1.Optimization of Water Extraction Technology of Medicinal Material in Lian-wei Anacyclus Formulation

OBJECTIVE:To optimize the water extraction process of medicine in lian-wei anacyclus prescription. METHODS:Composite score of polysaccharide content and raw polysaccharide were set as investigation index,the different adding water amount,extracting times and extraction duration were set as investigation factors,and L9(34)orthogonal test were conducted to op-timize the water extraction technology of medicine in lian-wei anacyclus prescription and verification test was carried out. RE-SULTS:Extracting times had the most significant effect on the investigation index;the most optimized process was as follows as 12 times of adding water amount,2.0 h of extracting times,3 times of extraction. The average content of polysaccharide was 36.34μg/ml(RSD=0.24%,n=3)in the verification test,and the average content of raw polysaccharide was 10.18%(RSD=0.65%, n=3). CONCLUSIONS:The optimized technology is reasonable,feasible and reproducible,and can provide reference for the dos-age form transformation.
2.Investigation of the Use of Antibiotics in Three Two-Level Hospitals in Urumqi City in 2006

Objective:To investigate the state of the use of antibacterial drugs in three two-level hospitals in Uru- mqi city.Method:According to information provided by the system,the statistics of antibacterial drugs amount purchased in three Urumqi hospitals,DDDs and daily cost were retrospectively analyzed.Result:The amount of the antibacterial pur- chases decreased compared with that in 2005.The antibiotics accounted for 16.14 in the Construction Hospital,25.68 in the Children's Hospital and 33.99 in the Coal Hospital.Cephalosporins accounted for more than 30% of the amount of the purchase of antibacterial drugs in every hospital.Conclusion:The application of antibacterial drug proportion was basically reasonable.Cephalosporins were still the most widely used antibacterial drugs.Through further standardization of clinical application of antibacterial drugs,their share will continue to decline hut the proportion of the use of antimicrobials impor- ted and produced in joint ventures will grow.
3.Qtracker-labeled rabbit osteoblasts in vitro

Objective To explore the feasibility of labeling in vitro rabbit osteoblasts with Qtracker and the features of Qtracker-labeled rabbit osteoblasts. Methods A healthy male rabbit, 3 months old, weighing 2 kg, was used in this study. After bone marrow was aspirated, bone marrow stromal cells (BMSCs) were isolated and cultured using the adherence method in vitro. The third passage of BMSCs was induced into osteablasts before incubation with Qtracker at concentrations of 1, 2, 4, 8, 16, 32 nmol/10~6 cells (Groups A, B, C, D, E, F respectively). Cells not labeled by Qtracker served as negative control (Group G). The following parameters were measured: induction, differentiation and determination of rabbit osteoblasts; the optimal mass concentration of Qtracker labeling by fluorescence microscopy and flow cytometry; the cell sur-vival rates at various concentrations of Qtraeker labeling by trypan-blue exclusion; Qtracker-labeled cell pro-liferation by MTr. Results The primary and the passage rabbit BMSCs were chiefly of fusiform shape. Rabbit BMSCs differentiated into osteoblasts following induction. The osteoblasts cytoplasm showed green fluorescence under fluorescence microscopy after being labeled by Qtracker. The mean labeling rate increased with the increased concentration of Qtracker, reaching up to (93.58±2.08) % after incubation at 8 nmol/ 10~6 cells by fluorescence microscopy, and (95.24±1.31) % by flow cytometry. There were no significant differences between Groups D, E, F(P>0.05), but significant differences were found between Groups A, B, C and Groups D, E, F (P<0.05). The labeling rate for Group G was 0. The cell survival rates were all above 96% (P>0.05) . No significant differences were found in the cell proliferation among various con-centrations (P>0.05). Conclusions Qtraeker can be used as a labeling marker for rabbit osteoblasts. When the concentration is at 8 nmol/10~6 cells, optimal labeling effect can be achieved. Rabbit osteoblasts labeled with Qtracker are of high efficiency and safety.
4.Alterations of Whole Brain Networks Degree Centrality in Patients with Primary Insomnia after Acupuncture Therapy:A Voxel-based Resting-state fMRI Research

