1.Pathogenesis of brain damage in infant rats induced by corticosteroids

Objective To explore the possible pathogenesis of brain damage in rats induced by prednisone or corticotrophin. Methods The doses of prednisone or corticotrophin were determined by a primary experiment to obtain corresponding plasma cortisol or corticosterone level as same as that in sick children after drug taking. Then 192 healthy infant (at the age of 7 d) and 192 adult (at the age of 2 months) male SD rats were divided into 4 groups as infant prednisone group, adult prednisone group, infant corticotrophin group and adult corticotrophin group (n=96 in each). then every group was further subdivided into 12 subgroups (n=8 in each). The subgroups were divided according to the dose (therapeutic or low doses), the course [short (10 d) or long (3 weeks)], the sacrificed time (24 h or 4 weeks after withdrawal), and their corresponding controls. Serum neuron-specific enolase (NSE) concentration was quantified by ELISA. Expressions of apoptosis-related proteins, Bax and Bcl-2 in the brain were detected by immunohistochemical assays. Neuronal apoptosis was detected by TUNEL. Results Our primary experiment indicated that the therapeutic dose was 4 mg?kg-1?d-1 or 150 U?m-2?d-1 for prednisone or corticotrophin, and the small dose was 2 mg?kg-1?d-1 or 38 U?m-2?d-1 for them. In infant rats treated with prednisone or corticotrophin at therapeutic-dose for short or long term, their serum NSE concentration were increased significantly by 50.6% to 103.2%. The data analysis of food supervision needs to reflect the integration of food safety and possible trend based on the fun-damental test data of stressed foods and their products. With the assessment and analysis on potential resources of hazard elements, the integrate management support of food safety could be achieved. The data management could be organized and evaluated through plan, collection, treatment and display, which might be assisted to establish data-link with process mining to enforce the prospect of food safety background and trend and to provide the support of food safety management.
3.Short-term effects of Aidi injection adjuvant GP methods chemotherapy in treatment for advanced non-small cell lung cancer

Objective:To study the short-term effects,toxic side effects and immune function of Aidi injection adjuvant GP methods chemotherapy for advanced non-small cell lung cancer(NSCLC).Methods:130 patients with advanced NSCLC were divided into study group and the control group according to treatment methods.The study group were given Aidi injection +GP,and the control group only treated with GP.The short-term effects,toxic side effects gradeⅢ-Ⅳand immune function changes were compared between the two groups.Results:The overall response rate was 40.28%in study group,higher than 37.93% in control group(P>0.05).The two groups showed differences in the rates of leucocyte decrease,gastrointestinal reaction and platelets decrease(P<0.05).The levels of CD4,CD8 and CD4/CD8 didn′t changed obviously in study group(P>0.05),while in the control group the levels of CD4,CD8 and CD4/CD8 changed more significantly than before(P<0.05).The two groups also produced differences in CD4,CD8 and CD4/CD8 after treatment( P<0.05 ) .Conclusion: The short-term effects of Aidi injection adjuvant GP chemotherapy regimen for advanced NSCLC is similar to only GP chemotherapy,but it can obviously reduce the incidence of toxic side effects and prevent the immune func-tion.
4.Effect of Clinically Equivalent Doses of Xuesaitong and Ginaton Injections on Cerebral Ischemia Reperfusion Injury of Rats

