1.Method for QRS complex self-assortment based on mathematical morphology

This paper presents a new method for QRS complex self-assortment based on mathematical morphology, which measures the comparability of QRS complex with weighted Hausdorff distance. Beginning with the traditional Hausdorff distance, this paper introduces the weighted Hausdorff distance of planar object and the detailed algorithm of self-assortment of QRS complex. The evaluation results of this algorithm by MIT-BIHECG database show that this method has high self-assortment capability.
2.Effect of advanced age on sepsis-caused heterogeneity of nicotinic acetylcholine receptors in cytomembrane of skeleton muscle in rats

Objective To evaluate the effect of advanced age on sepsis-caused heterogeneity of nicotinic acetylcholine receptors (nAChR) in the cytomembrane of skeleton muscle in rats.Methods Twenty SPF adult rats (aged 4-5 months,weighing 250-280 g) and 20 aged male Sprague-Dawley rats (aged 18-20 months,weighing 550-600 g),were obtained from the Experimental Animal Centre of Chongqing Medical University.The adult rats were randomly divided into control group (CAd group,n =10) and sepsis group (SAd group,n =10).The aged rats were randomly divided into control group (CAg group,n =10) and sepsis group (SAg group,n =10).Sepsis was produced by cecal ligation and puncture.The specimens of anterior tibial muscle were obtained at 24 h after operation for determination of the expression of neuronal nAChR (α7-nAChR) and fetal nAChR (γ-nAChR) using immunohistochemistry and Western blot.Results The expression of γ-nAChR and α7-nAChR in the cytomembrane of anterior tibial muscle was significantly higher in CAg and SAd groups than in CAd group,and in SAg group than in CAg and SAd groups.Conclusion Advanced age can aggravate sepsis-induced heterogeneity of nAChR in the cytomembrane of skeleton muscle in rats.
3.Gene Chip Screening of Related Genes of Shenqi Compound Recipe in Preventing Diabetic Skeletal Muscle Disease

Objective To investigate the correlation of diabetic skeletal muscle disease with macroangiopathy, and to explore the related genes of Shenqi Compound Recipe (SCR) in preventing and treating diabetic skeletal muscle disease by using gene chip technique, thus to reveal the molecular mechanism. Methods KKAy mice were fed with water containing nitri oxide synthase inhibitor of Nω-nitro-L-arginine methyl ester ( L-NAME) and high fat diet to induce the macroangiopathy complicated with type 2 diabetes. The experimental animals were divided into normal c57BL/GJ group, KKAy group, model group, SCR group (in the dosage of 14.4 g·kg-1·d-1) and rosiglitazone group ( in the dosage of 1.33 mg·kg-1·d-1) , 15 in each group. The medication groups were administered the corresponding agents for 8 consecutive weeks just as the modeling began. During the experiment period, blood glucose was monitored. At the end of the experiment, the abdominal aorta and skeletal muscle of mice were taken out for the observation of morphological changes, and differentially expressed genes of skeletal muscle between SCR group and model group, and between model group and KKAy group were detected by gene chip technique. Results SCR had an effect on relieving the atrophy, edema, fracture, and inflammatory changes in the skeletal muscle. There were 198 genes differentially expressed between model group and KKAy group, including 119 up-regulated genes and 79 down-regulated genes. There were 70 genes differentially expressed between SCR group and model group, including 33 up-regulated genes and 37 down-regulated genes. In the two comparison groups, 7 genes ( Celsr2, Rilpl1, Dlx6as, 2010004M13Rik, Anapc13, Gm6097, Ddx39b) showed reversed differential expression. Conclusion Diabetic skeletal muscle disease is associated with macroangiopathy. SCR has preventive effect on diabetic skeletal muscle lesion, and the mechanism may be related to the regulation of Celsr2, Rilpl1, Dlx6as, 2010004M13Rik, Anapc13, Gm6097, Ddx39b gene expression.
4.cDNA cloning and prokaryotic expression of human pancreatic alpha-amylase

