Primary liver cancer is one of the common malignant tumors.However, the efficacy of traditional treatments is not satisfactory.RNA interference is a new treatment method, which develops rapidly in recent years.It has been found that RNA interference can exert the anti-tumor effect through inhibiting the growth and proliferation of hepatocarcinoma cells, promoting apoptosis, inhibiting the invasion and metastasis of hepatocarcinoma cells, inhibiting angiogenesis and overcoming the resistance to chemotherapy.

3.Expression and significance of long non-coding RNA MALAT1 and interleukin 6 in premature infants with bronchopulmonary dysplasia

Objective:To investigate the expression of long non-coding RNA MALAT1, interleukin 6(IL-6) and apoptosis induced factor(AIF) in peripheral venous blood of premature infants with bronchopulmonary dysplasia (BPD) and its clinical significance.Methods:Preterm infants admitted to the Department of Neonatology, Shanghai Children′s Hospital from January 2015 to December 2016 were enrolled.The selection criteria included gestational age (GA) ≥28 weeks and ≤32 weeks, and birth weight (BW) < 1 500 g. According to the diagnosis, they were divided into BPD group (20 cases) and control group (20 cases). The clinical data of the two groups of premature infants were collected and analyzed, and the levels of MALAT1, IL-6 and AIF in the blood of 40 premature infants were detected by real-time polymerase chain reaction (RT-PCR). T test was used to compare gestational age, birth weight, MALAT1, IL-6 and AIF between the two groups. Results:(1)There was no significant differences in sex ( χ2=1.76), gestational age ( t= 0.17) and birth weight ( t=1.25) of premature infants in BPD group, compared with the control group (all P >0.05). (2)Compared with the control group, the expression of MALAT1 in the peripheral blood of premature infants in BPD group were significantly increased (0.273 4±0.067 3 vs. 0.375 5±0.081 9, P<0.05). (3)Compared with the control group, the expression of IL-6 in the peripheral blood of premature infants in BPD group were obviously decreased (1.448 8±0.191 8 vs.4.444 6±0.165 7, P<0.05). (4)Compared with the control group, the expression of AIF in the peripheral blood of premature infants in BPD group were remarkably decreased(0.006 8±0.002 0 vs.0.004 5±0.001 9, P<0.05). Conclusions:MALAT1 and IL-6 levels of long non-coding RNA in BPD and non-BPD preterm infants are different, which may be related to the incidence of BPD.IL-6 may be a predictor of BPD, and MALAT1 may protect premature infants with BPD.
4.Correction of secondary unilateral cleft lip nasal deformity by anatomical restoration and cartilage of nasal septum

Objective To explore the clinical methodology and effects of correction of secondary unilateral cleft lip nasaldeformity by anatomical restoration and cartilage of nasal septum.Methods With the Goodman's incision in the columella and nasal vestibule,we cut out the cartilage of nasal septum and built a new support structure of tip to reshape the nasal shape after the adequate anatomical restoration under the magnifier of 2.5 times.Results The operation corrected alanasi collapse skewed columella and nostrils symmetry,etc.21 cases were treated and followed up for a month and fifteen months,showing stablized effect with columella centerno alanasi collapse,symmetric nostril and no recurrence.Conclusions On the base of nasal anatomy,a good therapeutic effect has been archeived by the adequate anatomical restoration,cutting out the cartilage of nasal septum and builting a new support structure of tip to reshape the nasal shape.
5.Accurate Detemination of Isotopic Abundance of Intracellular Metabolites of Saccharopolysporaerythraea Based on Ultra Performance Liquid Chromatography-Triple Quadrupole Mass Spectrometry

A method for measuring 13C isotopic abundance of intracellular metabolites of Saccharopolysporaerythraea by ultra-high performance liquid chromatography (UPLC)-triple quadrupole mass spectrometry was established.First, the chromatographic conditions of UPLC were optimized, and then the MS conditions such as unique tube lens voltage, collision energy, and ion pair were optimized.On the bases of length of the parent and daughter ions carbon chains and whether the daughter ions contain 13C atoms, the one-to-one method, one-to-many method and SIM method were established for measuring 13C isotopic abundance.Then these methods were used to measure naturally labeled intracellular metabolite standards and 13C labeled samples, and according to the gap between the experimental value and the theoretical value, the best method was established for each metabolite of different characteristics.The results showed that one-to-one method was most effective for measuring the metabolites of daughter ions not containing 13C atoms represented by sugar phosphates, one-to-many method was the best for measuring the metabolites of both parent and daughter ions containing 13C short carbon chains represented by carboxylic acids, SIM method could play a role in measuring the metabolites of both parent and daughter ions containing 13C long carbon chains represented by coenzyme A.This method had a good measurement precision and could be applied to the measurement of Saccharopolysporaerythraea intracellular metabolites, which contributed to the consequent study of metabolic mechanism and the efficient expression of erythromycin.
6.Relationship between amount of literacy and behavior problems in primary pupils

