1.Regulating effect of PGE2 on mechanosensitivity of osteoblasts

Objective To explore the relation between prostaglandin E2(PGE2) and the mechanosensitivity of osteoblasts and to analyze its action mechanism in cellular level .Methods Osteoblasts cell line MC3T3-E1 cells were performed the pre-treatment by using 16 ,16-dimethyl prostaglandin E2(dmPGE2) .Then the regulating effect of PGE2 on intracellular calcium ion signal was ob-served;the activator agent and inhibitor of the protein kinase A (PKA) signal path were adopted to detect the participating effect of the PKA signal path in the regulating process .Results Compared with the blank treatment group ,dmPGE2 pre-treatment signifi-cantly increased the calcium ion signal intensity induced by hypotonic swelling stimulation in MC 3T3-E1 cells .This effect of dmPGE2 was mimicked by 8-bromo-cAMP(8br-cAMP) ,an activator of PKA pathway ;but blocked by PKI ,an inhibitor of PKA pathway .Conclusion dmPGE2 is able to up-regulate the calcium response induced by hypotonic swelling stimulation by activating PKA pathway ,which provides an important cellular mechanism for explaining PGE2′s anabolic effect in bone tissue .
2.RELATIONSHIP BETWEEN BLOOD AND URINE NUTRITIONAL PARAMETERS AND BLOOD PRESSURE AMONG FIVE POPULATION GROUPS IN CHINA

Five population groups, including farmers living in Veining (a cold hilly southern county), farmers and labourers in Shijiazhuang (a northern district), coastal fishermen in Zhoushan and "Yi" nationality farmers in Veining (where the incidence of hypertension is very low in China) were selected randomly (467 male subjects, 40-59 years old) to explore nutritional parameters-BP relationships. The results showed; (1) Pulse rate, QI and BP were lowest in Veining "Han" and "Yi" nationality groups. (2) Values of blood and urine nutritional parameters were in close accord with the diet customs of the different population groups. (3) Urine Na, Ca, Ca/Mg, urea nitrogen and serum cholesterol, total protein, albumin, uric acid were higher, while urine K was lower in hypertensives. (4) Urine Na, Na/K, Ca/ Mg and serum albumin, total protein, cholesterol, triglyceride, uric acid levels correlated positively, while K inversely with BP on multivariate stepwise regression and/or discrimination analyses. These results suggested high Na (Ca)-intake might be hypertensive, while high K(Mg)-intake an-tihypertensive, the serum protein - BP relationship could not be ascribed as a secondary effect of hemoconcentration in hypertension; a high protein intake, regardless of nature, could be hypertensionogenic; hyperlipidemia seemed to be a risk factor of hypertension, not being the secondary consequence to high BP. In designing a strategic protocol for primary prevention of hypertension, the above should be given due consideration.
3.CORRELATION ANALYSIS OF SERUM AMINO ACID AND BLOOD-PRESSURE AMONG 8 CHINESE POPULATIONS

Eight populations including 4 Chinese districts and 4 nationalities 484 subjects (male, 40~59 years) were conducted for exploring amino acid (22 sorts)-BP relationship by multi-variate regression and discrimination analyses. Results showed:(1)The different amino acid levels found among "Ka-zak", "Han" and "Uygur" nationalities living in one of the same Xinjiang region might be a reflection of their diverse dietary patterns; while the difference seen between "Han" and "Yi" nationalities living in one of the same Quizhou area, taking similar diet, might be due to the inherited metabolic influence. (2) The levels of aspartic and (or) glutamic acids were the the highest in the Quizhou "Han" and "Yi" groups living in cold hilly area, while other amino acids were comparatively lower, in keeping with their BP, Quetelet's indices and serum urea levels to be the lowest, tentatively denoting that the tissue metabolic turnover rate might be also lower as well in these groups. (3) The serum levels of sulfur containing amino acids in those populations taking much animal foods, were all higher, therefor ethe lower incidences of cardio-cerebro-vascular diseases seen in fishmen can not be thus wholly ascribed to the high intake of fish meat rich in sulfur containing amino acids. (4) From BP regression equations, valine, cysteine, alanine, threonine were found to be the prohypertensives while glycine, leucine, methionine and phenylalanine,the antihypertensives.
4.Identification of genes related to induced resistance to ceftriaxone in Neisseria gonorrhoeae using suppression subtractive hybridization and DNA microarray