Objective To obtain recombinant human pancreatic alpha-amylase protein (AMY2A). Methods Human pancreatic AMY2A cDNA was synthesized by RT-PCR using total RNA from human pancreatic tissues and a couple of primers designed according to the know sequence of human pancreatic alpha-amy lase gene, then digested with BamH Ⅰ and Kpn Ⅰ and inserted into the prokaryotic expression plasmid pGEX-5T vector. Construct The prokaryotic expression vector pGEX-5T-AMY2A was constructed and transformed into E. coli BL21 cell . Protein expressed under the induction of IPTG. The inclusion bodies were isolated and solubilized with urea and washed denatured and refolded. The fusion protein wes purified by affinity chrom atography with glutathione agarose. Results Sequence and restrction analysis revealed AMY2A gene was cloned in frame into pGEX-5T, SDS-PAGE profile showed a clear protein band with a relative molecular weight of 84000 and western blot indicated that the expressed product specifically reacted to polyclonal anti -human pancreatic AMY2A genes. Conclusion Human pancreatic AMY2A gene was successfully cloned,expressed and purification.
5.Method for molecular diagnosis of hereditary methemoglobinemia

Objective To investigate molecular diagnostic method for hereditarymethemoblobinemia. Methods The cDNA coding sequence of NADH-cytochrome b5 reductase (b5R) from 3 patients with hereditary methemoglobinemia was analyzed by direct sequencing of RT-PCR products and the genomic DNA of b5R gene by PCR-restriction endonuclease digestion or PCR-sequencing. Results The b5R cDNA of patient A was T/C heterozygous at nucleotide 527 and G/A heterozygous at nucleotide 608. The b5R cDNA of patient B was G/A heterozygous at both nucleotide 170 and nucleotide 179. The b5R cDNA of patient C was G/A heterozygous at nucleotide 608 and C/T heterozygous at nucleotide 791. Result of genomic DNA analysis was in agreement with that of cDNA approach. Conclusion The method for molecular diagnosis of hereditary methemoglobinemia was established and 3 novel b5R gene mutations were identified in compound heterozygosity in 3 Chinese patients.
6.Diagnosis and treatment of cerebral schistosomiasis:a report of 166 cases

Objective To discuss the clinical features diagnosis and treatment of cerebral schistosomiasis. Methods A to?tal of 166 patients with cerebral schistosomiasis were treated and their clinical data were collected and analyzed retrospectively. Results In 166 cases of cerebral schistosomiasis the confirmative diagnoses of 156 cases were diagnosed according to the clin?ical manifestation etiology immunology and auxiliary examination CT MRI .In among 74 cases were confirmed by pathologi?cal examination 10 cases were diagnosed through to the diagnostic treatment. Totally 102 patients received the oral medication of praziquantel and they all improved and discharged 14?16 days later 64 patients received the craniotomy and praziquantel medication after the operation and 48 patients significantly improved others did not improve or aggravated. There was no opera?tive mortality. Conclusions Neuroimaging and laboratory tests are valuable in the diagnosis of cerebral schistosomiasis. The praziquantel treatment is selected firstly when the diagnosis was established. However in the case of serious intracranial hyper?tension intractable epilepsy and praziquantel treatment fails the surgical treatment is required.
7.Anti-apoptotic effect of NGF on H9 c2 cardiac myocytes in a hypoxia/reoxygenation injury model

Aim To investigate the anti-apoptotic effect of NGF on H9 c2 cardiac myocytes in a hypoxia / reox-ygenation injury model and its mechanism. Methods The H9 c2 cardiac myocytes were randomly divided into five groups:control group ( C group) , hypoxia/reoxy-genation group ( H/R group) , NGF group ( N group) , NGF+LY294002 group ( N+L group) and LY294002 group( L group) . Each group received the correspond-ing treatment. Cell survival rate was tested by cell counter kit-8 methods. Apoptotic rate was evaluated by propidium iodide ( PI ) staining and flow cytometry (FCM). The levels of Caspase-12, p-Akt/Akt were e-valuated by Western blot. Results The NGF group could significantly protect the H9 c2 cardiac myocytes under the hypoxia / reoxygenation injury with increased cell survival rate. It also decreased the apoptotic per-centage, upregulated the level of p-Akt/Akt and inhib-ited the expression of Caspase-12 . As the specific in-hibitor of PI3k receptor, LY294002 decreased the level of p-Akt. Conclusion NGF has the effect of anti-ap-optosis on H9 c2 cardiac myocytes exposed to hypoxia /reoxygenation injury via PI3k-Akt signal pathway.