Objective To observe the curative effect of clinically equivalent doses of Xuesaitong and ginaton injections on cerebral ischemia reperfusion (I/R) injury of rats.Methods Male rats were randomly divided into five groups:normal control group,sham-operation group,model control group,Xuesaitong group and ginaton group.The cerebral ischemia rat model was established by middle cerebral artery occlusion (MCAO).Rats in the Xuesaitong group were given 20 mg·kg-1 of Xuesaitong injection,and rats in the ginaton group were intravenously injected with 7.5 mg· kg-1of ginaton immediately after I/R injury and once daily for 7 days.Rats in the sham-operation group and model control group were given the same volume of 0.9% sodium chloride solution.The score of ethology,volume of cerebral infarction,mortality,superoxide dismutase (SOD),malondialdehyde (MDA),xanthine oxidase (XOD),nitrogen oxide (NO) and NO synthase (NOS) in seruu were examined.Results Compared with model control group,Xuesaitong and ginaton effectively reduced behavioral score 96 h (P < 0.05),120 h (P<0.01),144 h (P<0.01) and 168 h (P<0.01) after I/R injury,the volume of cerebral infarction 168 h after I/R injury and NO content (P < 0.05).But they had no effects on NOS,SOD,MDA,and XOD contents.Conclusion Curatively injecting Xuesaitong and ginaton can effectively reduce cerebral I/R injury,but no significant difference in curative efficacy is observed between Xuesaitong and ginaton at clinically equivalent doses.
5.Application of Quality Metrics in Food Inspection Laboratory Management

Because of the particularity and the complexity of integrated management of food, government regulators must perform the whole process of quality control and trace to evaluate and judge the merits of food instead of only depending on product standards.Quality metrics is a new concept for the food industry, and can be regarded as the comprehensive evaluation and measurement for product quality and the standard for judging the quality of food products.Request for Quality Metrics Guidance for Industry was issued, which not only established a new model for the quality control of pharmaceutical industry, but also provided a new way of quality management for the inspection organization.By introducing the contents of Request for Quality Metrics Guidance for Industry issued by the US FDA, on the basis of summarization and thought, the quality management and evaluation of pharmaceutical industry was compared, and the feasibility and applicability of quality metrics management in inspection and testing field, especially in food testing field was evaluated.In this paper, the concept of quality metrics was extended to the level of food inspection laboratory management with hope to provide ideas for the future development of food quality management model.
6.Relationship between matrix metalloproteinase-9 gene-1562C/T polymorphism and hemorrhagic transformation from in Chinese Han population in Henan province

Objective To investigate the relationship between matrix metalloproteinase-9 (MMP-9) gene-1562 C/T polymorphism and hemorrhagic transformation (HT) from (IS) in Chinese Han population in Henan province.Methods The promoter-1562 C/T gene polymorphism in 84 IS patients with HT and 138 age-and sex-matched IS patients without HT were tested by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).The genotypes and alleles frequencies of-1562 C/T polymorphism were analyzed.Results The frequency of CT and TT genotype and T allele was lower in IS patients with HT than in IS patients without HT (17.86% (15cases) vs.30.43% (42 cases),8.93% (15 cases) vs.15.94% (44 cases),x2=4.326 and 4.458,P=0.037 and 0.035).The relative risk for HT from IS was decreased in patients with T allele than those withCTandTTgenotype[0.517 (95%CI:0.278-0.961) vs.0.497 (95%CI:0.255-0.967)].Conclusions The MMP-9 gene-1562C/T polymorphism may be related to hemorrhagic transformation of in Chinese Han population in Henan province.The T allele is a protective factor for hemorrhagic transformation of in the population.

8.PET/CT imaging of striatal dopamine transporters in a newborn piglet model of hypoxic-ischemic brain injury

