1.Expressions and clinical significances of microRNA-126 and vascular endothelial growth factor in gastric carcinoma

Objective To observe the expressions of microRNA-126 (miR-126) and vascular endothelial growth factor (VEGF) in gastric carcinoma tissues,and their associations with clinicopathologic features in gastric carcinoma.Methods Fifty cases of gastric carcinoma and matched tumor adjacent tissue specimens were collected from January 2012 to March 2014 in Shangqiu First People's Hospital of He' nan Province.Expression levels of miR-126 and VEGF were examined using reverse transcriptase-polymerase chain reaction.Their correlations and the relationships between them and the clinicopathologic features in gastric carcinoma were analyzed.Results The expression levels of miR-126 in the tumor tissues and in the adjacent tissues were 0.652 ± 0.102 and 0.379 ± 0.069,and the expression levels of VEGF in the two kinds of tissues were 1.523 ± 0.142 and 0.604 ± 0.152,with statistical differences (t =8.374,P =0.001;t =29.508,P =0.000).The expressions of miR-126 and VEGF were significantly positively correlated (r =0.735,P =0.001).The expressions of miR-126 and VEGF were associated with TNM stage (t =22.981,P =0.001;t =18.053,P =0.001),depth of tumor invasion (t =26.792,P =0.001;t =20.234,P =0.001) and tumor node metastasis (t=26.668,P=0.001;t=14.857,P=0.001).Conclusion miR-126 is high expressed in the gastric carcinoma tissue,which may up-regulate the expression of VEGF,and lead to the formation of new blood vessels,then to promote gastric carcinogenesis and the development process.
2.Influence of psychological intervention on life quality of patients with non-erosive reflux disease complicated with depression

Objective To study influence of psychological intervention on life quality of patients with non-erosive reflux disease(NERD) complicated with depression. Methods Patients with NERD and depression(95 cases) were divided randomly into the observation group(48 cases) and the control group(47 cases).The control group received oral administration of rabeprazole,10 mg,bid.The observation group was given psychological intervention based upon routine medication.Tbe reflux diagnostic questionnaire (RDQ), self-rating depression scale(SDS) and quality of life-index(QL-INDEX) scale were adopted for evaluation before and 8 weeks after treatment. Results The score of SDS was (41.58±6.85) in the observation group,which was lower than that of the control group,(50.42±9.26), after 8 weeks of treatment (P<0.01). The score of QL-INDEX was (8.56±1.38) in the observation group,which was higher than that of the con-trol group,(7.98±1.42), after 8 weeks of treatment (P<0.05). Conclusions Psychological intervention for patients with non-erosive reflux disease (NERD) complicated with depression can ameliorate their de-pression state and improve their life quality.
3.Effect of psychological behavior intervention on patients with functional constipation

Objective To investigate the effect of the psychological behavior intervention on the mental status and clinical symptom of patients with functional constipation. Methods All of 78 patients with functional constipation diagnosed by Rome m diagnostic code were randomly divided into study group (n =40) and control group (n = 38). The psychological behavior intervention was used in the study group, the caccagogue and symptomatic treatment was used in the control group. The self-rating anxiety scale(SAS),self-rating depressive scale (SDS) was analyzed and the improving of clinical symptom, adverse effect was compared before and after 4 weeks of treatment respectively. Results The scores of SAS and SDS in the study group significantly decreased after treatment [ (36.25±8.61 )scores vs (42.97±9.25 )scores, P<0.01 ;(38.81±6.53)scores vs (43.36±7.18 )scores ,P < 0.01].There was significant difference between the two groups after treatment (P <0.05).The effective rate of study group was higher than that of control group .(97.50% vs 84.21%,P <0.05). The rate of celiodynia, diarrhea and the recrudescence after the course of treatment in control group was 23.68%, 15.79% and 89.47% respectively, while this numerus in study group was 5.00%,2.50% and 5.00%. Conclusions Psychological behavior intervention plays an important role in the treatment of functional constipation. It can improve the mental status, clinical symptom, and decrease the adverse effect.
4.Molecular and genetic diagnosis of male infertility:practices and challenges

Genetic factors are important causes of male infertility and account for about 30%infertility cases.Therefore, it is necessary to carry out molecular and genetic detections in the diagnosis and treatment of male infertility.Here the most commonly used techniques for molecular and genetic diagnosis of male infertility in recent years are discussed , including chromosomal abnormities analysis , Y chromosome microdeletions detection , gene mutation screening and sperm quality and function examination.
5.PTEN inhibited the invasion of glioma cells with a common mutant epidermal growth factor receptor expression by dephosphorylating FAK

Background and purpose:PTEN mutation has been found in 20%-40% of malignant gliomas.The common mutant epidermal growth factor receptor(EGFR vⅢ)was reported to coexpress in PTEN-deficient EGFR-expressing tumor.PTEN has been shown to interact directly with FAK and reduce its tyrosine phosphorylation levels to inhibit cell invasion.The invasion of glioma cells with EGFRvⅢ expression and PTEN deficiency is increased.This study was to observe whether PTEN inhibits glioma cell invasion even in the presence of strong pro-invasive signals provided by constitutive EGFR activity.Methods:U87?EGFR cells were transfected with pcDNA3.1 constructs encoding PTEN and the cells invasion levels were detected by transwell invasion assay.The expression of FAK was detected by immunoblotting.FAK expression vector was transfected into U87?EGFR-wtPTEN cells and the change of cells invasion was documented.Results:PTEN and PTEN(G129E)could inhibit cell invasion induced by EGFRvⅢ.PTEN and PTEN(G129E)could decrease the FAK phosphorylation at Tyr397.Over expression of FAK in U87?EGFR-PTEN abrogated PTEN-induced down-regulation of the phosphorylation status of FAK and rescued cell invasion.Conclusions:PTEN could inhibit cell invasion induced by EGFRvⅢ by dephosphorylating FAK.
6.Study of anti-lipid A antibodies of bacterial endotoxin from phage antibody library

