1.Advances in the study of tumor suppressor genes in gliomas

Glioma is the most malignant tumor in CNS, the effect of present therapy is poor. A important discovery in tumor molecular biology is that the activation of oncogene and inactivation of tumor suppressor gene play an important role in the mechanism of tumorgenesis. The advances in the study of tumor suppressor genes in gliomas were reviewed.
2.Bibliometric analysis of hot issues of pediatrics from 2012 to 2016

Objective To conduct a bibliometric analysis of pediatric literature in recent 5 years in Pubmed database,and to explore the focus issues of pediatrics.Methods Bibliographies from research literature of pediatrics in PubMed database from 2012 through 2016 were retrieved.The publication years,journals,the prolific authors and frequency of Mesh major topics were counted.MeSH heading/subheading matrix were formed.SPSS 19.0 statistical software was applied for clustering analysis.Results A total of 9375 articles were included.There were 45 MeSH heading/subheadings,which were clustered into 4 categories.Conclusion The research focuses in pediatrics are as follows:the clinical competence training and education research;the related pediatric issues from the perspective of social medicine;medical error prevention and the related legislation;the situation of Chinese pediatricians and doctor-patient relationship.
3.Fugene 6——a new approach to induce the gene transfection in high efficiency for eukaryotic cells in vitro

Objective To develop a new method to induce the gene transfection in high efficiency for eukaryotic cells in vitro. Methods Four kinds of p14ARF gene primarily deleted human carcinoma cell line including H460,A549,U251,and PC-3 were transfected with the human p14ARF expression vector (pCI-neo-p14ARF) by using the new nonliposomal transfection reagent Fugene 6. The efficiency of gene transfer was determined by screening the cells in G418. Results After 21 days' selection, G418-resistant clones were shown in all the transfected plate. PCR product of p14ARF gene was positive in all the G418-resistant clones. Cytotoxicity of Fugene 6 was detected. The cell proliferation activity was not affected when it was cultured in a high dose of Fugene 6. Conclusion These results demonstrate that Fugene 6 is a rapid, feasible, reproducible, and noncytotoxic gene transfection approach for eukaryotic expression vector in vitro.
4.Percutaneous puncturing drainage for the treatment of liver abscess: analysis of its curative effect and influence factors

Objective To evaluate the curative effect of percutaneous puncturing drainage in treating liver abscess,to analyze the factors affecting curative effect,and to discuss the methods ior reducing mortality and complication rate as well as for shortening hospitalization time.Methods Clinical data of 121 patients with liver abscess,who were admitted to authors' hospital during the period from January 2011 to January 2016,were retrospectively analyzed.For the patients with confirmed liver abscess,adequate antiinfective therapy was adopted,at the same time CT scan was performed to evaluate the liquefaction of lesion,and under CT guidance percutaneous puncturing drainage was carried out.The mortality,complication rate,hospitalization time and the factors affecting curative effect were analyzed.Results A total of 121 patients with liver abscess were enrolled in this study.Two patients died after percutaneous puncturing drainage,the mortality was 1.6%.The factors affecting mortality included old age,underlying disease,the diameter and solid components of abscess.Two patients developed peripheral hepatic abscess and abdominal wall abscess,the complication rate was 1.6%,and clinical cure was achieved after active treatment in these two patients.The main factor affecting complication rate was inappropriate surgical manipulation.Clinical cure was achieved in all 119 patients,with a cure rate of 98.3%,and the average hospitalization time was (15.1±6.0)days.The risk factors that affected hospitalization time included the number of abscess X6 (r=0.232,P=0.021),abscess size X7 (r=0.26,P=0.005) and white blood cell count X8 (r=0.238,P=0.009).Multiple linear regression equation analysis indicated that statistically significant correlation existed between the above influence factors and hospitalization time (P<0.05).The multiple regression equation was as follows:Y=-3.438+3.055X6+0.527X7+0.297X8,F=5.819,R2=0.416.No statistically significant correlation existed between the hospitalization time and other factors,including gender,age,diabetes mellitus,pathogenic bacteria and location of abscess (P>0.05).Conclusion Percutaneous puncturing drainage is an effective treatment for liver abscess,it carries lower mortality and lower complication rate,and its hospitalization time is short.(J Intervent Radiol,2017,
5.Progress on GSK-3βparticipating in neuropsychiatric disorders

Glycogen synthase kinase-3β( GSK-3β) is a serine-threonine kinase,which was originally identified as a key regula-tor of glycogen metabolism. In nervous system, GSK-3β partici-pates in many central and peripheral nervous system physiological processes through transcription,cell proliferation and differentia-tion. The abnormal activity of GSK-3βinduces downstream signa-ling molecules and gene expression abnormalities,which are in-volved in some neuropsychiatric disorders such as schizophrenia, bipolar disorder,Alzheimer's disease,and pain. In recent years, lots of basic and clinic researches focus on GSK-3β,which might gradually become critical theraptic target of many diseases. The article mainly reviews the mechanisms involved in abnormal acti-vation of GSK-3β related neuropsychiatric disorders.
6.Determination of Six Isomers of Silibin in Silymarin Capsules by HPLC

