1.Effect of Shenqi Fuzheng injection on serum CRP,IL-6 and IL-10 levels in elderly patients with coronary heart disease

Objective To investigate the effect of Shenqi Fuzheng injection on serum CRP,IL-6 and IL-10 levels in elderly patients with coronary heart disease.Methods 64 elderly patients with coronary heart disease from Wenling First People's Hospital hospital were selected and randomly divided into the control group and the experiment group.32 cases in each group, the control group were treated by conventional symptomatic treatment, the experiment group were treated with Shenqi Fuzheng injection on the base of control group.The serum ( C-reactionprotein,CRP) ,white blood cell mediated IL-6 and leukocyte mediated element IL-10, systolic blood pressure, diastolic blood pressure(DBP),glycerin,total cholesterol(TC),triglyceride(TG), low density lipoprotein(LDL-C) and high density lipoprotein(HDL-C) levels were recorded,and the clinical efficacy and adverse reaction conditions were compared.Results The effective rate of treatment in the control group was 68.75% lower than that in the experiment group 90.63%(P<0.05). Compared with the control group after treatment,the serum CRP, IL-6, IL-10, systolic blood pressure, diastolic blood pressure, TC,TG and LDL-C levels were lower,theHDL-C level was higher,has statistical significance(P<0.05).Conclusion Shenqi Fuzheng injection in the treatment of elderly patients with coronary heart disease has a significant effect,can reduce serum CRP, IL-6 and IL-10 levels.
2.Aripiprazole in patients with bipolar disorder D2 receptor and 5 -HT1A receptor partial agonists and analysis of 5-HT2A receptor antagonism

Objective To explore the efficacy of aripiprazole in patients with bipolar disorder D2 receptor and 5 -HT1A receptor partial agonists and analyze 5 -HT2A receptor antagonism.Methods From January 2013 to January 2015,113 patients came to our hospital for treatment of bipolar disorder,in accordance with the order of admission,were divide into aripiprazole group (47 cases)and control group (66 cases).The control group was given venlafaxine,aripiprazole group was given aripiprazole treatment on the basis of the control group.SDS,BRMS score and therapeutic effect were compared between the two groups before and after treatment.Results The SDS,BRMS scores were decreased after treatment in the two groups,compared with before treatment,the differences were statisti-cally significant (t =31.3587,36.1207;all P <0.05 );and the aripiprazole group decreased more significantly than the control group,the SDS,BRMS scores of the two groups after treatment had statistically significant differences [SDS (31.8 ±4.3)points vs (28.7 ±3.6)points,BRMS (6.5 ±0.2)points vs (5.5 ±0.2)points,t =4.110 7,26.197 0, all P <0.05 ].The total effective rate of the aripiprazole group was 97.9%,which was significantly higher than 89.4% of the control group (u =3.365 9,P =0.000 8).The incidence rate of adverse reactions such as nausea, vomiting,drowsiness,anxiety and heart rate in the aripiprazole group was significantly lower than the control group (all P <0.05 ).Conclusion Aripiprazole for bipolar disorder patients with D2 receptor and 5 -HT1A receptor has partial agonism,of 5 -HT2A receptor with role constraints,can effectively improve the effect of treatment of patients,reduce the incidence of adverse reactions.
3.Research and Analysis Result of Recheck Blood Type in Part of Beijing Blood Bank

OBJECTIVE To investigate the degree of risk of viral prevalence of HBV,HCV and HIV through blood transfusion in Beijing Hospital in China,and to assess the need of a national Haemovigilance System. METHODS Retrospectively,7883 blood bank specimens (collected from 2004 to 2007) were re-examined using 8 indicators (including 5-item Hepatitis B,anti-HCV,anti-HIV and Syphilis) for the prevalence of most common viral infection. RESULTS From the blood bank specimens,the prevalence of HBsAg was 0.88% (69),the anti-HBc positive blood,only the anti-HBc was found in 2.65% of the specimens,while both the anti-HBc and the anti-HBe were found in 2.09% of the specimens. The prevalence of anti-HCV was 0.09% (7). CONCLUSIONS We need to establish the national Haemovigilance System to strengthen the monitoring of the above HbsAg,HBcAb and HCV indicators to prevent the transfusion-transmitted infection. Only in this way can the public confidence in blood safety be improved.