Objective To elucidate the molecular basis for induced resistance of N. gonorrhoeae to ceftriaxone in vitro. Methods The reference strain ATCC49226 and clinical isolate ZSSY00205 of N. gon-orrhoeae were exposed to subinhibitory concentration of ceftriaxone for the induction of resistance. Then,suppression subtractive hybridization was performed with the pre-induction parent strains as drivers and post-induction mutant strains as testers to create a subtractive cDNA library. Following that, a total of 192 clones were randomly selected from the library, and arrayed by spotting onto nylon membranes. Finally, dif-ferentially expressed genes were screened by hybridization with labeled-RsaI restriction fragments from the sensitive and resistant N.gonorrhoeae strains respectively, and analyzed by sequencing and homology research using Blast program. Results A subtractive library for these resistant N.gonorrhoeae strains was generated by SSH technique. Microarray analysis and homology research confirmed 5 genes related to ceftriaxone resistance, i.e. mtrR, mtrC, gyrB, rpsJ and PJD1. Conclusions The induced resistance of N. gonorrhoeae to ceftriaxone may be associated with mtrR, mtrC, gyrB, rpsJ and PJD1 genes which probably mediate the resistance by enhancing the activity of efflux pump system.
5.Onychomycosis in patients with chronic viral hepatitis: an preliminary survey in Guangzhou city

Objective To investigate the incidence, clinical patterns, and species distribution of pathogenic fungi of onychomyeosis in patients with chronic viral hepatitis (CVH), and to analyze the relationship between onychomycosis and CVH. Methods From November 2005 to October 2006, direct microscopy and fungal culture were performed on nail samples from CVH patients with clinically suspected onychomycosis in the two largest institutions for communicable disease control in Guangzhou city. The incidence, clinical patterns, and species distribution of pathogenic fungi of onychomycosis were assessed based on the findings in mycologic examinations. Results The study randomly recruited 995 patients with CVH, and onychomycosis was diagnosed in 116 patients. The incidence of onychomycosis was 11.66% in total, 6.20%, 8.59%, 14.09%, 19.67% in patients with mild, moderate, severe and extremely severe CVH respectively, 7.09%, 17.29%, 19.13% and 27.27% in patients with a clinical course of CVH of 0.5-9 years, 10-19 years, 20-29 years, ≥30 years respectively. The most common clinical pattern was distal and lateral subungual onychomycosis (DLSO, 69.83%), followed by total dystrophic onychomycosis (TDO, 14.66%). Among the pathogenic fungi, dermatophytes amounted to 71.43%, yeasts 21.43%, moulds 7.14%, and Trichophyton rubrum was the most frequently isolated fungus (42.86%). Conclusions The incidence of onychomycosis in patients with CVH is correlated with the severity and course of CVH. Among these patients, the most common clinical pattern is DLSO with the most frequent fungal species being dermatophytes and predominant fungal isolate being Trichophyton rubrum.
6.Inflammatory myofibroblastic tumor of long bone:a clinicopathologic analysis and review of literatures

Purpose To study the clinicopathologic features of inflammatory myofibroblastic tumor (IMT) of long bone.Methods HE and immunohistochemistry of EnVision two-step were used to observe the clinical,radiological,histological and immunophenotype features of IMT of bone.The literatures were reviewed.Results 4 cases of IMT of bone were respectively located in the tibia (2 cases) and femur (2 cases).Histologically,the lesions were characterized by collagen-rich and spindled to plump myofibroblast-like cells and a variable admixture of inflammatory cells.Immunohistochemical study showed that the vimentin,SMA,actin,H-caldesmon and CD34 were positive.Conclusion The IMT is a rare and locally aggressive tumor.The diagnosis should combine the histological characters with immunohistochemical results and should be differentiated from the other tumors and tumor-like lesions.
7.TRACE, ORDINARY ELEMENTS AND HYPERTENSION——Comparative analysis of 4 districts, 6 populations, 3 nationalities

Men aged 40-59 years (325 subjects) from 4 districts, 6 populations, 3 nationalities were investigated to explore the relationship between element and blood pressure (BP) . Fe, Mo, Ni, Pb, Si, Sn, Sr, V, Cd, Co, Cr, Cu, Mn, Zn, Ca, and Mg -were measured by ion coupled plasma spectroscopy, Se by catalytic pulse polarography. Results showed; (1) The element levels were different among districts and nationalities. (2) The correlation coefficients among serum Pb, Ni, Si, Sn, V, Cd, Cr were rather high, while Sr behaved relatively independently. (3)Using multivariant stepwise regression and discrimination analysis, serum Sr was found to associate positively with BP in normotensive and hypertensive and total pooled groups without exce-piton. (4) Serum Ca (Ca/Mg), Fe, Co were positively associated, while Cr, Sn inversely associated with BP. (5) The harmful element Cd was unexpectedly found to be inversely associated with BP and it was lower in those populations with high BP levels, which might be ascribed to the non-parallel distribution of certain elements in body tissues and blood. Results also indicated; (1) The levels of essential element Ni, V, Co, Cr were lower in hypertensive group. (2) Serum Ca and urinary Ca were non-parallel, and the former could not accurately reflect Ca-intake. In short, the relationship between element and BP still remains to be elucidated.
8.Exploration on the Famous Old TCM Doctor Wang Xingkuan’s Rules of Syndrome and Treatment of Chest Blocking and Heartache Based on Data Mining