Objective To study the treatment of sepsis caused by G - bacteria, anti-lipid A antibodies of bacterial endotoxin were screened from phage antibody library. Methods The mRNA was extracted from human B-lymphocytes against lipid A of bacterial endotoxin, reversely transcripted and amplified by polymerase chain reaction using general primers scanning Fd and light chain of IgG. The amplified fragments were inserted into pCOMB3 vector and electrotransfected competent E.coli XL 1-blue cells. Furthermore, the recombinant phage was lysed by coculture with helper VCSM13. Results Fab displayed on the surface as fusion protein with the N terminal of coat protein Ⅲ, and 4.8?10 6 clone library was established. Antibodies against lipid A of bacterial endotoxin were screened. Specific antibodies against lipid A of bacterial endotoxin were enriched by 100 times after three rounds of panning with lipid A.Conclusions Three clones exhibited specific binding to lipid A is identified by direct and competitive ELISA methods. The succcess of isolating anti-lipid A proves the usefulness of phage display system in human McAb preparation. The result shows that we have got the recombinant phage antibody.
7.Expression of inositol 1,4,5-triphosphate receptor in airway smooth muscle cells of rats and its roles in chronic asthma formation

Objective To detect the expression of inositol 1,4,5-triphosphate receptor (IP 3R) subtypes in normal rat airway smooth muscle cells (ASMCs) and changes during chronic asthma formation. Methods ASMCs were cultured by collagen enzyme digestion method. The expressions of subtypes of IP 3R were detected by RT-PCR and the purified PCR products were linked with pGEM-T vector for DNA sequencing. Chronic asthma model was established with egg albumin. 8.Immunogenicity of a novel vaccine with VLP as vector for Wilm's tumor

Objective To develop a vaccine for Wilm's tumor in order to lay a foundation for effective treatment and substitute for the radiotherapy and chemotherapy of this tumor. Methods DNA fragments encoding the WT1 antigens (including, HLA-A * 2402 and HLA-A * 2402x), and DNA fragments encoding the couple antigens HLA-A * 2402-HBc and HLA-A * 2402x-HBc were cloned and inserted into the eukaryotic expression vector pcDNA3.1(+) in sense orientation and baculovirus expression pFactbac respectively, then transfected into COS-7 cells and Sf 9 cell respectively. The expression of the target proteins in the cells were identified by CPE, SDS-PAGE and electron microscopy. Virus-like particles were observed under electron microscopy. Results The fusion gene of HLA-A * 2402 and HLA-A * 2402x and HBc express virus-like particle protein. Conclusions This preliminary result shows a hopeful future in developing an effective immunotherapy to Wilm's tumor.
9.Preliminary screening of different permeation enhancers in transcutaneous immunization with inactivated human highly pathogenic avian influenza vaccine

Objective To screen the potent permeation enhancers used in transcutaneous immunization with inactivated highly pathogenic avian influenza vaccine. Methods Five different permeation enhancers, ethanol, propylene glycol, dimethyl sulfoxide, ratinoic acid, oleic acid, were used to treat the skin of BALB/c mice before transcutaneous immunization. Sera were collected before the flist transcutaneous immunization and every two weeks post immunization. The titers of influenza virus-specific humoral IgG and IgA were assayed in serum, lung and nasal lavages by ELISA. The titers of hemagglutination inhibition ( HAI), IFN-γand IL-4 produced by splenic lymphocytes were also detected. Except that, clinical symptom of the skin in different time points and skin pathological changes were observed. Results The serum IgG titers, HAI titers and the influenza virus-specific lgA and IgG in lung and nasal lavages in the groups of HA +CT + DMSO, HA + CT + RA and HA + CT + OA were significantly higher than those of HA and HA + CT groups( P <0.05). Moreover, the numbers of splenic lymphocytes producing IFN-γ and IL-4 were increased in the above three groups than those in control groups. In addition, no evident clinical symptoms were observed, but stratum corneum of the skin in different groups showed different changes. Conclusion DMSO,RA and OA are potent permeation enhancers in mouse model inoculated with inactivated high pathogenic avian influenza vaccine transcutaneously.
10.The expression of osteopontin and its nucleating role in gallbladder bile of patients with cholesterol gallstone

Objective To investigate the role of osteopontin (OPN) in cholesterol gallstone formation.MethodsGallbladder bile was obtained from patients with cholelithiasis (n=36,the experimental group) and from donors of liver transplantation (n=19,the control group).OPN,calcium ion and lipid were analysed quantitively.The nucleating role of OPN in bile was evaluated using nucleating time (NT) approach.ResultsOPN inhibited cholesterol nucleation in a dose dependent manner.OPN (50 μg/ml and 100 μg/ml) prolonged NT by 48.90% (91.51%) and 17.07% (32.93%) in lithogenic and control bile,respectively.OPN (100 μg/ml) also inhibited the nucleating effect induced by calcium ion.Furthermore,a combination of OPN (50 μg/ml) and calcium prolonged NT by 75.78% and 33.96% in lithogenic and control bile,respectively.A combination of OPN (100 μg/ml) and calcium prolonged NT by 125.9% and 62.26% in the 2 groups.The contents of osteopontin and calcium were significantly lower in lithogenic bile than control bile (P<0.05).On the other hand,the cholesterol saturation index and the contents of cholesterol,phospholipid and bile acid were significantly higher (P<0.05).ConclusionsOPN inhibited cholesterol gallstone formation.It may be involved in the pathogenesis of cholelithiasis.