5.Effects of Zibu Piyin Recipe Serum on endoplasmic reticulum(ER)stress-induced neuron apoptosis and its mechanism

Objective To explore the inhibitory effects of Zibu Piyin Recipe(ZBPYR)serum on neuron apoptosis induced by tunieamyein(Tm,5 μg/ml)and its mechamsm in vitro by using sero-pharmacological method.Method Totally 12 healthy adult male SD rats(220~250 g)(SPF)were divided randomly into control group and ZBPYR group,6 in each group,then the blank and ZBPYR serum were prepared.The mouse.neuroblastoma cell line Neum2a cells were treated with Tunicamycin(Tin,an inhibitor of N-glycoslytion)to establish the endoplasmic reticulum(ER)stress model.The cells treated by ZBPYR aerum of different concentrations were interventional groups,and the cells treated by blank serum were control group.The viability of Neuro2a cells was meusurcdd by MTT assay.Flow cytometry wus applied to observe the apoptosis of Neuro2a cells.Western blotting was utilized to detect the protein expressions of two molecules,ER molecular chaperone-ucose regulated protein 78(CRP78)and transcriptional factor CCAAT/enhancer-binding protein-homologous protein(CHOP).The results were analyzed by sNK-q test.Results Compared to Tm group(cell viability 0.1673±0.0213,apoptotic rate 62.7050±1.4056),The cell viability of interventional groups(5%0.5295±0.0373,10%0.5843±0.0428,15%0.6274±0.0324)increased significantly(P<0.05);and the apoptotic rate(5%47.8733±2.8166,10%46.3366±1.2748,15%39.8833±1.0524)reduced significantly(P<0.05).The protein expressions of GRP 78(5%2.1228±0.2251,10%1.3293±0.9443,15%;15%0.0931±0.1168)and CHOP(5%1.1776±0.2927,10%0.7290±0.1708,15%0.6577±0.1883)of interventional groups reduced significantly compared with Tm group(GRP78 2.9149±0.5355;CHOP 1.6611±0.2913)P<0.05.Condusions ZBPYR serurn could increase the cell viability of Neuro2a cells treated with Tm and inhibit cell apoptosis.Thereby it may have neuroprotective effects,and the mechanism may be associated with the inhibition of ER stress and apoptosis pathway.
6.Effects of Electroacupuncture at Sanyinjiao and Yinlingquan Acupoints on Muscle Repair and Expression of Growth Factors in Gastrocnemius Muscle Damaged Rats

Objective To investigate the effects of electroacupuncture at Sanyinjiao (SP6) and Yinlingquan (SP9) on muscle repair in the gastrocnemius muscle damaged rats and its possible mechanism. Methods 30 male Sprague-Dawley rats were randomly divided into blank group (n=10), model group (n=10) and electroacupuncture group (n=10). The latter 2 groups were modeled with gastrocnemius muscle crush injury. The electroacupuncture group received electroacupuncture at Sanyinjiao and Yinlingquan for 14 days. Their gastrocnemius was observed with HE staining, and the expressions of basic fibroblast growth factor (bFGF) and insulin-like growth factor-1 (IGF-1) in gastroc-nemius tissues and the serum were determined with immunochemical assay and enzyme-linked immunosorbent assay (ELISA). Results Compared with the model group, there was less damage in the electroacupuncture group, with more expression of bFGF and IGF-1 in tis-sues (P<0.01) and serum (P<0.05). Conclusion Electroacupuncture can promote muscle repair after gastrocnemius muscle damage in rats, which may be related to the increase of the bFGF and IGF-1 expression in gastrocnemius tissue and serum.
7.Nuclease activity and cytotoxicity to host cells of toxic protein VapC produced by Leptospira species

Objective To determine the function of toxic protein VapC in toxin/antitoxin system of Leptospira species and the cytotoxicity to host cells of the toxic protein.Methods Using genomic DNA of pathogenic L.interrogans serogroup Icterohaemorrhagiae serovar Lai strain Lai as the template,several PCRs were performed to amplify entire vapB,vapC and vapBC genes.Subsequently,the prokaryotic expression systems of vapB,vapC and vapBC genes were constructed.Expression of the target recombinant proteins rVapB and rVapC was detected by SDS-PAGE and the expressed rVapB and rVapC were extracted by NiNTA affinity chromatography.Activity of rVapB and rVapC to lyse the DNAs or RNAs from L.interrogans strain Lai and THP-1 cells were then determined.The changes of transcription and expression of vapB and vapC genes of L.interrogans strain Lai before and after infection of THP-1 cells were detected by real-time fluorescent quantitative RT-PCR and Western blot assay.The eukaryotic expression vectors of the vapB and vapC genes were generated for transfection of host cells and CCK-8 agent was used to detect the effect of leptospiral VapB and VapC proteins on activity of host cells.Results The nucleotide and putative amino acid sequences of the cloned vapB and vapC genes were completely identical with the reported corresponding genes.The constructed prokaryotic expression systems could express rVapB and rVapC,respectively.rVapC displayed RNase avtivity but did not lyse DNA.When L.interrogans strain Lai infected THP-1 cells,the transcription and expression of vapB and vapC genes were upregulated and partial VapC protein was secreted from the leptospiral cells.The mass mortality was observed in HEK293 human renal tubular epithelial ceils containing the vapC gene through transfection.Conclusion VapC protein of L.interrogans strain Lai is a RNase and is secreted during infection of host cells with obvious cytotoxicity.
8.Aberrant function connectivity and grey matter volume within default mode network in patients with type 2 diabetes mellitus by functional MRI