Objective To explore Wang Xingkuan’s rules of syndrome and treatment of chest blocking and heartache (Xiongbixintong).Methods Collection of professor Wang Xingkuan’s 267 consilia of patients with Xiongbixintong for outpatients. Chinese medicine terminology was regulated and Excelldatabase was established. Symptom, syndrome element, pathogenesis and treatment were statistically described by using Weka3.6 software, and Apriori algorithm was adopted for the main pathogenesis→treatment analysis of association rules.Results Symptoms include:chest pain, heart palpitations, shortness of breath, pale tongue (dark) red, etc. Syndrome elements include:in liver, and heart, and blood stasis, phlegm, qi stagnation, etc. The key pathogenesis is liver-heart imbalance, including stagnation of liver qi, heart and blood stasis, deficiency of heart qi-ying, disturbing heart-mind, etc. The principle of treatment is liver-heart Tongzhi, so the treatment is of“liver” with Shu gan-mu;treatment of“heart” contains freeing channels, eliminating phlegm and blood stasis, quiet the heart, replenishing qi-ying, etc. The main pathogenesis related credibility→treatment was higher than 0.50;with high reliability, the liver-heart imbalance→liver-heart Tongzhi was 0.71. Medication includes catharsis and tonic,“catharsis” to salvia, allium macrostemon, pseudo-ginseng, bupleurum, etc;“tonic” to white ginseng, ophiopogon japonicus, radix paeoniae alba, poria with hostwood, polygala tenuifolia, etc. Conclusion “Xintongzhigan, liver-heart Tongzhi, catharsis and tonic” is Wang Xingkuan’s thoughts and experience in treating Xiongbixintong.
9. Dedifferentiated chordoma of sacrococcygeal region: a clinicopathologic analysis and review of literature

Objective:
To study the clinicopathologic features of dedifferentiated chordoma.
Methods:
Four cases of dedifferentiated chordoma of sacrococcygeal region were collected at Beijing Jishuitan Hospital, from 2009 to 2014. HE and immunohistochemistry (EnVision method) were used to observe the clinical, radiological and histological features of dedifferentiated chordoma and to make the diagnosis and differential diagnosis. The literature was reviewed.
Results:
Four cases of dedifferentiated chordoma were all located in the sacrococcygeal region. The mean age at diagnosis was 57 years (range 49-64 years). There were 1 female and 3 males. Histologically, there were two components of conventional chordoma and dedifferentiated sarcoma with or without transitional area between them. The histology of dedifferentiated components includes undifferentiated sarcoma and fibrosarcoma. Immunohistochemical study showed that cytokeratin, epithelial membrane antigen, S-100, Brachyury were positive in the portion of chordoma while p53 were positive in the portion of undifferentiated sarcoma. INI1 and vimentin were both positive.
Conclusions
The dedifferentiated chordoma is very rare. The diagnosis should combine the histological characters and immunohistochemical results and should be differentiated from the sarcomatoid chordoma, poorly differentiated chordoma and other high grade sarcoma. p53 may play a role in the malignant transformation mechanism of chordoma.
10.Preliminary investigation on the relation between clinical progress and anti-small monomolecular peptides antibody in individual infected with HIV.

OBJECTIVETo study the quantity of anti-R7V in individuals infected with HIV and AIDS patients and its relation with the progression of disease.
METHODSELISA and precipitation and other methods were used to investigate the quantity of anti-R7V in asymptomatic long-term survivors and AIDS patients.
RESULTSPositive rate and quantity of anti-R7V were higher in the HIV active ones and AIDS. It showed that the quantity and positive rate of anti-R7V were rather high in dissolving test.
CONCLUSIONSIt is strong suggestion for anti-R7V to obstruct the replication of virus by interfering the connection between HIV with CCR5 or CXCR4 and so it impossible HIV entering to CD4+ T cells.
