Objective:To observe the transfection efficacy and targeting efficiency of hepatitis B virus envelope(HBVE) as a gene transfer vector for liver cancer cells.Methods:HBVE was obtained from the supernatant of HepG2.2.15 cells with a PEG8000 system and ?-propiolactone.The pIRS2-EGFP was packed with HBVE to obtain HBVE-GFP and was packed with liposome to obtain Liposome-GFP.HBVE-GFP and Liposome-GFP were used to transfect human hepatoblastoma cell line HepG 2 to study the transfection efficiency.HepG 2,A 549,HeLa and FB cells were transfected with HBVE-GFP to appraise the targeting ability of HBVE-GFP.GFP protein expression was observed under a fluorescent microscope and the ratio of GFP positive cells was determined by flow cytometry.Results:(1) Transfection efficiency:The GFP protein was observed in both the liposome group and the HBVE group under the fluorescent microscope;the fluorescent intensity in the HBVE group was 3-4 times that of liposome group as determined by flow cytometry(P

Objective To explore cancer factor exhausted during chemotherapy intermittent for drinking, food and oxygenic sport interference to patients suffered from ovarium cancer. Method 92 patients suffered from ovarium cancer with drinking, food and oxygenic sport interference and taking self-comparison method. Simple tired Assessment Form, European cancer treatment and research origination questionnaire (EORTC-QLQ-C30) are as-sessed for the patients suffered from cancer tied and life quality situation before interference and after interference of 30 d so as to observe the tired relieving degree and life quality of the patients before and after interference. Results Using nursing interference and then the patients relive their tiredness greatly to have statistical significance (P <0.01). The comparison before and after interference and the life quality has statistic significance(P <0.01). Con-clusion The drinking, food and oxygenic sport interference can relieve the fired degree of the patients suffered from ovarium cancer effectively to improve the life quality of the patients.
5.Influence of psychological nursing intervention for quality of life in patients with ovary cancer

Objective To know the influence of psychological nursing intervention for quality of life in patients with ovary cancer. Methods Divided 300 patients with ovary cancer into the intervention group and the control group randomly, there were 150 cases in the each group. Routine nursing cares was ued in the control group, the psychological nursing intervention was used in the intervention group in addi-tion. Compared the quality of life between the two groups by interviewed questionnair. Results After the nursing intervention, the indexes which can indicated the quality of life in the intervention group were bettez than those of in the control group significantly. Conclusions Psychological nursing intervention can ef-fective promote the quality of life of patients with ovary cancer.
6.Effects ofⅠ,ⅡCrystal and Amorphous Forms of Lercanidipine Hydrochloride on the Preparation

OBJECTIVE:To study the effects of Ⅰ,Ⅱ crystal and amorphous forms of lercanidipine hydrochloride on the preparation,and provide theoretical basis for its development and consistency evaluation. METHODS:X-ray powder diffraction (XRD),infrared spectrophotometry(IR)and differential scanning calorimetry(DSC)were adopted to identify the 3 crystal forms of lercanidipine hydrochloride. XRD was used to compare the effects of crushing,grinding,pressing technology,wetting granula-tion,adhesive solvents(water,ethanol)and drying temperature(50,60,70℃)on stability of 3 crystal forms of lercanidipine hy-drochloride;the dissolution in vitro in water,hydrochloride,pH 4.5 acetate buffer,pH 6.8 phosphate buffer were compared among 3 crystal forms of Lercanidipine hydrochloride tablet. RESULTS:XRD showed both Ⅰ,Ⅱ crystal forms had characteristic diffrac-tion peak with inconsistent 2 θ values,amorphous had no characteristic diffraction peak;IR showed 3 crystal forms had different absorption intensity and absorption peak number;DSC showed Ⅰ,Ⅱ crystal forms had obvious endothermic peak in 194.6 ℃, 207.3 ℃,respectively,amorphous had obvious endothermic peak in 86.1 ℃ and exothermic peak in 299.8 ℃. Crushing,grinding, pressing and drying temperature had no effects on the stability of 3 crystal forms;water had no effect on the stability of crystal in wetting granulation,ethanol may cause the change of Ⅰcrystal form. Except for the comparison between Ⅰ,Ⅱ crystal forms in hydrochloride (f2=68),the dissolution f2 of 3 crystal forms in 4 kinds of medium were lower than 50. CONCLUSIONS:XRD, IR,DSC methods can identify the 3 crystal forms of Lercanidipine hydrochloride tablet. When preparing lercanidipine hydrochlo-ride by Ⅰcrystal form,wetting granulation should avoid using ethanol as a adhesive solvent,instead of water. Different crystal forms can affect the dissolution in vitro of prepared Lercanidipine hydrochloride tablet.
7.Study on correlations between CT perfusion parameters and tumor angiogenesis of cervical cancer.

