Simple sequence repeat (SSR) was used to investigate the genetic diversity and structure of Dendrobium officinale. A total of 15 primer pairs with stable and repeatable polymorphism were screened out from 60 SSR primer pairs developed by the method of microsatellite enrichment by magnetic beads. Forty-eight samples of Dendrobium officinale were analyzed in genetic polymorphism. These loci were polymorphic and displayed 3 to 9 alleles per locus with a mean number of 6.1. The observed and expected heterozygosities ranged from 0.60 to 0.85 and from 0.49 to 0.85 respectively. The polymorphic information content (PIC) of each SSR locus varied from 0.437 to 0.829 with an average of 0.702. Fifteen primer pairs were used in Dendrobium cross-species amplification and totally 13 primer pairs were proved to have the transferability in D. officinale related species. In addition, 500 tissue culture plantlets of D. officinale were tested for purity identification by means of PCR amplification with four SSR primer pairs. The results showed that SSR technique is a feasible, simple and inexpensive method for determining adulterants in germplasm identification.

BACKGROUND: The biological behaviors of preadipocytes in adipose tissue of young animals have been closely linked to the onset and prevention and treatment of obesity. Observing mechanical oscillation effects on biomechanical behaviors of preadipocytes using biomechanical stimuli would provide more direct experimental evidence for treatment of simple obesity using manipulation and massage therapy.OBJECTIVE: Different frequencies of mechanical force stimulation were performed on preadipocytes from young rats cultured in vitro to observe the changes in cell viability, proliferation, and apoptosis.METHODS: Preadipocytes from SD young rats were in vitro cultured. Following identification, preadipocytes were dynamically, mechanically stimulated through the use of constant temperature oscillator. According to different treatment frequencies, three groups were set: 0 Hz (blank control), 1.5 Hz, and 3 Hz. A 30-minute oscillation, once every 12 hours, total 3 days, was performed in each group. Following dynamic mechanical stimulation, cell viability, proliferation, and apoptosis in young rats were observed. RESULTS AND CONCLUSION: In the initial stage of culture, cells exhibited the morphology similar to fibroblasts. After oil red O staining, red particles appeared in the cells, indicating that the young mouse cells cultured in vitro were preadipocytes. With stimulation of oscillation force, the viability and proliferation of preadipocytes were significantly inhibited (P < 0.05 or P < 0.01). There wasno significant difference in effects of mechanical oscillation on preadipocyte apoptosis between 0 Hz and 1.5 Hz, 3 Hz groups (P > 0.05). These findings indicate that thecell biological mechanism underlying preventing simple obesity in adolescents is to inhibit the viability and proliferation of preadipocytes

(2)In immunohistochemistry assay and image analysis, collagen was positive in light to deep brown areas. In the relaxation group, positive units of collagen types I and III in cardinal ligaments were 13.8±2.1 and 9.6±2.4 respectively.Positive units of coHagen types I and Ⅲ of cardinal ligaments in the control group were 27.4 ±3.5 and 17.7 ±4.0 respectively.Difierences between these two groups were statistically significant (P

Objective To investigate the value of multiple sequential MRI in the differential diagnosis of vater ampulla carcinoma (VPC).Methods MRI data of 53 patients with VPC confirmed by surgery and pathology were analyzed retrospectively, which included 17ampullary carcinomas,15 pancreatic head carcinomas,10 duodenal papillary carcinomas and 11 distal common bile duct carcinomas. All patients underwent routine MRI plain scan,MRCP and multi-phase dynamic enhancement scan for the upper abdomen. The morphological characteristics and model of multi-phase dynamic enhancement of the tumor, and the morphological changes of pancreaticobiliary duct were focused, and relevant data were also measured. Results The differences of tumor size, central location, and signal homogeneity among the four different histological types showed statistical significance(P<0.05),and their patterns of multi-phase dynamic enhancement also had differences.Double-segment sign was most found in VPC (26/53),four-segment sign was easy showed in pancreatic head carcinoma (8/15)and three-segment sign in distal common bile duct carcinoma (8/11).Beak sign was common seen in ampullary carcinoma(10/17),rat-tail sign was easy discovered in pancreatic head carcinoma (7/15) and truncated sign in distal common bile duct carcinoma(10/11).Diameter of major pancreatic duct in pancreatic head carcinoma was maximum and minimum in distal common bile duct carcinoma, and their difference had statistical significance(P<0.05).Distance between obstructive end of bile common duct and major papilla of duodenum, and between obstructive end of major pancreatic duct and major papilla of duodenum as well as the pancreaticobiliary conjunction angle in pancreatic head carcinoma and distal common bile duct carcinoma were greater than that in ampullary carcinoma and duodenal papillary carcinoma. Conclusion MRI can prominently demonstrate various imaging characteristics of VPC with different histological origins. It is very valuable in the differential diagnosis of VPC with different histological types by comprehensive analysis of these findings.

Aim To investigate the effect of total flavonoids of Chrysanthemum indicum on proliferation of synoviocytes and TRAIL/TNF-? expression of synovial tissue with adjuvant arthritis rats. Methods SD rats were divided randomly into six groups including normol,model,TFC(84,168,336 mg?kg-1)and control drug Tripterygium glycosides(30 mg?kg-1)groups.Adjuvant arthritis rat model was induced by a single intradermal injection of 0.1 ml of the complete Freund's adjuvant into the the right hind feet pads of the SD rats. The proliferation of synoviocyte was measured by MTT;The expression of TRAIL and TNF-? on synovial tissue were detected by means of immunofluorescence. Results TFC was shown to suppress the excessive synoviocyte proliferation; The expression of TRAIL was lower in model group than that of the normal group.TFC can increase the expression of TRAIL protein; The contrary to TNF-? protein. Conclusions TRAIL play a role in the development of AA.TFC can increase the lower expression of TRAIL protein, that maybe one of the mechanisms of TFC improvement AA.