Objective To explore early aberrant change of the function and grey matter volume within default-mode network in patients with type 2 diabetes mellitus (T2DM) by Independent component analysis and voxel-based morphometry.Methods Twenty-nine T2DM patients and thirty-three healthy control subjects were recruited in this study between May 2014 and January 2015,and the demographic and clinical data (fasting plasma glucose and HbA1c level) were obtained.General cognition was assessed by MMSE and MoCA in all subjects.Resting-state fMRI and T1-weighted imaging were performed in all subjects.Then the z-maps of default-mode network were obtained from Resting-state fMRI by independent component analysis,and the grey matter volumes were extracted by VBM.Finally,two-sample t-test was performed in the function and grey matter volume of DMN between two groups,and pearson correlation coefficients among zFC values extracted from significantly different brain regions,grey matter volumes and durations of T2DM patients were calculated.Results Patients with T2DM showed decreased Z value around the right anterior cingulated cortex and the right medial pre-frontal cortex(cluster=121,t=-3.81,P< 0.05).There was no significant difference in grey matter volume between the two groups,and no correlation between Z value or duration and grey matter volume was found.Conclusions It suggests the abnormality of connectivity of default-mode network may precede the abnormality of grey matter volume in type 2 diabetes mellitus patients.The DMN might be a new potential tool in monitoring the change of cognition in patients with type 2 diabetes mellitus.
9.Intravascular Ultrasonic Features of Unstable Atherosclerotic Plaques in Unstable Angina patients

Objective To investigate the characteristics of plaque morphology of the patients with unstable angina(UA)by intravascular ultrasound(IVUS). Methods24 patients with stable angina pectoris(SA)and 33 patients with unstable angina(UA)were examined by coronary coronary arteriograph(CAG)and intravascular ultrasound(IVUS).The culprit lesions about the characteristic of the plaque were studied,the external elastic membrane cross-sectional area and lumen cross-sectional area were measured,the plaque area,plaque burden,eccentricity index were calculated,and the remodeling index was examined. ResultsExternal elastic membrane area,plaque area and vascular remodeling ratio were significantly greater at target lesions in UA patients than in patients with SA(P<0.05).Positive remodeling and soft plaque were predominantly in UA patients(P<0.05). ConclusionThe morphological features of coronary artery plaques in UA group were significantly different from those in SA group.More soft plaque,plaque rupture,thrombus formation,and positive remodeling were found in UA patients compared with SA patients.
10.Research on expression and function of phosphorylated DARPP-32 on pentylenetetrazol-induced epilepsy model of rat.

The present study is to explore the change process and distribution of phosphorylated DARPP-32 (p-DARPP-32) in rat brain including cortex, hippocampus and striatum and to further deduce whether p-DARPP-32 was possibly involved in epilepsy induced by repetitive low doses of pentylenetetrazol (PTZ). PTZ-induced epilepsy model in rat was established with 30 male SD rats randomly divided into 6 groups, control group and five trial groups [PTZ 1 h, PTZ 6 h, PTZ 24 h, PTZ 48 h and PTZ 72 h respectively, after onset of status epilepticus (SE)]. Immunohistochemistry and immunofluorescence double-labeling were used to detect the temporal time change and distribution of p-DARPP-32 expression and to analyze the coexpression of DARPP-32 and p-DARPP-32 in rat brain after the onset of PTZ-induced generalized SE. The results showed that there was a temporal time change of p-DARPP-32 expression in rat brain after the onset of SE. The number of p-DARPP-32-positive cells increased significantly and reached the peaks at the ends of 1 hour and 6 hours after the onset of SE, but decreased at the end of 24 hours. The moderate to strong p-DARPP-32-immunopositive neurons were observed in cortex, hippocampus and striatum, and located in cell cytoplasm and cell nucleus. Further immunofluorescence double-labeling revealed that denser colocalization of p-DARPP-32 and DARPP-32 in the neurons existed in the area mentioned above. Therefore, PTZ-induced SE may cause phosphorylation of DARPP-32 in rat brain. The temporal time change and distribution of p-DARPP-32 suggest that phosphorylation of DARPP-32 may be involved in PTZ-induced epilepsy in rat brain including cortex, hippocampus and striatum, and p-DARPP-32 may play a central role in the onset of SE.