[Objective]To investigate the value of degree centrality(DC),a novel resting-state fMRI parameter,in voxel-wise whole-brain functional networks analysis in primary insomnia(PI)after acupuncture therapy.[Methods]The resting state fMRI were performed in 29 PI patients and 22 age,education,and sex-matched normal healthy subjects. Analysis of DC map changes between the two patient groups and the control group were performed by two sample t test.(threshold at P<0.05).[Results]Compared with the control group,patients with PI showed significantly reduced DC value in middle temporal gyrus(MTG. R);hippocampus(HIP. B);parahippocampal gyrus(PHG. R);putamen(PUT. R);cuneus(CUN. L).[Conclusions]Changes of DC value occurred in some region of brain in the PI patient groups when compared with the control group. It was indicated that DC ,as a novel resting-state fMRI parameter in the voxel-wise whole-brain functional networks ,might be an appealing alternative approach for further study on pathologic and neuropsychological states of PI.
5.Clinical study on the treatment of Helicobacter pylori positive chronic superficial gastritis with modified Liqi-Hewei Decoction combined with conventional western medicine therapy

Objective:To evaluate the clinical efficacy of modified Liqi-Hewei Decoction combined with conventional western medicine therapy in the treatment of Hp-positive chronic superficial gastritis (CSG). Methods:A total of 96 patients with Hp-positive CSG in the First People’s Hospital of Lianyungang and Changshu No.1 People’s Hospital who met the inclusion criteria between January 2017 and January 2019 were divided into two groups according to the random number table method, with 48 in each group. The control group was treated with conventional western medicine, and study group was additionally given modified Liqi-Hewei Decoction on the basis of control group. Both groups were treated for 8 weeks and followed up for 12 weeks. The TCM symptom score, gastric mucosa and pathological grading were scored before and after treatment. Levels of motilin, gastrin and somatostatin were detected by immunoturbidimetry, and levels of CD3 +, CD4 + and CD8 + were detected by automatic flow cytometry and the ratio of CD4 +/CD8 + was calculated. The Hp negative conversion rate was observed, the adverse reactions were recorded, the clinical efficacy was evaluated and the recurrence rate was statistically analyzed. Results:The Hp negative conversion rate was 89.6% (43/48) in study group and was 72.9% (35/48) in control group, where the difference was statistically significant ( χ2=4.376, P=0.036). The total effective rate was 95.8% (46/48) in study group and was 83.3% (40/48) in control group ( χ2=5.031, P=0.025). After treatment, the scores of epigastric pain, upper abdominal fullness, acid reflux, belching and poor appetite in the study group were significantly lower than those in control group ( t=8.919, 3.971, 7.949, 8.171, 9.865, all Ps<0.01). The scores of gastric mucosa and pathological grading were significantly lower than those in control group ( t=13.705, 15.495, all Ps<0.001). After treatment, the levels of gastrin [(126.15 ± 14.36) ng/L vs. (152.38 ± 17.51) ng/L, t=8.025], motilin [(93.59 ± 11.87) ng/L vs. (102.48 ± 14.68) ng/L, t=3.263] and somatostatin [(36.76 ± 8.97) ng/L vs. (40.84 ± 10.68) ng/L, t=2.027] in the study group were significantly lower than those in the control group ( P<0.01 or P<0.05). The levels of CD3 +, CD4 + and CD4 +/CD8 + were significantly higher than those in the control group ( t=6.883, 6.720, 4.306, all Ps<0.001). The recurrence rate was 4.3% (2/46) in the study group and was 17.5% (7/40) in the control group ( χ2=3.950, P=0.046). During treatment, the incidence rate of adverse reactions was 8.3% (4/48) in the study group and was 12.5% (6/48) in the control group ( χ2=0.446, P=0.504). Conclusion:Modified Liqi-Hewei Decoction combined with conventional western medicine therapy can effectively improve the levels of gastrointestinal hormones and clinical symptoms, enhance the immunity and Hp negative conversion rate, and reduce the recurrence rate of patients with Hp-positive CSG.
6.Analysis of predictors of systemic lupus erythematosus with intestinal symptoms as the first manifestation

