Academic Journal of Second Military Medical University 1985;0(05):-
Effects of anaesthesia and sinoaortic denervation (SAD) on the hypotensive action of , ketanserin were studied in rats. It was found that the dose-effect curves of kelanserin on blood pressure were the same between SAD and sham-operated rats under anaesthesia. However, in concious free ly moving rats, SAD sharpened the dose-effect curves. It is concluded that the lack of hypotensive effect of ketanserin in conscious normotensive animals is due in large part to the integrity of the arterial baroreflex.

Objective To evaluate the significance of color Doppler EUS in the pancreatic pseudo-cysts drainage. Methods EUS - guided cystoenterostomy was performed in 3 patients. A needle and drainage tube was inserted in a one - step procedure into the cyst through the accessory channel of echoendoscope under EUS. The nasal plastic drainage tube was released immediately after insertion. Results When gastric ulcer and bleeding or the bulge of stomach were not found endoscopically in all patients . The needle with its stylet inserted through the gastric wall into the cyst at the site with minimal distance between them . The mean size of the pseudocysts was approximately 8 cm located at pancreatic tail ,body or the junction of them. In 3 cases but one, the blood flow signal around the cyst was not found . No significant echo of laminated structure between gastric and cyst wall was found . After aspiration or installation of naso-cystic drainage catheter, abdominal pain remitted at 2 days, 2 days and 3 days; the sonography demonstrated that cyst size decreased began from 3rd day ,and the cyst obliterated at 14 and 30 days after drainage. The only complication was found in one patient with gastric mucosal ooze, which recovered after noradrenaline spray . Infection or perforation never occurred. No recurrence of pseudocysts observed at a mean follow - up period of 3. 6 months (range 2-7 months). Conclusions The EUS-guided drainage of pancreatic pseudocysts in one-step is a rather simple and practical procedure with minimal risk of hemorrhage and infection.
8. The effect of TET1 on the proliferation of renal cancer 786-O cells and its related mechanism
Tumor 2012;32(12):962-968
Objective: To silence the expression of TET 1 (ten-eleven translocation 1) gene by siRNA (small interfering RNA) and to investigate its impact on the proliferation of human renal cancer 786-O cells and explore its possible mechanism. Methods: The TET1-siRNA targeting human TET 1 gene was transfected into 786-O cells by LipofectAMINE 2000.Three groups were designed as blank control, negative control and siRNA interference (TET1-siRNA) in this study. The expression levels of TET1, SUZ12 (suppressor of zeste 12), EZH2 (enhancer of zeste homolog 2) and EED (embryonic ectoderm development) mRNAs and proteins after transfection with TET1-siRNA were determined by real-time fluorescence quantitative-PCR and Western blotting, respectively. The MTT method, colony formation assay and flow cytometry were performed to detect the proliferation ability, cell colony formation rate and cell cycle distribution of 786-O cells, respectively. Results: As compared with the blank control, the expression levels of TET1 mRNA and protein in 786-O cells were reduced by (85.13±0.03)% and (56.41±0.09)% respectively after transfection with TET1-siRNA (P < 0.05). The proliferation of 786-O cells was inhibited and the ability of colony-formation was also weakened. The cell cycle of 786-O cells was arrested at G0/G1 phase (P < 0.05). The expression levels of SUZ12 mRNA and protein were reduced significantly (P < 0.05), meanwhile the expression levels of EED and SUZ12 proteins had no change (P > 0.05). Conclusion: TET1-siRNA can down-regulate the expression levels of TET1 mRNA and protein in human renal cancer 786-O cells, suppress the proliferation of 786-O cells, and block the cells at G1 phase. This mechanism may be associated with the recruitment of PRC2 (polycomb repressive complex 2) influenced by TET1 which regulates the expression of SUZ12. Copyright © 2012 by TUMOR.

Objective To investigate the protective effect of sodium tanshinone ⅡA sulfonate(STS) on adriamycin-induced H9c2 cell apoptosis. Methods H9c2 cells were divided into four groups: the control group, the ADR group, the STS group, and the AS(ADR+STS) group. MTT assay were used to detect cell proliferation and cell death. The cell cycle of H9c2 cells were determined by flow cytometry assay. Protein expression of Bcl-2, Bax and Caspase-3 was detected by Western blot assay. Results Compared with the control group, cell proliferation was significantly reduced , and the number of the cells was significantly increased in the G0/G1 phase in the ADR group(P < 0.01, P < 0.05). At the same time, the expression of Caspase-3 was dramatically enhanced and the ratio of Bcl-2/Bax was significantly reduced (P < 0.05, P < 0.01) in the ADR group. However, pretreatment of STS increased H9c2 cell proliferation, decreased the number of the arresting cells, inhibited the expression of Caspase-3, and improved the ratio of Bcl-2/Bax (P < 0.05, respectively). Conclusions STS can attenuate ADR-induced myocardial cell apoptosis.

[ABSTRACT]OBJECTIVETo investigate the expressions of Endoglin (CD105) and Galectin-3 protein in laryngeal squamous cell carcinoma (LSCC) as well as the relationship between their expressions and the clinicopathological factors of LSCC.METHODSThe expressions of CD105 and Galectin-3 protein were detected in 76 samples of LSCC and 25 normal laryngeal tissues (NLT) by immunohistochemical staining (S-P).RESULTS 1.The mean of Microvessel density (MVD) value marked by CD105 in LSCC was 10.33±2.29, which was significantly higher than that in NLT (1.20±1.04) (P<0.05). The expression of MVD marked by CD105 (CD105-MVD) was correlated with histological grading, T stage, clinical stage, lymph node metastasis, recurrence and prognosis in LSCC (P<0.05). 2.The positive expression rate of Galectin-3 protein in LSCC was 86.84%, which was significantly higher than that in NLT (36%)(P<0.05). The expression of Galectin-3 was correlated with T stage, clinical stage, lymph node metastasis, recurrence and prognosis in LSCC (P<0.05). 3.There was a positive correlation between CD105 and Galectin-3 protein. 4.Survival analysis indicated that the expressions of CD105 and Galectin-3, histological grading, lymph node metastasis,T stage and recurrence were independent factors for tumor prognosis in LSCC (P<0.05). CONCLUSIONThe expressions of CD105 and Galectin-3 protein have a positive correlation in LSCC. They may play important roles in the tumorigenesis, malignant progression and poor prognosis of LSCC. Combined detection of them may be great value in diagnosis and predicting prognosis of LSCC.