9.Preparation of 68Ga-PSMA-617 and its microPET imaging in BGC-823 cell bearing mice

Objective To prepare 68Ga-PSMA-617 and perform its microPET imaging on both normal BALB/c mice and BGC-823 (PSMA expression) tumor bearing mice.Methods 68GaCl3 was eluted from 68Ge-68Ga generator by 0.05 mol/L HCl,then added to the DKFZ-PSMA-617 and heated at 85 ℃ for 5 min.The labeling efficiency and in vitro stability of 68Ga-PSMA-617 in sodium chloride solution and HAS were analyzed by radio-HPLC.Water partition coefficient and plasma protein binding rate were also evaluated.MicroPET imaging was performed in normal female BALB/c mice and human gastric tumor (BGC-823) bearing mice at 60 min post-injection of 68Ga-PSMA-617.18F-FDG was also injected to BGC-823 tumor bearing mice to acquire microPET imaging for contrast.Results The labeling yield of 68Ga-PSMA-617 was 97.9%,and it could be used directly without purification.68Ga-PSMA-617 showed good in vitro stability in sodium chloride solution and 5% HAS,the radiochemical purities were 94.9% and 81.0% respectively at 80 min post-incubation.68Ga-PSMA-617 was water-solubility substance,and it cleared mainly through the kidneys.MicroPET imaging showed that 68Ga-PSMA-617 could be accumulated in tumor (T/NT=2.28),which was better than 18F-FDG.Conclusions Preparation of 68Ga-PSMA-617 is convenient and has a high labeling yield.It can specifically target to PSMA expression tumors and has a promising prospect in clinical application.
10.An analysis of efficacy of intensity-modulated radiotherapy with concurrent chemotherapy for stage T1-2 N1 nasopharyngeal carcinoma

Objective To retrospectively analyze the efficacy and toxicity of intensity?modulated radiotherapy ( IMRT) alone and IMRT with concurrent chemotherapy ( CRT) in the treatment of early?stage nasopharyngeal carcinoma ( NPC) using pairwise group comparison. Methods A total of 98 patients with stage T1?2N1M0 NPC were treated with IMRT alone or CRT from 2009 to 2010, and 39 pairs out of them were selected for comparison of efficacy and toxicity. The survival rates were calculated using the Kaplan?Meier method and analyzed using the log?rank test. Results The 3?year follow?up rate was 95%. There were no significant differences in the 3?year overall survival ( OS ) , progression?free survival ( PFS ) , local recurrence?free survival ( LRFS ) , and distant metastasis?free survival ( DMFS ) rates between the IMRT alone group and the CRT group ( 97% vs. 95%, P=0?411;97% vs. 92%, P=0?301;97% vs. 97%, P=0?606;100% vs. 92%, P=0?082) . The incidence rates of leucopenia, anemia, and thrombocytopenia were significantly higher in the CRT group than in the IMRT alone group ( P=0?000;P=0?000;P=0?000 ) . There were no significant differences in the incidence rates of grade 3 oral mucositis and hearing loss between the IMRT alone group and the CRT group ( 26% vs. 23%, P= 0?093;41% vs. 62%, P= 0?100 ) . Conclusions CRT fails to increase the OS, PFS, and LRFS rates and reduce the DMFS rate in patients with stage T1?2 N1 NPC. Moreover, CRT results in higher incidence rates of hematotoxicity, grade 3 mucositis, and hearing loss than IMRT alone.