Objective:To analyze the predictors of systemic lupus erythematosus (SLE) with intestinal symptoms as the first manifestation, and to provide evidence for the diagnosis and differential diagnosis of the disease.Methods:From January 2013 to June 2020, the clinical data of 165 patients diagnosed with SLE and treated at the First Affiliated Hospital of Zhengzhou University were retrospectively analyzed. According to whether the intestinal symptoms were the first manifestations, they were divided into intestinal symptoms as the first manifestations group ( n=50) and intestinal symptoms not as the first manifestations group ( n=115). The baseline data, imaging findings, orgen involvement and laboratory indicators of the two groups were compared and analyzed. Independent sample t test, non-parametric test and chi-square test were used for statistical analysis. Logistic regression analysis was used to establish a prediction model of SLE with intestinal symptoms as the first manifestation. Receiver operating characteristic curve (ROC) and Hosmer-Lemeshow test were used to evaluate the predictive value of the model. From July 2020 to May 2021, the data of 72 SLE patients treated at the First Affiliated Hospital of Zhengzhou University were collected (22 patients with intestinal symptoms as the first manifestation and 50 patients with intestinal symptoms not the first manifestation), and the predictive power of the model was validated. Results:Compared with intestinal symptoms not as the first manifestation group, the proportions of patients with fever, muscle involvement and joint involvement in intestinal symptoms as the first manifestation group were lower, while the proportions of patients with polyserositis, ascites, edema and dilatation or thickening of intestines, hydronephrosis or dilatation of the ureter, kidney involvement, blood system involvement were higher, and the level of complement C3, level of complement C4, absolute lymphocyte value and albumin level were lower (67.8%, 78/115 vs. 32.0%, 16/50; 24.3%, 28/115 vs. 4.0%, 2/50; 68.7%, 79/115 vs. 14.0%, 7/50; 27.8%, 32/115 vs. 86.0%, 43/50; 16.5%, 19/115 vs. 78.0%, 39/50; 13.9%, 16/115 vs. 86.0%, 43/50; 4.3%, 5/115 vs. 62.0%, 31/50; 29.6%, 34/115 vs. 48.0%, 24/50; 30.4%, 35/115 vs. 52.0%, 26/50; 0.76 g/L, 0.43 to 0.97 g/L vs. 0.48 g/L, 0.40 to 0.57 g/L; 0.14 g/L, 0.08 to 0.23 g/L vs. 0.09 g/L, 0.06 to 0.15 g/L; 0.90×10 9/L, 0.51×10 9 to 1.28×10 9/L vs. 0.64×10 9/L, 0.44×10 9 to 1.08×10 9/L; (34.07±7.30) g/L vs. (28.77±5.43) g/L), and the differences were statistically significant ( χ2=18.246, 9.699, 41.776, 47.567, 57.781, 78.833, 67.903, 5.195 and 6.955, Z=-4.053, -3.295 and -2.204, t=-4.606; all P<0.05). The results of multivariate logistic regression analysis showed that low level of complement C3 and low albumin level were risk factors of SLE with intestinal symptoms as the first manifestation (odds ratio 0.136, 95% confidence interval 0.031 to 0.590; odds ratio 0.923, 95% confidence interval 0.871 to 0.977; P=0.008 and 0.006). The established prediction model for SLE with intestinal symptoms as the first manifestation was p=1/(1+ e - Y), in which Y=2.906-1.994×complement C3 (g/L) -0.08×albumin (g/L). The area under the ROC was 0.761 (95% confidence interval 0.687 to 0.834, P<0.01). The result of Hosmer-Lemeshow test showed the model had good calibration ability ( χ2=13.024, P=0.111). The result of validation analysis showed that when p≥0.255 to predict SLE with intestinal symptoms as the first symptoms, the sensitivity of the model was 72.7% (16/22), the specificity was 76.0% (38/50), and the accuracy was 75.0% (54/72). Conclusions:The symptoms of SLE with intestinal symptoms as the first manifestations are obscure and easily misdiagnosed. When the imaging examination of patients with intestinal symptoms as the first manifestations shows edema and dilatation or thickening of intestines, hydronephrosis or dilatation of the ureter, or laboratory examination indicates low level of complement C3 and low albumin level, be wary of the possibility of SLE. Early diagnosis and intervention can greatly improve the prognosis of patients.
7.Genetic analysis of a Chinese pedigree affected with Congenital coagulation factor XII deficiency due to a c.1A>G start codon variant of F12 gene.

