Objective To study the effect of lgG Fc gene on JEV DNA vaccine immunity. Methods Gene encoding IgG Fc was amplified by nested-RT-PCR technique from BALB/c murine spleen cells. JEV prME protein gene was obtained with restriction endonuclease BamH Ⅰ/EcoR Ⅰ from the eukaryotic recombinant named after pJME, which was constructed by us before. Recombinant, named after pJME/IgG Fc, with above two genes encoding JEV prME protein and BALB/c murine IgG Fc was constructed, and was tested by restriction enzymes analysis and DNA sequencing, then was transfected into China hamster ovary (CHO) cells by Lipo-fectAMINE 2000. Distribution and expression of the fusion proteins encoded by JEV prME protein and BALB/c murine IgG Fc genes in transfected CHO cells were detected by immunofluorescence and Western blot. The BALB/c micc were vaccinated with pJME/IgG Fc via intramuscular injection. Then the cytotoxic T lymphocyt (CTL) activity were assessed by lactic dehydrogenase (LDH) and the neutralizing antibody titer were assessed by 80% plaque reduction neutralization test. Results Molecular weights (2001 bp, 2730 bp) of the two in- serts released from pJME/IgG Fc with two group of restriction analysis associated with BamH 1/EcoR I and BamH Ⅰ/Not Ⅰ were correlated to the expected theoretic results respectively. It was estimated that molecular weight (Mr) of the fusion protein was 101 x 103. The expression of the above fusion protein was mainly distribu- ted in endochylema of transfected CHO cells, and not much in membrane of transfected CHO cells. CHO cells transfected with pJME/IgG Fc could express the fusion protein at the 32th cell passage. After immunization, the CTL activity and the neutralizing antibody titer in the pJMF/IgG Fc vaccinated group increased significantly compared with other vaccinated groups(P <0.05). Conclusion The recombinant pJME/IgG Fc was construc- ted and transfected into CHO cells successfully, and CHO cellular lines expressed fusion protein encoded by JEV prME protein and BALB/c murine lgG Fc genes stably were obtained. IgG Fc gene could reinforce the cellular immunity and humoral immunity of JEV DNA vaccine.

Robotic catheter minimally invasive operation requires that the driver control system has the advantages of quick response, strong anti-jamming and real-time tracking of target trajectory. Since the catheter parameters of itself and movement environment and other factors continuously change, when the driver is controlled using traditional proportional-integral-derivative (PID), the controller gain becomes fixed once the PID parameters are set. It can not change with the change of the parameters of the object and environmental disturbance so that its change affects the position tracking accuracy, and may bring a large overshoot endangering patients' vessel. Therefore, this paper adopts fuzzy PID control method to adjust PID gain parameters in the tracking process in order to improve the system anti-interference ability, dynamic performance and tracking accuracy. The simulation results showed that the fuzzy PID control method had a fast tracking performance and a strong robustness. Compared with those of traditional PID control, the feasibility and practicability of fuzzy PID control are verified in a robotic catheter minimally invasive operation.

BACKGROUND: The biological behaviors of preadipocytes in adipose tissue of young animals have been closely linked to the onset and prevention and treatment of obesity. Observing mechanical oscillation effects on biomechanical behaviors of preadipocytes using biomechanical stimuli would provide more direct experimental evidence for treatment of simple obesity using manipulation and massage therapy.OBJECTIVE: Different frequencies of mechanical force stimulation were performed on preadipocytes from young rats cultured in vitro to observe the changes in cell viability, proliferation, and apoptosis.METHODS: Preadipocytes from SD young rats were in vitro cultured. Following identification, preadipocytes were dynamically, mechanically stimulated through the use of constant temperature oscillator. According to different treatment frequencies, three groups were set: 0 Hz (blank control), 1.5 Hz, and 3 Hz. A 30-minute oscillation, once every 12 hours, total 3 days, was performed in each group. Following dynamic mechanical stimulation, cell viability, proliferation, and apoptosis in young rats were observed. RESULTS AND CONCLUSION: In the initial stage of culture, cells exhibited the morphology similar to fibroblasts. After oil red O staining, red particles appeared in the cells, indicating that the young mouse cells cultured in vitro were preadipocytes. With stimulation of oscillation force, the viability and proliferation of preadipocytes were significantly inhibited (P < 0.05 or P < 0.01). There wasno significant difference in effects of mechanical oscillation on preadipocyte apoptosis between 0 Hz and 1.5 Hz, 3 Hz groups (P > 0.05). These findings indicate that thecell biological mechanism underlying preventing simple obesity in adolescents is to inhibit the viability and proliferation of preadipocytes

Chinese Journal of Practical Nursing 2016;32(12):885-889
Objective To explore the correlation between humanistic care ability of nursing students and their family environmental factors. Methods A sample of 198 nursing students was recruited with cluster sampling method. Demographic data, humanistic caring capacity, and family environmental factors were collected using a self-designed questionnaire, Caring Ability- Inventory (CAI) and Family Environment Scale (FES- CV) and then an analysis was conducted. Results Han nationality students accounted for 32.83%(65/198), minority students accounted for 67.17%(133/198) . The total average score of humanistic care ability was 190.79±18.84, the average score of items was 5.16±0.99. There were significant differences between our participants and those regarded as international norms on the three dimensions of humanistic care ability (t=-16.27--8.12, P < 0.01). The total average score of family environment was 42.57±8.88. Significant differences were also presented between our participants and those of domestic norms on the ten factors of the family environment (t=-10.94-30.04, P < 0.01). The significant correlation was not documented between the total scores of humanistic care ability and family environment. However, the correlations were documented between the six domains of family environment (intimacy, emotional expression, knowledge, entertainment, contradiction, sense of organization) and the total score and each dimension of humanistic caring capacity (P < 0.05 or 0.01). Conclusions The humanistic care ability of nursing students in minority area is relatively low. The humanistic care ability and family environment are correlated on the multi- dimen-sional facets. School educational strategies should be combined with family environment factors of nursing students to jointly promote the humanistic care ability of nursing students.

BACKGROUND:At present, the common filing materials used to correct secondary unilateral cleft lip nasal deformity include conchae cartilage, costal cartilage, Medpor implants, expanded polytetrafluoroethylene (ePTFE), alogenic acelular dermal matrix. OBJECTIVE:To analyze the therapeutic effects of comprehensive procedures with alogenic acelular dermal matrix or ePTFE for secondary unilateral cleft lip nasal deformity. METHODS: Thirty-six patients with secondary unilateral cleft lip nasal deformity were enroled, including 19 males and 17 females, aged 15-32 years. Alogenic acelular dermal matrix (n=22) or ePTFE (n=14) was used to correct nasal base colapse deformities. Anthropometry method was employed to make measurements. Fixed-point measurement was performed based on patient's pictures before and after correction. Long-term effects of these two kinds of filing materials were analyzed and assessed objectively and quantitatively. RESULTS AND CONCLUSION: After the folow-up of 6 months, al the patients were satisfied with their results, and no infection and no exposure occurred. The treatment effect of the alogenic acelular dermal matrix group was excelent in 16 cases and good in 6 cases; there were 10 cases of excelent and 4 cases of good in the ePTFE group. The objective indicators in the two groups were al improved at 6 months after correction (P < 0.05), but there was no difference between the two groups (P > 0.05). These findings indicate that alogenic acelular dermal matrix or ePTFE is useful to correct secondary unilateral cleft lip nasal deformity.