This study was aimed to analyze the correlation between CT perfusion parameters, e. g. perfusion (PF), blood volume (BV), peak enhancement image (PEI), time to peak (TTP), and the microvessel density (MVD) of cervical cancer. CT perfusion scans were carried out in 31 patients with cervical cancer. After their surgical resections, we obtained 31 cases of cervical cancer specimens, and 15 cases of adjacent normal cervical specimens as control group, then these 46 specimens were used in MVD detection through immunohistochemical CD34 staining. The MVDs of the tumor group were significantly higher than those in the control group, and the former seems to increase with the clinical stages with a positive correlation. The PF and PEI values have positive correlations with corresponding MVD values. The PF value of squamous carcinoma group has correlation with corresponding MVD value. The PF and PEI values of poorly differentiated group have correlations with corresponding MVD values. The PF, PEI and BV values of II-III stage group are positively correlated with corresponding MVD values. The PF value of lymph metastasis or non-lymph metastasis group both have correlations with corresponding MVD values. We found that CT perfusion parameters were positively correlated with corresponding MVD values in cervical cancer. Our results suggest that CT perfusion is a new method which could reflect tumor angiogenesis, histological malignant grade, invasion, and clinical stage. It will help us determine the tumor staging and prognosis.

8.Clinical application of pocket-sized ultrasound localization on radial artery cannulation

Objective To assess the clinical application effect of pocket-sized ultrasound-guided radial artery cannulation.Methods Two hundreds and ten patients undergoing elective surgery,146 males and 64 females,aged 18-70 years,ASA physical status Ⅰ-Ⅲ,were scheduled for radial arterial catheterization.The patients were randomly divided into three groups (n =70 each):control group (group C),pockevsized ultrasound group (group U1) and conventional portable ultrasound group (group U2).In group C,the radial artery catheter placement were performed by traditional palpation method.In group U1,the radial artery cannulation were achieved by GE Vsacn pocket-sized ultrasound guidance.In groups U2,the ultrasound-guided radial artery catheterization were completed by conventional portable ultrasound.Before radial arterial cannulation,the diameter of radial artery and the distance between skin to artery in ultrasound images were measured and recorded.The first attempt success rate,cases that punctured less than 3 times,the number of attempts,successful cannulation time and potential complications were recorded.Results There was no statistic significant difference in the hemodynamic parameters,the diameter and the depth of radial artery among three groups.Compared with group C,patients in the other groups had higher first attempt success rate and greater proportion of cases that punctured less than 3 times,fewer placement attempts,shorter catheterization time,and lower incidence of the complication of hematoma (P<0.05).There was no significant difference between groups U1 and U2 among these characteristics.Conclusion Ultrasound is a valuable adjunct to radial arterial catheter insertion.The pocket sized ultrasound may be a good alternative option to guide radial artery catheter placement.
9.Efficacy of preoperative transversus abdominis plane block for analgesia after kidney transplantation