OBJECTIVE:
To explore the clinical characteristics and genetic etiology of a consanguineous Chinese pedigree affected with Congenital coagulation factor XII (XII) deficiency.
METHODS:
Members of the pedigree who had visited Ruian People's Hospital on July 12, 2021 were selected as the study subjects. Clinical data of the pedigree were reviewed. Peripheral venous blood samples were taken from the subjects. Blood coagulation index and genetic testing were carried out. Candidate variant was verified by Sanger sequencing and bioinformatic analysis.
RESULTS:
This pedigree has comprised 6 individuals from 3 generations, including the proband, his father, mother, wife, sister and son. The proband was a 51-year-old male with kidney stones. Blood coagulation test showed that his activated partial thromboplastin time (APTT) was significantly prolonged, whilst the FXII activity (FXII:C) and FXII antigen (FXII:Ag) were extremely reduced. The FXII:C and FXII:Ag of proband's father, mother, sister and son have all reduced to about half of the lower limit of reference range. Genetic testing revealed that the proband has harbored homozygous missense variant of c.1A>G (p.Arg2Tyr) of the start codon in exon 1 of the F12 gene. Sanger sequencing confirmed that his father, mother, sister and son were all heterozygous for the variant, whilst his wife was of the wild type. By bioinformatic analysis, the variant has not been included in the HGMD database. Prediction with SIFT online software suggested the variant is harmful. Simulation with Swiss-Pbd Viewer v4.0.1 software suggested that the variant has a great impact on the structure of FXII protein. Based on the Standards and Guidelines for the Interpretation of Sequence Variants: A Joint Consensus Recommendation of the American College of Medical Genetics and Genomics (ACMG), the variant was rated as likely pathogenic.
CONCLUSION
The c.1A>G (p.Arg2Tyr) variant of the F12 gene probably underlay the Congenital FXII deficiency in this pedigree. Above finding has further expanded the spectrum of F12 gene variants and provided a reference for clinical diagnosis and genetic counseling for this pedigree.

8.Study on Improvement Effects of Ethanol Extract of the Root of Anacyclus pyrethrum on Cough Variant Asthma Model Rats

OBJECTIVE: To observe the improvement effects of ethanol extract of the root of Anacyclus pyrethrum (EEAP) on cough variant asthma (CVA) model rats. METHODS: Male SD rats were randomly divided into control group, model group, prednisone acetate group (positive control, 250 mg/kg), EEAP low-dose, medium-dose and high-dose groups (160, 320, 640 mg/kg, by the weight of EEAP), with 10 rats in each group. Except for control group, other group was given 1 mg/mL ovalbumin (OVA)-Freunds adjuvant complete solution subcutaneously, and aerosol inhalation of 1% OVA-normal saline (once a day, 20 min each time, 15 d) to induce CVA. After last inhalation, control group and model group were given constant volume of water intragastrically; administration groups were given relevant medicine intragastrically, once a day, for consecutive 30 d. General symptoms of rats were observed in each group during experiment. The airway sensitivity of rats in each group was investigated by capsaicin cough provocation test, and the cough times were recorded. The contents of SOD and TNF-α in serum were determined by ELISA. The morphological characteristics of lung tissue were observed by HE staining. The number of eosinophils and leucocytes in alveolar lavage fluid was recorded by Rayleigh staining. RESULTS: Rats in the control group breathed smoothly, responded quickly and had glossy coat. The rest of the groups showed restlessness, cough, shortness of breath and other symptoms after antigen stimulation. Compared with control group, the congestion and edema of bronchial wall and infiltration of inflammatory cells in the lung tissue were observed in model group; the cough times increased significantly; serum content of TNF-α, eosinophil and leukocyte counts in alveolar lavage fluid increased significantly, and serum content of SOD decreased significantly (P<0.05). After treatment, above symptoms of rats were alleviated to varying degrees in administration groups, and the cough times were significantly reduced; the serum contents of TNF-α as well as eosinophil and leukocyte counts in alveolar lavage fluid were significantly reduced; the serum contents of SOD was increased significantly, but the cough times of EEAP groups were significantly higher than that of prednisone acetate group (P<0.05). CONCLUSIONS: EEAP may show the anti-inflammatory and antiasthmatic effects by inhibiting the secretion of TNF-α, increasing the content of SOD and inhibiting inflammatory cell infiltration.
9.Added value of T 1-weighted StarVIBE sequence for PET/MR image quality

