Development of liver fibrosis is closely associated with angiogenesis and abnormal vascular remodeling. Recent studies have highlighted the importance of angiogenesis and vascular remodeling in fibrogenesis, the results that inhibition of angiogenesis is effective in suppression of liver fibrosis demonstrate that therapies with several molecular targets against angiogenesis, inflammation and fibrosis might be beneficial for the treatment of cirrhosis. However, there is some evidence that inhibition of angiogenesis can even worsen fibrosis. This article outlines recent advances regarding the interplay between inflammation, angiogenesis and fibrogenesis in terms of cellular and molecular mechanisms, and suggests a requirement of greater understanding to intervene in these key processes, such as liver sinusoidal endothelial cell fenestration and impact distinct chemokine actions driving monocyte migration and differentiation, for therapeutic benefit in the future.

3.Effects of hydrogen-rich saline on postoperative cognitive dysfunction in aged mice

Objective To investigate the effects of hydrogen-rich saline on postoperative cognitive dysfunction (POCD) in aged mice and the possible mechanism.Methods Seventy-two male C57BL/6 mice,aged 18 months,weighing 45-55 g,were randomly divided into 4 groups (n =18 each):control group (group C),hydrogen-rich saline group (group H),POCD group and POCD + hydrogen-rich saline group (group PH).POCD was induced by open tibial fracture.Hydrogen-rich saline 10 ml/kg was injected intraperitoneally twice a day for 7 days before surgery.Six mice were chosen at day 1,3 and 7 after surgery and cognitive function was assessed by Morris water maze test.The mice were then sacrificed and hippocampus was isolated for determination of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) contents (by ELISA) and nuclear factor-κB (NF-κB) activity.Results Compared with group C,the escape latency was significantly prolonged,the time of staying at the original platform quadrant was shortened,the frequency of crossing the original platform was decreased at 3 and 7 days after surgery,and the contents of NF-α and IL-1β and NF-κB activity were increased during 1,3 and 7 days after surgery in group POCD (P < 0.05),and no significant change was found in the parameters mentioned above in group H (P > 0.05).Compared with group POCD,the escape latency was significantly shortened,the time of staying at the original platform quadrant was prolonged,the frequency of crossing the original platform was increased at 3 and 7 days after surgery,and the contents of NF-α and IL-1β and NF-κB activity were decreased at 1,3 and 7 days after surgery.Conclusion Hydrogen-rich saline can alleviate POCD via inhibiting NK-κB activity in hippocampus and reducing inflammatory response.
4.Clinical Observation of Year Rhythm of Laryngeal Cough and Its Relation with TCM Syndromes

Objective To investigate the year rhythm of laryngeal cough.Methods To record the attack months,the syndromes of TCM and degrees of symptoms in detail and analyze the data by percentage and Ridit analysis.Results The periods of laryngeal cough are in spring(March to May) and in autumn(September to November).The syndromes of TCM such as wind violating lung are commonly coughed in spring and primarily mild degree.The syndromes of TCM such as fire hyperactivity are commonly coughed in autumn and primarily moderate degree.Conclusions There is significant year rhythm in laryngeal cough.To master the rhythm can help the diagnosis and treatment.
5.Primary culture of bovine retinal capillary endothelial cells and pericytes in vitro

