Objective To study the relationship between expression of matrix metalloproteinases-7 (MMP-7) and clinicopathological characteristics in patients with primary non-smaU cell lung cancer(NSCLC). Methods MMP-7 in 20 normal people and 60 advanced NSCLC patiens were detected with reverse-transcription-polymerase-chain-reaction. Gelatum image analysator analyzed the result. Results The amount of MMP-7 was less in normal people (30.000) than in NSCLC patients(41.231) significantly(P<0.05); the level of MMP-7 was no correlated with gender, age, pathology pattern, tumor size, was inverse correlation with differentiation, and was positive correlation with clinical stages(P <0.05). Conclusion The level of MMP-7 is closely correlated with tissue differentiation and clinical stages of NSCLC, which may serve as a parameter for determining tumor invasion and metastatic.

Objective This study aimed to investigate the incidence and the cause for duodenalbiliary reflux and reflux cholangitis after metallic stent placement in distal common bile duct Methods After percutaneous transhepatic bile duct puncture and biliary outside drainage was performed, 16 cases with malignant distal biliary stricture underwent metallic stent placement in distal common bile duct Before stent placement, the routine laboratory studies including leukocyte, neutrophil percentage and the levels of total bilirubin and direct bilirubin in blood were performed for all patients. Two to five days [ an average of (3.3 ±0. 9) days ] after stent implantation, the above indexes were tested again, and 1 ml of water containing 185 MBq of 99Tcm-DTPA was given orally before extubation, then 99Tcm radioactivity in the bile was detected 2 hours later. For the measurement data obtained from the experiment, t test or Wilcoxon signed rank test was adopted to compare them, and P < 0. 05 was considered to be statistically different Results In 14 cases, radioactivity was successfully detected in the bile 2-5 days after stent implantation. Twelve of them was detected to have radioactivity in the bile 2 hours before extubation with duodenal-biliary reflux. The technetium count in the bile accounted for 1.82% of the total intake dose. There was no radioactivity in the bile in 2 cases. In 14 patients, there were no symptoms of cholangitis such as high fever, chills, increased jaundice, and so on after stent implantation. The mean of white blood cell count was (7.59 t2. 62) × 109/L, and the median of neutrophil percentage was 0. 74. Compared with those before stent implantation, the difference did not reach statistical significance ( t = 0. 423, Z = 1. 036, P > 0. 05 ).After stent implantation, the median of total bilirubin and direct bilirubin were significantly lower, which were 92. 2 and 74. 3 μmol/L. Compared with those before stenting, the difference was statistically significant (Z= -3. 170, -3. 170, P <0.05). Conclusions There is a high incidence of duodenal-biliary reflux after stent implantation in distal common bile duct in the early stage. However, there is no simultaneous cholangitis caused by duodenal-biliary reflux.

[ ABSTRACT] AIM:To study the antiproliferation and proapoptotic effects of zoledronic acid ( ZOL) on human a-cute myeloid leukemia cell line U937.METHODS:The viability of U937 cells was detected by CCK-8 assay. The cell cy-cle of the U937 cells was analyzed by flow cytometry with PI staining.Apoptotic rate was assessed by flow cytometry with Annexin V-PI and Hoechst 33342 staining.Mitochondrial membrane potential was detected by JC-1 assay.Methylcellulose was used to assess colony formation. The protein levels of p21, Bcl-2 and Bax were determined by Western blot.RE-SULTS:ZOL inhibited the viability of U937 cells.ZOL induced S-phase cell cycle arrest in the U937 cells. The results of flow cytometry analysis with Annexin V-PI and Hoechst 33342 staining showed that ZOL also induced apoptosis in a dose-and time-dependent manner.Mitochondrial membrane potential assay was also used to verify the apoptosis. The apoptotic rate was consistent with the reduction of mitochondrial membrane potential.Colony formation assay showed that ZOL signifi-cantly inhibited the colony formation capacity of the U937 cells. This was achieved by the induction of S-phase cell cycle arrest, and up-regulation of Bax and p21, and down-regulation of Bcl-2.CONCLUSION:ZOL inhibits cell proliferation by regulating the expression of cell cycle-related protein, and induces apoptosis via the mitochondrial apoptotic pathway.

Objective To investigate the relationship between cardiac Tei index and plasma amino terminal brain natriuretic peptide precursor (NT- proBNP), glycosylated hemoglobin (HbA1c) in type 2 diabetes mellitus and its value in predicting the occurrence of major adverse cardiovascular events (MACE). Methods The clinical data of 160 patients with type 2 diabetes mellitus from March 2015 to October 2017 were retrospectively analyzed. The patients were divided into MACE group (45 cases) and non-MACE group (115 cases). The clinical indexes were compared between 2 groups, and the relationship between cardiac Tei index and NT-proBNP, HbA1c was analyzed. Logistic regression analysis was used to analyze the risk factors of MACE in patients with type 2 diabetes mellitus. Results The systolic blood pressure and diastolic blood pressure in MACE group were significantly lower than those in non-MACE group: (109.10 ± 14.53) mmHg (1 mmHg = 0.133 kPa) vs. (120.76 ± 18.74) mmHg and (64.29 ± 9.82) mmHg vs. (73.58 ± 12.11) mmHg, the heart rate, Killip grade > Ⅱ rate, cardiac Tei index and NT- proBNP were significantly higher than those in non- MACE group: (77.56 ± 13.36) times/min vs. (71.68 ± 11.39) times/min, 28.89% (13/45) vs. 2.61% (3/115), 0.63 ± 0.12 vs. 0.52 ± 0.08 and (1193.20 ± 145.69) μg/L vs. (823.87 ± 89.22) μg/L, and there were statistical difference (P<0.01). Pearson regression analysis result showed that cardiac Tei index was positively correlated with NT- proBNP and HbA1c (r = 0.817 and 0.793, P < 0.05), and there was positive correlation between NT-proBNP and HbA1c (r = 0.649, P < 0.05). Logistic regression analysis result showed that cardiac Tei index, NT-proBNP and HbA1c were the independent risk factors of MACE in patients with type 2 diabetes mellitus (OR = 1.015, 1.041 and 1.027; 95% CI 1.005 to 0.213, 1.012 to 7.036 and 1.002 to 4.222; P<0.01 or <0.05). Conclusions The cardiac Tei index has positive correlation with NT- proBNP and HbA1c in patients with type 2 diabetes mellitus, and both are risk factors for the occurrence of MACE. The combination of the 3 indexes can effectively predict the occurrence of MACE and assess the prognosis.
