1.Comparison of the Content of Tanshinone Ⅱ_A in Different Dosage Form of Guanxin Danshen Preparations

OBJECTIVE: To determine and compare the contents of tanshinone Ⅱ A in difference dosage form of Guanxin danshen preparations(tablets,capsules,granules and dripping pills).METHODS: The tanshinone Ⅱ A in different Guanxin danshen preparations was extracted by ultrasonic extraction with methanol as solvent,and the content of tanshinone Ⅱ A was determined by HPLC on Agilent Zorbax C8(150 mm?4.6 mm,5 ?m) column with mobile phase consisted of methanol-water(72∶28)under a detection wavelength of 270 nm.RESULTS: The linear ranges for tanshinone Ⅱ A stood at 2.76~138 ?g?mL-1(r=0.999 9).The average recoveries of Guanxin danshen tablets,Guanxin danshen capsules,Guanxin danshen granules and Guanxin danshen dripping pills were 99.29%,100.37%,100.18% and 98.71%,respectively with RSD at 1.78%,2.10%,1.55%,and 1.09%(all n=6),respectively.CONCLUSION:The content of tanshinone Ⅱ A in differed greatly in different Guanxin danshen preparations;therefore it is urgent to establish the quality standard for Guanxin danshen preparations as soon as possible.

3.Wheelchair Taijiquan for Mental Health of Patients with Spinal Cord Injury:A Case Study

Objective To observe the effect of Wheelchair Taijiquan on mental health of patient with spinal cord injury. Methods One42-year-old male patient with spinal cord injury practiced wheelchair Taijiquan, 90 min everytime, 3 times a week for 10 weeks. SCL-90was used to assess the relevant indicators before and 10 weeks after the experiment. Results 10 weeks after Wheelchair Taijiquan exercises,the patient improved in sleep quality, appetite and physical health, and the symptoms as antagonism, paranoid, psychosis and obsessive-compulsivedisorder as well as intervening with interpersonal sensitivity, depression, anxiety and fear. Conclusion Wheelchair Taijiquan mayfacilliate to improve the mental health of patient with spinal cord injury.
4.Significance of change in thyroid hormone in patients with heart failure

Thyroid hormone levels were fourd to be changed significantly in 93 patients with heart failure. Their T3 and FT3 decreased, while rT3 increased. The severer heart failure, the more evident change in thyroid hormone. The results suggest that the change in thyroid hormones might be an index in reflecting the severity of heart failure.
5.Comprehension of Daytime Prolonged Online-HDF on the Treatment of Refractory Edema

[Objective] To evaluate the clinical efficiency of refratory edema based on daytime prelonged Online-HDF.[Methods] Six patients include four females and two males whose age ranging from 28 to 74 years old and average 55 years old.Online-HDF treatment for 3-9 times,vascular access was percutaneous femoral or internal jugular catheterization.One patient's temporary access was changed to permanent access because who needed continous hemodialysis.The machine version was Ganbro AK-100S.To filter by u2000 and replace fluid online for 6-10 hous,blood flow was 200-250ml/min,uf 1200-8600,the total fluid used for replacement was 20-42 liter,during the process,regular heparin or low molecular was used for anticoagulation.[Results]Three patients' clinical symptoms resulted from edema resolved,one patient accepted continous hemodialysis treatment for inpaired kidney function,one patient left hospital for expensive medical cost,one patient accepted ventilation treatment for respiratory failure in ICU.[Conclusion]Daytime prelonged Online-HDF is effective on the trentment of refratory edema,especially effective on dyspnea,tachypnea,chest distress that resulted from edema.
6.Differentiation of human umbilical cord-derived mesenchymal stem cells into male germ cells

ObjectiveTo study the possibility of differentiation of human umbilical cord mesenchymal stem cells(HUCMSCs) into human male germ cells,and to explore a new source of cells for the treatement of male infertility.MethodsHUCMSCs were transplanted into the seminiferous tubules of the testis of infertile mice by microinjection.The survival rate,migration and germ cell markers of HUCMSCs in the mice testis were detected via immunohistochemistry,immunofluorescence and confocal laser scanning. ResultsHUCMSCs can survive in the mice testis for at least 120 days,and they can migrate from the lumens to the basement membrane.Immunofluorescence showed that HUCMSCs can further differentiate in the mice testicular environment,and express the germ cell marker.ConclusionsHUCMSCs can survive,migrate and differentiate into early male germ cell-like cells in the infertility mice testis after transplantation.
7.The real-time fluorescence quantitative PCR assay for detection of bacterial contamination in platelet concentrates