Objective To probe a selective cultural method for bovine retinal endothelial cells (BREC) and pericytes (BRP) in vitro. Methods With the isolation of active retinal blood vessels, BREC were cultured in a fibronectin coated substrate and Dulbecco′s Modified Eagle′s Medium (DMEM) supplemented with 10% human serum and 100 ?g/ml heparin, while homogeneous cultures of retinal pericytes were obtained when isolated microvessels were seeded to uncoated dishes and grown in DMEM supplemented with 20% fetal bovine serum. BREC were identified by acetylated-low density lipoprotein (Dil-Ac-LDL) incorporation and immunohistochemical method of Von Willebrand factor, while BRP were identified by immunohistochemical method of ?-isoform of smooth-muscle actin. Results The purity of selectively cultured BREC and BRP was more than 98%, being reproducible. BREC got together around the microvessel fragments with the small-cyprinoid-like configuration at first,and could phagocytize Dil-Ac-LDL with the expression of fluorescence in cytoplasm. The expressions of Von Wllebrand factor and ?-isoform of smooth-muscle actin were positive and negative respectively in BREC, while were negative and positive respectively in BRP. Conclusion BREC and BRP with high purity can be obtained by using selective culture and coated-dishes respectively which are simple and repeatable methods.
6.Relationship between retinal protein kinase C and endothelin system in early diabetic rats

Objective To investigate the alteration of protein kinase C (PKC) and endothelin system in early diabetic rats, and the effect of specific PKC inhibitor on the expression of retinal endothelin-1 (ET-1). Methods The rats model with streptozotocin(STZ)-induced diabetes were set up. The expression of retinal PKC was detected by enzyme-linked immunoabsorbent assay (ELISA). The expression of retinal ET-1, ET-3, ET-A and ET-B receptor mRNA was determined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The alteration of retinal ET-1 mRNA after intravitreal injection of PKC inhibitor GF109203X in diabetic rats was also observed. Results The activities of membranous PKC were significantly increased in 2-week diabetic rats compared with that in normal rats(t=3.296, P=0 008), while activities of cytosolic PKC were unchangeable(t=0 138, P=0 894). The expression of retinal ET-1 mRNA was significantly increased(P=0 008), while no change was found in expression of ET-3, ET-A and ET-B mRNA(P=0 918,P=0 889,P=0 500). After intravitreal injection of 10 -5、10 -6、10 -7 mol/L PKC inhibitor GF109203X in diabetic rats, the expression of retinal ET-1 mRNA was decreased in a dose-dependent manner compared with the control rats. Conclusion Activation of PKC and increased expression of ET-1 could be found in the retina of early diabetic rats, and PKC inhibitor could inhibit the expression of retinal ET-1.
7.Effects of advanced glycation end products on the survival and the morphology of bovine retinal endothelial cells and pericytes

Objective To study the effects of advanced glycation end (AGEs) products induced by bovine serum albumin (BSA) on the survival and the morphology of bovine retinal endothelial cells (BREC) and pericytes (BRP). Methods BSA with the final concentration of 50 mg/ml was incubated in PBS, containing 500 mmol/L D-glucose, for 12 weeks under 37℃. AGEs-BSA was purified by Sephacryl S-300 column chromatography and was confirmed by sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE). The concentration of AGEs-BSA was determined by the method of commassie protein assay. In order to detect the toxic effects of AGEs-BSA on cultured BREC and BRP, groups of AGEs-BSA and BSA with different concentration and untreated control were set up. Phase contrast microscope was used to observe the effect of AGEs-BSA and BSA (with the concentration of 500 ?g/ml and actuation duration of 48 hours) on morphology of BREC and BRP. Results[WTBZ] As the dosage of AGEs-BSA increased, the number of inhibited cells increased. When the concentration of AGEs-BSA was 500 ?g/ml, the inhibited BREC in AGEs-BSA group was (72.8?15.9)% of which in untreated control group, and the inhibited BRP was (64.8?9) % of which in untreated control group. AGEs-BSA with low concentration promoted the proliferation of endothelial cells, but there was no significant difference between AGEs-BSA and the control group (P=0.231). Inhibited proliferation and abnormal morphology were seen under the phase contrast microscope while the normal morphology of cells was found in BSA and control group. Conclusion AGEs-BSA with the high concentration may inhibit the growth of both BREC and BRP, which leads the loss of BRP and damage of vascular function. These results suggest that nonenzymatic glycosylation plays a major role in diabetic complications.
8.Insulin has no effect on vascular endothelial growth factor expression in the retina of streptozotocin-induced diabetic rats