Objective To investigate changes of striatal DAT following hypoxic ischemic (HI)brain injury in newborn piglets using 11C-N-2-carbomethoxy-3-(4-fluorophenyl)-tropane (CFT) PET/CT,and to evaluate the value of 11C-CFT PET/CT in brain injury.Methods Newborn piglets with HI brain injury (n =20) were taken as a model group,and five piglets were used as a control group.Radioligand 11CCFT (55.5-74.0 MBq) was injected through the jugular vein,and PET/CT imaging was performed to observe the changes of striatal DAT in newborn piglets.The ST/occipital lobe (OC) ratio was calculated.Model group was divided into 0-6 h,20-24 h,44-48 h and 68-72 h sub-groups after HI in accordance with the imaging time.The piglets were sacrificed immediately after 11C-CFT PET/CT scanning,and then the brains were removed for pathological analysis.Data analysis was performed with one-way analysis of variance and Pearson linear correlation analysis.Results After intravenous injection of 11C-CFT,the radioactivity accumulation in cortical,striatum,and cerebellum was shown clearly in the control and model groups.The radioactivity accumulation was lower in the white matter.The radioactivity in cortical and cerebellum exhibited decreased with time,while the striatum was still clear.After HI,the ST/OC activity ratio in the striatum was initially increased,and the ratio of 0-6 h group (1.34 ± 0.04) was statistically significant compared with that of thecontrol group (1.18 ± 0.06 ; F =4.658,P < 0.05),followed by a gradual decrease.ST/OC ratios of other HI subgroups were 1.27 ±0.01,1.27 ±0.10 and 1.18 ±0.05,respectively.There was a positive correlation between the number of DAT positive neurons ((13 ± 3),(13 ± 4),(8 ±3) and (4 ±4)/high power field) and 11C-CFT ST/OC activity ratios (r =0.844,P <0.05).Conclusion 11C-CFT PET/CT study can accurately reflect the changes of DAT in the striatum,and the amount of DAT is related to the severity of the ischemic insult in a newborn piglet model of HI.
9.Expression and bioactivity of OSF-1 in Pichia pastoris.

In order to research the biologic activity of osteoblast-stimulating factor 1 (OSF-1), the pPIC9K/osf-1 yeast expression vector was constructed to express and purify OSF-1. Firstly, the osf-1 gene sequence was obtained by artificial synthesis and cloned into Pichia pastoris expression vector pPIC9K to generate pPIC9K/osf-1. The recombinant plasmid was linearized by Sac I and transformed into P. pastoris GS115 by electroporation. Recombinant P. pastoris GS115/ pPIC9K/osf-1 was screened by MD and G418-YPD plates and further identified by PCR. The positive P. pastoris was induced with 1% methanol at 25 degrees C for 96 h. The target protein was analyzed by SDS-PAGE showing a special band about 18 kDa. The target protein was successfully purified from the supernatant of the broth using ion exchange chromatography of SP-Sephadex C-50. The purity of target protein was above 98%. Western blotting appeared a good antigenicity of the purified protein. Bioassay results show that the recombinant protein OSF-1 can promote the differentiation and proliferation of osteoblasts MC3T3-E1. We successfully expressed OSF-1 by recombinant P. pastoris for further development of anti-osteoporosis of research and industrial production of OSF-1.

10.The role of autophagy-related genes in the evaluation of therapeutic effect of ET regimen neoadjuvant chemotherapy in breast cancer

Objective To identify the role of autophagy‐related genes in prediction of therapeutic effect of ET regimen neoadju‐vant chemotherapy in breast cancer .Methods Breast cancer tissues were retrospectively collected from 41 cases of breast cancer pa‐tients that treated by ET regimen neoadjuvant chemotherapy with different therapeutic effects .Expressions of Atg3 and Beclin1 ,and the autophagy‐related genes ,were detected by Western blot ,Real‐time PCR and immunohistochemistry in these tissues to observe their expressions before neoadjuvant chemotherapy .Statistical test was used to analyze the relationship between Atg3 ,Beclin1 ex‐pressions and therapeutic effects of ET regimen neoadjuvant chemotherapy .Results Western blot ,Real‐time PCR and immunohis‐tochemistry revealed that expressions of Atg3 and Beclin1 were up‐regulated in 24 breast cancer tissues before neoadjuvant chemo‐therapy ,including 23 cases of complete response(CR) or partial response (PR) (95 .8% ) and 1 case of progressive disease (PD) . Atg3 and Beclin1 were not expressed or lowly expressed in the other 17 cases ,including 3 cases of CR or PR (17 .6% ) and 14 cases of no change and PD .The effective rate of ET regimen neoadjuvant chemotherapy in breast cancer patients with over expression of Atg3 and Beclin1 was significantly higher than those with low expression or without expression of Atg3 and Beclin1 (P<0 .05) . Conclusion Over expression of Atg3 and Beclin1 could be new indicators for therapeutic effect of ET regimen neoadjuvant chemo‐therapy in breast cancer .
