1.Clinical significance of CD+4CDHi25CDLo127 regulatory T cell in the peripheral blood of patients with lung cancer

Objective To evaluate the changes and clinical significance of CD+4CDHi25CDLo127 regulatory T cell(Treg) in peripheral blood of patients with lung cancer. Methods 30 patients with lung cancer and 20 heathy volunteers were included in this study. The proportion of Treg population in CD4+ T cells stained with three colors was analysed by flow cytometry. The serum level of IL-10 and TGF-β were measured by ELISA. Results The proportion of Treg in patients with squamous cell careinoma(n=20), adenocarcinoma (n=10) were all significantly higher than that of healthy controls (P <0.05), but there was not obvious difference between the two groups with different pathological types(P0.05). Increased serum level of IL-10 and TGF-β was also detected in lung cancer patients. Conclusion The proportion of Treg is increased in lung cancer patients, which may result in the inhibition of host anti-cancer immune response by excreting IL-10 and TGF-β.

Objective To observe the alteration of bone metabolism and to study the pathogenesis of osteoporosis and factors in elderly patients with chronic obstructive pulmonary diseases (COPD). Methods The biochemical markers of bone metabolism, bone mineral density (BMD) of the lumbar spine and the right femur, parameters of calcaneal quantitative ultrasound(QUS), blood partial pressure and pulmonary functions in 39 male patients with COPD and 30 controls were measured. Results The broadband ultrasonic attenuation (BUA) and the speed of sound (SOS), BMD of the lumbar spines and the femur were significantly lower than that in control group. 3.Construction of obese mouse models with high fat diet feeding:relationship between nutritional factor and metabolic syndrome

BACKGROUND:Metabolic syndrome greatly harms the human body, and is affected by many factors. Through constructing diet-induced animal models, we can better analyze the relationship between nutritional factor and metabolic syndrome, and provide reliable references for the clinical treatment of this disorder. OBJECTIVE:To construct obese mouse models with high-fat diet feeding and discuss the relationship between nutritional factor and metabolic syndrome. METHODS:Thirty mice were selected and randomly divided into model group (n=20) and control group (n=10), and were fed with high-fat and normal animal feeds for 10 consecutive weeks. RESULTS AND CONCLUSION:Compared with the control group, after 1 week of feeding with high-fat animal feeds, body weight of mice in the model group raised, and differences gradualy increased with the feeding time increased. After 8 weeks of feeding, body mass index of mice in the model group significantly raised (P < 0.05). After 4 weeks of feeding, fasting venous blood glucose level of mice in the model group significantly raised, and showed a gradual rise trend with feeding time. After 5 weeks of feeding, fasting insulin level of mice in the model group also began to rise. The oral glucose tolerance test showed that mice in the model group showed a gradual downward trend of glucose tolerance with feeding time. After 8 weeks of feeding, serum levels of total cholesterol and high density lipoprotein cholesterol in the model group significantly raised (P < 0.05). After 10 weeks of feeding, serum levels of triacylglycerol, total cholesterol, low density lipoprotein cholesterol and high density lipoprotein cholesterol in the model group raised (P< 0.05). The results demonstrate that obese mouse models were successfuly constructed with high-fat diet feeding, which can simulate the natural progression of metabolic syndrome in human, moreover, the nutritional factor is closely related to metabolic syndrome.
4.The comparison of coronary arteries imaging features between Uygur and Han populations in Xinjiang with 64-slice spiral CT

Objective To analyze the imaging findings of coronary angiography using 64-slice row CT and investigate the difference of coronary artery's morphological characteristics between Uygur and Han populations.Methods A retrospective study was made to coronary CT angiographic images of 88 Uygur cases matched with 88 Han cases.The data were analyzed with X~2 test and paired Wilcoxon test.Results The coronary CT angiographic findings were different between Uygur population and Han population in the following aspects: there were 62, 18, and 8 cases with the left coronary artery originating from intra-sinus, para-sinus and extra-sinus location respectively in Uygur population, while there were 73, 14, and 8 cases in Han population respectively (t=8319, P<0.05).And there were 78,7, and 3 cases with the right coronary artery originating from intra-sinus, para-sinus and extra-sinus location respectively in Uygur population, while there were 82, 1 and 1 case respectively in Han population (t=6936, P<0.05).The incidence of the sharp marginal branch were 52 (52.09%) and 67 (76.13%) in the two populations respectively (X~2=5.8381, P<0.05).The cases with various malformations of coronary arteries were 16 and 6 cases in the two populations respectively(X~2 =5.1948,P<0.05).The cases with LCA variations were 28 and 49 cases in the two populations respectively(t =2692,P<0.05) and the number with RCA variations were 33 and 27 cases in the two populations respectively(2 =968,P<0.05).Conclusions There are lots of differences of the coronary artery morphology between the Uygur and Han populations.Firstly, these differences may be related to different patterns in coronary angiography.Secondly, these differences may be related to differences between Uygur and Han populations in the incidence and severity of coronary heart disease.
5.Effect of Grp75 on the alteration of apoptosis related gene Bax and NF-κB induced by glucose deprivation