Objective To establish a real-time PCR assay to detect bacterial contamination in platelet concentrates(PCs).Methods PCs were spiked with serial dilutions of Staphylococcus aureaus and Eschetichia coli,and the bacterial genomic DNA was isolated by modified Chelex-100 method and then quantitatively detected by Tagman real-time PCR.Filtration was performed to avoid contamination from other bacterial DNA in the reagents of extraction kit as well as the PCR mixture.Results The real-time PCR targeting conservative 16S rRNA gene showed high sensitivity and specificity in detecting bacterial contamination in PCs,and no cross-reaction with human genomic DNA and viruses was found.The Ct value of the lowest bacterial dose groups after filtration in both S.aureaus and E.coli were statistically different from the negative controls,with a minimal detection quality of 0.3CFUs/PCR for S.aureaus,and 0.1CFUs/PCR for E.coli.,respectively.Conclusion The modified Chelex-100 method to extract bacterial genomic DNA and the subsequent real-time PCR are convenient,sensitive and specific,and may be applied to the large scale clinical detection of bacterial contamination in PCs.
8.The role of serum uric acid in different types of coronary heart disease

Objective To detect the mechanism of serum uric acid (UA) in different types of coronary heart disease (CHD).Methods 88 patients were divided into three groups:the control group,stable angina (SA) group and acute coronary syndrom (ACS) group.The levels of UA,alpha-granule membrane protein 140 (GMP-140),von Willebrand factor(vWF),plasminogen activator inhabitor typed (PAI-1),Thromboxane B_2 (TXB_2) and C-reacting protein(CRP) were measured.Results ①UA and CRP in ACS group were higher than that in SA group and control group [(392.1±68.57) μmol/L and (42.2±39.4) mg/L vs (370.50±58.80) μmol/L and (18.9±17.1) mg/L vs (286.00±65.31) μmol/L and (2.5±0.7) mg/L,P<0.05)].For UA,there was no difference between ACS and SA group(P>0.05);CRP was higher in ACS group than in SA group (P<0.05).②vWF and TXB_2 were higher in ACS[(1.65±0.48)%,(19.73±18.66)ng/L]and SA group[(1.35±0.49)%,(11.18±10.71) ng/L]than in control group[(1.07±0.26)%,(6.46±5.41) ng/L,P<0.05],and those were higher in ACS group than in SA group (P<0.05).③GMP-140 and PAI-1 were higher in ACS [(13.04±0.99) μg/L and (65.65±14.76) μg/L]and SA group[(12.55±0.74) μg/L and (62.69±12.24) μg/L]than in control group [(12.32±0.29) μg/L,(50.78±13.88) μg/L,P<0.05].There were no differences between ACS and SA group (P>0.05).④Comparing hyperuricemia group and non-hyperuricemia group in CHD patients:the CRP(71.3±18.9) mg/L,vWF(1.08±0.52) %,GMP-140(13.57±1.11) μg/L,TXB_2 (57.26±47.84)ng/L,PAI-1 (72.12±9.23) μg/L in ACS group possessing hyperuricemia were higher than non-hyperuricemia group [CRP (20.7±17.9) mg/L,vWF (0.84±0.54) %,GMP-140 (13.23±1.07) μg/L,TXB_2 (26.70 + 23.83) ng/L,PAI-1 (61.30±12.07) μg/L](t=7.394,0.008,0.227,7.605,0.421,P<0.05);CRP(31.1±18.9)mg/L and TXB2 (21.54±3.90) ng/L in SA group possessing hyperuricemia group were higher than non-hyperuricemia group[(10.9±10.1)mg/L and (5.02±4.93) ng/L,t=0.494,8.669,P<0.05].Logistic stepwise regression analysis indicated that the related factors with ACS were UA(OR=1.046),CRP(OR=7.615),PAI-1(OR=1.301),PT(OR=0.300)and TG(OR=2.243) (P<0.05).Conclusions UA is an important risk factor in CHD patients.UA can induce different types of CHD by damaging blood vessel endothelium function,activating platelet,changing coagulation and causing inflammatory.
9.Electro-microscopic observation of trabecular bone remodeling in ovariectomized rats

