1.Study on immunosensor of immobilized HRP-HBV antibody on Au colloid

Objective To form Hepatitis B electrochemical immunosensor for the clinic detection. Methods Using the gold electrode as the fundus,the thionine and the HBV horseradish peroxidase(HRP) antibody were electrostaticly bounded to the self-assemble monolayer competitively to form the Hepatitis B electrochemical immunosensor. Cyclic voltammetry and linear scan voltammetry were used to investigate the chemical characteristics of the immunosensor. Results The linearity of Hepatitis B in the range of 1:30~1:200 with the correlation coefficient of 0. 994 was obtained. Conclusion The Hepatitis B electrochemical immunosensor formed by this way can be used to the clinic detection.
2.Effects of Telmisartan on the expression of Toll-like receptor 4 in monocytes of ApoE-deficient mice

ObjectiveTo observe the effects of telmisartan on toll-like receptor (TLR) 4 expression in monocytes of mice atherosclerosis (AS)model, and explore the mechanism of inflammation.MethodsAS mice model were established in ApoE-deficient mice fed with high-fat diet,and treated with telmisartan as intervention group. After 8 weeks, serum low density lipoprotein,triacylglycerol and total cholesterol were detected in group fed with high-fat diet and intervention group.TLR 4 expressions in two groups were analyzed by flow cytometry. Results There was no significant difference in levels of blood lipids between the two groups.TLR4 expression in monocytes in telmisartan intervention group was lower than group fed with high-fat diet (P<0.05).ConclusionsTelmisartan may intervene the inflammation during atherosclerosis by downregulating TLR4 expression in monocytes.

4.The effect of placenta derived tumor necrosis factor-α and myostatin in patients with preeclampsia and fetal development

Objective To investigate the effect of placenta derived tumor necrosis factor (TNF)-α and myostatin (MSIN) in patients with preeclampsia (PE) and fetal development.Methods One hundred and twenty pregnant women who delivery from October 2008 to October 2013 were enrolled in this study.In them,40 healthy pregnant women was normal control group,40 PE and fetal growth in normal pregnant women was PE group,40 PE and fetal growth restriction (FGR) of pregnant women was PE + FGR group.The immunohistochemical localization of SABC method was used to detect for TNF-α and MSIN protein in placenta tissue in each group respectively.Real-time fluorescent quantitative PCR and Western blotting were used to detect for TNF-α and MSIN mRNA and protein in placenta tissue.Results The TNF-α mainly located in placental blood vessels surrounding stroma,decidual cells,trophoblastic cells and MSIN mainly located in placental blood vessels surrounding stroma,decidual cells and terminal villi.The TNF-α and MSIN mRNA expression quantity in PE group was 3.65 ±0.86,1.80 ±0.32 ; in PE + FGR group was 3.88 ± 0.71,2.01 ± 0.55 ; in normal control group was 1.32 ± 0.21,0.77 ± 0.39.The TNF-α and MSIN mRNA expression quantity in PE group and PE + FGR group were significantly higher than those in normal control group(P < 0.01),and there were significant differences between PE group and PE + FGR group(P < 0.05).The TNF-α and MSIN protein expression in normal control group was 0.56 ±0.13,1.31 ± 0.23;in PE group was 1.67 ±0.25,1.55 ±0.34 ;in PE + FGR group was 2.78 ±0.41,3.07 ±0.51.The TNF-α and MSIN protein expression in PE group and PE + FGR group were significantly higher than those in normal control group(P < 0.01),and there were significant differences between PE group and PE + FGR group(P < 0.05).Conclusions The placenta derived TNF-α and MSIN have more important roles in the pathogenesis of PE and normal development of fetal.It can provide coping strategies by detecting the level of placenta derived TNF-α and MSIN.
5.Expression of laminin and fibronectin in the growth cycle of human hair follicles

Objectives To investigate the role of laminin (LM) and fibronectin (FN) in the growth cycle of human hair follicles. Methods The expression of LM and FN was detected by streptavidin-peroxidase (SP) staining. Results In the anagen phase, LM was expressed in dermal papilla, basement membrane and outer root sheath; FN was expressed in dermal papilla, basement membrane and connective tissue sheath. In the catagen phase, LM showed a lower expression in the dermal papilla and a linear expression in the basement membrane; the expression of FN in the dermal papilla and basement membrane was less intense than that in anagen phase, but was still positive. In the telogen phase, LM was only expressed in the basement membrane while FN was negative. Conclusion The difference between LM and FN expression in hair growth cycle indicates that LM and FN may play important roles in the regulation of human hair follicle growth cycl.
6.Ag85A DNA vaccination boosting enhances BCG primed-mice anti-tuberculosis T cell responses