Objective To study the effect of glucose regulated protein 75(Grp75) on the alteration of Bax and NF-κB induced by glucose deprivation through the stably transfected PC12 cells with Grp75. Methods The cells of Grp75-overexpressing group and control group incubated in glucose-free DMEM medium for indicated time (6, 12, 24 and 48hours). The expression level of Grp75, Bax and the activity of NF-κB were determined by Western blotting, and the expression level of Bax was determined by semi-quantitative RT-PCR and Western blotting. Immunocytochemistry was performed using a conformation specific anti-Bax (6A7) antibody to detect the activation of Bax. Results The activation of Bax and the decline of NF-κB activity played important roles in the apoptosis of PC12 cells induced by glucose deprivation. Grp75 inhibited the apoptosis induced by glucose deprivation through inhibition of the activation of Bax and the decline of NF-κB activity. There was no change in Bax expression level under glucose deprivation in two groups. Conclusion The activation of Bax and the decline of NF-κB activity were associated with apoptosis of PC12 cells induced by glucose deprivation, and Grp75 provided protection to PC12 cells through inhibition of activation of Bax and maintaining activation of NF-κB.

Objective To investigate the effects of fluvastatin (Flu) on the levels of estradiol(E2 ) ,tumor necrosis factor-α(TNF-α) ,insulin-like growth factor-1(IGF-1) and bone glaprotein(BGP) in experimental osteoporosis rats .Methods The model of osteo-porosis was established ,all rats were ovariectomized .The rats were given Flu(20 ,10 ,5 mg/kg) through intragastric administration (ig) .After treating for 12 weeks ,weighed all rats ,then sacrificed all rats ,removed the uterus and weighed its wet weight ,and calcu-lated uterus index ;the levels of E2 ,TNF-α,IGF-1 and BGP in serum were detected .Results Flu did not affect the uterus weight in experimental osteoporosis rats ;20 mg/kg Flu could increased the osteoporosis rat′s serum levels of E2 and IGF-1(P<0 .05) ,de-creased the levels of TNF-αand BGP(P<0 .01);10 mg/kg Flu could decreased the osteoporosis rat′s serum levels of BGP .Conclu-sion Flu could increased the levels of E2 ,IGF-1 ,and decreased the levels of TNF-α,BGP in experimental osteoporosis rats .

The medical mobile learning platform was constructed according to the information need of teachers and students in Shanxi Medical University, Changzhi Medical College, and Fenyang Medical College.The teaching and learning resources in Shanxi Medical University were integrated into the platform which could thus provide a variety of interactive learning ways for its users and users could rapidly find out their interested information resources. However, the platform construction needs the implementation of incentive measures, and regulations and rules for the protection of intellectual property rights.
8.Role of N-cadherin in cranial neural crest delamination during chick embryogenesis

AIM: To investigate the role of N-cadherin in the delamination of neural crest cells. METHODS: The normal expression of N-cadherin in neural tube was identified using in situ hybridization. The cells with N-cadherin over expression were obtained by transfection of wild-type N-cadherin (wt-N-cadherin) ,and the cells with N-cadherin silencing expression were obtained by transfection of dominant-negative N-cadherin (dn-N-cadherin). The migration of cranial neural crest cells was determined by the technique of immunohistochemistry. RESULTS: Either overexpression or down-regulation of N-cadherin significantly affected the migration of cranial neural crest cells. CONCLUSION: Delamination and migration of the cranial neural crest cells rely on the relative N-cadherin expression in the neural tube during neurulation.
9.Mechanism of action of direct-acting antiviral agents in treatment of chronic hepatitis C

With the development and launch of direct-acting antiviral agents (DAAs) in the world in recent years, therapeutic regimens for chronic hepatitis C are constantly evolving. DAAs will also be launched in China in the near future. DAAs mainly target at the non-structural proteins of HCV and can inhibit HCV RNA replication. This article introduces the targets, mechanism of action, and resistance characteristics of different DAAs, as well as their current research and development in China and the results of phase Ⅲ clinical studies, in order to provide a reference for combined therapeutic strategies with DAAs in the treatment for chronic hepatitis C.
10.Anti-tumor and immunoenhancement effects of effective combination of extraction from edible fungi on C26 xenograft mice

Background and purpose:Edible fungi has shown powerful potential in health care. This study aimed to explore the anti-tumor and immunoenhancement effects of combination of extraction from edible fungi. Methods:C26 cells has been xenografted into mice, and the animal models were randomly divided into control group, prevention group, treatment group and cyclophosphamide group. Mice were given the combination of extraction from edible fungi every day after vaccination of C26 cells into mice, except the prevention group which has been given the combination of extraction from edible fungi two weeks before vaccination. Two weeks after being planted, all the mice were killed and the tumor inhibition rates were studied. The immune function was measured by T lymphocyte transforming assay and NK killing assay. Results:There were signif icant difference in terms of tumor weight between prevention group and control group. The prevention group mice display improved T lymphocyte transforming assay and NK killing ability. Conclusions:The combination of extraction from edible fungi has remarkable inhibitory effects on C26 carcinoma in mice and can enhance immunity against mice with C26 carcinoma.
