Recently, in the field of pathogenesis and treatment of type 2 diabetes, incretin is a major breakthrough. Based on the knowledge about physiological mechanism, pathological characters, and clinical application, this review article summarized the mechanism of nutrients ( glucose, fat, and protein) mediated incretin secretion, the impairment of incretin secretion and action in type 2 diabetes and glucagon-like peptide-1 's use in type 2 diabetic patients.

6.Progress in the mechanisms of glucocorticoid resistance in ectopic ACTH syndrome

High level of eetopie serum ACTH can not be suppressed by endogenous or exogenous glucocorticoid, this is the cardinal characteristic of ectopic ACTH syndrome (EAS), that is to say there exists glucocorticoid resistance in EAS. However, the mechanism of glucocorticoid resistance remains unclear. Identifying the mechanism can help us to diagnose and treat the disease. This review focuses on the mechanism of glucocorticoid resistance from several aspects, including the pre-glucocorticoid receptor (GR), GR and post-GR.

The mRNA expression of CAR in P3, P6 and P8 was similar, and the same change was observed in the protein expression of CAR in P3 and P8 BMSCs . CONCLUSION: Ad-CMV-GFP is transferred to BMSC effectively and sustained about 28 days. It is suspected that BMSCs in mitotic phase are easy to be transferred by Ad-CMV-GFP and different passages of BMSCs from P3 to P8 BMSCs can be as high-effectively g ene vehicle.

The biodistribution and the radio activity in blood clearance of 99m Tc AMLCA were determined in 5 normal dogs by whole body imaging and measuring the radio activity in blood samples at 2,4,8 and 24h after 99m Tc AMLCA injection. The feasibility of imaging of the site of myocardial infarct was determined in 2 of the experimental MI dogs by demonstration of the left ventricle blood pool imaging (LVBPI) with 99m Tc AMLCA and by subsequent imaging of the excised heart. The results showed that the plasma concentration of 99m Tc AMLCA decreased rapidly from 51 5%?5 2% at 2nd hr to 27 3%?3 1% at 4th, 12 3%?1 8% at 8 hr and 5 6%?0 6% at 24th after the injection. The LVBPI in one MI dog showed that the region of the radionuclide concentration of 99m Tc AMLCA corresponded to region of absence of 99m Tc MIBI. The scintigraphy in another MI dog heart slices showed that the region of the radionuclide concentrate of 99m Tc AMLCA corresponded to the region of triphenyltetrazolium chloride (TTC) staining. The measurement of double radionuclides in the double interesting region in the MI dog heart slices indicated that the infarct myocardium uptook 99m Tc AMLCA specifically. These findings suggested that 99m Tc AMLCA scintigraphy might be a new approach for detecting and localizing MI

Objective:To induce an anti-hCG? antibody response in reproductive tract and the peripheral by oral inoculation with a recombinant alive Lactobacilli expressing hCG? in different strains of mouse.Methods:The 108,109,1010 Lb.hCG?, a recombinant Lactobacillus expressing hCG?, were inoculated orally into 6-8-week-old female BALB/c and C57BL/6 mice. The immunized mice were all boosted with the same dose at interval of 3 weeks. An indirect ELISA was used to determine anti-hCG? IgG and IgA antibodies in vaginal lavage and serum that were got at the end of 2-8 weeks after the primary immunization. The B and T lymphocytes of spleen, uterus and vagina were prepared respectively, and their proliferation was analyzed by flow cytometry. The numbers of the cells secreting anti-hCG? IgG or IgA antibody were detected by ELISpot.Results:Inoculation of 109 or 1010 alive Lb.hCG? could induce similar response while 108 induce a lower attitude of response. The antiserum of either 109 or 1010 Lb. hCG? immunization after booster could neutralized more than 100 ng/ml of hCG antigen both in BALB/c and C57BL/6 mice. 10.Determination of Gastrodin in Choudongning Capsule by HPLC

OBJECTIVE:To establish an HPLC method for the determination of gastrodin in Choudongning capsule. METHODS:The sample was determined on Elite C18 column(250 mm?4.6 mm,5 ?m);the mobile phase consisted of methanol -water(3∶97) at a flow rate of 1.0 mL?min-1;the detection wavelength was set at 221 nm. RESULTS:The linear range of gastrodin was from 0.051 ?g to 0.357 ?g(r=0.999 9) and its average recovery was 98.99%(RSD=1.91%,n=6).CONCL-USION:The method is convenient,accurate and reproducible,and it is applicable for the quality control of Choudongning capsule.
