Background The ubiquitin-proteasome pathway(UPP)involves 80%-90% degradation of all in- tracellular proteins.Both ubiquitin and rat component 3 of proteasome(RC3)are hence considered to be central me- diators for cell biology.Objective To evaluate the effect of simvastatin on neointimal hyperplasia and the expres- sion of ubiquitin and rat component 3 of proteasome(RC3)after balloon injury in carotid artery.Methods Thirty- two male SD rats were randomly to receive,low dose(0.4 mg/ng,n=8),or moderate(4 mg/ng,n=8),or high- dose(40 mg/ng,n=8)of simvastatin treatment for 28 days.Common carotid aortic artery was injuried rat ballon. The ratio of intima-media(I/M)thickness was determined.The expression level of ubiquitin and RC3 mRNA were assessed by RT-PCR.The expression level of ubiquitin protein was examined with immunohistochemistry. Objective To observe the structure changes of the synapse of the neurons in hippocampus CA3 of young rats and study the basis for the mechanism of polyene phosphatidyl in providing learning and memory and the effect on synaptic plasticity.Methods A total of 20 Wistar rats with 5 months were randomly divided into polyene phosphatidyl group and normal control group.Each group had 10 rats.After 4 weeks feeding,Water maze training was peformed in all the rats for 1 weeks.The immue expressions of synapsin(SYN) of the rats in polyene phosphatidyl group and control groups were observed with immunohistochemical method and analyzed by MOTAC imagine analyzing system.The change of synapse of hippocamal CA3 was observed with electron microscope.And the other 24 to 26 months rats were selected as aged group,and fed in the same condition.Moreover,the ultrastructures of hippocamal CA3 of aged rats were observed by transmission electron microscopy(TEM).Results The SYN in polyene phosphatidyl group(0.430 0?0.022 4) was higher than that in control group(0.3567?0.0209) (P

Objective:To isolate breast cancer stem/progenitor cells from human breast cancer and study their proliferation and differentiation biological characteristics over long-term passages in vitro. Methods:Human breast cancer stem/progenitor cells were enriched in suspension cultures as nonadherent mammospheres(MS). Serial sphere formation assay was performed to determine self-renewal ability of mammosphere-derived cells (MSDC). Differentiation was induced by culturing MSDC in DMEM-F12 supplemented with serum but without growth factors. The ratio of CD44~+/CD24~(-/low) cell population was evaluated by flow cytometry(FCM). Results:The mammospheres were formed after inoculation of primary breast cancer cells into culcutre medium with growth factors but without serum. The mammospheres contained undifferentiated cells similar to stem cells, which had self-renewal and extensive proliferation capabilities. With increasing passages, the cells tended to adhere and differentiate. The number of adhering and differentiating cells increased, and the amount and size of mammospheres decreased. The CD44~+/CD24~(-/low) cell population was enriched in the basal-like molecular subtype of human breast tumors. The biological behaviors of mammospheres varied between different specimens.Conclusion:Cancer cells with stem cell properties of self-renewal, indefinite proliferation and multi-lineage differentiation widely existed in human breast cancer tissues. The biological behaviors varied because of different origin of specimens and changed under the effects of environmental factors.

Objective To study the clinical value of anti-ribosomal P0 protein antibodies (anti-P0). Methods Line immuno-assay and immunoblotting methods were used to detect anti-P0 and rRNP antibodies of 49 SEE patients and 61 patients with other rheumatic diseases.The expressions of anti-P0 were compared with those of rRNP antibodies.Possible relations between anti-P0 and clinical features and other antibodies in SLE were explored.Results 36.7%and 6.1% of SEE patients showed positive anti-P0 and rRNP antibodies respectively.Anti-P0 was always negative in patients with other rheumatic diseases (P＜0.01).Among SLE pa tients,incidence of skin rash was 77.8%(35.5%) ff anti-P0 was found to be positive (negative) (P＜0.05). Besides,for patients with positive and negative anti-P0,the chance of positive anti-SmD1 was 61.1% and 19.4% respectively (P＜0.01).Sensitiviy of anti-P0 for SLE diagnosis was 36.73% with specificity and positive predictive value as high as 100%,and 66.30% as the negative predictive value.Conclusion Sera anti-P0 is highly specific for SLE and it is associated with the occurence of rash and the expression of anti-StuD 1 anti bodies in SEE.

During 14-17th days, the duodenum of the rat fetus was composed of stratified epithelium and peripheral mesenchyme. On 18th day, the mesenchyme began to protrude toward the basal aspect of the epithelium, resulted in the formation of primary villi. At the sametime, goblet cells and the primordia of intestinal glands appeared. The number of goblet cells,which located at the surface of the villi, increased gradually with the fetus age, whereas after birth the goblet cells decreased with the development and maturation of intestinal glands. From 19th day till 3-4 weeks after birth, the shape of villi changed continuously, and attained to the adult shape in 4th week.
