4.Effect of angiotensin on the hypertrophy of myocardial cells in culture
Chinese Journal of Pathophysiology 1986;0(01):-
Experiments were carried on cultured neonatal myocardial cells of rat to study the effects of angiotensin Ⅰ(AngⅠ) and Ⅱ (AngⅡ) on the synthesis of DNA, RNA, protein and on the cardiac hypertrophy. It was shown that the incorporation of both ~3H-TdR and ~(14)C-uR increased over a set of time periods after addition of AngⅠ and AngⅡ to the medium. The mean protein content of the single myocardial cell was also enhanced, and the volume of myocardial cell was increased significantly after a continuous stimulation of AngⅠ and AngⅡ for 7 days. No stimulation effect was observed either in the synthesis of DNA, RNA, and protein or in the volume of myocardial cells after addition of AngⅠ together with the angiotensin converting enzyme inhibitor(ACEI, captopril). These results indicated that: (1). AngⅠ acts as a regulator by its indirect stimulation effect on the synthesis of DNA, RNA and protein of myocardial cells, since it must be changed into AngⅡ by the ACE present in myocardial cells. The ACE can be inhibited by the ACEI: (2) The hypertrophy of cultured neonatal myocardial cells may induced by a continuous stimulation of AngⅠ and AngⅡ.
5.Effect of AngⅡ on Insulin Gene Expression in RIN-m Cells and Its Molecular Mechanism
China Pharmacist 2015;18(12):2025-2029
Objective:To discuss the effect of AngⅡ on insulin gene expression IN RIN-m cells and its molecular mechanism. Methods:RIN-m cells were cultured and divided into three groups, including the control group, 100 nmol·L-1 AngⅡ group and losartan pretreatment group. After 24-hour incubation, insulin gene expression in RIN-m cells was detected by RT-PCR, the mean flu-orescent intensity of 2', 7'-dichlorofluorescein ( DCF) was detected by flow cytometry, PDX-1 and MafA mRNA expression were detec-ted by RT-PCR and the protein expression was detected by Western-blot. Results: Insulin expression in RIN-m cells, cellular ROS level, PDX-1 expression and MafA expression in 100 nmol · L-1 AngⅡ group were significantly different from those in the control group and losartan pretreatment group (P<0. 05). The later two groups had no significant differentces in those indices (P>0. 05). Conclusion:AngⅡ can down-regulate PDX-1 and MafA expression inβ-cells through oxidative stress pathway, and then inhibit insu-lin gene expression. Pretreatment with losartan can antagonize the effect of AngⅡ, and has protective effect onβ-cells in the aspect of insulin gene expression.
6.Clinical Analysis of Drug Fever Induced by Piperacillin Sodium and Tazobactam Sodium for Injection
China Pharmacy 2015;(18):2503-2505
OBJECTIVE:To provide reference for the timely and accurate judgment of drug fever induced by Piperacillin sodi-um and tazobactam sodium for injection and rational drug use in the clinic. METHODS:The medication, clinical manifestations, laboratory examination results and the physician treatment of 30 inpatients with drug fever induced by Piperacillin sodium and tazo-bactam sodium for injection from Sept. 2013 to Sept. 2014 were analyzed retrospectively and statistically. RESULTS:Most of the drug fever induced by Piperacillin sodium and tazobactam sodium for injection occurred in continuous 7 to 14 d medication, and the cumulative dosages were between 1.3 to 2.7 g/kg;73.3% of the 30 patients had fever during the intravenous drip, with body temperature mainly≥38.5 ℃;the elevation of eosinophil and slightly increase of serum C-reactive protein (CRP) and blood cell sedimentation rate (ESR) could be used as observation indexes of drug fever, but the leucocyte reduce could’t be suitable;the body temperature dropped to normal within 24 to 48 hours after stopped using it. CONCLUSIONS:The drug fever induced by Piperacillin sodium and tazobactam sodium for injection has no correlation with the patients’gender or age,and no special diagnos-tic criteria. But it has certain correlation with duration of medication, cumulative days and dosages and it can be used as reference of judgment with the combination of hematological examination index. Clinicians should improve the understanding and attention about drug fever to stop using suspicious drug in time.

9.MicroRNA in platinum-resistance of ovarian cancer
Journal of International Oncology 2011;38(5):384-386
MircoRNAs (miRNAs) are widely existing inorganisms, non-coding small RNA. MircoR-NAs play important role in cell proliferation, apoptosis, stress resistance and fat metabolism. In vitro studies indicate that miRNAs are important factors in platinum resistance of ovarian cancer cell lines. However, the pathway of platinum resistance of ovarian cancers is complex. Studies find miRNA expression play important roles in the sensitivity of platinum resistance of ovarian cancer cell lines , and can be used as an early alert for platinum resistance. Changing the expression of miRNA even can reverse the platinum resistance . But the regular role of miRNAs in target genes network is still unconfirmed. Thus, more studies about miRNA in vivo and animal tumor models are needed to identify the function of miRNAs in platinum resistance of ovarian cancer.