Objective To investigate if insulin can affect the expression of vascular endothelial growth factor (VEGF) in the retina of streptozotocin-induced diabetic rats. Methods A total of 60 male Sprague-Dawley rats were randomly divided into sodium citrate buffer control group (CIT-CON, n= 30) and STZ-induced diabetic group (STZ-DM, n=30). At the 16th week, 24 rats from CIT-CON group at random were randomly divided to group A (sodium citrate buffer control group, n = 12) and group B (sodium citrate buffer plus insulin group, n= 12). The remaining 6 rats from as CIT-CON group served as negative control. At the same time, 24 rats from STZ-DM group at random were randomly divided to group C (STZ-induced diabetic group, n= 12) and group D (STZ-induced diabetic plus insulin group, n= 12). The remaining 6 rats from STZ-DM group also served as negative control. 4 IU of insulin was injected subcutaneously to rats of group B and D. Immunohistochemistry, Western blot and Real-time polymerase chain reaction (RT-PCR) were used to measure the expression level of VEGF protein and mRNA respectively. RESULTS Insulin significantly increased the VEGF mRNA (7.71 ± 0.25 vs 5.36 ±0. 37, t test P< 0. 05) and protein expression (0. 4925 ± 0. 0122 vs 0. 4272 ± 0. 0110, t test P< 0. 05) in the retina of CIT-CON rats.However, in retina of STZ-DM rats, insulin had no effect on VEGF mRNA (8. 92±0. 27 vs 9. 05±0. 28, t test, P>0. 05) and protein expression (0. 5152±0. 0109 vs 0. 5099±0. 0100, t test P>0.05). Conclusions Insulin had no effect on VEGF expression in the retina of STZ-DM rats.
9.Study on material base of corydalis rhizoma.

Corydalis Rhizoma, the dried tuber of Corydalis yanhusuo (Papaveraceae) distributed traditionally mainly in south-eastern and now cultivated in northwestern and other district in China, is one of the commonly used and well-known traditional Chinese medicine. It has been widely used to treat spastic pain, abdominal pain, pain due to injury, and promote blood circulation. Its main chemical constituents were alkaloids, which were divided into the two types of protoberberines and aporphines. Among them, some alkaloids were found to elicit profound effects on the dopaminergic system in the central nervous system, which plays an important role in regulating nociception. In this article, the chemical composition and structure-types, new methods of qualitative and quantitative analysis as well as characteristics of biotransformation, absorption, distribution, metabolism, excretion, pharmacokinetic, and drug-drug interaction for the alkaloids were revealed. These results would greatly contribute to the establishment of bioactive material base of Corydalis Rhizoma.

10.Quality Investigation of Four Kinds of Cefoperazone and Sulbactam Sodium injection

OBJECTIVE To investigate different kinds of cefoperazone and sulbactam sodium of different manutactures.METHODS RP-HPLC methods was used to determinate of the concerntrations of 4 categories cefoperazone and sulbactam sodium injections.Agar-dilution method was used to determinate the minimal inhibitory concentration(MIC) of 60 clinical bacterial strains.RESULTS Cefoperazone contents of sample 1 to 4 were 89.11%,100.49%,95.04% and 91.09% respectively;sulbactam contents 90.98%,75.93%,98.98% and 78.09% respectively.The number of the peaks of impurity were 5,4,4,5 respectively;and the total area of impurity peaks were 8.01%,2.94%,2.04% and 10.76%.MIC,MIC50,MIC90 of 4 samples have 2 to 3 difference grades.CONCLUSIONS The products of different manufacturs have different contents and impurities of cefoperazone and sulbactam sodium injections,and the MIC also has different results.In clinical experience,we should surveillance same kinds of antibiotics which produced by different manufacturer and try to reduce the side effect of antibiotics.
