Objective To investigate the effect of amyloid on the expression of kinesin superfamily(KIFs) genes in cortex of Wistar rats. Methods The animal model of Alzheimer's disease was prepared by injection of amyloid into brain ventricle of rats.The learning impairments were verified by water maze test.The levels of mRNA for six kinesin superfamily genes,kinesin,KIFlb,KIFlc,KIF3a,KIF3c,and KIF4,in cortex of rats,were estimated by real-time RT-PCR. Results Two weeks after amyloid injection,the rats showed significant learning impairments.In cortex,the expression of KIF genes was increased after amyloid injection.Conclusion\ The increase of KIF gene may be an evidence for the excitotoxity of amyloid to neurons.

BACKGROUND: The denervated skeletal muscle is chiefly characteristic as the loss of neurotrophic factor, which may cause the muscle atrophy degeneration and fibrosis. OBJECTIVE: To investigate the effect on the muscle satellite cells and muscle atrophy by the implantation of basic fibroblast growth factor (bFGF) released through silicon tube in the gastrocnemius. DESIGN, TIME AND SETTING: Randomly control animal experiment was finished at the Animal Experiment Laboratory of Shenyang Medical College Fengtian Hospital in November 2006. MATERIALS: Twenty-eight Wistar healthy male rats (weighing 250-300 g) were adopted. Silicon tube was prepared by the encapsulation with bFGF or physiological saline, and sealed with 502 glue. METHODS: The sciatic nerve of left lower limb was cut off, and then gastrocnemius was taken out. All of 28 Wistar rats were divided into experiment group and control group randomly, each group involved 14 rats. The experimental gastrocnemius was encapsulated with silicon tube containing bFGF, while the control group with physiological saline. MAIN OUTCOME MEASURES: At day 30 postoperatively, the following indicators were evaluated: wet weight of gastrocnemius and wave amplitude of muscle fibrillation potential; proliferating cell nuclear antigen positive nuclei on surface of muscle fibers, diameter and section area of gastrocnemius fiber under light microscope; ultrastructure of gastrocnemius under electron microscope. The incidence rate of nonalcoholic fatty liver disease (NAFLD) has been increasing in China in recent years and has reached 11％-15％.At present,the multiple-hit hypothesis is considered the main pathogenesis of NAFLD,yet the specific pathogenesis remains unknown.This article elaborates on the roles of carcinoembryonic antigen-related cell adhesion molecule 1 highly expressed in the liver,vitamin D/vitamin D receptor axis,and psychological factors in the development and progression of NAFLD,in order to provide new directions for the research on the pathogenesis of NAFLD in future.

Objective To investigate the effect of isochlorogenic acid B(ICAB)on CCl4-induced acute liver injury(ALI)in mice. Methods The animal model of CCl4-induced ALI in mice was established and then the protective effect of ICAB was evaluated using this model. Serum levels of alanine transaminase(ALT)and aspartate aminotransferase(AST),hepatic levels of malondialdehyde (MDA)and the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were measured by spectrophotometric methods. Liver cell morphology was observed by hematoxylin and eosin staining method and the effects of ICAB on the protein expres-sion of nuclear factor erythroid-2-related factor 2(Nrf2),heme oxygenase-1(HO-1)and quinone oxidoreductase 1(NQO1)in mice he-patocyte were determined by Western blot method. Results ICAB(5,10 and 20 mg/kg)significantly protected against CCl4-induced liver injury by reducing the elevated levels of serum aminotransferases and hepatic MDA and remarkably restored the impaired antioxi-dants. Meanwhile,the histopathological changes were also attenuated in mice. In addition,ICAB could induce the protein expression of Nrf2 and promote its nuclear translocation,and further increase the protein expression of HO-1 and NQO1. Conclusion ICAB has protective effect against CCl4-induced ALI in mice,which is mainly due to its ability to promote the nuclear translocation of Nrf2 and decrease the oxidative stress.

Objective:To establish a method for the determination of chloride and sulfate in lithium carbonate crude drug by sup-pressed ion chromatography. Methods:The analysis column and the guard column was Thermo IonPac AS11(250 mm × 4 mm) and IonPac AG 11(50 mm ×4 mm), respectively. Potassium hydroxide solution(10 mmol·L-1) generated by an eluent generator(EG) was used as the eluent at a flow rate of 1. 0 ml·min-1 . And the temperature of column and conductivity cell was 30℃ and 35℃, re-spectively. The suppressor electric current was 36 mA, and the injection volume was 10 μl. Results:The linear range of chloride ion and sulfate ion was 1. 0-20μg·ml-1 with r of 0. 995 1 and 0. 997 3,respectively, and the recovery was 106. 6%(RSD=2. 6%,n=6) and 100. 3%(RSD=1. 9%,n=6), respectively. Conclusion:The method is accurate and reliable, and can be used in the determina-tion of chloride and sulfate in lithium carbonate crude drug.

Objective:To explore the effects of antisense oligonucleotide(ASODN)of connective tissue growth factor (CTGF)on the apoptosis of human hypertrophic scar fibroblasts(HSF)and the related mechanisms.Methods:The hypertrophic scar fibroblasts were cultured in vitro and were divided into the following 3 groups:CTGF-ASODN-treated group, liposome control group and blank control group.The distribution of CTGF-ASODN in HSF of different phases was observed under fluorescent microscope in 3 groups.Flow cytometer was employed to detect the apoptotic rate of HSF and the expression of Fas.The expression of CTGF,Fas and bcl-2 mRNA in HSF was assessed by RT-PCR.Results:24 h after transfection,large quantity of yellow-green fluorescence was seen in the HSF cytoplasm of CTGF-ASDON group in a scattered or doted manner, but not found in that of the other 2 groups.The apoptosis rate of HSF in CTGF-ASDON group was significantly higher than those in the other 2 groups(P
