AIM:To study the effect of Taxol on the proliferation, apoptosis, and mRNA expressions of α2,6-sialic acid (SA) and α2,6-sialyltransferase (ST6Gal) in mouse cervical cancer cell line U14.METHODS:After the U14 cells were treated with Taxol, the IC50 value of Taxol to U14 cells was detected by MTT assay.The expression of α2,6-SA and apoptosis-related factors (Bcl-2, Bax, caspase 8 and caspase 3), the apoptosis rate and cell cycle were determined by flow cytometry.The mRNA expression of ST6Gal1 and ST6Gal2 was detected by qPCR.RESULTS:As compared with control group, Taxol induced obvious U14 cell growth inhibition, reduced α2,6-SA expression, up-regulated Bax, down-regulated Bcl-2, decreased the ratio of Bcl-2/Bax, enhanced caspase 8 and caspase 3 activity, increased the apoptotic rate and cell proportions of Sub-G1 and S phases, and induced G2/M phase arrest.Taxol also down-regulated the mRNA expression of ST6Gal1, and slightly up-regulated the mRNA expression of ST6Gal2.CONCLUSION:α2,6-SA and ST6Gal are involved in the multiple effects of Taxol on modulation of the cell cycle and apoptosis in U14 cells.

The calcium antagonist action of arctigenin (ACT) was studied in order to verify the ef-fect of Fructus Arctii for the relieve of exterior syndrome. Muscular contraction of isolated rat trachea,colon, pulmonary artery and thoracic aorta induced by KC1, that of guinea pig trachea induced by CaCI2,before and after the addition of ACT were assessed and their contraction-response curves drawn and PD'2calculated according to Scott. The inhibition rate of two-phase contraction of guinea pig trachea induced byacetylcholine chloride (Ach) in comparison with verapamil (VER) was calculated. Results of the studyshowed that ACT could non-compatitively antagonize the muscular contraction of the test specimens withPD'2 of 4.01, 5.11, 5.98 and 6.05 respectively. Similar to VER, ACT could non-competitively antagonizethe isolated guinea pig trachea with PD'2 of 4.04 and 5.62 respectively. Both of them could inhibit thefirst phase contraction induced by Ach with inhibition rates of 66.14% and 81.42% respectively. It wasconcluded that ACT, as the active constituent of Fructus Arctii, relaxed smooth muscle contraction byblocking the potential dependant Ca2+ channel and the internal release of Ca2+.

Objective To study the effect of extract of cockroach on animals with acute hepatic injury and its antiviral action in vitro. Methods The animal models with hepar injury were established by CCl4, bcg vaccine and lipopolysaccharide (LPS) . And the activities of ALT and AST in serum were measured. Serum of ducks was separated after cockroach extract was administrated. The serum was added to the culture fluid of HBV-DNA transfected hepatic cancer cells-HepG2. Results After the cockroach extract was administrated, the activities of ALT and AST in mouse serum were reduced. Both the therapeutic indexes of cockroach extract and lamivudine were larger than 2. Conclusions Cockroach extract can improve the liver function of animals with acute hepatic injury, and inhibit the secretion of ALT and AST in serum.

Objective To clarify if estrogen increases gastric mucosal injury in portal hypertensive rats and its role in the pathogenesis of portal hypertensive gastropathy. Methods Forty SD rats were divided into 4 groups:P + E, P, S + E and S groups. P + E and P groups received portal vein ligation and the S + E and S groups underwent sham operation. P + E and S + E groups were given estrogen intramascularly. All rats were maintained on their indiuidual treatment for 14 days. One hour before the sacrifice rats were orally lavaged with 2 ml 99% ethanol. Gastric mucosal blood flow, degree of gastric mucosal injury and mucosal NO production were determined. Objective To evaluate the predictive value of morphology of superficial depression in estimation of histology and invasive depth in colorectal neoplasia by using magnifying chromo-endoscopy.Methods Flat or depressed and sessile eolorectal lesions which were indicated for endoscopic mucosal resection (EMR) were consecutively collected. Depressed lesions were classified into type 1 (star-like) and type 2 (round) according to the morphology of depressive areas in the colorectal neoplasia with magnifying ehromoendoscopy. The relationship between morphologic classification with histology and invasive depth was studied with reference to pathological diagnosis after EMR. Results Ninety lesions including 25 sessile and 65 flat were resected with EMR. Lesions with central depression (54. 4%,49/90) were more likely to have high-grade dysplasia (HGD) or cancer than those without ( 51.0% vs. 17. 1%,P＜0. 001 ). Depressive lesions of type 2 were more susceptible to have HGD or cancer than those of type 1 (89. 5% vs. 26. 7%,P＜0. 001 ). The overall accuracy of depression morphology for distinction between lesions of superficial submueosal (m-sm1) and deep submucosal (sm2-sm3) invasion was 83. 7% (41/49). Conclusion The morphology of superficial depression on colorectal neoplasia is highly correlated with the histology and invasive depth,which facilitates the EMR treatment.

Objective To assess the local anatomy of the Jing Bi with high-frequency ultrasound, and to evaluate the clinical significance of high-frequency ultrasound in acupuncture. Methods A total of 206 healthy subjects were observed with the high-frequency ultrasound. Two common location points of Jing Bi were selected and acupunctured under guidance of high-frequency ultrasound. The sensation of subjects was observed. Brachial plexus diameter, distance between skin and membrana pleuralis, as well as the brachial plexus and the pleura were measured. Results The filiform needle tip to lung, lung tissue and the brachial plexus were all showed, and the brachial plexus diameter, distance between skin and membrana pleuralis, the distance between the brachial plexus and the pleura were obtained. The patients felt when the needle reached the membrane of brachial plexus. All subjects got feeling within 10 seconds after acupuncture, and no pneumothorax occurred. Conclusion The local anatomy of dangerous acupoints can be accurately observed with high-frequency ultrasound. High-frequency ultrasound has important value and significance for acupuncturing the dangerous acupoints.

It is the measurement of high-loss dielectric to measure the permittivity and conductivity of biologic tissue.But now it is difficult to measure them accurately.Good result is got by measuring complex permittivity of phantom model at2450MHz using perturbation method,which is meaningful for the research on biologic tissue simulation by phantom model.
