cases of active peptic ulcer with Ranitidine during the same time. 80 patients with active peptic ulcer were divided into two groups. The age, scx and size of ulcer between the two groups have been compared. The ulcer diameter was from 3 to 20mm. The number of the ulcer was from 1 to 5.40 patients were treated with Omeprazole 20~40mg p. o. everyday. After a course of two weeks trcatment, the healing rate was 95%; 40 patients were treated with Ranitidine 300mg p. o. everday. Age-related macular degeneration (AMD) has become a leading cause of irreversible visual loss in senior population and a serious issue of public health as the aging of society worldwide,especially wet AMD.Wet AMD is characterized by choroidal neovascularization(CNV)with a rapidly developing process andsevere impairment of visual acuity.The effective therapy that can destroy choroidal neovascularization and improve visual acuity for very long time is lack.In the recent 20 years,more attentions focuse on the treatment of choroidal neovascularization.From bench to bedside,the research on targeted treatment of angiogenesis brings the bloom of wet AMD medical therapy.This paper will summarize the update information of wet AMD.
5.Application of invasive intracranial pressure monitoring in treatment of traumatic brain injury

Objective To discuss the effect of invasive intracranial pressure (ICP) monitoring in treating traumatic brain injury (TBI).Methods A total of 116 patients with TBI (including 112 with severe TBI,3 with moderate TBI and 1 with slight TBI) were monitored by using invasive ICP monitoring device in Tiantan Hospital from July 2003 to December 2007.All patients underwent ICP monitoring within first 24 hours after admission and treated with corresponding therapy including drug therapy and surgical treatment.Results Of all,74 patients survived but 42 with severe TBI died.Of 75 patients with GCS 3-5,33 died,with morbidity rate of 44%.Of 37 patieats with GCS 6-8,nine died,with morbidity rate of 24%.Four patients with GCS 9-15 survived.Conclusions Continuous ICP monitoring can help timely understanding of ICP changes for early diagnosis and correct treatment of TBI and is useful for judgment of prognosis.Low GCS and high ICP are predictors for bad outcome.
6.Clinical and imaging analysis of corpus callosum infarction

Objective To investigate the clinical and imaging characteristics in patients with corpus callosum infarction.Methods The clinical data of 416 patients meetingthe diagnosis of cerebral infarction Were collected,in which,8 patients Were confirmed as corpus callosum infarction by MRI.Results Corpus callosum infarction accounted for 1.9% of all patients with cerebral infarction.CT scan did not show the corpus callosum infaretiom in 7 patients.The nonenhanced MRI revealed the lesions.The enhanced MRI revealed the lesion in another patient.The infarction foci were not only involved in the corpus callosum(knee,body or splenium),but also cornplicated with frontal lobe,occipital lobe and thalamus infractions.The clinical manifesta tions of the corpus callosum infarction were different due to the specific lesion sites.The simple infarction in the body of the corpus callosum mainly presented as contralateral paraparesis.atria,and left limb apra.xia;the infarction in the knee of the corpus callosum mainly presented as lower limb paralysis or contralateral paraparesis;the infarction in the splenium of the corpus callosum presented as limb paralysis and dizziness;the lacunar infarct in the corpus callosum had no obvious clinical manifestations.The patients who complicated with frontal lobe and thalamus infarction had behavioral and psychological syrnptonm,including mental retardation,language abnormalities.and incontinence.Conelusions Corpus callosum infarction is not common.MRI is the basis of diagaosis.The clinical manifestation is lack of specificity.and it is agsociated with the location of corpus callosum infarction and whether it comolicates with the infarction on other part.

Objective Toinvestigatetheentropyoflocalfieldpotentials(LFPs)recordedinratmedialprefrontal cortex during a Y-maze working memory (WM) task, to provide computing support for neural coding mechanism.Methods Sixteen-channel LFPs were recorded from SD rats while they performed a Y-maze WM task.The data came from 4 rats, 20 trials (10 correct trials and 10 incorrect trials) per rat provided by laboratory of neurobiology in medicine,Tianjin Medical University.Original LFPs were preprocessed to remove 50 Hz power line noise and baseline drift.Multi-taper Fourier transform was applied to calculate spatial distributions of LFPs and band pass filter were used to extract characteristic signal.The entroy coding of 16 channel LFPs was as follows: the physiological window was set to be 500 ms, the step length of physiological window was set to be 125 ms, windows were added to LFPs data, and then LFPs entropy of each sliding window was computed and averaged to get the trend of multichannel entropy values duringthe WM task.Results The power of θ band (4-12 Hz) in LFPs increased.The averaged entropy value ofmultichannel θ band LFPs in correct trials was 0.939±-0.020, which were larger than those in the resting state, 0.795±0.031 (P<0.05).Those during wrong WM task had no significant difference, which didn't encode the WM task.Conclusions The principal frequency band related to WM is the θ band and LFPs entropy encodes the WM effectively.
9.Changes in endothelial glycocalyx in ischemia-reperfusion injury and their relationship with microvascular permeability

AIM: To determine the role of the glycocalyx in microvascular permeability. METHODS: Dextran was intravenously injected and quatitatively examined in the rat transient cerebral ischemic model. At the same time, endothelial glycocalyx (anionic sites) was labelled with the probe cationic gold colloid (CGC) using post-embedding technique and examined with electron microscope. RESULTS: The labeling of CGC decreased significantly following ischemia, meanwhile, microvascular permeability to dextran increased. CONCLUSION:Endothelial glycocalyx is very sensitive to ischemia or anoxia. Its disruption may be the initiator of the dysfunction of endothelium and the determinant of increased permeability.
10.Clinical Study on Acupuncture and Ischemic Stroke

Objective:To study the clinical efficacy of acupuncture for ischemic stroke and its effect on ET-1 levels in IS cases.Method:The 63 cases were randomized into a treatment group (31 cases),receiving acupuncture and routine method,and control group (32 cases),receiving routine method alone.Seven days constitute a course of treatment,a 2-day interval between two courses.The clinical efficacy and ET-1 level in two groups were compared after four weeks.Result:The total effective rate in the treatment group and control group were 96.8% and 75% respectively,with a statistical significance (P<0.05);the ET-1 level in blood plasma also with a statistical significance after the treatment (P<0.05);the pre-treatment and post-treatment ET-1 level in the treatment group showed a statistical significance (P<0.01),whereas the control group didn't.Conclusion:Acupuncture is effective for ischemic stroke and can lower the ET-1 level in ischemic stroke cases.