Objective To construct DNA vaccine expressing Mycobacterium tuberculosis(Mtb) immunodominant antigen Ag85A and analyze its anti-tuberculosis T cell responses in BCG primed-mice after DNA vaccination boosting.Methods The coding gene of Ag85A mature fragment was amplified by PCR with H37Rv genomic DNA as template,and then cloned into the eukaryotic expression vector pVAX1 to construct Ag85A DNA vaccine.After purification,Ag85A DNA vaccine was injected intramuscularly twice in BCG primed-mice with BCG vaccination and DNA vaccination alone as control.Eight weeks post-vaccination,spleen lymphocytes were separated and were then used to analyze Mtb antigen specific effector T cell response and polyfuntional IFN-γ/TNF-α/IL-2 secreting CD4+ T cell frequencies and intensities,and CD8+T cell responses by IFN-γ ELISPOT assay and intracellular staining,respectively.Results Compared to BCG vaccinated-and DNA vaccinated-mice,Ag85A DNA boosting not only enhanced significantly BCG primed-mice IFN-γ+TNF-α+IL-2+,IFN-γ+ IL-2+,TNF-α+IL-2+ and IL-2+ CD4+ T cell frequencies and IL-2 secretion,but also improved significantly IFN-γ-secreting and IL-2-secreting CD8+ T cell frequencies.Condusion Ag85A DNA vaccine was constructed successfully and was demonstrated to enhance significantly BCG primed-mice Mtb antigen specific CD4+ and CD8+ T cell responses when boosting,which is beneficial to improve BCG immunogenicity and its waning immune protection against Mtb.
7.Effect of dose rate of X-ray on clonogenic formation in human lung cancer cell line A549

Objective To explore the effects of different dose rates of X-ray under the same dose on cell clonogenic formation in non-small-cell lung cancer cell line A549 in order to provide experimental basis for clinical radiotherapy plan. Methods The A549 cells were cultured at low density and irradiated with X-rays at dose of 4 Gy and selected dose rates of 1, 2, 4 and 6 Gy/min, respectively, from a linear accelerator. The 8th day after irradiation, the cells were fixed and stained with Giemsa solution, and colonies containing at least 50 cells were counted. The plating efficiency and surviving fraction were calculated. Results The clonogenic number in non-irradiated cells was 88.6±4.6. The numbers were significantly reduced in irradiated cells at dose rate 1, 2, 4 and 6 Gy/min (12.3±3.4, 9.0±0.8, 5.6±1.0, 11.5±1.7, respectively) than that in non-irradiated control cells (F=678.799, P<0.05). The plating efficiencies were decreased in irradiated cells, especially in 4 Gy/min irradiated cells, which was lower than that in any of the other three dose rate groups (P< 0.05). Conclusions Though at same radiation dose, cancer cells have different clonogenic formation efficiency when irradiation with X-ray at different dose rates. Thus, treatment with optimal dose rate may improve the radiotherapy efficacy.
8.Building a comprehensive health assessment indicators systems for health management of military officers

In line with the new WHO definition for health and reference of comprehensive health assessments of elderly overseas, the paper screened and set the weight for comprehensive health assessment indicators for military officers, establishing a general health assessment model as a result 402 military officers were subject to Comprehensive Health Assessments, and the outcomes were compared with subjective assessment of experts. The results of both methods are highly coincidental. It proves Comprehensive Health Assessment as a scientific quantitative health assessment method, with promising perspectives in health management in the future.
9.Clinical Study on Long-snake Moxibustion for Asthma Due to Deficiency of Lung and Spleen

Objective To observe the clinical efficacy of long-snake moxibustion in treating asthma due to deficiency of lung and spleen. Method Seventy patients were randomized into a treatment group and a control group. The treatment group was intervened by using long-snake moxibustion, while the control group was by regular acupuncture treatment. The therapeutic efficacy was evaluated after 1-month treatment. Result In the treatment group, 22 subjects showed marked efficacy, 27 showed effective, 6 failed, and the total effective rate was 89.1%; in the control group, 18 subjects showed marked efficacy, 25 showed effective, 12 failed, and the total effective rate was 78.2%. The therapeutic efficacy of the treatment group was significantly higher than that of the acupuncture group(P<0.05). Conclusion Long-snake moxibustion can produce a higher therapeutic efficacy than regular acupuncture in treating asthma due to deficiency of lung and spleen.
10.Therapeutic Observation of Gastrodin Acupoint Injection for Depression

Objective To observe the clinical efficacy of Gastrodin acupoint injection in treating depression.Method Eighty depression patients were randomized into a treatment group and a control group, 40 cases in each group. The treatment group was by gastrodin acupoint injection plus medication, while the control group was by acupuncture plus medication. The Hamilton Rating Scale for Depression (HAMD-17) was observed before and after intervention, and the clinical efficacies were compared. Result Respectively after 2 and 3 treatment courses, the HAMD scores were significantly different from that before intervention in both groups (P<0.05). Respectively after 1, 2, and 3 treatment courses, the HAMD scores in the treatment group were significantly different from that in the control group (P<0.05). The recovery rate and total effective rate were respectively 70.0% and 100.0% in the treatment group, versus 45.0% and 90.0% in the control group, and the between-group differences were statistically significant (P<0.05).Conclusion Gastrodin acupoint injection is an effective approach in treating depression.