Objective:To know the amount of literacy and scores of behavior problems of the grade 2-5 pupils in the primary school,and explore the correlation between them.Methods:A total of 673 pupils from grade 2 to 5 in primary school of Nanhai distric,Foshan City were enrolled.Their amount of literacy was assessed with the Primary School Literacy Assessment Scale,and their teachers were assessed with the Conners Teacher Rating Scale.Results:The amount of literacy in different grades,genders and parents' education levels were significantly different among the primary school students (P <0.05),girls's scores were higher than boys's [(2312 ±719) vs.(2184 ±734),P < 0.05],students whose parents of high education level were higher than those of low education level (P < 0.05).Scores of hyperactivity,inattentive-passive behaviors and conduct problems in different grades and genders were significantly different.Scores of Grade 2 pupils were higher than others,and boys's scores were higher than girls' s (P < 0.05).Three factors of CTRS,including conduct problems (r =-0.31),hyperactivity (r =-0.43) and inattentive-passive behaviors (r =-0.36) and hyperactivity index(r =-0.38) had significantly negative correlation with the amount of literacy (P < 0.001).Regression analysis showed that hyperactivity (β =-22.27,P < 0.01) and conduct problems (β =-17.69,P < 0.01) could significantly explain the amount of literacy (R2 =0.81).Conclusion:It suggests that hyperactivity and conduct problems are moderately associated with the amount of literacy in primary pupils.
7. Effect of hyperoxia exposure on expression of microRNA-125b and tumor necrosis factor alpha, interleukin-6 in lung tissues of premature newborn rats

Objective:
To observe the expression of microRNA-125b (miR-125), tumor necrosis factor alpha (TNF-α) and interleukin-6(IL-6) in premature rats exposed to hyperoxia.
Methods:
Eighty 1-day old Sprague Dawley (SD) rats were randomly divided into an air group and a hyperoxia group.The rats in the hyperoxia group were continuously exposed to oxygen chamber for 1-3 L/min, oxygen volume fraction was maintained at (800±50) mL/L, and the rats in air group were placed in the same room with the oxygen volume fraction at 210 mL/L.The feeding conditions were same in 2 groups.Lung tissues of premature rats were extracted at different time (1, 4, 7, 10, 14 days). The pathologic changes in the lung tissues were observed by hematoxylin-eosin (HE) staining.The levels of miR-125b and TNF-α, IL-6 in lung tissues were detected by reverse transcription polymerase chain reaction (qRT-PCT) and enzyme-linked immunosorbent assay (ELISA).
Results:

8.Expression changes of miR -335 in focal cerebral ischemia tissues in rats and their related mechanisms

Objective To investigate the expression changes of miR-335 in rats with focal cerebral ischemia and their mechanisms. Methods Fifty adult healthy male SD rats were randomly divided into sham-operated group, model group, miR-335 transfection group, negative control group and blank plasmid group (n=10). The focal cerebral ischemia rat models in the later 4 groups were constructed by Longa method. After model making, rats in the miR-335 transfection group were injected recombinant plasmid pcDNA3.1-miR-335 and stearamide (SA) liposome mixture into the lateral ventricle, rats in the negative control group were injected pcDNA3.1-negative control and SA liposome mixture into the lateral ventricle, rats in the blank plasmid group were injected pcDNA3.1(+) and SA liposome mixture into the lateral ventricle, and rats in the sham-operated group and model group were injected the same volume of saline. Twenty-four h after ischemia, the neurological deficit of these rats were assessed by Zea-Longa scale. These rats were sacrificed, and the brain infarct volumes were measured by TTC staining; the miR-335 expression in cerebral ischemia tissues of rats was detected by using real-time fluorescence quantification PCR (RTFQ PCR); the protein expressions of cell adhesion molecule 1 (CD31) and vascular endothelial growth factor (VEGF) in cerebral ischemia tissues of rats were detected by Western blotting. Results As compared with those in the sham-operated group, the neurological function scale scores and brain infarct volumes of rats in model group, miR-335 transfection group, negative control group and blank plasmid group were significantly increased (P<0.05); the neurological function scale scores and brain infarct volumes of rats in miR-335 transfection group were significantly decreased as compared with those in the model group (P<0.05). As compared with those in the sham-operated group, the relative miR-335 expression level in the cerebral ischemia tissues of the model group, miR-335 transfection group, negative control group and blank plasmid group were statistically decreased (P<0.05); miR-335 transfection group had significantly increased relative miR-335 expression level as compared with the model group (P<0.05). As compared with those in the sham-operated group, the CD31 and VEGF protein relative expression levels in the cerebral ischemia tissues of the model group, miR-335 transfection group, negative control group and blank plasmid group were significantly decreased (P<0.05); as compared with those in the model group, the CD31 and VEGF protein relative expression levels in the miR-335 transfection group were statistically increased (P<0.05). Conclusion Up-regulation of miR-335 expression might improve cerebral ischemic tissue injury; it might be related to promote the angiogenesis in cerebral ischemia tissues.
9.Expression changes of soluble urokinase-type plasminogen activator receptor and soluble triggering receptors expressed by myeloid cell-1 in serum of children with primary nephrotic syndrome and their clinical signifi-cance

