1.Determination of Residual Solvents in α-Ketophenylalanine Calcium by Gas Chromatography

To develop a method for the determination of residual solvents inα-ketophenylalanine calcium by capillary gas chromatography. Methods:The residual solvents were separated on a DB-624(30 m × 0. 32 mm, 0. 25 μm) capillary chromato-graphic column with temperature programming. The column temperature was maintained at 40℃ for 1 min,and then raised to 180℃at a rate of 10℃·min-1 and maintained for 2 min. N2 was used as the carrier gas, and FID was used as the detector with temperature of 250 ℃. The injector temperature was 200 ℃ and the split ratio was 10∶1, and direct injection was adopted. Methanol, ethanol, ethyl acetate and tetrahydrofuran in α-ketoleucine calcium were detected using an external standard method. Results:The four solvents were separated completely. There was a good linear relationship between the peak area and the concentration of each solvent ( r=0. 997 2-0. 999 5). The average recovery of the four solvents was 95. 47%-100. 26%(RSD≤4. 7%, n=9). Conclusion:The method is rap-id, simple, accurate and sensitive, and can be used in the determination of residual solvents in α-ketophenylalanine calcium.

Objective To study the effect of hyaluronidase and VEGF on human breast cancer invasion and metastasis. Methods By means of ELSA and immunohistochemistry, hyaluronidase and VEGF were detected in breast cancer. Results Hyaluronidase were (3.99?1.91) mU/g, (8.01?2.59) mU/g and (12.00?3.96) mU/g in gradeⅠ, Ⅱ, Ⅲ of breast cancer respectively. Hyaluronidase levels were significantly elevated in grade Ⅱ,Ⅲ as compared to grade Ⅰ, Hyaluronidase levels were significantly elevated in grade Ⅲ as compared to grade Ⅱ(P
3.ANTHROPOMETRY AND FAT STORE DURING PREGNANCY

Weight gain, skinfold thickness, circumference and body density were measured in 30 primiparous women aged from. 23-31 years (averaged 26.3 years old).The average body weight gained 13kg from 6 to 37 weeks of pregnancy.The suprailiac skinfold thickness in 24-37 weeks and the subscapular skinfold thickness in 37 weeks increased significantly from the 6th week of pregnancy, but the triceps and biceps skinfold thickness did not change significantly. The density of the body in 24th and 37th week of pregnancy decreased significantly.The change and the body fat (%) calculated by means of 4 skinfolds increased in 3.7% (5.7kg of fat). The mean values of the hip circumference in 24th and 37th week were significantly different from that in 6th week of pregnancy. The upper thigh in the 24th and 37th week also increased slightly but not significantly different as compared with the 6th week. The mean values of arm, mid-thigh and calf circumference did not change during pregnancy.
4.EFFECT OF VITAMIN A AND IRON SUPPLEMENTATION ON IRON STATUS DURING PREGNANCY

Objective: To observe the effect of vitamin A and iron supplementation on iron status during pregnancy. Methods: 167 pregnant women aged 21~33 years in the second trimester were recruited and randomly allocated to four groups: placebo; vitamin A (1.1 mg/d); iron(60 mg elemental iron); vitamin A and iron, for 10 weeks. Results:The incidences of vitamin A deficiency, anemia and iron depletion were 0.6%,7.8% and 6.2% in the second trimester respectively. Serum VA level was not changed in three supplemented groups but decreased in placebo group. Hemoglobin in all groups before and after supplementation was in normal range, with no significant differences. The improvement in serum ferritin, free erythrocyte proporphyrin and transferrin saturation was better in the group supplemented with VA+Fe than VA or Fe alone. Conclusion: The simultaneous supplementation of vitamin A and iron is better in improvement of iron status than supplementation of vitamin A or iron alone during pregnancy. Improvement of iron status by simultaneous supplementation of pregnant women with vitamin A and iron was better than supplementation with iron or vitamin A alone.
5.The clinical characteristics and cognitive function of Parkinson's disease patients associated with pain

Objective To explore the type and etiology of chronic pain in patients with Parkinson's disease (PD),and to assess the association between pain and cognitive function.Methods A total of 116 PD patients were enrolled in the study and assessed with the following rating scales:Unified Parkinson's Disease Rating Scale (UPDRS),Hoehn-Yahr Scale (H-Y),Hamilton Depression Rating Scale for Depression(HRSD),Visual Analogue Scale (VAS) and Montreal Cognitive Assessment (MoCA).Results Compared with PD patients without pain,PD patients with pain had higher scores of UPDRS,H-Y and HRSD rating scales.Depression(scores of HRSD) was the only factor associated with pain showed by the Logistic regression model (P =0.007).PD patients with pain had lower scores of delayed recall (P =0.020).PD patients with pain happened before their motor symptoms had a lower score of delayed recall (P =0.015).Conclusions Musculoskeletal pain is the most common type in PD patients with pain.Depression is probably an independent risk factor for pain in PD patients.Delayed recall is the dominant impaired cognitive function.
6.Generation of monoclonal antibodies against complexed prostate specific antigen and development of an antibody-based chemiluminescence immune quantifica-tion assay