Objective To evaluate the efficacy of preoperative transversus abdominis plane block (TAPB)for analgesia after kidney transplantation. Methods Forty American Society of Anesthesiologists physical status ⅡorⅢpatients, aged 18-64 yr, with body mass index of 18-24 kg∕m2, scheduled for e-lective kidney transplantation, were divided into 2 groups(n=20 each)using a random number table:TAPB group and routine analgesia control group(group C). After induction of anesthesia, ultrasound-guided TAPB was performed on the operated side with 0.5% ropivacaine 20 ml in group TAPB. Patient-controlled intravenous analgesia with sufentanil was provided to all the patients after surgery. Tramadol 50 mg was intravenously injected as a rescue analgesic to maintain visual analogue scale score within 24 h after surgery≤3. The consumption of anesthetics during surgery and amount of sufentanil consumed and require-ment for rescue analgesic within 24 h after surgery were recorded. Ramsay sedation scores were recorded at 2, 4, 6, 12 and 24 h after surgery. The occurrence of nausea and vomiting, pruritus and respiratory de-pression within 24 h after surgery were recorded. TAPB-related complications were also recorded in group TAPB. Results Compared with group C, the consumption of remifentanil was significantly reduced during surgery, and the amount of sufentanil consumed within 24 h after surgery was decreased(P<0.05), and no significant change was found in the incidence of Ramsay sedation scores, nausea and vomiting, pruritus or respiratory depression in group TAPB(P>0.05). No patients required rescue analgesic in two groups. TAPB-related complications were not found in group TAPB. Conclusion Preoperative TAPB reduces the perioperative opioid consumption and enhances the efficacy of postoperative analgesia in the patients under-going kidney transplantation.
10.Comparison of analgesic effects of ultrasound-guided continuous iliac fasicia block using different approa-ches after total hip arthroplasty

Objective To compare the analgesic effect of continuous iliac fasicia block with dif-ferent approaches for patients undergoing total hip arthroplasty (THA).Methods Forty patients,21 males and 9 females,ASA physical status Ⅰ or Ⅱ,scheduled for elective hip arthroplasty surgery in Tongji Hospital from June to December 2016 were enrolled and randomly divided into two groups:parallel group (n =20)or vertical group (n =20).Patients received ultrasound-guided continuous iliac fasicia block with a short-axis in-plane technique in the parallel group and a long-axis in-plane tech-nique in the vertical group after the surgery.All patients had a 10 ml bolus of lidocaine 1% via the catheter followed by an infusion of ropivacaine 0.2% at 4 ml/h for 48 h.The ultrasound imaging time,procedure time and the depth of catheter were recorded.Cumulative frequency of patient-con-trolled nerve blockade analgesia (PCNA),ropivacaine consumption,pain scores at rest,the success rate of block,satisfaction scores and postoperative complications were recorded at every six hours during 48 h after the surgery.Results The ultrasound imaging time and procedure time in the vertical group were longer than in the parallel group (P <0.05).The depth of catheter in the vertical group was significantly greater than in the parallel group (P <0.05).The cumulative frequency of PCNA in the vertical group was significantly less than the parallel group (P <0.05 ).The success rate of the lateral femoral cutaneous nerve block in the vertical group was significantly higher than the parallel group at 6,12,18,24,30,36,42,48 h.The cumulative ropivacaine consumption at 24,30,36 h in the vertical group was significantly less compared with the parallel group (P <0.05).The pain inten-sity at rest was less in the vertical group compared with the parallel group at 24,30,36,42,48 h (P<0.05).There were no significant differences in the success rate of the femoral nerve block,satisfac-tion scores and related complications between the two groups.Conclusion Continuous iliac fasicia block with the short-axis in-plane and long-axis in-plane technique can effectively provide the postop-erative analgesic effects for patients undergoing THA.The long-axis in-plane technique in the vertical group with a higher success rate of the lateral femoral cutaneous nerve block can significantly reduce the ropivacaine consumption and pain scores.