Objective To establish a method for the determination of 6 isomers of silibin in silymarin Capsules.Method The RP-HPLC method was applied.Separation and determination of silibinin was performed on a C18 column(125 mm? 4.0 mm) with the flow rate of 0.8 mL/min,temperature at 25 ℃ and detection wavelength at 288 nm.The mixed solvent of A(85 % phosphoric acid ∶ methnol ∶ water=5 ∶ 35 ∶ 65) and B(85 % phosphoric acid ∶ methnol ∶ water=5 ∶ 50 ∶ 50) served as the mobile phase(gradient elution).Results A good linearity of silibinin was in the rang of 0.508 8~ 2.544 0 ? g(r=0.999 8).The average recovery was 99.5 % and RSD was 2.2 %(n=5).Conclusion With silibinin as as the control,this method is simple and accurate and can be used for the determination of six isomers of silibin and can be used for quality control of Silymarin Capsules.
7.HO-1 mRNA expression in brain of rats with experimental allergic encephalomyelitis

Objective:To investigate the role of heme oxygenase-1(HO-1) in molecular mechanism of experimental allergic encephalomyelitis (EAE). Methods: Seventy-eight healthy female Wistar rats were randomly divided into 3 groups(n=26): rats in control group received no treatment; rats in EAE group were induced with complete Freund's adjuvant and Guinea pig spinal cord homogenate(CFA-GPSCH); and rats in pyrrolidine-dithiocharbamate (PDTC) group received PDTC (100 mg/kg), a specific inhibitor of NF-kB, 1 h before and after(once a day for 7 d) CFA-GPSCH treatment. HO-1 mRNA expression were analyzed with reverse transcription polymerase chain reaction(RT-PCR) on 1 d,7 d,14 d,and 21 d after EAE induction, respectively. The relationship between HO-1 and symptoms of EAE was also investigated. Results: The expression of HO-1 mRNA was very low in the brains of the control group (0.27?0.05), whereas enhanced gradually with onset and development of EAE in EAE group, peaked on d 7 (1.11?0.12), kept at a high level till d 14(1.06?0.18) and decreased on d 21 (0.37?0.1). HO-1 mRNA expression change was in parallel with severities of EAE. In PDTC group,the EAE symptoms were mitigated markedly and the expression of HO-1 mRNA reduced noticeably compared with that of EAE group. Conclusion: Brain HO-1 mRNA expression may play an important role in the pathogenesis of EAE,and application of some inhibitors of NF-kB may be one of the potential therapies for prevention and treatment of EAE.
8.Effect of cold underwear on acute radioactive dermatitis in cervical cancer patients undergoing radiotherapy

Objective To investigate the effect of cold underwear on acute radioactive dermatitis in the cervical cancer patients undergoing radiotherapy.Methods Ninety-seven patients with stage Ⅱb and above Ⅱb cervical cancer receiving radiotherapy were divided into the control group (n =48) and the experiment group (n =49) according to the random digit table.In the control group,Orgotein was sprayed on the local skins and the experiment group was treated with wearing cold underwear for 20 minutes in addition to local spraying of Orgotein.The two groups were compared in terms of dermatitis on the early stage,middle stage and final stage.Result On the early stage there was no statistical significant difference between two groups on dermatitis (P>0.05),but the dermatitis in the experiment group was statistically less than that in the control group at the middle stage and final stage (P<0.01).Conclusions The cold underwear for the cervical cancer patients undergoing radiotherapy can effectively prevent dermatitis or reduce its severity.It is designed suitable for patients prom anatomical perspective and simple for application.
10.Effect of energized fusion protein anti-CD2o (Fab)-LDM on proliferation and DNA damage of human lymphoma cell line BJAB

Objective To investigate the growth inhibition and DNA damage of energized fusion protein anti-CD20(Fab)-LDM on B JAB cells in vitro.Methods The binding activity of fusion protein anti-CD20 (Fab)-LDP to B JAB cells was studied by flow cytometry and confocal laser scanning microscopy.MTT assay was used to study the energized fusion protein anti-CD20(Fab)-LDM on cell growth of B JAB cells.Comet assay was employed to detect DNA damage in B JAB cells.The cell growth cycle of BJAB was analyzed by FACS.Results The recombinant fusion protein anti-CD20 (Fab)-LDP possessed an significant target affinity towarded BJAB cells.The energized fusion protein anti-CD20(Fab)-LDM showed obvious inhibition on proliferation,as well as induced potent DNA damage in B JAB cells in vitro compared with lidamycin.B JAB cells treated with energized fusion protein anti-CD20 (Fab)-LDM showed S phase cell cycle.Conclusion The energized fusion protein anti-CD20 (Fab)-LDM could target binding to BJAB cells and significantly inhibit the proliferation of B JAB cells by inducing DNA damage and S phase arrest.