Objective:To explore the added value of T 1-weighted stack-of-stars volumetric interpolated body examination (StarVIBE) sequence on PET/MR image quality. Methods:A retrospective analysis was performed on 60 patients (42 males, 18 females; age 11-86 (58±12) years) who underwent 18F-FDG PET/MR examination and with positive PET results in Ruijin Hospital, Shanghai Jiao Tong University School of Medicine from April 2020 to April 2021. All patients completed StarVIBE sequence collection, and volumetric interpolated body examination (VIBE) sequence was used as control. StarVIBE and VIBE sequence images were evaluated independently using five-point method by two physicians. The evaluation was carried out from six aspects: lesion display, lesion boundary display, vascular around lesions display, fusion level with PET image, image artifact and overall image quality. Wilcoxon signed rank test was used to compare the image quality of the two sequences, and Kappa test was performed to assess the consistency of the image quality scores between the two physicians. Results:There were 26 cases with cervical lesions, 14 cases with chest lesions, 7 cases with abdomen lesions and 13 cases with pelvic lesions. The scores of lesion display (4.0(3.8, 4.5) vs 3.5(3.0, 4.0)), lesion boundary display (4.0(4.0, 4.0) vs 3.0(3.0, 3.5)), vascular around lesions display (5.0(4.0, 5.0) vs 4.0(3.5, 4.5)), fusion level with PET image (5.0(5.0, 5.0) vs 4.5(4.0, 5.0)), image artifact (4.5(4.0, 5.0) vs 4.5(4.0, 5.0)) and overall image quality (5.0(4.0, 5.0) vs 4.0(4.0, 4.0)) of StarVIBE sequences were better than those of VIBE sequences ( z values: 3.77-6.54, all P<0.001). On the vascular around the lesions display, the scores of StarVIBE were significantly better than those of VIBE sequence in the neck (5.0(4.5, 5.0) vs 3.0(2.7, 3.5); z=4.49, P<0.001) and chest (4.5(4.3, 4.7) vs 4.0(3.6, 4.3); z=3.10, P=0.002). As for image quality, the scores of StarVIBE were also significantly better than those of VIBE in neck (5.0(4.5, 5.0) vs 4.0(3.7, 4.5); z=4.36, P<0.001) and chest (5.0(5.0, 5.0) vs 4.0(4.0, 4.5); z=3.02, P=0.003). In abdominal lesions, the score of StarVIBE was higher than that of VIBE in blood vessels (4.5(3.5, 5.0) vs 4.0(3.5, 4.5); z=2.07, P=0.038), and there was no difference between score of overall image quality (4.0(3.7, 4.5) vs 4.0(3.5, 4.5); z=0.27, P=0.785). The score of overall image quality of pelvic StarVIBE sequence was better than that of VIBE sequence (5.0(4.5, 5.0) vs 4.0(4.0, 4.5); z=2.12, P=0.034). Kappa value of image quality score between two physicians was 0.554, indicating moderate consistency. Conclusion:In whole-body PET/MR imaging, StarVIBE sequence can significantly improve the image quality of cervical, thoracic and pelvic lesions when comparing with VIBE sequence.
10.How Job Stress and Psychological Adaptation Predicting Interpersonal Needs Among Female Migrant Manufacturing Workers in China: A Structural Equation Model

Background:
This study aimed to explore relationships between job stress and psychological adaptation and how they related to interpersonal needs through mood states among female migrant manufacturing workers.
Methods:
A cross-sectional survey was conducted in 16 factories in Shenzhen, China. Sociodemographic, job stress, psychological adaptation and other psychological information of was collected. Structural equation modeling was performed to delineate the internal relationship between variables.
Results:
The hypothetical structural equation model exhibited acceptable model fit among female migrant manufacturing workers (χ2 = 11.635, df = 2, χ2/df = 5.82, p = 0.003, RMSEA = 0.090, CFI = 0.972, SRMR = 0.020). Job stress was directly associated with mood states and interpersonal needs; Psychological adaptation was directly associated with mood states and indirectly associated with interpersonal needs; Bootstrapping tests demonstrated mediation effect of mood states in the way from psychological adaptation to interpersonal needs.
Conclusion
Female migrant manufacturing workers who suffered stress from job and the process of psychological adaptation may have worse mood states and workers with worse mood states are more likely to develop unmet interpersonal needs, a proximal factor of suicidal ideation.