Objective To investigate the expression changes of soluble urokinase-type plasminogen activator receptor(suPAR)and soluble triggering receptors expressed by myeloid cell-1(sTREM-1)in serum of children with primary nephrotic syndrome(PNS)and their clinical significance.Methods A total of 92 cases of newly diag-nosed PNS children were selected in Central Hospital of Yidu Affiliated to Weifang Medical College from June 2014 to September 2016.According to presence or absence of acute tubular necrosis,they were divided into acute renal injury group(27 cases)and non-acute renal injury group(65 cases).According to pathology type,they were divided into mesangial proliferative glomerulonephritis(30 cases),focal segmental glomerulosclerosis(23 cases),membranous ne-phropathy(18 cases),minimal change disease(14 cases)and membrane proliferative glomerulonephritis(7 cases).In the same period,45 healthy children were selected as the healthy control group.The clinical data were collected.The serum levels of suPAR and sTREM-1 were measured by adopting enzyme-linked immunosorbent assay(ELISA). Results The levels of total cholesterol(TC),triglycerides(TG),uric acid(UA),urinary protein/creatinine,24 h urinary protein,urinary N-acetyl-β-glucosaminidase(NAG)and β2-microglobulin(MG)in children with PNS were higher than those in the healthy control group,while serum albumin(ALB)was lower than that in the healthy con-trol group,and the differences were statistically significant(all P<0.05).The serum levels of suPAR and sTREM-1 in PNS patients were(133.09 ± 62.48)ng/L and(79.29 ± 34.68),respectively,which were significantly higher than those in the healthy control group[(31.11 ± 11.61)ng/L and(25.08 ± 8.10)ng/L](t=51.714,49.435;all P=0.000).The serum levels of suPAR and sTREM-1 in acute renal injury group were(188.82 ± 32.21)ng/L and (109.11 ± 24.78)ng/L,respectively,which were significantly higher than those in non -acute renal injury group [(75.96 ± 28.69)ng/L and(52.23 ± 14.07)ng/L]and healthy control group[(31.11 ± 11.61)ng/L and (25.08 ± 8.10)ng/L](F=16 739.607,10 487.256,all P=0.000).The serum levels of suPAR and sTREM-1 in children with focal segmental glomerulosclerosis and membrane proliferative glomerulonephritis were higher than those with minimal change disease,membranous nephropathy and mesangial proliferative glomerulonephritis,and the differences were statistically significant(all P<0.05).Pearson correlation analysis results showed that the serum levels of suPAR and sTREM -1 were positively correlated with TC,TG,urinary protein/creatinine,24 h urinary protein, urinary NAG and β2-MG(all P <0.05),while negatively correlated with ALB(P <0.05). Conclusions The serum levels of suPAR and sTREM-1 are elevated in children with PNS,and which are related with acute renal injury and pathological type,which can reflect the degree of renal tubular disease and kidney function to a certain extent.
10.Establishment and application of intelligent evaluation system for scientific research performance

Objective The study built a scientific performance assessing model by using information technology to construct an intelligent reviewing system,in order to provide intelligent and normalized evaluation services for scientific research performance.Methods A comprehensive and novel practical scientific performance reviewing system was established to provide objective evaluation of scientific output,which was based on accumulated experience in Fuwai Hospital and ample discussion by the experts of academic committee.Results Scientific performance assessment service,which carried out by the collaboration of scientific research management department and informational technology center,were including senior professional title self-assessing system,annual scientific research performance assessing system on both the department basis and individual basis.All the service received satisfactory effect making the assessing service from manual to intelligent.It helps the assessment result turn into openness,fairness and justice.The establishment of the intelligent scientific performance assessing system broaden the mind and provide the basis for the further scientific reward self-evaluation,scientific project application conditions filtering,technology professionals intelligent screening,supervising qualification self-evaluation and scientific project self-reviewing etc.Conclusions The establishment of scientific performance reviewing system effectively avoided the subjective influence,provided timely,highly efficient and accurate evaluating service and comparative analysis,enabling the assessment result much more objective and fair.It merits the promotional value.