BACKGROUND: The changes in trabecular bone microarchitecture in osteoporosis have aroused much attention. The decrease in the number of trabecular nodes and increase in the number of free ends are found in osteoporosis, but the mechanism is still unclear.OBJECTIVE: To observe the trabecular remodeling process in ovariectomized rats as the osteoporosis models electron microscopically, and to explore the reasons for the decrease in the number of trabecular nodes and increase in the number of free ends.DESIGN: Randomized and controlled animal trial.SETTING: Department of Orthopedics, Third Hospital of Peking University.MATERIALS: The experiment was conducted in the Animal Laboratory, Third Hospital of Peking University from September 1999 to February 2000. Thirty-six female Wistar rats of 3 months old and 240-280 g were selected and randomly divided into ovariectomized group and control group with 18 rats in each group. The rats were observed at 4,8, and 12 weeks postoperatively with 6 rats at each time point.METHODS: The rats of ovariectomized group were subjected to ovariectomy 1 week after feeding, but the control group was not. The changes of proximal tibia trabecular microarchitecture was observed under scanning electron microscope at 4, 8 and 12 weeks, respectively, and the osteoclast, osteoblast, and structure of cell organs were observed under transmission electron microscope.MAIN OUTCOME MEASURES: [1]The re modeling process after ovariectomy by electron microscope; ②morphological changes of trabecular bone.RESULTS: [1]Scanning electron microscope observation showed that trabecular bone remodeling was distributed in every region of trabecular microarchitecture, especially St and Nd-St region. After ovariectomy, the transverse trabecular was easier to be perforated and broken; the trabecular network was almost intact at 4 weeks, but gradually damaged at weeks 8 and 12; moreover, the collagen fibers on the surface of trabecular bone were scrappy, disorder and thinner. ②By the transmission electron microscopic study, the tibial osteoclast were found active at 12 weeks. When absorbing cancellous bone, osteoclast closely adhered to its surface, and digitations stretched into the cancellous bone. The shape and size of digitations were significantly different, and around them, lucent area was observed. Osteoclast was polynucleation with abundant kytoplasm, and there were plenty of Golgi complex, smooth endoplasmic reticulum and mitochondrium. Lysosome inclusion compounds with different sizes and electron density were found in cells. Osteoblast was rarely found, and cell edge was rough, with bone lacuna.CONCLUSION: Bone remodeling is significantly active in St and Nd-St region of trabecular bone in ovariectomized rats.This may be the reason for the decrease in the number of trabecular nodes and increase in the number of free ends.
10.Comparative study of recent adverse reactions caused by 131I-Metuximab treatment of advanced hepatic celluler cancer via two different routes

Objective To compare the recent adverse reactions caused by 131I-Metuximab (licartin) treatment via two different routes and to assess the safety and advantages of peripheral intravenous bolus of licartin for the treatment of advanced HCC.Methods Clinical data of 54 patients (45 males,9 females,age 33-80 years) with advanced HCC treated with Licartin in Tianjin Medical University Cancer Institute and Hospital from October 2010 to March 2013 were collected and analyzed.The patients were divided into vein group (n=33) with Licartin injected through peripheral vein and artery group (n =21) with Licartin injected through hepatic artery.The results of blood routine examination,liver and kidney function and thyroid function between the two groups (1 week before treatment,4 and 12 weeks after treatment) were compared.The adverse reaction rate (ARR) and adverse reaction progression rate (ARPR) were also compared between the 2 groups.Ten days after Licartin treatment,all patients underwent gamma imaging to access the drug distribution in vivo.x2 test and two-sample t test were used to analyze data.Results There were no significant differences on age,gender and TNM staging between the vein group and the artery group (t =0.721,x2=0.561 and 4.769,all P>0.05).The vein group showed temporary drug-related leucopenia (x2=7.041,P<0.05) and increased level of serum total bilirubin (STB;x2 =10.297,P<0.05) 4 weeks post-treatment.Twelve weeks later,the above parameters returned to baseline.In artery group,no influence on liver and kidney functions was observed,but the numbers of WBC and PLT decreased significantly (x2 =8.949 and 8.778,both P<0.05) and returned to baseline 12 weeks post-treatment.The ARR in patients who had normal ALT levels before treatment between the two groups was significantly different(3.33%(1/30) vs 5/19,x2=5.718,P<0.05).No significant difference was observed on ARR in patients with normal level of other parameters,and on ARPR in patients with abnormal preoperative parameters between the two groups (x2 =0.000-2.500,all P>0.05).The drug's in vivo biodistribution and the thyroid function between the 2 groups showed no significant difference.Conclusion The peripheral intravenous bolus administration of Licartin is safe to treat patients with advanced HCC.