Objective:To construct a chemiluminescense immune quantification assay based one paired mAbs against complexed prostate specific antigen ( c-PSA).Methods:Six week-old female BALB/c mice were immunized with the commercial c-PSA antigen.After serum titer reaching a platform stage ,the spleen was immunized and fused with mouse myeloma cell lines ( Sp2/0 ) .The hybridoma were screened by indirect ELISA ,and eight generated antibodies were paired to obtain a quantitative analysis of the chemical luminescence.Results:7D6 specifically recognized c-PSA,while 1A10 recognized total PSA(t-PSA).And the paired antibody 1A10/7D6 were determined to successfully construct a chemiluminescense immune response quantitative detection method through the detection of c-PSA standard and clinical serum samples .had,positive samples have statistically significant difference ( P<0.000 1 ) with negative samples.And the correlation coefficient R 2 was 0.97 between our c-PSA quantitative results and that of the Siemens c-PSA chemiluminescense immunoassay kit .The detection linear range was 0.1-100 ng/ml,and the sensitivity was 0.005 ng/ml.Conclusion:The paired monoclonal antibodies specifically detecting c-PSA were generated and a c-PSA chemiluminescense immunoassay were developed in this study .The detection capability of our method was comparable with that of the international commercial kit .
7.Determination of Benzyl Alcohol in Metamizole Sodium Injection by HPLC

Objective: To establish an HPLC method to determine the content of benzyl alcohol in metamizole sodium injection. Methods: The determination was performed on an Xtimate C18(250 mm×4. 6 mm,5 μm)column. Phosphate buffer (dissolving 6. 0 g sodium dihydrogen phosphate in 1000 ml water, adding 1ml triethylamine, adjusting pH to 7. 0 with sodium hydroxide solution) -methanol (75: 25) was used as the mobile phase at a flow rate of 1. 0 ml·min-1. The detection wavelength was 254 nm. The column tamprture was 30 ℃ and the sample size was 5 μl. Results: The linear range of benzyl alcohol was 76. 88-269. 08 μg·ml-1( r=0. 998 5)with the average recovery of 98. 77% (RSD=0. 77% ,n=9). Conclusion: The method is accurate and reproducible for the content determination of benzyl alcohol in metamizole sodium injection.
8.Serum C-type lectin-like receptor-2 combined with insulin resistance predicts the outcome of patients with acute ischemic stroke after intravenous thrombolysis

Objective:To investigate the predictive value of serum C-type lectin-like receptor 2 (CLEC-2) combined with insulin resistance in the outcome of patients with acute ischemic stroke (AIS) after intravenous thrombolysis.Methods:Patients with AIS received alteplase intravenous thrombolytic therapy in the Department of Neurology, the Second Affiliated Hospital of Nantong University from October 2019 to March 2021 were enrolled retrospectively. According to the modified Rankin Scale score at 90 d after onset, they were divided into good outcome group (0-2) and poor outcome group (>2). Homeostasis model assessment of insulin resistance (HOMA-IR) was used to evaluate insulin resistance. Person correlation analysis was used to determine the correlation between CLEC-2 and HOMA-IR. Multivariate logistic regression analysis was used to determine the correlation between serum CELC-2, HOMA-IR and the outcome after intravenous thrombolysis. Receiver operating characteristic (ROC) curve was used to determine the predictive value of serum CLEC-2 combined with HOMA-IR for poor outcome after intravenous thrombolysis. Results:A total of 100 patients were enrolled (56 males, 56.0%; aged 70.6±10.86 years, range 49-83 years). The baseline National Institutes of Health Stroke Scale (NIHSS) score was 10.00±6.36. Senenty-four patients (74.0%) had a good outcome and 26 (26.0%) had a poor outcome. Person correlation analysis showed that there was a significant positive correlation between serum CLEC-2 and HOMA-IR ( r=0.523; P<0.001). Multivariate logistic regression analysis showed that after adjusting for confounding factors (C-reactive protein, baseline NIHSS score, onset-to-needle time), the highest quartile of serum CLEC-2 (compared with the lowest quartile: odds ratio [ OR] 4.836, 95% confidence interval [ CI] 1.105-21.169; P=0.036) and the highest quartile of HOMA-IR (compared with the quartile 1-3: OR 15, 95% CI 2.647-30.722; P=0.002) were the independent risk factors for the poor outcome in patients with AIS after intravenous thrombolysis. ROC curve analysis showed that the area under the curve for serum CLEC-2 combined with HOMA-IR to predict poor outcome was 0.785 (95% CI 0.688-0.883; P<0.001), the optimal cut-off value was 0.72, and the sensitivity and specificity were 76.0% and 95.0%, respectively. Conclusion:CLEC-2 combined with insulin resistance has a certain predictive value for the poor outcome of patients with AIS after intravenous thrombolysis.
9.Fast volume rendering of echocardiogram with shear-warp algorithm.

Shear-warp is a volume rendering technology based on object-order. It has the characteristics of high speed and high image quality by comparison with the conventional visualization technology. The authors introduced the principle of this algorithm and applied it to the visualization of 3-D data obtained by interpolating rotary scanning echocardiogram. The 3-D reconstruction of the echocardiogram was efficiently completed with high image quality. This algorithm has a prospective application in medical image visualization.

10.Determination of Sodium Glycerophosphate and Phosphate Salts in Concentrated Divitamins and Sodium Phosphate Syrup by Ion Chromatography

Objective:To establish ion chromatography(IC) method for determination of the content of sodium glycerophosphate and phosphate salts in concentrated divitamins and sodium phosphate syrup. Methods:An AG19 -HC (50 mm× 4 mm) guard col-umn and an IonPac AS18 -HC (250 mm × 4 mm) ion analysis column were used, potassium hydroxide solution was applied as the mobile phase with gradient elution,and a conductance detector was used with suppression conductometric detection. Results:Sodium glycerophosphate and phosphate could be separated well under the chromatographic conditions. The linear range was 1.054-84.35 μg ·ml-1(r=0.999 4)for sodium glycerophosphate and 0.316 8-25.35 μg·ml-1(r=0.999 8)for phosphate. The average recovery was 99.35% (RSD=1.16%,n=9) and 98.96%(RSD=2.61%,n=9), respectively.Conclusion: The method is simple, rapid and accurate,and can be used for the quality control of concentrated divitamins and sodium phosphate syrup.
